Modern language-lab management software provides an increasingly sophisticated range of functions that appear to have been designed to change or supposedly improve certain aspects of traditional classroom language teaching. However, using management software in a way that regularly puts a computer between the teacher and student may create an unnatural or unnecessary ‘electronic go-between’ in the dynamics of traditional teacher-student interaction. Human communication contains many rich visual cues that have always been an important part of the learning experience, and it is this age-old ‘human to human’ learning experience that should remain vitally important in the language lab.

The paper may be of interest to new and/or existing LAMS users who would like to read more about current issues in LAMS learning design literature and may be of use to the more adventurous computer-savvy LAMS user who aspires to exploit further the potential of certain LAMS tools. The version of LAMS described in this paper is Version 2.1.1 . It is hoped that study of this extended paper and its preceding introductory paper (Alexander 2008a, An overview of LAMS) will be of particular value to those wishing to gain more confidence in the proficient utilisation of LAMS and to those intending to engage in debate on current LAMS issues. The paper’s thematically diverse core sections may also heighten reader awareness of a pressing issue: the need to bring together the treasures of progressively more hard-core and swiftly changing computer technologies with the beliefs of an increasingly discerning education community.

W artykule przedstawiono przegląd analizatorów DNA wykorzystujących technikę real-time PCR i lab-chipy. Zaprezentowano opis opracowanej metodologii i instrumentarium do detekcji fluorescencji z wykorzystaniem miniaturowych komponentów optoelektronicznych i "inteligentnego" oprogramowania analizującego. Przedstawiono dwa przykłady miniaturowych analizatorów real-time PCR/DNA opracowanych w ramach projektów europejskich i krajowych wykorzystujących lab-chipy i nowatorską metodę detekcji fluorymetrycznej.
The paper presents a novel miniaturized optical instrumentation for fluorescence excitation and detection for miniaturized real-time PCR analyzers using lab-on-a-chip (LOC) devices. Application of miniaturized semiconductor laser to fluorescence excitation, CCD-minicamera as fluorescence photodetector and specialized software for optical signal conditioning led to development of low-cost and highly sensitive optical instrumentation. To carry out full characterization of the optical instrumentation, miniaturized thermocycler co-working with LOC was built. The optical instrumentation was tested with three LOC made of different materials and technologies, giving proper detection of fluorescence signals during real-time PCR. Finally, two miniaturized devices for real-time PCR detection and identification of DNA and utilizing described here novel optical instrumentation were briefly described. The scheme and technical realization of the novel instrumentation in laboratory version is shown in Fig. 4. The detection limit of DNA was about 0,1 ng/ml (Fig. 5). It was also found that the optical instrumentation co-works with different constructions of lab-on-a-chips for DNA real-time PCR amplification (Fig. 6). The developed optical instrumentation was successfully applied to two miniaturized devices for DNA detection and identification by real-time PCR. Technical realizations of the devices and views of the lab-on-a-chips are shown in Figs. 7 and 9. In both cases, proper detection of fluorescence signals generated during real-time PCR of Campylobacter j. DNA (Fig. 8) or complementary DNA (Fig. 10) were observed. It confirmed high sensitivity of the developed optical instrumentation.

One of the main scopes of modern cell engineering is development of cellular models that can replace animals in drug screening and toxicological tests, so called alternative methods. Construction of the alternative model is a very challenging task due to a richness of factors creating the in vivo environment. The monolayer cell culture — cultivation of adhesive cells on artificial surfaces such as glass or polymer — lack most of the in vivo-like interactions, but still is the only tool for the majority of applications. One of the most prospective approaches on mimicking in vivo environment is “Lab-on-a-chip” technology. Microfluidic devices offer lots of advantages over traditional in vitro culture, e.g. much higher cell volume-to-extracellular fluid volume ratio or possibility of regulation of hydrodynamic stress. This presentation aims to introduce latest advances of our team in microfluidic cell culture devices. Our novel approach is to cultivate three dimensional multicellular aggregates (spheroids) in microenvironments arranged in a microfluidic system. The geometry and materials of the system allow for cultivation, observation and analysis of multicellular spheroids. The results presented concern multicellular tumor spheroids (MCTS) rising from human cancer cells, which are considered to represent most of the conditionings of cancer tumor in vivo. The fully developed MCTS microdevice will be a reliable tool for anticancer drug screening, as the results most likely will be in a close accordance with the results obtained in vivo.

Lactic Acid Bacteria (LAB) belong to the normal fl ora of human alimentary tract and vaginal epithelium. The main products of LAB metabolism are short chain fatty acids (SCFA), which are known to have signifi cant infl uence on human health. The aim of this study was to examine the suitability of direct pyrolytic methylation in GC/MS profi ling of SCFA in the selected pharmaceutical preparations of LAB. For method optimization, standard SCFA samples consisting of acetic, propionic, butyric and lactic acids were pyrolyzed under various temperature conditions in the presence of methanolic solution of tetramethylammonium hydroxide (TMAH) as a derivatizing reagent. It was demonstrated that pyrolytic derivatization of standard SCFA to methyl esters was the most effi cient with the use 10% TMAH and the pyrolytic fi laments with Curie temperature of 480°C, when the pyrolysis cell was kept at 150°C. It was shown that the method is suitable for GC/MS qualitative analysis of SCFA profi le in the pharmaceutical preparations that contain LAB in lyophilized form.
Bakterie kwasu mlekowego (Lactic Acid Bacteria – LAB) należą do naturalnej flory bakteryjnej zasiedlającej przewód pokarmowy oraz nabłonek pochwy człowieka. Głównym produktem metabolizmu LAB są krótkołańcuchowe kwasy tłuszczowe (short-chain fatty acids – SCFA) wywierające istotny wpływ na zdrowie człowieka. W pracy poddano ocenie przydatność techniki bezpośredniej metylacji pirolitycznej w oznaczaniu profi lu SCFA w wybranych preparatach farmaceutycznych LAB techniką GC/MS. W celu optymalizacji metody, wzorcowe próbki SCFA zawierające kwas octowy, propionowy, masłowy i mlekowy poddano pirolizie w obecności metanolowego roztworu wodorotlenku tetrametyloamoniowego (TMAH) w różnych warunkach temperaturowych. Stwierdzono, że derywatyzacja analizowanych kwasów tłuszczowych do estrów metylowych zachodziła z największą wydajnością pod wpływem 10% TMAH, gdy zastosowano druty pirolityczne o temperaturze Curie 480°C, a komorę pirolizera utrzymywano w temperaturze 150°C. Wykazano, że zoptymalizowana metoda jest przydatna w prowadzonej techniką GC/MS analizie jakościowej profi lu SCFA w wybranych preparatach farmaceutycznych zawierających LAB w formie liofi lizatu.

W artykule porównano dostępność zakładu przemysłowego produkującego rury w przypadkach występowania jednoczesnych i niezależnych uszkodzeń podsystemów. Badania prowadzono przy stałych intensywnościach uszkodzeń podsystemów i zmiennych intensywnościach napraw . Konstytutywne równania różniczkowe systemu rozwiązano przy użyciu metody Lagrange'a. Oceny wydajności systemu dokonano na podstawie długotrwałej dostępności z wykorzystaniem pakietu oprogramowania Matlab 7.0.4. Przedstawiono tabele dla różnych parametrów, które mogą być wykorzystywane przez osoby zarządzające produkcją przy poprawianiu i planowaniu harmonogramów przeglądów.
The paper discusses the comparison between availability of a pipe manufacturing industry when the sub systems are subjected to simultaneously and independent failure. The failure rates of the sub-systems are constant and the repair rates are variable. The governing differential equations of the system are solved using Lagrange's Method. The performance evaluation of system is done by means of long run availability making use of software package Matlab 7.0.4. The tables for various parameters are given which can be useful to the plant management for improving and planning the maintenance schedule.

Opis:
The fluorometric detection in lab-on-a-chip devices for life-sciences is one of the main detection methods. Well know from “large-scale” analytical chemistry methodologies and instrumentation has been applied in lab-on-a-chip solutions. In most cases, optical instrumentation for fluorescence induction and detection is based on configuration and components borrowed from large laboratory instruments. As a result, optical instrumentation surrounding lab-on-a-chip is bulky, expensive and dedicated for operation only inside laboratory. In this paper a discussion on fluorescence detection in lab-on-a-chip is carried out. A novel image sensor – based detection instrumentation co-working with a “clever” software is described. Instrumentation is dedicated for operation in portable and low-cost devices for different life-science applications. Examples of applications of the novel method and instrumentation co-working with various lab-on-a-chips are presented on the base of author’s works.

Microfluidic devices, such as lab-on-a-chip systems, are highly advantageous for cell engineering and cell based assays. It is a particularly useful approach for development of the in vitro cellular systems mimicking the in vivo environment. In this paper, a novel lab-on-a-chip device for three-dimensional human cell culture and anticancer drug testing is presented. Cells were cultured as Multicellular Tumor Spheroids (MCTS) — the best cancer tumor model developed so far. Diff erent designs were tested and novel technique of microfabrication in poly(dimethylsiloxane) was developed. MCTS were cultured in a system of polymeric microwells, with the network of microfluidic channels for culture medium flow. Design included optimal shear stress and proper nutrients supply for cultured cells. Final design provided MCTS culture for four weeks with the homeostasis-like state achievement, which is characteristic for the in vivo situation.

This paper presents the new-generation test bench SETHI, developed by ONERA, the French Aerospace Lab. SETHI is a medium range platform dedicated to environmental, scientific and security applications. The first part of this paper describes the system architecture, the development state and the future capabilities. A set of recent significant results are presented: these results cover various applications, such as high spatial resolution imaging, change detection between two acquisitions, biomass measurement in the rain forest, bistatic imaging and innovative measurements, such as air-to-air imaging or circular imaging.

Artykuł prezentuje projekt aplikacji zarządzania pracownią radiologiczną, która organizuje prace wykonywane przez systemy zewnętrzne pracowni. Wymogiem jej działania jest poprawna komunikacja z pozostałymi systemami wykorzystywanymi w pracowni, oraz szpitalnym systemem informacyjnym. Aplikacja ma na celu usprawnienie działania placówki obsługującej dużą liczbę pacjentów, która wymaga szybkiej obsługi danych z pracowni.
The paper presents a radiology lab management application, which organizes activities performed by peripheral systems of a laboratory. The prerequisite for its operation is its correct communication with other systems used in the laboratory as well as the hospital IT system. The purpose of this application is to improve the institution activity handling a large number of patients and requiring quick data handling within the laboratory. The work on development of the structure of the reports involved a large emphasis being placed on the adherence to the IHE recommendations. The model put forward assumes a multiple sending process for the very same information in order to maintain synchronization. Those decisions were important from the point of view of programming the report handling procedures. The exact project of data exchange improves the process of application programming. The use of XML format for coding the information allows the data exchange to be traced with use of a sniffer. The communication mechanics adopted by us allows for efficient exchange of information. There are more than a dozen commercially available HL7 protocol interfaces. The lack of availability of a specification and the ambiguous mechanics of data organization, used within Polish hospital applications has led us to use our own protocol. The working solution utilizes only a few communicates based on HL7. The work puts a higher emphasis on the informational content of those communicates. Our own protocol is also used to code DICOM communicate equivalents, which convey text messages or serve as event reminders.

During the past few years, the process of miniaturisation in the field of biochemical laboratory equipment led to the introduction of the so-called "lab-on-a-chip" microdevices, which combine separate functional units into a complex, multifunctional apparatus. Fluid dynamics plays an essential role in the development of such equipment, since frequently the major part of chemical analysis is based on soluble analytes. In this work, we consider a device for the analysis of cell growth under different conditions. In this device, dozens of cell spots absorb the nutrient (analyte) from the liquid medium. The concentration of the analyte must be strictly controlled to maintain a specific microenvironment. A two-dimensional model for the flow field and the distribution of concentration of the analyte is developed taking into account the geometrical shape of the spot with a simplified absorption model. The dependence of the results on the controlling parameters is investigated in order to determine the influence of the presence of the cell spot on the distribution of the analyte.

Lysosomal storage disorders (LSDs) represent a group of more than 45 genetically inherited diseases caused by the absence or deficiency of one or more specific lysosomal enzymes. Nowadays, there is a lack of reports on fast, reliable methods for the diagnostics of LSDs. Currently applied diagnostic approaches generate many false-negative and false-positive results, which results in classification of patients to inappropriate therapeutic groups. Moreover, these methods are time-consuming (even 20 hours), and are carried out only in a few laboratories in the world. The goal of this work was to develop a method and a tool, a Point-of-Care system, for diagnostics of LSDs. The polymeric microdevice consists of a cell lysis module, a mixing microchannel and an optical detection module. The system enables to determine the activity of α-galactosidase (deficient in Fabry disease), and to reduce the time of analysis to 10 min. Due to its easy fabrication steps and low price, it seems to be a prospective tool for a point-of-care approach.

Microfluidic devices called lab-on-a-chips utilize two kinds of characterization of a biosample, which are based on spectrofluorimetric and spectrophotometric methodologies. Lab-on-a-chips are equipped with an optical instrumentation and a software system that allow detecting of the optical signals and their processing into spectral characteristics. In the paper, technology of lab-on-a-chip (cytometer microdevice) and miniature silicon-glass field-emission light source (FELS) is described. Electrons emitted from a silicon cathode covered with carbon nanotubes excite light emission from nanocrystalline rare earth doped yttrium oxide phosphor (anode). The emitted light can be precisely tailored to the main absorption line of a fluorescent marker. The presented system has been successfully used for testing of the cow embryos as well as fluorescent-marked porcine oocytes.

Opis:
Artykuł prezentuje opis modelu przetwarzania badań, jakie wykonywane są w pracowniach radiologicznych. Wyniki wykonanych badań w postaci opisów tekstowych - standard HL7 oraz obrazów - standard DICOM powiązane są następnie ze strukturą szpitalnego systemu informacyjnego. Proponowana aplikacja zarządzająca pracowniami radiologicznymi może zostać zaimplementowana w placówce o dowolnej wielkości z wdrożonym systemem informacyjnym oraz z dowolną liczbą aktywnych pracowni obrazowania medycznego. Opracowana aplikacja może być testowana poza obrębem sieci szpitalnej.
Applications managing a radiology laboratory organize works per-formed by the laboratory peripheral systems. Proper communication with other systems used in the laboratory and a hospital IT system is required for realizing this task. The paper presents description of a model of processing the tests performed in radiology laboratories. The test results in the form of text descriptions - HL7 standard and images - DICOM standard are subsequently affiliated with the hospital system structure. The proposed application which manages radiology laboratories may be implemented in the facility of any size with the IT system implemented and with any number of active medical image laboratories. A simulator is also proposed to enable testing applications outside the hospital network. A communication model assumes the usage of HL7 and DICOM standards. Due to different versions of these protocols and large freedom during creating messages, the application uses the protocol which sends the messages in the XML format. The messages are patterned on the HL7 and DICOM equivalents, sending the same information, however encoded in the manner which is unambiguously read out by all elements of the application (Fig. 2). The creation of software which may be implemented in the productive environment requires application of the developed testing process. In the case of medical facilities the process is even longer. The next step is to prepare communication interfaces for HL7 and DICOM protocols. It will allow conducting tests with participation of the software which stimulates events signaled by means of these protocols and will be the next stage for testing the application in a real radiology laboratory.
