Opis:
Embryo development of Lupinus albus L. and L. mutabilis Sweet. after pollination with L. angustifolius L. was studied. Observations of ovules revealed non-typical embryo development. From the 5th day after pollination, high number of degenerated ovules was found; the highest number was observed on 15 and 20 days after pollination. The period between the 10th and 15th day after pollination was the most suitable for isolation of embryos in all the examined combinations, with exception L. albus x L. angustifolius where optimal isolation time was between 15th and 20 th day after pollination. Embryos were obtained in each cross-pollination. Some of them developed in vitro into plantlets and grew in an artificial medium for some time, but most of them died due to non-typical development of the root system, stem, or both. The hybrid character of the survived plants requires confirmation.

Opis:
A characteristic trait of lupins is a production of alkaloids, which are a toxic and bitter taste compound ofseeds. Due to the lack of fast, sensitive and inexpensive screening techniques to identify and reject high alkaloidplant material, development of suitable tools is important challenges for lupins breeding and seed production.The aim of this study was to compare two alkaloid content estimation methods in Lupinus angustifoliusL. and Lupinus albus L.During the Wagner’s colorimetric test, which is recommended by the UPOV, seed halves were stained onfour colors depending on the alkaloid content but only the level of 0.5% – 0.6% showed clear color change.Gas chromatography allowed accurate quantification and qualification of alkaloid content.Since safe alkaloid content for consumption is 0.02% of seed dry weight, colorimetric method is less usefulfor dividing lupin cultivars into sweet and bitter, than gas chromatography but can be used as a screeningtechnique.

The pollen grain germination and the growth of pollen tubes after intra- and interspecific reciprocal pollination of Lupinus albus L., L. mutabilis Sweet, and L. angustifolius L. were examined. Results of microscopic observations of pollen grain germination and pollen tube growth enable to deny the existence of certain barriers that prevent fertilization after pollination of L. albus flowers with pollen of L. angustifolius and vice versa, as well as after pollination of L. mutabilis flowers with pollen of L. angustifolius and vice versa. Some delay in pollen grain germination and a slower growth of pollen tubes were observed in all combinations, as related to intraspecific ones, though in the case of each interspecific combination pollen tubes reached the ovary. Moreover, a different level of pollination efficiency (the number of growing pollen tubes) and various rates of pollen grain germination and pollen tube growth, depending on the direction of pollination, were found. The noted pollination efficiency may generally be considered to be low.

The aim of the studies was to establish in vitro conditions for the culture of pea and lupin embryos as the first step in the development of an in vitro assisted single seed descent technique for the attainment of homozygous populations. Materials for the study included of pea, and narrow-leafed and yellow lupin cultivars. Embryos dissected from mature but still-green seeds were cultured in vitro on two modified MS media and under three temperature regimes. Shoot and root lengths of regenerated plants were measured after 7, 14 and 21 days of culture. For pea plants full-strength MS medium with 4 g l−1 agar and temperature 22/ 20°C (day/night) appeared to be the most conducive to shoot and root development, whereas for lupin plants lower temperatures were more propitious: 12°C in the dark for narrow-leafed lupin and 16/ 12°C (day/night) for yellow lupin. Almost all the cultured embryos developed into plants, but not all the regenerated plants survived acclimation to ex vitro conditions.

Opis:
Fusarium wilt is the most harmful soil-borne disease of lupins in Russia and in the Ukraine. The aim of the studies was to find broad resistance to disease in collection accessions and to breed lupin forms resistant to Fusarium wilt for the future breeding program. Lupin cultivars and lines (547 accessions) from the genebank of the N.I. Vavilov All Russian Scientific Research Institute of Plant Industry, St. Petersburg, Russia, were tested for Fusarium resistance under different environmental conditions of three regions in Russia and in the Ukraine on plots with artificially infested soil. So, differences in the disease susceptibility of the same accessions were found in contrasting environments. Resistant forms from one region were crossed with accessions showing their resistance in two other regions to accumulate resistance genes in new genotypes. As a result of hybridization two transgressive forms were obtained in F₄ of the crosses cv. Frost × cv. Apendrilon (L. angustifolius) and line G-413 × line 85 (L. luleus). Their resistance in all three regions appeared to be higher than that of their parental forms. These two lines were found suitable for the breeding programme of Fusarium resistant forms in Russia and in the Ukraine.

In view of the predicted increase in sulphur deficiency in crop production, a study was undertaken to assess the effect of fertilization with this nutrient on criteria determining the biological value of narrow-leafed lupin seeds (total and true protein, their ratio, nitrates and amino acid composition). The study was based on a three-year, three-factor field experiment, in which the Elf cultivar of narrow-leafed lupin (Lupine Angustifolius L.) was grown on light soil with low sulphur content. The experiment included foliar and soil application of sulphur and its elemental and ionic forms. The application doses were 0, 20, 40 and 60 kg S ha-1. In all years of the study, the sulphur application dose was found to significantly determine the total and true protein content as well as their ratios in the lupin seeds. Application of just 20 kg S ha-1 caused a significant increase in true protein in the lupin seeds and in the proportion of true protein in the total protein, as compared to the control. The CS index for all the essential amino acids showed that the first limiting amino acid in the lupin seed protein was sulphur-containing methionine. As the dose of applied sulphur increased, there was an overall increase in CSmeth. and EAAI. Given the positive effect of sulphur, particularly in the elemental form, on the protein content and amino acid composition of lupin seeds and in reducing the content of harmful nitrates, the use of this nutrient in fertilization of this species is recommended.

In F₂, F₃ and F₄ generations of one hybrid line of Lupinus varius × L. digitatus, segregation into infertile and fertile plants in the ratio 3:1 was observed. Cytoembryological analyses showed that sterility was caused by irregularities in megasporocyte formation, in megasporogenesis and megagametogenesis. The following abnormalities lead to female sterility: no megasporogenic cells separated in nucelluses; in other nucelluses with megaspore mother cells, these cells underwent vacuolation and died before reduction division. In the megaspore mother cells, in which probably meiosis occurs, the megasporocyte division is irregular; a restitution nucleus is frequently formed after reduction division and such megasporocyte develops into an embryo sac. In 8-nucleate embryo sacs a change in the nucellus polarization was observed, while in rarely encountered embryo sacs the embryo cells underwent vacuolation and then died. Female sterility in the studied segregants is determined genetically, whereas sterilization of reproduction cells is a developing process throughout the period of sporocyte and female gametophyte formation.
