Both Globodera rostochiensis (Wollenweber, 1923) Behrens, 1975 and Globodera pallida (Stone, 1973) are considered the most economically important nematode pest. Annual regional survey conduct in Poland have revealed high infestation of pathotype Ro1 to the northwest of the country and presence of new pathotype Ro5 of G. rostochiensis. In 2009-2013 fifty eight soil samples were tested. parasitic root organisms cause significant losses in quantity and quality of potato crops. In many European countries PCN have become a major problem in potato production mainly in starch potato cultivars. The pathotype Ro1 was detected mostly in central and northern Poland and pathotype Ro5 in four separated provinces: malopolskie, lubuskie, lubelskie and pomorskie. The abundance of cysts in location where the new pathotype was detected suggests that is a thread of further spread of pathotype Ro5 of G. rostochiensis in the country. Consequently, producers and farmers need to grow resistant potato cultivars to keep nematode population below damaging level. These results indicate the urgent need to prevent the spread of PCN to non-infested areas. This report focused on identification of nematode pathotypes in soil samples from infested polish fields and presentation of G. rostochiensis distribution on territory of Poland.

The cyst nematodes belonging to the genus Globodera are big worldwide problem in countries were Sola-naceaous plants growing. Knowledge of species-composition in populations of Globodera rostochiensis and Globodera pallida is very important for selection of appropriate measure of nematode regulations occurrence. Inter- and intraspecific variability among species of Globodera rostochiensis and Globodera pallida were studied intensively with the use of molecular analyses of DNA methods. This review summarize and compare of methods chosen to distinguishing between Globodera, both pathotypes and species.

Opis:
Mątwik ziemniaczany i mątwik agresywny to dwa nicienie tworzące cysty na korzeniach ziemniaka. Zarówno w Polsce jak i w wielu innych państwach posiadają status organizmów kwarantannowych. Obecność mątwika ziemniaczanego może powodować straty w plonie bulw sięgające 50–80%. Metody zwalczania mątwika można podzielić na trzy grupy: agrotechniczną, biologiczną i chemiczną. Pozyskane wyniki będą stanowiły podstawę opracowania zaleceń dla rolników, które usprawnią eliminację szkodnika.
Potato cyst and aggressive cyst are two nematodes forming cysts on the roots of the potato. In Poland and in many other countries they have the status of quarantine organisms. The presence of potato cyst nematodes may cause losses in the tuber yield reaching 50–80%. Methods of fighting cyst nematode can be divided into three groups: agrotechnical, biological and chemical. The obtained results will be the basis for the development of recommendations for farmers, which will improve the elimination of the pest.

Opis:
The efficiency of breeding new potato cultivars may be increased by pre-breeding that is by developing parental lines, which have new traits, not present in the genetic pool available for breeders or have new combinations of genes, or have improved level of economically important traits. The use of parental lines in commercial breeding programs is expected to ensure the biological progress in the newly created cultivars of potato. At the beginning, the development of parental lines in Młochów Research Center of Plant Breeding and Acclimatization Institute – Nation-al Research Institute (IHAR-PIB) was focused on resistance to viruses and later on resistance to late blight and other pathogens. The procedures of selecting resistant parental lines are described. These procedures were initially based on purely phenotypic tests for resistance, which lately were supplemented with marker assisted selection (MAS) apply-ing molecular markers linked with some resistance genes.

Opis:
Globodera rostochiensis, as well as other cyst nematodes, induces formation of a multinucleate feeding site, called syncytium, in host roots. In tomato roots infected with a potato cyst nematode, the syncytium is initiated in the cortex or pericycle. Progressive cell wall dissolution and subsequent fusion of protoplasts of newly incorporated cells lead to syncytium formation. Expansion and development of a syncytium strongly depends on modifications of a cell wall, including its degradation, elongation, thickening, and formation of ingrowths within it in close contact with tracheary elements. Recent reports have demonstrated that during formation of syncytium, numerous genes of plant origin, coding for cell wall-modifying enzymes are up-re-gulated. In this research, we studied a detailed distribution and function of two tomato 1,4-β-endoglucanases in developing feeding sites induced by G. rostochiensis. In situ localization of tomato LeCel7 and LeCel8 transcripts and proteins demonstrated that these enzymes were specifically up-regulated within syncytium and in the cells adjacent to the syncytium. In non-infected roots an expression of LeCel7 and LeCel8 was observed in the root cap and lateral root primordia. Our data confirm that cell wall-modifying enzymes of plant origin have a role in a modification of cell wall within syncytia, and demonstrate that plant endoglucanases are involved in syncytia formation.

Opis:
The sequence of anatomical and ultrastructural events leading to the syncytium development in tomato roots infected with Globodera rostochiensis was examined. The syncytia were preferentially induced in cortical or pericyclic cells in the elongation zone of root. They developed towards the vascular cylinder by incorporation of new cells via local cell wall breakdown. After surrounding primary phloem bundle and reaching xylem tracheary elements syncytia spread along vascular cylinder. Roots in primary state of growth seemed to be the best place for syncytium induction as syncytia formed in the zone of secondary growth were less hypertrophied. At the ultrastructural level syncytial elements were characterized by strong hypertrophy, breakdown of central vacuole, increased volume of cytoplasm, proliferation of organelles, and enlargement of nuclei. On the syncytial wall adjoining vessels the cell wall ingrowths were formed, while the syncytial walls at interface of phloem were considerably thickened. They lacked of functional plasmodesmata and did not form any ingrowths. Using immunofluorescent-labelling and immunogold-labelling methods tomato expansin 5 protein was localized in nematode infected roots. The distribution of LeEXP A5 was restricted only to the walls of syncytia. The protein distribution pattern indicated that LeEXP A5 could mediates cell wall expansion during hypertrophy of syncytial elements.

The investigation covers a three-year (1993-1995) period of many years field experiments with six-coarse crop rotations with different proportions of potatoes: fodder cereal crop rotation 16,7%; potato-cereal crop rotation 33,3%; and potato crop rotation 50,0% G. rostochiensis, Ro 1 non-resistant cvs. Bronka and Lena and resistant cvs. Fala and Ibis were used. Potatoes followed both non-cereal and cereal forecrops. A static experiment has been continued since 1987 on a grey-brown podsolic soil, developed from sandy loam on fine sandy soil, rich in available nutrients. A negative reaction of potatoes to their percentage increase in crop rotation was found, higher in non-resistant than in resistant cultivars to G. rostochiensis. In the fodder-cereal rotation, after lucerne with red clover, all cultivars produced yields which approximated each other, averaging 38,6 t/ha. A drop in yields in non-resistant cultivars was 13,3% in the potato-cereal rotation and 20,2% in the potato rotation, and in resistant cultivars it was 1,1% and in non-resistentl2,0%, respectively. There was also a relationship between yield quality, population of G. rostochiensis and cultivars.
