Opis:
Telfairia occidentalis leaves and Cucumis sativus (cucumber) are vegetables that grow best in Tropical countries like Nigeria and have good nutritional properties but are highly perishable. Both vegetables were evaluated for wine production singly and in composite forms using palm wine and Baker’s yeast strains. In addition to wine production, the vegetables were screened for phytochemicals, anti-nutrients and nutrients (proximate composition, elements and vitamins) using standard methodologies. Proximate composition (g/100g dry matter) revealed that T. occidentalis leaves had moisture content, ash, protein, fat, fibre and carbohydrate contents of 86.60±0.10, 5.72±0.02, 4.30±0.10, 0.97±0.02, 6.30±0.10 and 82.45±0.02, respectively. In contrast, Cucumis sativus had a moisture content, ash, protein, fat, fibre and carbohydrate contents of 96.75±0.01, 5.40±0.02, 13.50±0.02, 10.30±0.10, and 66.12±0.01, respectively. Both plants contained various phytochemicals, including alkaloids, glycosides, saponins, tannins, flavonoids, reducing compounds and polyphenol, but not anthraquinones and phlabotanins. However, on quantification, reducing compounds and polyphenol recorded the highest concentrations. Vitamins A and C were detected in both samples. C. sativus had high amounts of K, P, and Mg and moderate amounts of Ca, Na, Cu, Fe and Mg. T. occidentalis leaves had moderate amounts of Ca, Zn, Fe, Mg, Ca and Na. Anti-nutrients levels were all below allowable regulatory limits for vegetables. Density, pH and alcohol values ranged from 0.20 - 0.99, 0.30- 4.6 and 1.95 - 9.94%, respectively. Sensory evaluation of the wine samples showed that wine produced from 70% fluted pumpkin and 30% cucumber had the best scores in terms of acceptability, taste, aroma and appearance. Given the findings in the study, wine production using tropical vegetables could double as a viable alternative to tropical fruits and also help to curb post-harvest losses commonly experienced in the tropics.

Palm wine in Nigeria is considered a beverage. Its usage in local and traditional events pre-dates history of man’s existence. Recently bio-mining of this local drink for high throughput microbial strains have not gained much attention to scientist. This study is aimed at characterizing yeasts isolated from freshly tapped palm wine in Alakahia, Rivers State, Nigeria. A total of 4 samples was analysed. Total yeast count was done on glucose yeast agar (GYA) using spread plate technique. The yeast isolates were identified by molecular characterization. DNA sequence analyses was achieved through DNA extraction using CTAB procedure, amplification of regions of rRNA/Internal Transcribed Spacer sequence (ITS) and purification of the PCR products. The amplified region was done using ITS1 and ITS4 which are recommended universal primers for fungi identifications. The total yeast count ranged from 6.5×104 to 3.2×108 cfu/ml. Gene amplification bands revealed that the molecular weight of the isolates were 500 bp. The BLAST and phylogenetic analysis of F2 and F3 amplicons had 80.04% similarity with Saccharomyces cerevisiae strain CS11 with 50% frequency of occurrence while F4 had 100% similarity with Candida ethanolica with 25% frequency of occurrence and F1 had 99.8% similarity with Pichia sp. feni 106 with also 25% frequency of occurrence. Statistical analysis using two-way ANOVA showed that the microbial quality of the palm wines is location specific. This investigation further suggests that raffia palm can be harnessed in bioprospecting of high-throughput yeast starter in biotechnological studies.

In the present study, we described the protein profile experimentally by 2D-PAGE and MALDI analysis to understand the stress mechanisms of cocoti sap and wine on E.coli Nissle 1917. We isolated one newly expressed protein from cocoti wine treated gel which is not present in both control and cocoti sap treated sample i.e. P21 prophage-derived head-stabilizing proteinVG03_ECOL6 (3n1) also called as Head protein gp3. This protein mainly activities related to the viral life cycle. It helps to attach the viral gene into host. The growth rate was delayed in cocoti wine treated E.coli Nissle 1917 when compared to control and cocoti sap treated samples. Stress mechanism induce many proteins they are involved in metabolic process, hydrolase activity, lyase activity, quinone binding, phosphotransferase system, carbohydrate metabolism, DNA binding, DNA repair, transferase activity, oxidoreductase, purine metabolism, transcription antitermination, transcription regulation and other related activities. We proved that the predicted protein structure quality, resolution, density and error plot values by QMEAN analysis. Based on these results, only two differentially expressed proteins under sap stress showed that the significant results, which were N-acetylgalactosamine-specific phosphotransferase enzyme IIB component 1, PTPB1_ECOLI and DinI-like protein Z3305/ECs2939 in prophage CP-933VDINI1_ECO57. In case of wine stress, the differentially expressed proteins were Transcription anti-termination protein RFAH- ECO57 NusA and PUR7- eco24- phosphoribosylamidazole-succinocarboxamide synthase showed significant results. ProtParam analysis indicating that the multiple physico-chemical characters of differentially expressed proteins were differed and compared. The phylogenetic tree represents the relationship in-between the differentially expressed proteins, were showed siblings (related) as well as monophytic clade.
