Monocytes are comprised of three phenotypically and functionally distinct subsets: classical CD14++CD16-, intermediate CD14++CD16+ and non-classical CD14+CD16++ cells that can play differential roles in regulation of both systemic and local inflammatory processes. In addition, these monocyte subsets represent differential developmental stages with CD16-positive monocytes being the most mature cells that can be considered direct precursors of tissue macrophages. Monocytes and, most significantly, monocyte-derived macrophages constitute an important component of both normal and asthmatic airways. Here we summarize the current knowledge on the roles of monocytes and macrophages in asthma pathogenesis. In addition, we discuss here the usefulness of standard and potential monocyte-directed anti-asthmatic therapeutic approaches.

Opis:
This study evaluated the in vitro leukocyte response to titanium implants in the presence of autologous or heterologous antimicrobial peptides extracts. Antimicrobial peptides (AMPs) appeared to be a new approach both against microorganisms and for regulation of inflammatory and repair processes. To evaluate their potential usefulness in regenerative medicine, we prepared different extracts of neutrophil-derived AMPs from rabbit, ovine or porcine blood which contained AMPs of different compositions, mainly defensins, cathelicidins and fragments thereof. Then, we assessed in vitro the influence of different AMPs extracts on the neutrophils and monocyte-derived macrophages (MDM) activity. For this purpose, these cells were obtained from experimental animals, rabbits, or sheep submitted to insertion of a titanium implant into the tibial defect. The cultured cells stimulation was autologous or heterologous, dependently on the AMPs extract origin and the experimental animal species. The neutrophil activity was assessed on the basis of the enzymes release from azurophilic and secondary granules and the free radicals generation. The MDM functional assessment was based on the NO and superoxide generation and arginase activity. Additionally, morphological changes were evaluated in the cell cultures. Our results indicated that the origin of AMPs extract is crucial for its activity. The autologous extracts stimulated anti-inflammatory responses, whereas the heterologous extracts displayed pro-inflammatory effect on neutrophils and macrophages. These results might be considered during the introduction of new preparations in regenerative medicine.

Introduction: Colorectal cancer (CRC) is one of the most common cancers in Poland. The aim of this study was to investigate the clinical significance of absolute monocyte count, neutrophil-to-monocyte ratio (NMR) and monocytetolymphocyte ratio (MLR) in pre- and postoperative blood samples of patients with CRC. Materials and Methods: We retrospectively reviewed medical records of 160 patients diagnosed with CRC who underwent surgery. Blood samples were obtained within 3 days before and after the surgical treatment. Venous blood samples were also obtained from 42 healthy controls. Results: Pre- and postoperative NMR were significantly higher than healthy controls (p<0.0001; p<0.0001). Moreover, MLR in pre-and postoperative blood samples were higher than voluntaries (p<0.001; p<0.001). The area under the ROC curve for pre and postNMR showed that the parameter exhibits strong diagnostic power (1.000). Pre- and postMLR had moderate diagnostic power amount 0.751 and 0.746. There is also correlation between monocyte count in samples obtained before and after surgery and, lymph node metastasis and size of lymph node metastasis in both cases. PreNMR value was significantly associated with venous and lymphatic invasion and the presence of cancer deposits. PostNMR was found to correlate with presence of distant metastasis and cancer cell deposits (R=0.633, p<0.001; R=0.158, p=0.040). Moreover, preMLR value was correlated with only perineural invasion. Conclusions: Analyzed hematologic markers may be useful as simply obtained parameters, next to histopathological examination, that determine a systemic immune response

Cel: Celem niniejszego badania było zbadanie związku, o ile on istnieje, między wartością MHR a idiopatycznym szumem usznym. Projekt badania: Prospektywne badanie kliniczno-kontrolne. Materiał i metody: Do badania włączono 89 pacjentów (grupa badana), u których od marca 2015 roku do czerwca 2016 roku w ambulatorium przyklinicznym zdiagnozowano idiopatyczne szumy uszne oraz 87 zdrowych osób, które zgłosiły się do szpitala w celu przeprowadzenia rutynowego badania lekarskiego i uzyskały prawidłowe wyniki badania audiometrycznego i otoskopowego (zdrowa grupa kontrolna). Próbki krwi pobierano od pacjentów podczas pierwszego badania. Współczynnik HDL (MHR – Monocyte/HDL rate) uzyskano przez prosty podział liczby monocytów przez poziom HDL. Współczynniki MHR uzyskane dla obu grup poddano analizie statystycznej. Wyniki: Liczba monocytów i wartość MHR były większe w grupie badanej, a poziom stężenia HDL był niższy w porównaniu z grupą kontrolną (p<0,05). W niniejszym badaniu wartość odcięcia dla MHR w odniesieniu do szumów usznych ustalono na 11. Wniosek: Współczynnik MHR okazał się wysoki u pacjentów z idiopatycznymi szumami usznymi. Uzyskany wynik wskazuje na udział zdarzeń miażdżycowych i stresu oksydacyjnego w etiologii szumów usznych.

Objective: In this study, our aim was to investigate whether Monocyte/HDL ratio is a marker of the prognosis of the idiopathic sudden hearing loss (ISHL). Study design: Retrospective, case-control clinical trial. Materials and Methods: 45 patients, who were diagnosed with idiopathic sudden hearing loss and were treated with the same therapy regime and 47 healthy volunteers, who applied to the hospital for routine controls and had audiological and laboratory examination between March 2014 and December 2015, were included in the study. Monocyte/HDL ratios of the patients in the study and control groups were calculated from the results of the blood counts and biochemical analysis. Additionally, the study group was divided into two sub-groups regarding their responses (responders and non-responders) to the treatment determined by the audiological examination, which was carried out after 3 months according to the Siegel criteria. The Monocyte/HDL ratios between the groups were statistically evaluated. Results: There was no statistically significant difference between the MHRs of the study and control groups (p=0.574). However, the MHR was significantly higher in the non-responders’ group compared with the responders’ group, although they were treated with the same therapy regimen (p=0.005). Conclusion: There was no difference in MHRs between study and control groups. However, as MHR was higher in the patients with good prognosis compared with the patients with bad prognosis, we believe that regarding the ISHL, MHR is not a predictive value but might have prognostic marker.

The objective of this study was to determine the effect of ß-l,3/l,6-D-glucan (Biolex-Beta HP) on the phagocytic activity and oxidative metabolism of peripheral blood granulocytes and monocytes in rats intoxicated by cyclophosphamide. The experimental material comprised 40 adult Wistar rats aged 14 weeks, divided into two equal groups, a control group and an experimental group, each of 20 adult rats, including 10 males and 10 females. In the course of 3 successive days, 20 rats from the experimental group were administered cyclophosphamide intramuscularly at a dose of 50 mg/kg body weight/day. On the 8th day of the experiment, 10 control group (K) rats and 10 experimental group (C) rats were sacrificed. Arterial blood samples were collected and diluted with heparin to determine and compare the phagocytic activity (Phatogest) and oxidative metabolism (Bursttest) of peripheral blood granulocytes and monocytes by flow cytometry. Starting on the 8lh day of the experiment, the feed of the remaining 10 rats from the experimental group (C+G) and 10 rats from the control group (G) was supplemented with Biolex-Beta HP at a dose of 50 mg/kg body weight/day for 14 consecutive days. On day 22, arterial blood samples were collected from all (C+G) and (C) group rats, diluted with heparin to determine and compare the phagocytic activity and oxidative metabolism of peripheral blood granulocytes and monocytes by flow cytometry. The results showed that Biolex-Beta HP modulated the phagocytic activity and oxidative metabolism of blood neutrophils and monocytes suppressed by cyclophosphamide in rats. The immunorestoring activity of Biolex-Beta HP was observed in this study.

Aim: The aim of this study was to evaluate the association between the monocyte/HDL ratio (MHR) and idiopathic tinnitus. Study Design: Retrospective case-control study. Material-method: Eighty-nine patients with idiopathic tinnitus diagnosed on an outpatient basis between March 2015 and June 2016 and 87 healthy individuals who presented to our hospital for a routine health examination and had normal audiometry and otoscopy results were included in the study. Blood samples were obtained from patients during the first examination. The MHR was calculated as the ratio of the monocyte count to the HDL level. MHR ratios were compared statistically between the groups. Results: The monocyte count and the MHR were higher and HDL was lower in the study group compared to the control group (p< 0.05). The cut-off value of MHR for diagnosing tinnitus was 11. Conclusion: The MHR ratio was high in patients with idiopathic tinnitus, which supports the fact that atherosclerotic events and oxidative stress are implicated in the etiology of tinnitus.
