Opis:
Fusarium wilt is the most harmful soil-borne disease of lupins in Russia and in the Ukraine. The aim of the studies was to find broad resistance to disease in collection accessions and to breed lupin forms resistant to Fusarium wilt for the future breeding program. Lupin cultivars and lines (547 accessions) from the genebank of the N.I. Vavilov All Russian Scientific Research Institute of Plant Industry, St. Petersburg, Russia, were tested for Fusarium resistance under different environmental conditions of three regions in Russia and in the Ukraine on plots with artificially infested soil. So, differences in the disease susceptibility of the same accessions were found in contrasting environments. Resistant forms from one region were crossed with accessions showing their resistance in two other regions to accumulate resistance genes in new genotypes. As a result of hybridization two transgressive forms were obtained in F₄ of the crosses cv. Frost × cv. Apendrilon (L. angustifolius) and line G-413 × line 85 (L. luleus). Their resistance in all three regions appeared to be higher than that of their parental forms. These two lines were found suitable for the breeding programme of Fusarium resistant forms in Russia and in the Ukraine.

A characteristic trait of lupins is a production of alkaloids, which are a toxic and bitter taste compound ofseeds. Due to the lack of fast, sensitive and inexpensive screening techniques to identify and reject high alkaloidplant material, development of suitable tools is important challenges for lupins breeding and seed production.The aim of this study was to compare two alkaloid content estimation methods in Lupinus angustifoliusL. and Lupinus albus L.During the Wagner’s colorimetric test, which is recommended by the UPOV, seed halves were stained onfour colors depending on the alkaloid content but only the level of 0.5% – 0.6% showed clear color change.Gas chromatography allowed accurate quantification and qualification of alkaloid content.Since safe alkaloid content for consumption is 0.02% of seed dry weight, colorimetric method is less usefulfor dividing lupin cultivars into sweet and bitter, than gas chromatography but can be used as a screeningtechnique.

This article present a comparison of soluble sugar levels in seeds of Lupinus atlanticus, Lupinus cosentinii, Lupinus palaestinus and Lupinus pilosus, Lupinus hispanicus subsp. hispanicus and Lupinus luteus of Juno variety. Considering that sugars are accumulated in embryos, only the embryonic tissues were used for biochemical analyses. Additionally, the share of testa and embryo in seed tissues was evaluated. The seed-coat thickness was measured using scanning electron microscopy. The seed coat had the largest share in seeds of Lupinus pilosus and Lupinus palaestinus, and the least share in seeds of Lupinus hispanicus subsp. hispanicus and Lupinus luteus of Juno variety. In the seed of Lupinus pilosus the thickness of the seed coat was 1100 µm, while in Lupinus luteus it was 300 µm. The analysed Lupinus seeds accumulated from 73 mg/g d.m. (dry matter of seed embryo) to 155 mg of soluble sugars/g d.m. The highest quantity of oligosaccharides of the raffinose series was found in seeds of Lupinus luteus, while the lowest amount in seed of Lupinus palaestinus. Galactosyl cyclitols appeared in largest amount in seeds of Lupinus palaestinus and Lupinus pilosus, appropriately 41.93 and 33.75 mg/g dm. The lowest amount of galactosyl cyclitols appeared in Lupinus atlanticus, Lupinus cosentinii and Lupinus hispanicus.

In F₂, F₃ and F₄ generations of one hybrid line of Lupinus varius × L. digitatus, segregation into infertile and fertile plants in the ratio 3:1 was observed. Cytoembryological analyses showed that sterility was caused by irregularities in megasporocyte formation, in megasporogenesis and megagametogenesis. The following abnormalities lead to female sterility: no megasporogenic cells separated in nucelluses; in other nucelluses with megaspore mother cells, these cells underwent vacuolation and died before reduction division. In the megaspore mother cells, in which probably meiosis occurs, the megasporocyte division is irregular; a restitution nucleus is frequently formed after reduction division and such megasporocyte develops into an embryo sac. In 8-nucleate embryo sacs a change in the nucellus polarization was observed, while in rarely encountered embryo sacs the embryo cells underwent vacuolation and then died. Female sterility in the studied segregants is determined genetically, whereas sterilization of reproduction cells is a developing process throughout the period of sporocyte and female gametophyte formation.

The aim of this study was to compare the accumulation of soluble carbohydrates in embryos of two lupin species: cultivated Lupinus luteus (cv. Juno) and wild L. pilosus, developing on plants grown under normal soil humidity and soil drought. All analysed seeds accumulated soluble carbohydrates, including: monosaccharides, sucrose, cyclitols, galactosyl cyclitols and raffinose family oligosaccharides. Soil drought caused a nearly two-fold increase of soluble carbohydrate contents in both species. L. pilosus embryos however, responded to water deficiency by increasing the accumulation of cyclitols and galactosyl cyclitols, whereas L. luteus embryos enhanced accumulation of cyclitols and raffinose family oligosaccharides.

A great progress had been made in narrow-leafed lupin in a comparatively short period. Despite elimination of basic disadvantages (hard seeds, high content of alkaloids, pod shattering) disease-resistant, unbranched and thermoneutral cultivars have been bred. Investigations on the variation range and inheritance mode of commercially important characters were followed by breeding achievements. The paper contains a review of references for 35 genes controlling alkaloid content, growth rhythm, branching, vegetation length, shape and color of leaves, color of flowers and seeds, seed hardiness, pod characters and disease resistance. Authors of characters and gene symbols from different research centres and from different years are given on the basis of priority. The importance of these genes for breeding improvement is discussed. Type lines for characters/genes and genotype of more important cultivars collected in the Polish narrow-leafed lupin genebank are presented.

Opis:
Some trace elements, for example zinc, play both a positive and a negative role in plant life, which requires their content in soil. If soil is excessively contaminated with zinc, an attempt should be made to reduce the negative effect of this element on plants and other living organisms. For this reason, a study was undertaken to determine whether it was possible to alleviate the effect of soil zinc contamination (0, 150, 300 and 600 mg Zn kg-1 of soil) on the yield and macronutrient content of yellow lupine (Lupinus luteus L.). Compost (3%), bentonite (2%) and zeolite (2% relative to soil mass) were used to reduce the effect of soil zinc contamination. Macro- and micronutrients were applied to the soil in the same amounts in all pots: 30 mg N, 30 mg P, 100 mg K, 50 mg Mg, 0.33 mg B, 5 mg Mn and 5 mg Mo per kg soil. Yellow lupine was harvested in the flowering phase and plant material samples were collected for laboratory tests. The induced soil zinc contamination reduced yellow lupine growth and development because a dose of 300 mg Zn kg-1 soil caused plant seedlings to wither. Compost and bentonite reduced the negative influence of soil zinc contamination on yellow lupine yield, especially on aerial parts. The most demonstrable effect of zinc on the macronutrient content of lupine plants was recorded for magnesium and calcium, whose content increased compared to the control in both the aerial parts and roots of yellow lupine. Among the neutralizing substances, the effect of zeolite on the phosphorus, magnesium and calcium content and bentonite on the sodium content in the plants was the most beneficial.

The aim of the study in the field experiment was to determine the effect of different nitrogen fertilization and growth phases on the nickel and chromium content, accumulation and translocation in yellow lupine. The test factors included nitrogen doses (0, 30, and 120 kg ha-1) introduced into the soil prior to seed sowing, as well as growth stages (BBCH 65 and 90) of tested plant. Mineral nitrogen was introduced to the soil as ammonium sulphate (NH4)2SO4. Plants harvested at 65 BBCH were divided into roots, steams, leaves and flowers, whereas those harvested at 90 BBCH were divided into roots, steams, leaves, pods and seeds. 30 kg N ha-1 application did not significantly impact on the content and uptake of Cr and Ni by lupine. The content of these heavy metals in lupine decreased after fertilization with120 kg N ha-1. This N dose did not significantly affected the amounts of Cr and Ni taken up by tested plants. Cr bioaccumulation coefficient did not significantly depend on N fertilization used, while the increase of N doses decreased the value of this coefficient for Ni. Under the conditions of growing lupine on soil with a natural chromium and nickel content, no tendency to hyperaccumulate these heavy metals was found.

The effects of different types of arbuscular mycorrhizal (AM) fungi on the root inoculation and plant elemental content of lupin (Lupinus albus L.) were investigated in the present study. The growth and development of lupin were examined to determine the species of AM fungi that can help to grow lupin with a high protein content and economic value. In this study, which was carried out as a pot experiment under controlled greenhouse conditions, first the pots were inoculated with Glomus geosporum, Glomus mosseae, Glomus caledonium, Glomus etunicatium mycorrhizal spores and then lupin (Lupinus albus) seeds were sown. The plants were watered with pure water during the experiment. The trial was terminated after a 60-day plant gowing period. In the study, inoculation occurred at lupin roots at rates varying between 13.3 and 30.0%. However, there was no statistically significant difference among the types of arbuscular mycorrhizal fungi applied to the plant in the inoculation rate. The examination of the effect of the application of different AM (Glomus geosporum, Glomus mosseae, Glomus caledonium, Glomus etunicatium) spore on the plant development showed that AM inoculation did not have an effect on the lupin development. The effect of AM inoculation on the plant’s nutrient content revealed no significant difference in the content of crude protein, P and K, while demonstrating a significant increase in the sulphur and magnesium content versus the control. The plant content of crude protein varied between 185.6 and 226.5 (g kg-1), phosphorus – 0.61-0.74 (g kg-1) and potassium – 9.6-11.1 (g kg-1). The concentrations of Zn, Cu, B and Mo in lupin did not show statistically significant modifications caused by the inoculation of different types of AM. However, the plant Mn content showed a decrease due to AM inoculation, whereas a significant increase was observed in the Na content after AM inoculation. AM fungi were observed in plant roots after the inoculation with any of the four different types of mycorrhiza. But no positive effects of mycorrhizal inoculation were not observed on crude protein and the uptake of plant nutrients.
