This paper presents a new solution for adding a new database server on an existing Oracle Multimaster data replication system. In normal conditions adding a new database server on this system we cannot insert, delete and update data during this time. So during this time the system is down and we can make only queries. The time for adding the new database server depends on the number of tables on the replication group and on the network conditions. To reduce this time we propose to replace the Replication group in more than one replication group based on logic of database schema. So we will have a replication group for base data, one replication group for most populated tables and another group for non most used tables. Using this method on the Oracle database schema with 250 replication tables and communication speed 1Mbit/s, the time reduced seven times. This research is implemented on border control system (Total Information Management System at Ministry of Interior) in Albania and using this solution we can add or remove replication sites with minimal down time.

Session guarantees define consistency properties of replicas in a distributed system from the point of view of a single, migrating client. Consistency is checked by ensuring execution of required operations by servers. Consistency protocols of session gua- rantees usually use version vectors for efficient representation of sets of writes. This paper presents performance evaluation of several consistency protocols of session guarantees that use different forms of version vectors. The evaluation was carried out by means of simu- lation experiments.

The concept of creative industries is becoming popular in recent years. Therefore, many publications deal with this topic. Nevertheless, fewer of them are dedicated to managerial issues. Most publications concentrate on the scope of creative industries and their influence on economy and society. The first aim of this paper is to present the specificity of management of organization in creative industry and its activity. The second aim is to collect and cite different definitions of knowledge replication to explain the meaning of the notion of knowledge replication. The third aim is to show the role of knowledge replication for organization’s activity in creative industries. The article also contains an introduction concerning different approaches to definition and classification of creative industries.

Database replication is a way to improve system throughput or achieve highavailability. In most cases, the use of an active-active replica architecture isefficient and easy to deploy. Such a system has CP properties (from the CAPtheorem: consistency, availability, and network-partition tolerance). Creat-ing an AP (available and partition-tolerant) system requires the use of multi-primary replication. Because of the many difficulties in its implementation, thisapproach is not widely used; however, ALICE’s deployment of CCDB (exper-iment conditions and calibration database) needs to be an AP system in twolocations. This necessity became the inspiration for examining the state-of-the-art methods in this field and testing the available solutions. The tests thatwere performed evaluated the performance of the chosen replication tools: Bu-cardo, and EDB Replication Server; these showed that the tested tools couldbe successfully used for the continuous synchronization of two independentdatabase instances.

To study the biochemical properties of SSB's from Deinococcus grandis (DgrSSB) and Deinococcus proteolyticus (DprSSB), we have cloned the ssb genes obtained by PCR and have developed Escherichia coli overexpression systems. The genes consist of an open reading frame of 891 (DgrSSB) and 876 (DprSSB) nucleotides encoding proteins of 296 and 291 amino acids with a calculated molecular mass of 32.29 and 31.33 kDa, respectively. The amino-acid sequence of DgrSSB exhibits 45%, 44% and 82% identity and the amino-acid sequence of DprSSB reveals 43%, 43% and 69% identity with Thermus aquaticus (TaqSSB), Thermus thermophilus (TthSSB) and Deinococcus radiodurans SSBs, respectively. We show that DgrSSB and DprSSB are similar to Thermus/Deinococcus SSBs in their biochemical properties. They are functional as homodimers, with each monomer encoding two single-stranded DNA binding domains (OB-folds). In fluorescence titrations with poly(dT), both proteins bind single-stranded DNA with a binding site size of about 33 nt per homodimer. In a complementation assay in E. coli, DgrSSB and DprSSB took over the in vivo function of EcoSSB. Thermostability with half-lives of about 1 min at 65-67.5°C make DgrSSB and DprSSB similar to the known SSB of Deinococcus radiodurans (DraSSB).

Telomeres are the specialized nucleoprotein structures associated with eukaryotic chromosomal ends, which are essential for maintaining the stability of the linear eukaryotic chromosomes. Progressive telomere shortening is an inevitable occurrence in normal somatic cells due to the end-replication problem leading to limited replication efficiency. The hallmark characteristics of human cancer cells include infinite reproductive potential, uncontrolled proliferation and immortality. These abilities of transformed cancerous cells are mainly due to the maintenance of their telomeres since degradation of chromosomal telomeric ends leads to cellular senescence or death. Thus telomere biology is important in the study of human cancer development. The mechanism by virtue the cancer cells are able to divide indefinitely is by maintaining telomeres. Activity of telomerase, a telomere-elongating ribonucleoprotein reverse transcriptase enzyme, is responsible for inducing the property of immortality to cancer cells. In humans nearly about 80% to 90% cancer cells activate telomerase and elongate their telomeres to overcome the end-replication problem. Telomere shortening suppresses cancer formation in contrast according to certain reports it sometimes promotes genomic instability which leads to enhancement of carcinogenesis and consequently the development of malignancy. Majority of cancer cells activate telomerase, but it remains mysterious as to find the reasons of the fact that certain cancer cells often show shorter telomeres in comparison to the cells in the surrounding normal tissues. This controversial role of telomerase associated with certain transformed cells leading to the cancerous state in relation to its role in normal cells is an interesting field to study which points out to the fact of development of cancer cells targeting drugs based on telomerase activities as an alternative weapon in combating against this dreaded human disease. The present review focuses on the activity of telomerase in telomere maintenance in the development of cancerous cells in humans, the use of telomerase as an assay technique for cancer detection as well as the anti-cancer therapeutic approach of targeting the telomerase in the current era of treatment of human cancer.

Alongside the growing number of systems accessible via Web services came the need for specialized data repositories that could offer effective means of searching the available services. The development of mobile systems and wireless data transmission technologies has allowed us to use distributed devices and computer systems on a greater scale. The accelerating growth of distributed systems might be a good reason to consider the development of distributed Web service repositories with built-in mechanisms for data migration and synchronization.

This paper summarizes recent advances in understanding the links between the cell's ability to maintain integrity of its mitochondrial genome and mitochondrial genetic diseases. Human mitochondrial DNA is replicated by the two-subunit DNA polymerase γ (pol γ). We investigated the fidelity of DNA replication by pol γ with and without exonucleolytic proofreading and its p55 accessory subunit. Pol γ has high base substitution fidelity due to efficient base selection and exonucleolytic proofreading, but low frameshift fidelity when copying homopolymeric sequences longer than four nucleotides. Progressive external ophthalmoplegia (PEO) is a rare disease characterized by the accumulation of large deletions in mitochondrial DNA. Recently, several mutations in the polymerase and exonuclease domains of the human pol γ have been shown to be associated with PEO. We are analyzing the effect of these mutations on the human pol γ enzyme. In particular, three autosomal dominant mutations alter amino acids located within polymerase motif B of pol γ. These residues are highly conserved among family A DNA polymerases, which include T7 DNA polymerase and E. coli pol I. These PEO mutations have been generated in pol γ to analyze their effects on overall polymerase function as well as the effects on the fidelity of DNA synthesis. One mutation in particular, Y955C, was found in several families throughout Europe, including one Belgian family and five unrelated Italian families. The Y955C mutant pol γ retains a wild-type catalytic rate but suffers a 45-fold decrease in apparent binding affinity for the incoming dNTP. The Y955C derivative is also much less accurate than is wild-type pol γ, with error rates for certain mismatches elevated by 10- to 100-fold. The error prone DNA synthesis observed for the Y955C pol γ is consistent with the accumulation of mtDNA mutations in patients with PEO. The effects of other pol γ mutations associated with PEO are discussed.

One of the most important parameters in the study of diffraction gratings is their optical efficiency. This paper analyzes the different manufacturing processes to cover gratings of Moiré interferometry and their influence on the quality and absolute efficiency of replicated gratings on the surfaces of specimens. The Moiré interferometry is a field measurement technique that has been used in many different fields such as applied mechanics, microelectronics, biomechanics or micromechanics, hence the importance of this study. The applied reflected coating was done by sputtering and aluminium vaporization processes. In this work different materials and thickness layers were analyzed. The obtained coatings have a high degree of reflectivity on the replicated gratings.

Recombinational repair is a well conserved DNA repair mechanism present in all living organisms. Repair by homologous recombination is generally accurate as it uses undamaged homologous DNA molecule as a repair template. In Escherichia coli homologous recombination repairs both the double-strand breaks and single-strand gaps in DNA. DNA double-strand breaks (DSB) can be induced upon exposure to exogenous sources such as ionizing radiation or endogenous DNA-damaging agents including reactive oxygen species (ROS) as well as during natural biological processes like conjugation. However, the bulk of double strand breaks are formed during replication fork collapse encountering an unrepaired single strand gap in DNA. Under such circumstances DNA replication on the damaged template can be resumed only if supported by homologous recombination. This functional cooperation of homologous recombination with replication machinery enables successful completion of genome duplication and faithful transmission of genetic material to a daughter cell. In eukaryotes, homologous recombination is also involved in essential biological processes such as preservation of genome integrity, DNA damage checkpoint activation, DNA damage repair, DNA replication, mating type switching, transposition, immune system development and meiosis. When unregulated, recombination can lead to genome instability and carcinogenesis.

The paper outlines geographic search technology as the future of research methodology. Due to the importance of geographical environment in the routine issues, the close connection between spatial extension and most of the phenomena running over the world, the geographic query process turns to be key factor in overcoming hazard decision-making and evalu-ation. We describe a two step process where derived output from spatio-textual research could be largely disseminated by spatial replication techniques. Spatial research differs in that it requires a combination of text and geographical data. The mathematical formula to calculate distances derives from the assertion that the Earth is nearly spherical (ignoring ellipsoidal effects) so great-circle distance formulas give the distance between points on the surface of the Earth correct to within 0.5% or so. Based on the search range and limit, we introduce the Cosine algorithm who offers better speed performance and data retrieval accuracy. Retrieved data exploit spatial replication technique potentiality. When it comes to spatial data we deal extensive amount of information. Web Gis Albania is an informative platform initially structured for academicals purposes further enriched with modules which support the spatial data circulation.

NHST (null hypothesis significance testing) is the most popular statistical paradigm in psychology. Mistakes in interpretation of its assumptions and their consequences are topic for methodological and statistical discussion for over fifty years. Article presents two problems associated with NHST that are particularly prevalent in psychology: identifying non-significant results with research failure and conducting underpowered research. They can contribute to increase in exploiting questionable research practices in order to obtain desirable, significant outcomes. Three practices: p-hacking, HARKing and selective publishing are described, along with analysis of their impact on replication crisis in psychological science.

Mutagenesis in Escherichia coli, a subject of many years of study is considered to be related to DNA replication. DNA lesions nonrepaired by the error-free nucleotide excision repair (NER), base excision repair (BER) and recombination repair (RR), stop replication at the fork. Reinitiation needs translesion synthesis (TLS) by DNA polymerase V (UmuC), which in the presence of accessory proteins, UmuD', RecA and ssDNA-binding protein (SSB), has an ability to bypass the lesion with high mutagenicity. This enables reinitiation and extension of DNA replication by DNA polymerase III (Pol III). We studied UV- and MMS-induced mutagenesis of λO(am)8 phage in E. coli 594 sup+ host, unable to replicate the phage DNA, as a possible model for mutagenesis induced in nondividing cells (e.g. somatic cells). We show that in E. coli 594 sup+ cells UV- and MMS-induced mutagenesis of λO(am)8 phage may occur. This mutagenic process requires both the UmuD' and C proteins, albeit a high level of UmuD' and low level of UmuC seem to be necessary and sufficient. We compared UV-induced mutagenesis of λO(am)8 in nonpermissive (594 sup+) and permissive (C600 supE) conditions for phage DNA replication. It appeared that while the mutagenesis of λO(am)8 in 594 sup+ requires the UmuD' and C proteins, which can not be replaced by other SOS-inducible protein(s), in C600 supE their functions may be replaced by other inducible protein(s), possibly DNA polymerase IV (DinB). Mutations induced under nonpermissive conditions for phage DNA replication are resistant to mismatch repair (MMR), while among those induced under permissive conditions, only about 40% are resistant.
