The present study was carried out with the aim to evaluate the relation of the content of proinflammatory and antiinflammatory cytokines with the sperm quality indicators. The study involved 45 patients with a pathological spermogram and 15 practically healthy men. A positive correlation (r = 0.45) between the content of IL-6 and the sperm concentration in the ejaculate and a negative correlation (r = -0.42) of this cytokine with the number of live sperm were revealed. The concentration of IL-8 was positively correlated with sperm density (r = 0.52) and it was negatively correlated with the number of live sperm (r = -0.55). A positive correlation between the content of IL-10 and the sperm concentration as well as the number of active forms was observed (r = 0.45 and 0.5, respectively), and a negative correlation (r = -0.7) was observed between the ejaculate volume and the concentration of IL-10. In the obtained ejaculates, a close positive correlation between the content of IL-8 and IL-6 (r = 0.74) and between the concentration of IL-10 and IL-6 (r = 0.72) was observed. Based on the completed study, a conclusion was made that the increased level of IL-6 and IL-8 may result in the reduction of the sperm quality, and, possibly, infertility, and that IL-10 plays an important role in improving sperm parameters.

Opis:
Interleukin 18 (IL-18) is a pleiotropic cytokine involved in the regulation of innate and acquired immune response. In the milieu of IL-12 or IL-15, IL-18 is a potent inducer of IFN-gamma in natural killer (NK) cells and CD4 T helper (Th) 1 lymphocytes. However, IL-18 also modulates Th2 and Th17 cell responses, as well as the activity of CD8 cytotoxic cells and neutrophils, in a host microenvironment-dependent manner. It is produced by various hematopoietic and nonhematopoietic cells, including dendritic cells and macrophages. In an organism, bioactivity of the cytokine depends on the intensity of IL-18 production, the level of its natural inhibitory protein - IL-18BP (IL-18 binding protein) and the surface expression of IL-18 receptors (IL-18R) on the responding cells. This review summarizes the biology of the IL-18/IL-18BP/IL-18R system and its role in the host defense against infections. The prospects for IL-18 application in immunotherapeutic or prophylactic interventions in infectious and non-infectious diseases are discussed.

Interleukin 1 (IL-1) and interleukin 6 (IL-6) are typical examples of multfunctonal pro-infammatory cytokines involved in the regulaton of the immune response, hematopoiesis, and infammaton. Both peripheral and intraventricular administraton of these cytokines causes acute phase symptoms, e.g. fever, actvaton of the hypothalamic-pituitary-adrenal axis and psychological depression. The amygdala belongs to the structures of the limbic system involved in the regulaton of the immune response. Increased actvity of immune system may lead to changes in the role of amygdala, medial prefrontal cortex, anterior cingulate cortex or insula. The aim of the study was to present the mutual interactons between the amygdala and pro-infammatory cytokines such as interleukin-1β (IL-1 beta) and interleukin 6 (IL-6). Most of the data included in this review comes from animal studies.

Opis:
The endometrium of women with endometriosis has a different expression of cytokines, angiogenic and hormonal factors compared to healthy women. Endometriosis is a disease occurring mostly in women at a reproductive age with a frequency of 7–10%. Despite many studies and hypotheses on the pathogenesis of the disease, marker specific to endometriosis has not yet been detected. Thus, laparoscopy remains the gold standard in the diagnosis, particularly in the case of peritoneal form that cannot be visualized by means of an ultrasound test. Other forms of endometriosis – ovarian (chocolate cysts) and deep infiltrating – can be identified in an ultrasound test. Scientists are currently searching for markers that in a specific combination would ensure maximum sensitivity and specificity of the non-invasive detection of endometriosis, even at early stages. The article presents selected factors, such as interleukin 8, vascular endothelial growth factor, platelet-derived growth factor, nerve growth factor, detected in the endometrium, which can be a potential diagnostic target. Diagnoses made at the level of endometrium would facilitate the identification of endometriosis and would significantly reduce costs associated with the necessity to conduct laparoscopy.
Endometrium pacjentek z endometriozą charakteryzuje się inną ekspresją cytokin, czynników angiogennych i hormonalnych niż u kobiet zdrowych. Endometrioza występuje głównie u kobiet w wieku reprodukcyjnym, z częstością około 7–10%. Mimo wielu badań i hipotez dotyczących patogenezy nie wykryto dotychczas swoistego markera dla endometriozy. Złotym standardem diagnostycznym, zwłaszcza w przypadku postaci otrzewnowej, której nie udaje się uwidocznić w badaniu ultrasonograficznym, pozostaje laparoskopia. Inne formy endometriozy – jajnikową (torbiele czekoladowe) i głęboko naciekającą – można rozpoznać w badaniu ultrasonograficznym. Obecnie poszukuje się markerów, które w określonej kombinacji zapewniłyby maksymalną czułość i swoistość nieinwazyjnego wykrywania endometriozy, nawet we wczesnych stadiach. Czułość oznaczeń wykonywanych z krwi nie jest niestety satysfakcjonująca w odniesieniu do otrzewnowej postaci choroby. W artykule zaprezentowano potencjalne cele diagnostyczne – wybrane czynniki wykrywane w endometrium, czyli interleukinę 8, czynnik wzrostu śródbłonka naczyniowego, płytkopochodny czynnik wzrostu i czynnik wzrostu nerwów. Pobranie endometrium do badań jest łatwe i może być wykonywane ambulatoryjnie. Diagnozowanie z poziomu endometrium ułatwiłoby rozpoznawanie endometriozy i znacząco zmniejszyło koszty związane z koniecznością przeprowadzania laparoskopii.

Objective: To determine transforming growth factor (TGF) β1, interleukin (IL) 4, and IL-10 concentrations in human milk and to assess the relationship between allergic disorders in mothers and the content of the interleukins in their milk. Material and methods: Thirty allergic and 46 healthy mothers were included in the study. Colostrum was collected 2-3 days after delivery. Cytokine concentrations were determined with commercial enzyme-linked immunosorbent systems. Results: TGF-β1was found in milk from 23 women in the control group (53.49%) and 11 in the allergy group (37.93%). When TGF-β1 was present, the median concentration was higher in the allergy group than in the control (61.5 and 30.4 pg/mL, respectively; P < 0.004). IL-10 was present in the colostrum of all the women and the median IL-10 concentration did not differ between the allergy (50.5 pg/mL) and control (51.5 pg/mL) groups. The probability of occurrence of a positive IL-4 value in the allergy group was greater than in the control group (chi-squared [df=1] = 2.60, P < 0.053). Median IL-4 level did not differ significantly between the two groups (0.5 and 0.5 pg/mL respectively). Conclusions: TGF-β1 was detected less often in the colostrum of allergic mothers than in that of mothers without allergy (but the difference was not statistically significant). IL-4 was found more often in the colostrum of allergic mothers than nonallergic ones. The allergy status did not correlate with IL-10 concentration.

Tkanka chrzęstna charakteryzuje się małą zdolnością regeneracji oraz niewielkim potencjałem do prawidłowej odbudowy, dlatego też, dotychczas stosowane metody leczenia uszkodzeń chrząstki są niewystarczające. Nadzieję na przełom w leczeniu wielu chorób chrząstki, jak również regenerację tej tkanki, daje inżynieria tkankowa wykorzystująca m.in. syntetyczne polimery z grupy poliestrów alifatycznych i poliwęglanów takie jak kopoolimery glikolidu, ε-kaprolaktonu, L-laktydu oraz trimetylenowęglanu zsyntetyzowane za pomocą nowatorskiej metody wykorzystującej jako inicjator polimeryzacji acetyloctan cyrkonu. W ramach niniejszej pracy dokonano oceny biozgodności wybranych biodegradowalnych materiałów w oparciu o poziom sekrecji prozapalnych cytokin IL-1α i IL-1β, IL-6 oraz IL-8 oznaczonych metodą ELISA. Spośród badanych polimerów biodegradowalnych pod-łoża o składzie PCLGA 92:8, PLAGA 85:15, PLATMC 70:30, PLATMC 30:70, PLATMC 50:50 wydają się charakteryzować największą biozgodnością, gdyż ich obecność nie powodowała istotnego wzrostu sekrecji cytokin prozapalnych przez chondrocyty rosnące na ich powierzchni. Pozostałe badane materiały polimerowe powodowały pewne niepożądane reakcje komórek związane ze stymulacją uwalniania niektórych spośród analizowanych cytokin prozapalnych przez chondrocyty. Uzyskane wyniki pozwoliły na wyciągnięcie wniosku, iż podłoża wykonane z analizowanych w pracy polimerów biodegradowalnych zsyntetyzowanych przy użyciu acetyloacetonianu cyrkonu zasługują na dalszą uwagę, gdyż niosą nadzieję na umożliwienie leczenia ubytków tkanki chrzęstnej.
Cartilage characterizes low potential to regeneration and proper reconstruction. Nowadays, methods of cartilage lesions treatment are unsatisfactory. Tissue engineering is very promising in therapy of many cartilage injures as well as regeneration of this tissue. Biodegradable aliphatic polyesters and polyesterocarbonates such as copolymers of glycolide, L-lactide, ε-caprolactone and trimethylene carbonate were synthesized using a novel method employing non-toxic zirconium acetylacetonate as initiator of polymerization. The aim of the study was to examine biocompatibility of selected biodegradable materials based on the level of secretion of proinflammatory cytokines IL-1α, IL-1β, IL-6 and IL-8. Our results show, that among the studied biodegradable polymers the PCLGA 92:8, 85:15 PLAGA, PLATMC 70:30, PLATMC 30:70, 50:50 PLATMC were characterized by highest biocompatiblity, because they do not cause a significant increase in secretion of proinflammatory cytokines by chondrocytes growing on their surface. Other polymers could cause up-regulation of some proinflammatory cytokines secretion by chondrocytes cultured on their surface. The other tested polymeric materials did not induce the release of proinflammatory cytokines by chondrocytes. Generally, it should be concluded that the studied biodegradable copolymers synthesized with the use of zirconium acetylacetonate as initiator of polymerization deserve further attention, as being promising for the treatment of cartilage defects.

ObjectivesVenom immunotherapy (VIT) is an effective treatment method and is addressed to patients with a history of an anaphylactic reaction to Hymenoptera stings. However, the immunological mechanisms of protection have not been explained yet. The objective of this study was to analyze neutrophils, interleukin 8 (IL-8) and interleukin 17 (IL-17) before and after the initial phase of the immunotherapy.Material and MethodsOverall, 40 individuals, including 20 wasp venom sensitized and 20 bee venom sensitized patients, were included in the study. The patients had had a history of severe allergic reactions type III and IV according to Mueller’s classification. An ultra-rush VIT protocol was used in this study. The concentration of serum IL-8 and IL-17A was determined using the ELISA enzymatic method.ResultsThe authors demonstrated a significant rise in the IL-8 level after the immunotherapy, compared to baseline (14.9 vs. 24.7, p < 0.05). The rise in the neutrophils level was also noticeable but proved to be barely out of the range of statistical significance (4.3 vs. 5.0, p = 0.06). The shift in IL-17A was negligent and not statistically significant in the paired samples t-test (1.6 vs. 1.5, p = 0.34)ConclusionsVenom immunotherapy induces neutrophils and IL-8 activity after 2 days. After the desensitization, the level of IL-17A did not change.

In this study plasma concentrations of IL-1α and IL-8 in 29 female dogs with malignant mammary gland tumours (19 without metastasis and 10 with metastasis) and in 10 healthy control animals were determined. Concentrations of IL-1α and IL-8 were analysed using a specific canine ELISA assay. Mean plasma concentrations of IL-1α and IL-8 were significantly higher (p<0.05) in female dogs with both non-metastatic and metastatic malignant tumours compared to the healthy animals. The concentrations of both tested cytokines were significantly increased (p<0.05) in the dogs with metastasis. In female dogs with mammary carcinomas, the plasma concentration of IL-1α was significantly higher (p<0.05) in the animals with grade 3 tumours compared to the dogs with grade 1 tumours. The concentration of IL-8 was significantly higher (p<0.05) in the dogs with grade 3 tumours compared to that found in the animals with grade 1 and grade 2 tumours. A moderate correlation (r=0.433) was found between IL-1α and IL-8 concentrations in the female dogs. These findings suggest that increased malignancy and invasiveness of canine mammary tumours is associated with an increased production of IL-1α and IL-8 in the tumour microenvironment, which, in turn, leads to an increase in their circulating levels. This may indicate that circulating levels of the cytokines investigated could be considered as diagnostic and prognostic markers in canine malignant mammary tumours. However, further studies in this fields are needed.

The main aim of this article is to analyze some aspects of the historical roots of the North Korean ideology and political system. This study argues that the North Korean ideology is to a certain extent rooted in the historical background of the Korean Peninsula but has also been purposely changed and manipulated in form to deal with at-the-time difficulty and circumstance that the regime faced. Meanwhile what can be observed in the North Korean ideology is that there is no direct referring to the antecedents. These claims are illustrated in this article by various primary material of a propagandistic nature published in North Korea and by research articles.
