Opis:
Interleukin-1 receptor antagonist (IL-1Ra) and vaccinia virus protein C10L share a VTXFYF motif, with X being Lys or Arg residue, respectively. Peptides of such sequence compete successfully with IL-1 for the cellular receptor. A pair of complementary peptides, based on the Siemion's hypothesis on the periodicity of the genetic code (QWLNIN and QWANIN), and another pair, in which, following the Root- Bernstein theory, Lys was used as complementary amino acid to Phe (QWLKIK and QWAKIK), were investigated for the peptide-antipeptide interactions using mass spectrometry (ESI-MS) and circular dichroism (CD) methods. The CD measurements indicated some conformational changes, more pronounced in the Siemion's pairs, however, no heterodimer formation was found by MS. In the region of IL-1 receptor situated close to the position of IL-1Ra in the IL-1Ra-receptor complex, a KQKL motif is present, suggesting a possibility of complementary recognition of the Root-Bernstein type in the IL-1 receptor. The biological activity of the complementary peptides is similar to that of the original ones. They efficiently compete with IL-1 and show moderate immunosuppressory activity in humoral and cellular immune response. The inhibition of the IL-1-IL-1 receptor interaction may result from the complementary peptides acting as mini-receptors with affinity for IL-1.

Opis:
Pro-inflammatory cytokines participate in the induction of ischemic stroke. So far, their participation in the cerebral ischemia was proven for the tumor necrosis factor TNF-α, interleukin-1 (IL-1), and interleukin-6 (IL-6). The release of the pro-inflammatory cytokines into the extracellular space causes the enlargement of the brain damage region, and consequently increases the neurological deficit and negatively affects the survival rate prognoses. That is confirmed by the increased concentration of pro-inflammatory cytokines in blood and the cerebrospinal fluid of patients with brain stroke, as well as by the research on the induced/experimental cerebral ischemia in animals. The pro-inflammatory cytokines participate in the migration of the reactive T lymphocytes to the regions of brain ischemia where they enhance the nerve tissue damage by down-regulation of microcirculation, induce the pro-thrombotic processes and release other neurotoxic cytokines. Also, in the early stage of cerebral ischemia, cytokines activate the axis hypothalamus-pituitary gland-adrenal cortex and increase the cortisol concentration in blood, what results in the decreased resistance to infectious diseases. Administration of the inhibitor of the interleukin-1 receptor (IL-1Ra) inhibits the inflammatory processes in the region of brain ischemia, and subsequently improves the prognosis for the size of the neurological deficit and the survival rate, as well as resistance to infectious diseases.

Opis:
Several relations between cytokines and pathogenesis of diabetes are reviewed. In type 1 and type 2 diabetes an increased synthesis is observed and as well as the release of pro-inflammatory cytokines, which cause the damage of pancreatic islet cells and, in type 2 diabetes, the development of the insulin resistance. That process results in the disturbed balance between pro-inflammatory and protective cytokines. Pro-inflammatory cytokines such as interleukin 1β (IL-1β), tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ), as well as recently discovered pancreatic derived factor PANDER are involved in the apoptosis of pancreatic β-cells. Inside β-cells, cytokines activate different metabolic pathways leading to the cell death. IL-1β activates the mitogen-activated protein kinases (MAPK), affects the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) and activates the inducible nitric oxide synthase (iNOS). TNF-α and IFN-γ in a synergic way activate calcium channels, what leads to the mitochondrial dysfunction and activation of caspases. Neutralization of pro-inflammatory cytokines, especially interleukin 1β with the IL-1 receptor antagonist (IL-1Ra) and/or IL-1β antibodies might cause the extinction of the inflammatory process of pancreatic islets, and consequently normalize concentration of glucose in blood and decrease the insulin resistance. In type 1 diabetes interleukin-6 participates in regulation of balance between Th17 and regulatory T cells. In type 2 diabetes and obesity, the long-duration increase of IL-6 concentration in blood above 5 pg/ml leads to the chronic and permanent increase in expression of SOCS3, contributing to the increase in the insulin resistance in cells of the skeletal muscles, liver and adipose tissue.

Opis:
Deltamethrin is a type II pyrethroid. Deltamethrin’s action is characterised by nephrotoxicity, hepatotoxicity and immunotoxicity. The aim of the study was to evaluate the effect of poisoning with deltamethrin on the levels of interleukin1ß and TNFα in the livers and kidneys of mice. A total of 24 female mice were divided into 3 groups of 8: - controls, - receiving deltamethrin i.p. at the dose of 41.5 mg/kg for 28 days - receiving deltamethrin i.p. at the dose of 8.3 mg/kg for 28 days. On day 29 the animals were euthanised, livers and kidneys were obtained, homogenised and centrifuged. The supernatant was used for measuring IL-1ß and TNFα concentration with ELISA tests. The results were analysed with Statsoft Statistica. The interleukin 1ß concentrations were significantly higher in the kidneys (18.30±16.85) of mice exposed to the higher dose of deltamethrin than in the controls (8.15±4.66) (p<0.05). In the livers of mice receiving 41.5mg/kg deltamethrin it was 203±71.63 vs 46.77±34.79 (p<0.05). In the livers of animals receiving the lower dose it was higher than in the control group (96.51±24.73) (p<0.05). The TNF α was elevated in the kidneys of mice exposed to the higher dose of deltamethrin (6.56±3.26 vs 2.89±1.57)(p<0.05). Conclusion: Deltamethrin produces a significant increase of interleukin 1ß in the livers and kidneys of mice and so the cytokine seems to be a good marker of hepatotoxicity and nephrotoxicity in the course of subacute poisoning.
Deltametryna jest pyretroidem typu II. Działanie deltametryny charakteryzuje się nefrotoksycznością, hepatotoksycznością i immunotoksycznością. Celem pracy była ocena wpływu zatrucia deltametryną na poziom interleukiny1ß i TNFα w wątrobie i nerkach myszy. W badaniach 24 samice myszy podzielono na 3 grupy po 8: - kontrole, - otrzymywanie deltametryny i.p. w dawce 41,5 mg/kg przez 28 dni - deltametryna i.p. w dawce 8,3 mg/kg przez 28 dni. W 29 dniu zwierzęta zostały uśmiercone, pobrano wątroby i nerki, które następnie homogenizowano i odwirowano. Supernatant wykorzystano do pomiaru stężenia IL-1ß i TNFα za pomocą testów ELISA. Wyniki analizowano za pomocą Statsoft Statistica. Stężenia interleukiny 1ß były znacząco wyższe w nerkach (18,30±16,85) myszy narażonych na wyższą dawkę deltametryny niż u kontroli (8,15±4,66) (p<0,05). W wątrobach myszy otrzymujących 41,5 mg/kg deltamtehryny było to 203±71,63 vs 46,77±34,79 (p<0,05). W wątrobach zwierząt otrzymujących niższą dawkę była ona wyższa niż w grupie kontrolnej (96,51±24,73) (p<0,05). TNFα był podwyższony w nerkach myszy eksponowanych na wyższą dawkę deltametryny (6,56±3,26 vs 2,89±1,57) (p<0,05). Wniosek: Deltametryna powoduje znaczny wzrost interleukiny 1ß w wątrobie i nerkach myszy, dlatego cytokina wydaje się być dobrym markerem hepatotoksyczności i nefrotoksyczności w przebiegu zatrucia podostrego.

Opis:
Objectives: To investigate cardiorespiratory and inflammatory responses in male workers following exposure to welding fumes and airborne particles in actual workplace conditions. Materials and Methods: We measured blood leukocytes and their differential counts, platelet count, hemoglobin, sensitive C-reactive protein, fibrinogen, E-selectin, IL-(interleukin)1β, IL-6, IL-8, tumor necrosis factor alpha (TNF-α) and endothelin-1 in blood samples of twenty workers before and after their working day. We also studied peak expiratory flow (PEF), forced expiratory volume in one second (FEV₁), and exhaled nitric oxide (NO). We assessed heart rate variability (HRV) by obtaining 24-hour ambulatory electrocardiograms. Results: The total blood leukocytes and neutrophils increased after the work shift, whereas IL-1β and E-selectin decreased significantly. There were no tatistically significant changes in exhaled NO, FEV₁, PEF or HRV. Conclusion: Occupational exposure to welding fumes and particles caused a slight, acute inflammatory effect estimated based on the increased values of leukocytes and neutrophils in blood and a decrease in the interleukin 1β and E-selectin values, but no changes in the pulmonary function (exhaled NO, FEV₁, PEF) or HRV during the working day were observed.

Opis:
IL-1 family of cytokines, is a group of several proteins which are involved in regulation of immune response. It has been shown that members of that family , especially: IL-1α, IL -1β, IL-18 and IL-33 act as mediators in nociceptive transmission and play a crucial role in pathogenesis of multiple diseases. The discovery of IL-1Ra, naturally occurring antagonist of I L-1 receptor (IL-1RI i IL-1RII) and IL-18 binding protein has led to many studies involving use of recombinant receptor antagonist in pain therapy or in treatment of such a diseases as: rheumatoid arthritis, multiple sclerosis, skin diseases or diabetes.
Rodzina Interleukiny-1 to rodzina kilkunastu, należących do cytokin, cząsteczek białkowych zaangażowanych głównie w regulację odpowiedzi odpornościowej. Udokumentowano, że członkowie tej rodziny, w szczególności: IL-1α i -1β, IL-18 oraz IL-33 pełnią istotne funkcje w mediowaniu transmisjii nocyceptywnej oraz w patogenezie wielu chorób. Odkrycie naturalnie występującego antagonisty (IL-1 Ra) receptorów z rodziny IL-1 (IL-1RI i IL-1RII) oraz białka wiążącego IL-18, doprowadziło do rozpoczęcia szeregu badań nad zastosowaniem zrekombinowanych antagonistów w leczeniu bólu czy zapobieganiu postępu choroby w takich schorzeniach jak, m.in.: reumatoidalne zapalenie stawów, stwardnienie rozsiane, choroby skóry czy cukrzyca.

Opis:
Gram-positive and nonpathogenic lactic acid bacteria (LAB) are considered to be promising candidates for the development of new, safe systems of heterologous protein expression. Recombinant LAB has been shown to induce specific local and systemic immune response against selected pathogens, and could be a good alternative to classical attenuated carriers. The main goal of our study was to express the avian influenza haemagglutinin (H5) and chicken interleukin 2 (chIL-2) in Lactococcus lactis. Results of this study were anticipated to lead to construction of lactococcal strain(s) with potential vaccine properties against the avian influenza A (H5N1) virus. Expression of the cloned H5 gene, its His-tagged variant and chIL-2 gene, under the control of the ptcB gene promoter was attested by RT-PCR on transcriptional level and Western or dot blot analysis on translational level, demonstrating that system can be an attractive solution for production of heterologous proteins. The results of the preliminary animal trial conducted in mice are a promising step toward development of a vaccine against avian bird flu using Lactococcus lactis cells as antigen carriers.

Opis:
WC1+ cells in cattle exhibit both regulatory and effector activities. However, it has not been elucidated whether they are so plastic that both activities co-exist in one cell or there are separate subpopulations of effector and regulatory cells. Since the production of IFN-γ and IL-10 seems to be related to WC1+ cells’ effector and regulatory function, respectively, the main aim of this study was to determine whether those cytokines are produced by separate subpopulations of WC1+, or are co-produced by the same cells. Due to increasingly frequent emphasised role of consumption of IL-2 in the mechanism of suppressor action of mouse CD25+CD4+ T regulatory cells, expression of the receptor’s α chain for interleukin 2 (CD25) on WC1+ lymphocytes has been evaluated. An average of 5.21% of WC1+ cells obtained from PBMCs of 12-month-old heifers show constitutive expression of the CD25 molecule, with CD25highWC1+ and CD25lowWC1+ cells accounting for 1.05% and 4.10% of WC1+ lymphocytes, respectively. For detection of intracellular cytokine production, PBMCs were stimulated with concanavalin A. Both IFN-γ- and IL-10-producing cells within the CD25-WC1+ and CD25+WC1+ subpopulations were mainly separate subpopulations. The average percentage of IFN-γ+IL-10-, IFN-γ-IL-10+ and IFN-γ+IL-10+ cells among CD25-WC1+ lymphocytes was 4.03%, 2.67% and 0.51%, respectively. A positive correlation was observed between the presence of the CD25 molecule on WC1+ lymphocytes and production of IL-10 and TGF-β, because the average percentage of IFN-γ-IL-10+ and IFN-γ+IL-10+ among CD25+WC1+ lymphocytes was 3 and 4.5 times higher as compared to the corresponding cells in the CD25-WC1+ subpopulation, whereas the percentage of IFN-γ+IL-10- cells in both the subpopulations was not significantly different. The percentage of TGF-β+ cells within the CD25+WC1+ subpopulation was 2.72 times as high as that of CD25-WC1+ lymphocytes. Therefore, with respect to the production of IFN-γ, IL-10 and TGF-β, CD25+WC1+ lymphocytes turn out to have a more suppressor profile than CD25-WC1+.
