    It has been investigated, with the aid of routine cytochemical methods, whether any changes of the activity of oxydoreductases of G. candidum which has been intluenced by chemical factors affecting its growth, and with which it can contact in human organism or his environment, occur. The factors were: antibacterial antibiotic from tetracycline group /chlortetracycline/, antifungal antibiotic representing polyenes derivatives /nystatine/, heavy metal salt /mercuric chloride/, aniline dye /malachite green/. To evaluate cytochemical preparates in which the fungus ce lis have been classified according to intensity of enzymatic reaction the chi² test has been suggested. It has been proved that all the investigated compounds have significant changes in the fungus metabolism showing in the oxydoreductases activity deviations.

    We have studied the capability of dissociation of 22 L-form aminoacids by 27 strains of fungi belonging to species: Candida, Cryptococcus, Geotrichum, Rhodotorula, Saccharomyces, Trichosporon. Fungal strains were cultured on agar medium with phenyl red and studied aminoacid. Disamination of an aminoacid by a fungus resulted in alkalisation of the medium and change of its colour from yellow to pink. Strains from genus Trichosporon and Geotrichum decomposed the most aminoacids (15-16), while fungi from genus Candida decomposed less aminoacid (1-8). Strains form species Candida famata, C. kefyr, C. lambica. C. rugosa, C. neoformans, R. mucilaginosa and S. cerevisiae did not show any ability to desaminate any of the 22 aminoacids used in the study. Aminoacids most frequently disaminated by the studied strains of fungi included acidic aliphatic aminoacids and their amides: L-aspartic acid, L-asparagine, L-glutaminic acid, L-glutamine and neutral aliphatic aminoacids: L-glycin, L-alanine, L-serine. Aromatic and sulphur aminoacids (L-cysteine, L-cystine) were not decomposed by the studied fungal strains.

    The aim of the study was to evaluate the ability of Galactomyces geotrichum MK017 for the biosynthesis of 2-phenylethyl alcohol and optimization of the culture medium composition in order to increase the yield of the product. The culture of Galactomyces geotrichum MK017 strain was carried out in the laboratory scale. For isolation of volatiles Solid Phase Microextraction (SPME) has been used. The identification and quantification of aroma compounds produced by examined microflora was determined by gas chromatography and mass spectrometry (GC/MS) system. The results showed that the tested mould Galactomyces geotrichum MK017 shows the potential for the production of 2-phenylethyl alcohol, but also for the synthesis of phenylacetaldehyde, phenylacetic and phenyllactic acids. For the optimization of the 2-phenylethyl alcohol production yield four different medium composition have been tested. The bioprocess of aroma compound production by tested microorganism was the most efficient on the medium composed of sucrose (80 g/l) and L-phenylalanine (21 g/l) and pH value of 5.0. Using this composition in a batch culture of 770 ml volume the highest concentration of 2-phenylethyl alcohol has been obtained – 6 mg/l. At the same time amount of phenylacetaldehyde, phenylacetic acid and phenyllactic acidhas reached 2.2 mg/l, 10.66 mg/l and 32.3 mg/l respectively.
    Celem pracy było określenie zdolności pleśni Galactomyces geotrichum MK017 do biosyntezy 2-fenyloetanolu oraz optymalizacja składników podłoża produkcyjnego w celu zwiększenia ilości uzyskanego produktu. Hodowlę pleśni Galactomyces geotrichum MK017 przeprowadzono w skali laboratoryjnej. Do identyfikacji związków lotnych zastosowano metodę mikroekstrakcji do fazy stałej (SPME). Identyfikacji oraz oceny ilościowej uzyskanego związku zapachowego dokonano z wykorzystaniem chromatografii gazowej sprzężonej ze spektrometrią mas (GC/MS). Uzyskane wyniki potwierdziły zdolność badanej pleśni Galactomyces geotrichum MK017 do produkcji 2-fenyloetanolu oraz do syntezy aldehydu fenylooctowego, kwasu fenylooctowego oraz kwasu fenylomlekowego. W celu optymalizacji produkcji 2-fenyloetanolu przetestowano cztery różne podłoża hodowlane. Największą produkcję 2-fenyloetanolu otrzymano na pożywce zawierającej sacharozę (80 g/l) i L-fenyloalaninę (21 g/l) oraz przy pH pożywki na poziomie 5.0. Uwzględniając powyższe zawartości poszczególnych składników pożywki, uzyskano w hodowli okresowej w 770 ml pożywki, najwyższą koncentrację 2-fenyloetanolu – 6 mg/l. Równocześnie w próbie otrzymano aldehyd fenylooctowy, kwas fenylooctowy oraz kwas fenylomlekowy odpowiednio w ilościach: 2.2 mg/l, 10.66 mg/l i 32.3 mg/l.

    The 50% trichomonocidal concentration of 7 examined ethers (T. vaginalis strain N° 1/86, Roiron medium, safranine staining, 30 min.) was estimated by means of dose-response curve. The minimal mycostatic concentration (Candida albicans L-45, Geotrichum candidum; 3% Sabouraud agar, 37°C, 24 hr) was calculated with the aid of regression equation. All examined ethers show in vitro a marked trichomonocidal effect comparable with activity of ornidazole or phenol. Mycostatic activity of new compounds is many times higher than activity of phenol, but considerably lower than that of klotrimazole. The strongest complex trichomonoddal and mycostatic effect-comparable to ornidazole or klotrimazole - shows the new derivative N,N-diethylaminoethyl oxime of 1-tioflavone (compound II), which is fairly toxic for mammal.

    Hydrolase activity of (enzymograms, biotypes) in Geotrichum candidum, one of the poorly described pathogenic fungi, was studied 81 strains were isolated from oral cavity and faeces of patients with gastrointestinal tract disorders. Axenic strains were differentiated with API 20C Aux and API ZYM tests. Then, enzymograms and biotypes were determined for all strains based on the activity of 19 hydrolases. High variability of enzymograms (17 different types) was found. The highest activity was noted in case of: e2-alkaline phosphatase, e6-lecucine arylamidase, e11-acid phosphatase. E5-lipase. e7-valine arylamidase, e12 - naphtol-AS-BI-phosphohydrolase and e17-β-glucosidase were used for biotyping procedures. Our own system of biotyping of 81 strains of G. candidum was based on the mathematical binominal distribution formula (1 : 4 : 6 : 4: 1) – all; ,,+”;one „-”, three „+”; two „-”,two „+”; three ,,-”,one ,,+”; all „-”. We have found: A (11.1 ± 3.5%), B1 (6.17 ± 2.67%). B2 (1.23 ± 1.22%), B4 (4.94 ± 2.41%), C1 (1.23 ±1.22%), C3 (63.0 ± 5.4%), D2 (9.88 ±3.31%), D3 (2.47 ± 1.72%). Among all strains from 8 various biotypes of G. candidum.

    Wzrost znaczenia wirusa Y jest wiązany głównie ze zmianą jego biologii, licznymi szczepami (rekom- binacjami, mutacjami) wskutek zmian pogodowych oraz szybkim przemieszczaniem się go w okresie wiosennym za pośrednictwem licznie występujących mszyc nieziemniaczanych (wektorów wirusów), które przy wcześnie rozpoczynającej się gorącej wiośnie zaczynają się nagle uskrzydlać i poszukiwać w przestrzeni rolniczej żywicieli pośrednich. Wzrost znaczenia chorób grzybowych, w tym wodnej zgnilizny przyranowej (Pythium sp.) oraz geotrychozy ziemniaka (Geotrichum candida) jest także związany ze sprzyjającymi infekcji warunkami pogodowymi – gorącym latem z lokalnie występującymi okresami ulewnych deszczów. Choroby te nie dają objawów diagnostycznych na roślinach w czasie wegetacji. Do zainfekowania bulw dochodzi najczęściej w trakcie zbioru, a potem następuje lawinowy rozwój chorób w nieodpowiednich warunkach w przechowalni. Wzrasta również znaczenie antraknozy ziemniaka (Colletotrichum coccodes), która kiedyś uważana była za sporadycznie spotykaną i nie miała znaczenia gospodarczego. Ostatnio wskutek zmian pogodowych powoduje ona czasami bardzo duże straty w uprawie. Wynika stąd potrzeba przyjrzenia się tym chorobom, oceny zagrożenia, umie- jętnego zdiagnozowania i wprowadzenia trafnych zabiegów agrotechnicznych i ochronnych na planta- cjach ziemniaka.

    This work was carried out during two successive seasons (2016 and 2017) on cucumber fruits from a plastic greenhouse and from open field cultivation in El Gharbeia and El Giza Governorates, Egypt. Isolation trials from spoilage fruit samples of plastic greenhouse cultivation recorded high frequency of Alternaria tenusinium, Fusarium spp. and Pleospora alli. The most common fungi of rotten cucumber fruits from an open field were Galactomyces spp. and Fusarium spp. Pathogenicity tests proved that, Fusarium solani from El-Gharbeia followed by A. tenusinium from El-Giza were the most frequent isolates responsible for rot of cucumber fruits from plastic greenhouse cultivation. Moreover, the most frequent isolates causing postharvest disease of cucumber fruits of the open field were Galactomyces candidium from El-Giza followed by Geotrichum sp. and F. fujikuroi from El-Gharbeia Governorates, respectively. This is the first report of several fungi causing postharvest fruit rot disease of cucumber i.e., G. candidium, Geotrichum sp., A. tenusinium, P. alli and Fusarium spp. (F. fujikuroi, F. verticiolides, F. solani, F. geraminearium and Fusarium incarnatum). Fungal isolates were identified according to cultural, morphological and molecular characterization based on sequencing of internal transcribed spacer1 (ITS1). All the ITS nucleotide sequences of fungi were applied and conserved in GenBank.
