The aim of the study was to investigate the changes in proliferation rate, cell cycle and apoptosis of normal skin fibroblasts during fractionated irradiation with a fraction dose of 2 Gy. Fibroblasts were irradiated 5 days per week for 12 days using gamma irradiation. Twenty four hours after each fraction, and for three days after finishing experiment the cells were harvested, fixed, and BrdUrd labelling index (BrdUrdLI), cell cycle and level of apoptosis and debris were assessed. It was found that fractionated irradiation caused disturbances in the proliferation rate and the cell cycle. Irradiation caused also constant, statistically significant increase in the number of G2M cells and level of apoptosis and debris, which was observed even during 3 days after irradiation. Data indicate non equal biological effect of each fraction dose. Block at G2/M phase suggests accumulation of sublethal damage and increased radiosensitivity, which was manifested by elevated level of cell death (apoptosis and debris).

In the present study, four new pegylated drugs were synthesized to investigate the anticancer activity. All the compounds were characterized on the basis of FTIR and 1HNMR. The new compounds were evaluated against HCT116 cell line by CellTiter-Glo® 3D cell viability assay. The sustained release pattern of newly synthesized compounds were also evaluated, slow released lasts for 240 h which was confirmed by in vivo and in vitro drug release profile. The sustained and controlled release behavior of pegylated drugs over the extended period of time showed a linear profile and this release pattern was not limited to diffusion process instead supporting chemically controlled release pattern which was dependent only on how fast the compound hydrolyzed. The newly designed pegylated drugs showed almost same killing effect against the cancer cells as the free drugs. The compounds with lower cLogP value showed more antiproliferative (lower IC50) effect due their higher cell permeability.

In the search for potential therapeutics, isoxazole derivatives are still objects of interest. Previously described immunoregulatory properties of 5-amino-3-methyl-4-isoxazolecarboxylic acid (AC) benzylamides prompted us to synthesize a new class of compounds of immunotropic activity. A series of new compounds containing the isoxazole moiety were synthesized using Passerini three-component reaction. The effects on phytohemagglutinin A (PHA)-induced proliferation of human peripheral blood mononuclear cells (PBMC), production of tumor necrosis factor alpha (TNF α) in human whole blood cultures stimulated with lipopolysaccharide (LPS) and two-way mixed lymphocyte reaction (MLR) of PBMC, were investigated. Also, the effect of 1-(cyclohexylcarbamoyl)cyclohexyl 5-amino-3-methylisoxazole-4-carboxylate (PUB1) on the expression of signaling molecules associated with cell apoptosis in Jurkat cells was also determined. The results showed that the compounds inhibited to various degree mitogen-induced PBMC proliferation in a dose-dependent manner and TNF α production at 10 μg/ml. PUB1 compound, selected on the basis of its strongest antiproliferative activity, was also shown to inhibit MLR. The molecular data suggest that immunosuppressive action of PUB1 depended on induction of Fas and elevation of caspase 8 expression. In summary, we revealed immunosuppressive properties of a new class of isoxazoles and established the mechanism of action of a representative PUB1 compound.

Morphometric procedures were used to examine peroxisome number and di-stribution in pea (Pisum sativum L.) root nodules under NaCl (50 mM) or HgCl2 (7.3 µM) treatment. Peroxisomes were visualized cytochemically in meristem, invasion zone and prefixing zone of pea root nodules by catalase (EC 1.11.1.6) activity. The observations using light and electron microscopy revealed that the peroxisomes were predominantly spherical in shape and showed catalase activity. In nitrogen fixation zone, catalase active peroxisomes were observed occasionally. Bacteroids of nitrogen fixing zone showed enhanced cata-lase activity probably as a response to higher level of oxidative stress. Fluorescence microscopy investigations revealed enhanced level of (homo)glutathione in prefixing and nitrogen-fixing zone of NaCl- and Hg2+treated nodules, which served as an indicator of antioxidative response. Morphometric measurements revealed that during differentiation of meristematic cells into central tissue (bacteroidal tissue) cells an increase in peroxisome number was observed in unstressed nodules. Peroxisomes located in meristem, invasion zone and prefixing zone of NaCl- and Hg2+-treated nodules outnumbered that in control nodules. A substantial enlargement of peroxisome profiles was detected in NaCl- and Hg2+treated nodules. Peroxisome divisions observed in meristematic and infection thread penetration zone were responsible for an increase in peroxisome number.

The purpose of this article is to examine the impact of the creation of nuclear-weapon-free-zone in Africa on the security of this continent, and the state of nuclear non-proliferation regime. First we present the idea of nuclear-weapon-free-zone in general and characterize Pelindaba Treaty in comparison with other arrangements of this type. Next we investigate how South Africa’s nuclear program contributed to the delay in the Africa’s denuclearization and indicate this program as one of the major obstacles to the creation of nuclear-weapon-free zone in Africa. Next we briefly review the factors that led to initiate, implementation and eventually to dismantle South African nuclear program, taking into account political changes inside and outside South Africa after the end of Cold War, especially changes in its security environment. Final remarks concern virtual potential of the African bloc in international forums on disarmament.

Immunosenescence is viewed as a remodeling process with the exhaustion of naïve T cells and filling up of the immunological space with memory cells. In this study some phenotypic changes of CD8+ human cells during in vivo ageing were compared with those observed in long term cultures of lymphocytes derived from cord blood or from peripheral blood from donors of different age. Both in vivo and in vitro a significant decrease of the fraction of CD8+CD28+ cells was observed. Comparing the proportions of other T cell subpopulations (the CD4/CD8+ ratio, CD56, CD57, CD27) made it possible to conclude that replicative senescence in vitro partially reflects in vivo ageing.

Ulcerative colitis (UC) is a inflammatory disease of large bowel. The amount of people suffering from UC increases from year to year. Pathogenesis of this affection is still not entirely clear. Mechanisms of proliferation and apoptosis in colonocytes in the course of the disease are defectedThe aim of the study was to assess the rate of proliferation and intensity of apoptosis in colonocytes in patients with diagnose UC.Material and methods. Colon pathological samples taken from patients with diagnosed ulceraive colitis were examined. Patients were in both clinical and endoscopic remission and were treated with mesalazin. They were patient of Department of General and Colorectal Surgery. To estimate proliferation index dye with monoclonal antibody against Ki67. To determine apoptosis level immunohistochemistry with antybody against Bax was used.Results. Average Ki-67 in the test group was 42,13%, the largest value amounted to 57% and the lowest of 33%. Average value of Bax was 1.47 and ranged between 0-3. High index of bax appear not only in the bottom of the crypt, but also at their outlet.Conclusions. In ulecerative colitis genetic and immunological disturbances occur despite treatment. Mesalazine acting only on certain routes associated with the UC holds the remission, without, however "the molecular remission". Thus, it appears that the results of our research are another proof of the necessary caution in weaning support treatment.

Opis:
The effect on cell viability and content of PhC of three grapes varieties – Moravian Muscat, Blue Burgundy and Lemberger is presented. The effect of polyphenols from wine and grapes was studied for many times, but the effect of pomace, the by-product of wine production, was neglected. Thus study is devoted to compare the effect of berries and pomace on cell viability in context of their utilization as source of bioactive compounds. Effect on viability of human keratinocytes (HaCaT) was investigated in vitro using following concentrations of PhC in cultivation medium: 25, 50, 75 and 100 µg·ml–1. The results show that the content of PhC in berries and pomace was similar and the cell viability decreased with increasing concentrations of PhC, in most cases. The impact on cell viability also depends on individual variety of grapes.

Proliferation of internal armed conflicts poses a challenge to the framework of international humanitarian law (IHL) as it was primarily designed to guide states’ behaviour while at war. Are the existing mechanisms available under IHL sufficient to ensure compliance with the law by armed groups? The purpose of this article is to examine how socialisation, by which I mean a process of internalising a norm or a rule so that external pressure is no longer necessary to ensure compliance, might contribute to producing long-term rule-consistent behaviour. I begin with the identification of the legal framework applicable to internal conflicts and challenges associated with its implementation. Following is the discussion of the ideas on socialisation and their utility in the context of IHL and armed groups. I then proceed to present a particular example of positive engagement with such groups under the so-called Deed of Commitment, launched by a Swiss NGO, Geneva Call. Finally, I return to my analytical framework, apply it to the system established under the Deed of Commitment, and assess to what extent socialisation and positive engagement may ensure greater respect for humanitarian rules. I conclude that socialisation is a valuable tool that may complement the existing mechanisms under IHL and, under certain conditions, contribute to long-term rule-consistent behaviour.
Rozprzestrzenianie się międzynarodowych konfliktów zbrojnych gromadzi wyzwania dla zakresu oddziaływania międzynarodowego prawa humanitarnego (IHL), którego założycielską ideą było kształtowanie zasad zachowania państw w czasie wojen. Dlatego należy zadać pytanie, czy istniejące mechanizmy w zakresie międzynarodowego prawa humanitarnego (IHL) są wystarczające, aby zapewnić zgodność w zakresie prawnym w odniesieniu do funkcjonowania ugrupowań zbrojnych? Ideą tego artykułu jest zbadanie, jak uspołecznienie, prze która autorka postrzega proces umiędzynarodowienia zasad lub reguł, tak aby zewnętrzny wpływ nie był dalej konieczny, w zapewnieniu zgodności, co w perspektywie może być źródłem do tworzenia długoterminowych spójnych zasad zachowań. We wstępie artykułu wyszczególniono ramy prawne mające zastosowanie w konfliktach zbrojnych oraz wyzwań towarzyszących ich wdrażaniu. W dalszych rozważania jest prezentowana dyskusja koncepcji uspołeczniania oraz jej przydatność w kontekście międzynarodowego prawa humanitarnego (IHL) oraz działania ugrupowań zbrojnych. W dalszej części autorka przechodzi do prezentowania wybranych przykładów pozytywnej współdziałania z takimi ugrupowaniami w ramach tzw.„Deed of Commitment”, czyli inicjatywa aktów dobrej woli zorganizowana przez Szwajcarskie Organizacje Pozarządowe (NGOs), Geneva Calls. Ostatecznie autorka powraca do analitycznego obszaru badawczego i odnosi wyniki badan do ram działań w systemie ustanowionym przez „Deed of Commitment”, aby ostatecznie ocenić w jakim zakresie uspołecznienie oraz pozytywne zaangażowanie mogą zapewnić większe zrozumienie dla stosowania humanitarnych zasad. W podsumowaniu, stwierdza się, że uspołecznienie jest znakomitym narzędziem, który może uzupełniać mechanizmy funkcjonowania międzynarodowego prawa humanitarnego (IHL) oraz w szczególnych warunkach wnosić wartość dodaną do długoterminowych spójnych zasad zachowań.

Plants of Schlumbergera truncata (Haw.) Moran were obtained by indirect morphogenesis from the segment section of shoots in vitro, they were multiplied and rooted. Also were determined the effect of the lighting regime, the composition of the nutrient medium on the consistency and frequency of callus formation. The studies were conducted during 2016–2018. The mode of effective sterilization (more than 90%) of S. truncata plant explants using 0.1% HgCl2 for 7–8 min was established. Optimal conditions for the induction of callus formation in stem node segments of S. truncata plants (rate more than 90% and significant growth) were created on MS (Murashige and Skoog 1962) nutrient medium supplemented with 1.0 mg/l BAP (6-benzylaminopurine) and 0.3 mg/l NAA (1-naphthylacetic acid) under conditions of placement on the nutrient medium and doing a significant number of cuts on the explants. The light intensity of 2.0–3.0 klx, obtained by a callus of dense consistency of dark green pigmentation, when using the thermostat condition without illumination, the callus had loose consistency, dark yellow pigmentation. It is established that the influence of the lighting regime and the composition of the nutrient medium on the frequency of callus formation is statistically significant. The largest number of shoots was obtained on the MS medium with the addition of 2.0 mg/l of BA. At the same times, shoot proliferation and root induction in such numbers were observed on MS culture medium with the addition of 0.5 mg/l BA and 0.5 mg/l kinetin (multiplication factor – 8.8±0.6 per 60-day cultivation cycle).

During 2015, samples from 22 apple trees showing proliferation symptoms were collected in southwest Bulgaria and Central and South Poland and tested for phytoplasma presence. ‘Candidatus Phytoplasma mali’ was identified in 18 samples based on results of restriction fragment length polymorphism (RFLP) analysis of the 16S rRNA gene amplified in nested PCR using primer pair P1/P7 followed by R16F2n/R16R2 and F1/B6 primer pairs. The nitroreductase and rhodonase-like genes and ribosomal protein genes rpl22 and rps3 were then analyzed using PCR-RFLP technique to study the genetic variability of the phytoplasma strains. Two restriction profiles, P-I or P-II, were obtained from fragments of 16S rDNA plus 16S-23S spacer region digested with HpaII enzyme. Restriction fragment length polymorphism analysis of nitroreductase and rhodonase-like genes using digestion with HincII endonuclease revealed that all ‘Ca. P. mali’ strains belonged to the subtype AP-15. Analysis of rpl22 and rps3 ribosomal protein genes digested with AluI enzyme resulted in classification of detected phytoplasma strains to rpX-A subgroup.

Opis:
Retinoic acid and transforming growth factor-β (TGF-β) affect differentiation, proliferation and carcinogenesis of epithelial cells. The effect of both compounds on the proliferation of cells of the hormone sensitive human breast cancer cell line (ER+) MCF-7 was assessed in the presence of estradiol and tamoxifen. The assay was based on [3H]thymidine incorporation and the proliferative activity of PCNA- and Ki 67-positive cells. The apoptotic index and expression of the Bcl-2 and p53 antigens in MCF-7 cells were also determined. Exogenous TGF-β1 added to the cell culture showed antiproliferative activity within the concentration range of 0.003-30 ng/ml. Irrespective of TGF-β1 concentrations, a marked reduction in the stimulatory action of estradiol (10-9 and 10-8 M) was observed whereas in combination with tamoxifen (10-7 and 10-6 M) only 30 ng/ml TGF-β1 caused a statistically significant reduction to aproximately 30% of the proliferative cells. In further experiments we examined the effect of exposure of breast cancer cells to retinoids in combination with TGF-β1. The incorporation of [3H]thymidine into MCF-7 cells was inhibited to 52 ± 19% (control =100%) by 3 ng/ml TGF-β1, and this dose was used throughout. It was found that addition of TGF-β1 and isotretinoin to the culture did not decrease proliferation, while TGF-β1 and tretinoin at low concentrations (3 × 10-8 and 3 × 10-7 M) reduced the percentage of proliferating cells by aproximately 30% (67±8% and 67±5%, P <0.05 compared to values in the tretinoin group). Both retinoids also led to a statistically significant decrease in the stimulatory effect of 10-9 M estradiol, attenuated by TGF-β1. In addition, the retinoids in combination with TGF-β1 and tamoxifen (10-6 M) caused a further reduction in the percentage of proliferating cells. Immunocytochemical analysis showed that all the examined compounds gave a statistically significant reduction in the percentage of cells with a positive reaction to PCNA and Ki 67 antigen. TGF-β1, isotretinoin and tretinoin added to the culture resulted in the lowest percentage of PCNA positive cells. However, the lowest fraction of Ki 67 positive cells was observed after addition of isotretinoin. The obtained results also confirm the fact that the well-known regulatory proteins Bcl-2 and p53 play an important role in the regulation of apoptosis in the MCF-7 cell line, with lowered Bcl-2 expression accompanying easier apoptotic induction. The majority of the examined compounds act via the p53 pathway although some bypass this important proapoptotic factor.
