Opis:
Cisplatin is an antineoplastic drug, used for the treatment of ovarian cancer. SMEDDS has many advantages such as enhanced bioavailability, lymphatic targeting and ease of manufacture. The main objective of this study was to prepare and characterize Cisplatin loaded SMEDDS formulation and to evaluate antitumoral activity with cell viability studies. Cisplatin SMEDDS formulation was prepared and characterized physicochemically. In vitro release studies and cell viability studies were performed and evaluated. The mean droplet size of Cisplatin SMEDDS was measured as 25,4±1,9 nm and PDI was 0,241±0,018. Refractive index of the formulation was measured as 1,471±0,001. pH values of Cisplatin SMEDDS (dilution ratio 1:10 w) were measured as 5,84±0,09 and (dilution ratio 1:10 pH 6,8 PBS) 6,51±0,14. Viscosity of formulation was measured as 284 mPa. According to in vitro release studies, %78,17 of Cisplatin were released from Cisplatin SMEDDS. The formulation performed cytotoxic effect to A2780 cells; vitality was found 20,26% at 0,02 µg/mL. It was concluded that, Cisplatin SMEDDS could be beneficial for the treatment of ovarian cancer and it could be promising alternative due to its enhanced bioavailability.

The antifungal activities of ethanolic extracts of three Saudi plants; camel thorn (Alhagi maurorum Medic.), caper (Capparisspinosa L.), and pomegranate (Punica granatum L.) were investigated in vitro against Alternaria alternata, Fusarium oxysporum, Phomadestructiva, Rhizoctonia solani, and Sclerotium rolfsii at concentrations of 0, 3, 6, and 9% (v/v). All tested plant extracts; seeds, roots, and rinds had different degrees of antifungal activity against the tested fungi. When compared with the control, the highest antifungal activity was recorded for camel thorn seeds extract at a concentration of 9%, while, pomegranate rinds extract at 9% came in second. Camel thorn rinds extract came in last even when used at a high concentration. The ethanolic extract of camel thorn seeds may be recommended as a potent bio-fungicide. Extensive studies should be undertaken for the ethanolic extract of camel thorn seeds as a strong antifungal agent against fungal plant diseases.

Poly(ADP-ribose) polymerase (PARP-1) is an abundant nuclear protein with a high affinity for single- and double-strand DNA breaks. Its binding to strand breaks promotes catalysis of the covalent modification of nuclear proteins with poly(ADP-ribose) synthesised from NAD+. PARP-1-knockout cells are extremely sensitive to alkylating agents, suggesting the involvement of PARP-1 in base excision repair; however, its role remains unclear. We investigated the dependence of base excision repair pathways on PARP-1 and NAD+ using whole cell extracts derived from normal and PARP-1 deficient mouse cells and DNA substrates containing abasic sites. In normal extracts the rate of repair was highly dependent on NAD+. We found that in the absence of NAD+ repair was slowed down 4-6-fold after incision of the abasic site. We also established that in extracts from PARP-1 deficient mouse cells, repair of both regular and reduced abasic sites was increased with respect to normal extracts and was NAD+-independent, suggesting that in both short- and long-patch BER PARP-1 slows down, rather than stimulates, the repair reaction. Our data support the proposal that PARP-1 does not play a major role in catalysis of DNA damage processing via either base excision repair pathway.

The Far East medicinal plant - Plantago camtschatica was propagated in vitro from tips of shoots (obtained in vitro) and from different explants of 4-week-old seedlings: seedling tips, hypocotyls, cotyledons, roots, first leaves. To our knowledge there is no information in literature about in vitro culture of this plantain. MS basal medium, supplemented with 0.6 pM IAA in combination with various cytokinins (BA, KIN, ZEA), was used. After 6 weeks of culture, micropropagation rate (MR) - mean number of buds and shoots per explant - was calculated. Our study proved that P. camtschatica species was amenable to propagation in vitro from different kinds of explants. However, multiplication by adventitious shoot regeneration from hypocotyl explants was found to be the most suitable method for the propagation of this plant. Adventitious shoots could root without stimulation what allows to omit the stage of rooting. The plants obtained as a result of micropropagation were not phenotypically changed.

A three-dimensional numerical model of the human spine including three vertebrae of the lumbar spine (L1, L2, L3) has been built using the ANSYS 5.2 FEM program. Mechanical properties of different elements of the spine have been taken into account, i.e., cortical bone, cancellous bone, posterior elements, etc., in the model. The model allows one to analyse spine displacement under various static and dynamical loads acting on the vertebral structure, as well as it enables one to select proper implants. For verification purposes, the numerical analysis results having the form of displacements under different loads have been compared with the in-vitro experiment results obtained by other authors. In addition, a test stand allowing for experimental investigation of displacements of different elements of the spine has been constructed. In the investigation into stiffness, the artificial vertebrae L1, L2, L3 have been used. A good agreement between tje results obtained from the numerical analysis and experiments has been achieved.
Wykorzystując komercyjny program ANSYS 5.2 oparty na MES, stworzono trójwymiarowy model numeryczny kręgosłupa ludzkiego składający się z trzech kręgów lędźwiowych (L1, L2, L3). W modelu uwzględniono własności mechaniczne poszczególnych elementów kręgosłupa (kość korowa, gąbczasta, wyrostki, pierścienie włókniste, jądro). Model ten pozwala na analizowanie przemieszczeń zachodzących w układzie kręgosłupa pod wpływem różnych obciążeń statycznych i dynamicznych oraz umożliwia badanie i dobieranie różnego rodzaju implantów. W celu zweryfikowania modelu numerycznego porównano otrzymane wyniki w postaci przemieszczeń, uzyskane pod wpływem różnego rodzaju obciążeń, z wynikami uzyskanymi przez różnych autorów w testach biomechanicznych na preparatach biologicznych in-vitro. Okazało się, że wyniki te są w dużym stopniu zbieżne. W celu dodatkowej weryfikacji stworzonego modelu numerycznego zostało zaprojektowane i wykonane stanowisko umożliwiające badanie in-vitro przemieszczeń fragmentu kręgosłupa ludzkiego. W badaniach stabilności wykorzystano sztuczny model kręgów L1, L2, L3. Wyniki, które uzyskano w modelowaniu numerycznym są bardzo zbliżone do wyników uzyskanych podczas testów na tym stanowisku.

In vitro screening techniques were used to evaluate 46 genotypes of Iranian potato collection for resistance to bacterial soft rot caused by Pectobacterium carotovorum subsp. carotovorum (Pcc). One month old in vitro rooted potato plantlets were inoculated by two inoculation techniques under in vitro conditions: 1) sterile toothpicks dipped into bacterial suspension and pressed into the crown of plantlets and 2) the freshly cut crown of plantlets were dipped into bacterial suspension of 108 cfu ∙ ml–1 for 10 min. Typical soft rot disease symptoms, including the percentage of wilted leaves were recorded on inoculated plantlets 3, 6, 9, 12 and 15 days post-inoculation. The potato genotypes which were examined responded differently to Pcc and varying levels of resistance were observed. Potato genotype AG showed the highest level of resistance. Results obtained from in vitro screening were then verified by classical tuber slice assay. The verifications were conducted using five representative cultivars: Milva, Ramus, Picaso, Marfona and Agria which responded similarly to both in vitro and classical evaluation systems. Similar results obtained from these tests indicated that the in vitro screening technique developed in this study could provide a simple and rapid whole plant assay in selecting resistant potato genotypes against bacterial soft rot.

The aqueous extracts of leaf of Citrus aurantifolia L were assessed in vitro for inhibitory activity against Macrophomina phaseolina isolated from dry root rot specimens of Gingelly. The antifungal activity was determined by poison food technique. The extracts have shown dose dependent inhibition of mycelial growth of test fungi. The extracts were more effective in inhibiting Macrophomina phaseolina. The extracts of Citrus aurantifolia were found effective against Gingelly dry root rot pathogens. Further field experiments are to be carried out to recommend the extracts against the disease.

Problematyka dotycząca zrodzenia i wychowania potomstwa zawsze stanowiła przedmiot szczególnej troski Kościoła. Przejawiało się to m.in. w prawnej regulacji tego zagadnienia. Uprawnienie-obowiązek zrodzenia i wychowania potomstwa łączyło się nieodzownie z ogólną wizją małżeństwa, jego celami i przedmiotem małżeńskiej zgody, stąd też wraz z ewolucją rozumienia samego małżeństwa w historii kanonistyki, zmieniało się także rozumienie terminu bonum prolis (dobro potomstwa). Normy zawarte w kodeksach Jana Pawła II (KPK/83 i KKKW) dotyczące zagadnienia wykluczenia dobra potomstwa, o których była mowa we wstępie niniejszego artykułu, w swojej treści są identyczne. Jedynie treść kan. 776 § 1 KKKW, stanowi swoiste dopełnienie treści kan. 1055 § 1 KPK/83, w którym mowa o strukturze małżeństwa. Z kolei kan. 1061 § 1 KPK/83, będący swoistą definicją małżeństwa dopełnionego i ważnie zawartego, nie znajduje swojego odpowiednika w KKKW. Zrodzenie i wychowanie potomstwa jest naturalnym celem i uwieńczeniem małżeństwa. Akty małżeńskie, powinny być ukierunkowane na prokreację i dokonywane w sposób ludzki i naturalny. Stąd też wykluczenie aktów małżeńskiego współżycia, otwartych na zrodzenie potomstwa, podjęte pozytywnym aktem woli skutkuje nieważnością samego małżeństwa.
The issue of procreation and education of offspring has always been the object of special concern for the Church. It appears, among others in legal regulation of this problem. The right and the duty to procreate and educate offspring was inevitably connected with general vision of marriage, it is purposes and property of matrimonial consent. Therefore, with the evolution of understanding of marriage itself in the history of canon law, the term of bonum prolis (good of the offspring) was also changed. Legal norms regarding the issue of exclusion of the good of the offspring contained in the 1983 Code of Canon Law (CIC/83) and in the 1990 Code of Canons of the Eastern Churches (CCEO), which was mentioned in the introduction of this article, are identical in their bodies. Only can. 776 § 1 CCEO is a kind of complement of can. 1055 § 1 CIC/83, which speaks about structure of marriage. In turn, can. 1061 § 1 CIC/83, which is a specific definition of consummated and valid marriage, has no equivalent in CCEO. The procreation and education of offspring is the natural purpose and the culmination of marriage. Marital acts should be directed towards procreation and must be performed in a natural and human fashion. Hence, the exclusion of acts of conjugal living which are open to procreation of offspring, taken with a positive act of the will results in the invalidity of the marriage itself.

Different Acanthamoeba species are amphizoic organisms distributed in wide range of habitats in natural and man-made environments; they are also detected on surfaces of equipment and accessories in health facilities. Some strains of the amoebae are causative agents of the vision-threatening human disease Acanthamoeba keratitis, mainly reported in contact lens wearers. An exceptional high resistance of Acanthamoeba trophozoites and particularly cysts to chemicals, disinfectants and drugs is believed as influencing difficulty resulting in unsuccessful therapeutic management. As Acanthamoeba keratitis is the serious medical problem worldwide, different chemicals with possible activity against environmental and clinical Acanthamoeba strains are tested. In our study, selected disinfectants used in health care settings and laboratories were tested and their efficacy against the corneal strains Acanthamoeba castellanii and A. polyphaga , and environmental A. castellanii Neff strain was assessed. Comparative assessment of results of the assays show that, apart from amoebistatic effects, the disinfectants indicated expected cysticidal efficacy.

The purpose of this study was to determine the in vitro susceptibility to 6 antifungal agents of 143 strains of Candida species isolated from 545 patients with suspicion of fungal infection. Test ATB Fungus (bioMerieux) was used. Among all Candida isolates 91.6% were susceptible to tested polyenes and 32.9% to all azoles. Less susceptible to polyenes isolates were noted in low percentage (0.7 to 4.2%) and to azoles more frequcntly (32.9 to 41.2%). Candida isolates resistant to antifungal agents tested were dependent from the kind of clinical samples and species of fungi. Monitoring of the susceptibility to antifungal agents by ATB commercial test seems to be uscful tool for therapeutic purposes.

Human activity has caused environmental transformations for millennia, most often driven by the desire to acquire natural resources. Unfortunately, the desire to acquire these resources in many cases results in a reduction in biodiversity, that is, a reduction in genetic diversity in a given area. In addition, the transformation of the environment results in the replacement of plant species that originally existed in a given area by other vegetation, including foreign ones. Environmental transformation can take various forms, such as agricultural intensification, the acquisition of new mining areas, or simply the occupation of more land for municipal or industrial purposes. Nor can we forget one of the most important problems of recent decades: global warming, which contributes significantly to climate change and leads to the disappearance of many plant species. The way to reduce the negative human impact is to carry out environmental protection activities, both passive and active. In this paper, we take a closer look at how in vitro cultures of plants can benefit biodiversity conservation thanks to the possibility of micropropagation and long-term storage of plants.
