Polyhydroxyalkanoates (PHAs) have gained much attention as biodegradable polymers, many efforts are being made to minimize the cost of PHAs by finding cheap carbon source depending on the type of microorganism and fermentation conditions. The aims of this study were to evaluate the effects of different glucose concentrations and other important conditions on the PHA production by Bacillus cereus isolated from soil. Polyhydroxyalkanoates PHAs accumulated by soil microorganisms were examined by screening the isolated bacteria using Sudan B Black and Nile Blue staining process. A Gram positive strain was identified using the 16s rRNA gene, deposited in the NCBI GenBank sequence database. Different growth conditions (favorite glucose concentrations 1-8 % (w/v), temperatures and pH) were tested and the growth parameters (sugar consumption, cell counting and Cell Dry Weight CDW) were studied. The extracted polymers were analyzed and characterized using an FTIR spectrophotometer followed by a GC-MS analysis. The pure bacterial strain isolated from soil was deposited in the NCBI GenBank database B. cereus strain ARY73, which showed significant black colored granules (or dark blue) using Sudan B Black stain, it also showed positive to Nile blue A as a high indicator stain for PHA accumulation. B. cereus ARY73 showed high production of PHA using (w/v): 2% glucose and 1% nitrogen source at 35 °C and pH7 yields 79% per Cell Dry Weight and 96 h of incubation. The extracted polymers were analyzed and characterized using an FTIR spectrophotometer confirming the PHA structure. The FTIR spectrophotometer, followed by a GC-MS analysis indicated the Scl-co-mcl PHA structure. This research demonstrates that the isolated strain B. cereus ARY73 was a good candidate for PHA production with a better quality for use in biomedical and other applications. The use of biopolymer in soil, enhanced the accumulation of the microorganisms (such as bacteria) capable of degrading biopolymer or biodegradation by-products yields by other species which were isolated in this study and demonstrated their efficiency in producing biopolymers.

The main aim of this study was to compare the reaction of quiescent and proliferating, i.e. phytohemagglutinin (PHA)-stimulated, human peripheral blood mononuclear cells (PBMCs) to γ-radiation, and analyse changes of proteins related to repair of DNA damage and apoptosis, such as γH2A.X, p53, p53 phosphorylation at serines-15 and -392, and p21 and their dose dependence. Freshly isolated PBMCs in peripheral blood are predominantly quiescent, in G0 phase, and with very low amounts of proteins p53 and p21. Using confocal microscopy we detected dose dependent (0.5-5 Gy) induction of foci containing γH2A.X (1 h after γ-ray exposure), which are formed around radiation-induced double strand breaks of DNA. Apoptosis was detected from 24 h after irradiation by the dose of 4 Gy onwards by Annexin V binding and lamin B cleavage. Seventy two hours after irradiation 70% of CD3+ lymphocytes were A+. Neither increase in p53 nor its phosphorylation on serine-392 after irradiation was detected in these cells. However, massive increase in p21 (cyclin-dependent kinase inhibitor 1A) was detected after irradiation, which can be responsible for late occurrence of apoptosis in these quiescent cells. PHA-stimulation itself (72 h) caused an increase in early apoptosis (A+PI-) in comparison to non-stimulated PBMCs (38% A+ resp. 13.4%). After PHA-stimulation also the amount of γH2A.X, p53, and p21 increased, but no phosphorylation of p53 on serine-392 or -15 was detected. Reaction to γ-radiation was different in PHA-stimulated lymphocytes: the p53 pathway was activated and p53 was phosphorylated on serines-15 and -392 4 h after irradiation by the dose of 4 Gy. Phosphorylation of p53 at serine-15 increased in a dose-dependent manner in the studied dose range 0.2-7.5 Gy. Also the amount of p21 increased after irradiation. Seventy two hours after irradiation of PHA-stimulated CD3+ T lymphocytes by the dose of 4 Gy 65% of cells were A+.

The subject of the study are space threats – Near-Earth Objects (NEO) and Potentially Hazardous Asteroids (PHA). The research methods employed in this article included the classic theoretical methods used in security sciences and a practical method – a quantitative study of social media. At present, space threat studies aim to resolve the terminological confusion related to NEOs, determine currently and potentially hazardous space objects and estimate the potential threats they could cause. Another expected result of the research is to present two literature methods for estimating NEO threats, the Palermo and Torino scales. The practical result is to evaluate public mood regarding NEO threats. Studies have shown that certain active space objects are capable of reaching the Earth's surface or impacting human-made in-space objects and devices, such as communication satellites. Should it happen, it could cause substantial social damage and destabilise state security, particularly if elements of critical infrastructure of the state were to be affected. Continuous monitoring of NEOs may play a central role in the provision of security. Furthermore, the public should be kept abreast of the threats.

Celem pracy był pomiar i poznanie zmian ilościowych polihydroksyalkanianów (PHA) w dwóch oczyszczalniach ścieków komunalnych województwa łódzkiego. Dodatkowo, porównano dwie metody oznaczania PHA, to jest metodę chromatograficzna i opatią na cyfrowej analizie obrazu. Zaobserwowano zgodność pomiędzy wynikami obydwu metod zastosowanych w pracy, choć zasada oznaczenia jest odmienna. Obie metody mogą być wykorzystane do monitorowania zmian PHA w komorach osadu czynnego oczyszczalni ścieków.
The aim of this work was to identify and measure the variations of polyhydroxyalkanoates quantity (PHA) in two wastewater treatment plants (WWTPs) in the Lodz voivodship. Two methods of PHA assay were compared, namely chromatographic method and method based on digital image analysis. The results obtained using both of them were compatible, despite the difference in the rule of analytical approach. Both methods can be used to monitor PHA in the activated sludge chambers of WWTPs.

Factors limiting polyhydroxyalkanoates (PHA) recovery from activated sludge were examined. The biosynthesis of the polymer was carried out for 3 months in 7-day cycles in a laboratory reactor inoculated with activated sludge from municipal wastewater treatment plant. The reactor was continuously aerated and PHA synthesis was stimulated by nitrogen limitation in the medium. PHA recovery at the end of each cycle was performed by biomass centrifugation, extraction of the biopolymer with chloroform and its precipitation with n-hexane. Excess organic carbon in the medium was utilized by microorganisms to accumulate PHA during sludge regeneration phase. Parameters of the biosynthesis were conducive to production of other biopolymers, which lowered efficacy of PHA recovery from biomass, despite presence of PHA granules in the bacterial cells in the amount of 10–19% (dry weight). The PHA recovery by extraction with chloroform and subsequent precipitation of biopolymer with n-hexane was inefficient. High carbon to nitrogen ratio in the medium stimulated growth of filamentous and slime-forming microorganisms. It resulted in the increased viscosity and density of the culture, difficulties during biomass separation and limited chloroform penetration into bacterial cells. However, the method of PHA biosynthesis and recovery proved efficient for the pure culture of Cupriavidus necator NCIMB 10442 (49% PHA per dry weight of cells).
Badano czynniki decydujące o skuteczności procesu pozyskiwania polihydroksykwasów (PHA) z osadu czynnego. Biosyntezę polimeru prowadzono przez 3 miesiące w cyklach trwających 7 dni w reaktorze laboratoryjnym zaszczepionym osadem czynnym z miejskiej oczyszczalni ścieków. Hodowlę napowietrzano w sposób ciągły, a syntezę PHA stymulowano poprzez ograniczanie zawartości azotu w podłożu. Pod koniec każdego cyklu przeprowadzano odzyskiwanie biopolimeru przez odwirowanie biomasy, ekstrakcję PHA chloroformem i jego wytrącenie w n-heksanie. Stwierdzono, że zastosowany nadmiar węgla organicznego w podłożu był wykorzystywany przez mikroorganizmy do kumulacji PHA w fazie regeneracji osadu. Warunki biosyntezy sprzyjały powstawaniu także innych biopolimerów, co obniżyło skuteczność odzyskiwania PHA z biomasy, pomimo obecności granulek PHA w komórkach bakterii w ilości 10÷19%. Sposób pozyskania polimeru metodą ekstrakcji chloroformem z późniejszym wytrącaniem w n-heksanie okazał się nieskuteczny. Wysoki stosunek węgla do azotu w podłożu stymulował rozwój mikroorganizmów nitkowatych i wytwarzających śluz. Spowodowało to zwiększenie lepkości i gęstości hodowli, trudności podczas sedymentacji biomasy i utrudnioną penetrację rozpuszczalnika w głąb komórek. Zastosowana metoda biosyntezy i odzysku PHA okazała się natomiast w pełni skuteczna w przypadku czystej hodowli szczepu Cupriavidus necator NCIMB 10442 – uzyskano 49% stopień odzyskania biopolimeru w przeliczeniu na zawartość suchej masy komórek.

For commercial success, enhanced poly(hydroxyalkanoate) (PHA) production must address both material performance and economic aspects. Conventional PHA production consumes expensive feedstocks dedicated to nutrition. Switching to carbon-rich (agro)industrial side-streams alleviates industrial disposal problems, preserves food resources, and can be economically superior. Processes developed in the recently performed EU-FP7 project ANIMPOL resort to lipid-rich surplus streams from slaughterhouses and the rendering industry; these materials undergo chemical transformation to crude glycerol phase (CGP) and biodiesel. The saturated biodiesel share (SFAE) counteracts its applicability as abiofuel but, in addition to CGP, can be converted biotechnologically to PHAs. Depending on the applied microbial production strain and the selected carbon source (SFAE or CGP), thermoplastic short chain length PHA (scl-PHA), as well as elastomeric to latex-like medium chain length PHA (mcl-PHA), can be produced from these inexpensive feed stocks. The article illustrates the biotechnological conversion of animal-based CGP and SFAE towards poly(3-hydroxybutyrate) (PHB) and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), respectively, by Cupriavidus necator strain DSM 545. SFAE conversion towards mcl-PHAs consisting of various saturated and unsaturated building blocks by two pseudomonades, Ps. citronellolis DSM 50332 and Ps. chlororaphis DSM 50083, are also shown. Together with the kinetics of the bioprocesses, the results from the characterization of isolated samples of these structurally diverse biopolyesters are compared; data demonstrate the high versatility of biopolymer properties making them applicable in various fields of the plastic market. In addition to the need for inexpensive carbon feed stocks, the article points to further hot spots of the PHA-production chain that must be considered in order to lower the overall PHA production costs, and to enhance product quality. The benefits arising from multistage continuous cultivation production set-ups, namely high-throughput production of PHA of predefined composition and constant quality, are especially discussed. Finally, contemporary approaches towards environmentally and ecologically sustainable PHA recovery from biomass are summarized.
Artykuł stanowi przegląd literatury dotyczącej biosyntezy poli(hydroksyalkanianów) (PHA) z wykorzystaniem jako surowców odpadów z przemysłu rolno-spożywczego. Omówiono wyniki uzyskane podczas realizacji projektu ANIMPOL (7. PR UE), w którym jako surowiec do syntezy PHA stosowano bogate w tłuszcze produkty uboczne z rzeźni i zakładów utylizacji odpadów zwierzęcych, przekształcone chemicznie w surową fazę glicerynową (CGP) i biodiesel (estry nasyconych kwasów tłuszczowych, SFAE). Zależnie od użytego szczepu bakterii oraz źródła węgla (SFAE lub CGP) otrzymano termoplastyczne krótkołańcuchowe PHA (scl-PHA) lub średniołańcuchowe PHA (mcl-PHA). Zaprezentowano biotechnologiczną konwersję CGP iSFAE pochodzenia zwierzęcego do poli(3-hydroksymaślanu) (PHB) i poli(3-hydroksymaślanu-co-3-hydroksywalerianu) (PHBV) za pomocą szczepu bakteryjnego Cupriavidus necator strain DSM 545, syntezę mcl-PHA zawierających nasycone i nienasycone elementy strukturalne za pomocą bakterii z rodzaju Pseudomonas (Ps. citronellolis DSM 50332 i Ps. chlororaphis DSM 50083). Omówiono kinetykę bioprocesów oraz charakterystykę otrzymanych biopoliestrów, przedyskutowano elementy cyklu produkcyjnego PHA kluczowe z punktu widzenia zmniejszenia kosztów i poprawy jakości produktów oraz korzyści wynikające z zastosowania układów ciągłej wielostopniowej hodowli w wysokowydajnej produkcji PHA o założonym składzie i stabilnej jakości. Omówiono też nowe metody odzyskiwania PHA z biomasy, zgodne z wymaganiami ochrony środowiska.

Polymers of biological origin have become a topic of interest due to growing concerns about the environmental impact of the disposal of plastics. In recent years, the production of ecobenign microbial polymer polyhydroxyalkanoates (PHAs) using inexpensive and renewable resources has gained significant interest as these compounds are highly biodegradable, biocompatible, and sustainable. This study used leaf endophytic isolate Bacillus cereus RCL 02, obtained from the oil-yielding plant Ricinus communis L., to achieve statistical optimization of culture variables for the enhanced production of PHAs utilizing sugarcane molasses as the sole carbon source. A threelevel and four-factor Box–Behnken design of response surface methodology was implemented to optimize the process variables, namely molasses (carbon substrate), ammonium sulfate (nitrogen source), initial pH, and incubation period, for improved biomass formation and PHA production. The highest growth (14.8 g/l) and PHA production (85.2%, dry cell weight) by the isolate were observed with 47 g/l molasses, 3 g/l ammonium sulfate, an initial pH of 6.7, and 62 h of incubation. Statistical optimization of the process allowed achieving a 1.6-fold increase in the PHA yield (7.8–12.6 g/l) compared with the conventional single-factor system of analysis. The biopolymer thus produced was confirmed as a copolymer of 3 hydroxybutyrate and 3-hydroxyvalerate [P(3HB-co3HV)] using 1 H nuclear magnetic resonance spectroscopic analysis and was found to contain 7.8 mol% 3 hydroxyvalerate. These findings clearly indicate the efficacy of the B. cereus RCL 02 isolate in the biotransformation of raw sugarcane molasses to P(3HV-co-3HV), without the need for supplementation with high-cost precursors.

Introduction. A wheelchair is a special vehicle designed specifically for people with walking limitations. One of the types is an active wheelchair. This is a manually propelled wheelchair, the design and structure of which maximise the mobility of its given user at the expense of accepted instability. Objective. The aim of this study was to identify and present a hierarchy of potential incidents causing a health hazard and reducing the mobility of individuals with ambulatory disability who use active wheelchairs in non-urban settings. Materials and methods. Information about incidents connected with wheelchair use was collected from interviews with disabled individuals, based on a questionnaire. Recorded data were analysed using the Preliminary Hazard Analysis (PHA). Results. The results comprise a list of hazards for wheelchair user with ascribed priority numbers metrising their significance. Conclusions. An ordered list of hazards connected with wheelchair operation situations is useful when designing active wheelchairs and when learning the technique of wheelchair riding.

Przedstawiono metodologię badań procesu biodegradacji prototypowych wyrobów wytworzonych z polilaktydu i jego kompozycji polimerowych z polihydroksyalkanianami w warunkach kompostowania przemysłowego. Próbki inkubowano w pryzmie kompostowej, systemie kontenerowym KNEER oraz systemie BIODEGMA na terenie Stacji Segregacji i Kompostowni w Zabrzu. Opracowana metodologia pozwala ocenić, czy testowane materiały i wytworzone z nich produkty końcowe o odpowiednich właściwościach użytkowych, po spełnieniu swojej funkcji, kwalifikują się do recyklingu organicznego.
This work presents a methodology for testing the biodegradation process of prototype products made of polylactide and its polymer compositions with polyhydroxyalkanoates under conditions of industrial composting. The samples were incubated in a composting pile, KNEER containers system or BIODEGMA system at the Segregation and Composting Station in Zabrze. The developed methodology allows the assessment of whether the tested materials and final products, with appropriate functional properties, are suitable for organic recycling.

Objective: The aim of this work was to study the in vivo effect of AAES on cellular (leukocyte-induced cutaneous angiogenesis, LIA test, and proliferative response to PHA) and humoral (anti-SRBC antibody response)immunity in mice. Methods: Balb/c mice were fed AAES from 0.5 to 75 mg/kg body mass for seven days before grafting their splenocytes intradermally to F1 (Balb/cxC3H) recipients (LIA test). Neovascular reaction was evaluated 72 h later in dissection microscope. Spleen cell cultures were incubated with 0.5, 1 and 2 µg/ml of PHA. After 48 h of incubation, tritiated thymidine was added. After further 24 h, cells were harvested (Skatron) and incorporation of tritiated thymidine was measured using Beta-scintillation counter. Balb/c mice were fed for 7 days with AAES, then immunized intraperitoneally with 5% SRBC suspension and 7 days later the antibody response was measured with hemagglutination test. Results: Neovascular reaction was significantly higher in groups grafted with splenocytes collected from all AAES fed donors than from the controls. The proliferation of splenocytes taken from mice fed AAES at doses ranging from 0.5 mg/kg to 7.5 mg/kg was stimulated in all cultures. Suppression of proliferation was observed in cell cultures derived from mice fed with higher doses of AAES. Stimulation of anti-SRBC antibody production was seen in mice fed both 2.5 and 7.5 mg/kg dose of AAES. Conclusion: Powdered form of Biostymina (AAES) might be useful in the treatment of patients with ischaemia of tissues and organs (myocardial infarction, stroke, necrosis) and in deficiency in the production of immune cells and growth factors (infections, chronic wound healing, ulceration and bone fusion).
