Novel 6-phenylselenenyl-5-propyluracils were synthesized from 5-propyluracil with the use of regioselective synthesis to give 1-[(2-hydroxyethoxy)-methyl]-6-phenylselenenyl-5-propyluracil (6), 1-ethoxymethyl-6-phenylselenenyl-5-propyluracil (9) and 1-benzyloxymethyl-6-phenylselenenyl-5-propyluracil (10). Interaction of these compounds with recombinant HIV-1 reverse transcriptase (RT) was evaluated using a non-isotopic colorimetric method. Compounds 9 and 10 exerted potent HIV RT inhibition (IC50 0.06 and 0.05 µM respectively) while compound 6 showed moderate inhibition (IC50 = 3.5 µM). Potent anti-HIV-1 activity in MT-2 cells inoculated by a syncythia-inducing HIV-1 (cat #3 strain) laboratory isolate was exerted by compounds 9 and 10 (EC50 0.62 µM and 0.025 µM, respectively), while compound 6 showed only moderate activity (IC50 = 4.1 µM). In addition, compound 10 showed very good in vitro therapeutic index (TI > 2046), indicating that it is a potential anti-HIV/AIDS drug.

From the seeds of Acacia confusa, a chitinase-like antifungal protein designated as acaconin that demonstrated antifungal activity toward Rhizoctonia solani with an IC50 of 30±4 µM was isolated. Acaconin demonstrated an N-terminal sequence with pronounced similarity to chitinases and a molecular mass of 32 kDa. It was isolated by chromatography on Q-Sepharose, SP-Sepharose and Superdex 75 and was not bound by either ion exchanger. Acaconin was devoid of chitinase activity. The antifungal activity against Rhizoctonia solani was completely preserved from pH 4 to 10 and from 0°C to 70°C. Congo Red staining at the tips of R. solani hyphae indicated inhibition of fungal growth. However, there was no antifungal activity toward Mycosphaerella arachidicola, Fusarium oxysporum, Helminthosporium maydis, and Valsa mali. Acaconin inhibited proliferation of breast cancer MCF-7 cells with an IC50 of 128±9 µM but did not affect hepatoma HepG2 cells. Its IC50 value toward HIV-1 reverse transcriptase was 10±2.3 µM. The unique features of acaconin include relatively high stability when exposed to changes in ambient pH and temperature, specific antifungal and antitumor actions, potent HIV-reverse transcriptase inhibitory activity, and lack of binding by strongly cationic and anionic exchangers.

Opis:
Multiple transmission of simian immunodeficiency virus (SIV) into the human population started the pandemic of human immunodeficiency virus (HIV-1). The extraordinary variability of human immunodeficiency virus HIV-1 resulting from the high rate of genome mutations and the recombination between different strains resulted in the emergence of a genetically diverse viral strains. Genetic differences between HIV-1 isolates are now the basis for the classification of the virus. Tracking the spread of the HIV-1 genetic variants allows for the monitoring of the development of the epidemic, and provides important information about the time and possible routes of virus introduction into different countries, geographical regions or populations with different transmission routes of infection. On the other hand, the vast viral genetic diversity complicates the diagnosis of infection, can affect the susceptibility of HIV-1 to drugs and is a major obstacle in research aimed at creating an effective vaccine. In this paper the current knowledge on the origin of virus, the genetic diversity of HIV-1, as well as its impact on the course of infection and development of the epidemic is presented.
Pandemia ludzkiego wirusa niedoboru odporności (human immunodeficiency virus – HIV) została zapoczątkowana przez wielokrotną transmisję małpiego wirusa upośledzenia odporności (simian immunodeficiency virus – SIV) do populacji ludzkiej. Niezwykła zmienność HIV-1 wynikająca z dużej częstości mutacji pojawiających się w genomie oraz możliwości rekombinacji między materiałem genetycznym różnych szczepów wirusa zaowocowała wykształceniem zróżnicowanych pod względem genetycznym wariantów. Genetyczne różnice między izolatami HIV-1 stanowią obecnie podstawę klasyfikacji wirusa, a śledzenie rozprzestrzeniania poszczególnych wariantów pozwala na monitorowanie rozwoju epidemii i dostarcza ważnych informacji na temat czasu i sposobu transmisji wirusa do różnych krajów, regionów geograficznych lub populacji. Z drugiej strony olbrzymia różnorodność form genetycznych wirusa utrudnia diagnostykę infekcji, może wpływać na wrażliwość HIV-1 na leki i stanowi poważną przeszkodę w badaniach zmierzających do stworzenia skutecznej szczepionki. W niniejszej pracy przedstawiono aktualny stan wiedzy na temat pochodzenia wirusa, genetycznego zróżnicowania HIV-1 i wpływu tego czynnika na przebieg zakażenia i rozwój epidemii.

This paper presents an algorithm for nonlinear adaptive control of the viral load in HIV-1 infection. The infection model considered is a reduced complexity nonlinear state-space model with two state variables, that represent the plasma concentration of uninfected and infected CD4+ T-cells of the human immune system. The viral load is assumed to be proportional to the concentration of infected cells. First, a change of variables that exactly linearizes the system is obtained. For the resulting linear system the manipulated variable is obtained by state feedback. To compensate for the uncertainty in the infection parameter of the model an estimator based on a Control Lyapunov Function is designed.

Despite the extensive research efforts over the past 25 years that have focused on HIV, there is still no cure for AIDS. However, tremendous progress in the understanding of the structure and biology of the HIV virus led to the development of safe and potent HIV-based transgene delivery vectors. These genetic vehicles are referred to as lentiviral vectors. They appear to be better suited for particular applications, such as transgene delivery into stem cells, compared to other viral- and non-viral vectors. This is because Lentivirus-based vectors can efficiently infect nondividing and slowly dividing cells. In the present review article, the current state of understanding of HIV-1 is discussed and the main characteristics that had an impact on vector design are outlined. A historical view on the vector concept is presented to facilitate discussion of recent results in vector engineering in a broader context. Subsequently, a state of the art overview concerning vector construction and vector production is given. This review also touches upon the subject of lentiviral vector safety and related topics that can be helpful in addressing this issue are discussed. Finally, examples of Lentivirus-based gene delivery systems and their applications are presented, with emphasis on animal transgenesis and human gene therapy.

HIV-1 or Human Immuno Deficiency Virus-1 is the main causative agent of Acquired Immuno Deficiency Syndrome (AIDS). Human host infected with HIV - 1 extensively harbours many viral variants but very little is known about the difference in pattern[17] of evolution of phylogenetic lineages of HIV-1 non recombinant, normal inter subtype recombinant and main two specific recombinant forms of HIV-1 i.e., Circulating Recombinant Forms (CRFs) and Unique Recombinant Forms (URFs). This study is mainly concerned with study of the difference in evolutionary lineages of non-recombinant and recombinant sequences of HIV-1 genome sequences and identification of geographically rich areas which has reported high degree of HIV-1 occurrence and variety. Total 1550 HIV-1 genome sequences were obtained from HIV Los Alamos Database. The sequences were aligned using MAFFT (Multiple Alignment using Fast Fourier Transform) web server tool. Alignment was carried out using 10 different set of alignment parameter values. After alignment the aligned file was used for constructing N-J phylogenetic tree using Clustal X2 tool. Phylogenetic analysis was performed keeping in mind the category to which the sequence belongs. Upon analysis it was observed that the clade containing the probable ancestor belongs remained constant in all cases of different alignment values. Non recombinant isolates, inter subtype recombinants, CRFs, URFs all followed different patterns of evolution. Non recombinant sequences were found geographically specific and subtype specific to some extent whereas, normal recombinants were subtype specific and less geographically specific. CRFs showed variation among the pattern of their evolution. At some instances the sequences occurred as sister taxa of non-recombinant or normal inter subtype recombinant sequences, while at some instances as sister taxa of other CRFs where they were geographically specific. Three CRFs existed as completely diverged sequences. URFs were four in number; two of them were Indian isolates of while other two were Japanese isolates. URFs were found to be totally geographically specific. Geography wise high rate of variation was observed in India and Japan as these two countries had sequences belonging to all of the above categories. Cameroon and South Africa have very large number isolates and a considerable amount of genetic variation among isolates but they lack URFs.
