Opis:
A new method facilitating the identification of the two most common alleles (A and B) of the bovine beta-lacoglobulin (LGB) gene is described. The method is based on two steps: PCR amplification of 240 bp fragment of LGB gene followed by the single stranded conformation polymorphism (SSCP) detection. AA, AB and BB genotypes of LGB were identified with this technique. The PCR-SSCP is simple, accurate and relatively inexpensive. Additionally, this method has a potential to detect new variants within the amplified gene fragment.

In the research carried out in the years 1999-2001 the following characteristics of three edible potato cultivars were determined: tuber yield, starch content, dry matter content, vitamin C content and starch yield. Genotype-environment interaction for the above characteristics was evaluated on the basis of main effects (genotypes and years) and interaction effects by means of the variance analysis for repeated experiments and according to the linear model for the design which confounds the type 33 interaction. Moreover, there were calculated the variance of genotype stability describing the environmental variation of the genotypes, and the ecovalence coefficient which describes the genotype x environment interaction. All the analysed characteristics were influenced by the cultivars (genotypes) and the environment. The genotype-environment interaction reflecting different response of the cultivars to changeable environmental conditions proved to be significant for all the examined characteristics. The analysis of results does not allow to unambiguously indicate the stabile genotype because the Muza cultivar was most stable for vitamin C and dry matter contents, and starch yield whereas Aster and Ania proved to be most stable as far as tuber yield and starch content, respectively, were concerned...

This study was carried out with the objective to estimate the genotypic variability and other yield related traits of Ethiopian mustard in North West Ethiopia. A total of 36 genotypes of Ethiopian mustard were considered for this study. Analysis of variance was computed to contrast the variability within the collected genotypes based on yield and other yield related traits. The results revealed highly significant values(p<0.01) for days to maturity, grain filling period, number of pod per plot, secondary branches per plant, harvest index, seed yield per plot, seed yield per hectare and oil content. Significant differences (p<0.05) were noted for days to flowering, plant height, primary branch per plant, biomass per plot, oil yield per plot differences among the genotypes. Genotypic coefficient of variation (GCV %) was lower than phenotypic coefficient of variation (PCV %) for all the traits studied. High genetic advance with heritability was observed in the following characters; plant height, biomass of the plant, number of secondary branch per plant and grain filling period. There are variations in the extent of genetic variability, heritability and genetic advance of traits which can facilitate selection for further improvement of important traits of Ethiopian mustard. Therefore, it can be concluded that the variability within Ethiopian mustard genotypes collected from different areas of northern Ethiopia is high and vital for better crop improvement.

Damping-off disease caused by Pythium ultimum can kill both germinating seeds and young seedlings and cause considerable damage in safflower cultivation. An estimation of heritability lets safflower breeders determine the most effective method for improving seedling emergence in soils  infected  with P. ultimum, the causal agent of seed rot and damping-off. Two cycles of selection were performed to estimate the realized heritability of resistance to the pathogen in five safflower populations. Undamaged seedlings were selected as resistant individuals and were kept to produce seed. The results showed that selection for two consecutive generations increased the emergence of seedlings in Pythium-infected soil from 46 to 53 %. The heritability estimates varied between 1.72 and 77.66 % over the genotypes and environments, in inverse proportion to the severity of the disease. Estimates of heritabilities showed that genes conferring resistance to P. ultimum in safflower are highly heritable and would respond to selection breeding, particularly in some of the studied genotypes, like Isfahan and Zarghan259. However,  different breeding methods must be explored for other genotypes.

The Katowice conurbation took shape from the end of the 18th century in the borderlands of two states, Poland and Germany. In this sense, it was an area on both their peripheries. Permanent and dynamic interactions between different national-ethnic groups resulted in cultural intermingling. From the middle of the twentieth century, this complex settlement system was already at the centre of socio-economic development on a supra-regional scale. As a result of multidirectional migration, influenced by intensive industrialisation, social relations evolved between the migrants and the local (indigenous) population, as well as between the migrants themselves. The aim of this paper is to analyse the early multicultural nature of selected cities in the Katowice conurbation against a methodological and terminological background and to indicate the extent to which this feature might be an asset in present day socio-economic transformations in this area. The temporal and spatial overlapping of various origins of socio-cultural links and relations leads the authors to formulate the concepts of cultural genotype and primary and secondary cultural genotype, and to exemplify the formation of cultural genotypes in the Katowice conurbation.

The studFigy aims to determine the TNF-α single-nucleotide polymorphism TNF-α (-308) and assess the association of TNF-a(-308) SNP with the risk of silicosis among workers directly exposed to silica dust in Vietnam. A study was undertaken among 78 cases with silicosis and 103 controls without silicosis in Vietnam. Blood samples were collected for genomic DNA extraction from each subject. The phenotyping of TNF-α(-308) was performed using polymerase chain reaction‐based restriction fragment length polymorphism (PCR‐RFLP) and dye termination sequencing. Results: The average exposure time of the case group was slightly higher than that of the control group (12.46 ± 6.732 years vs. 12.09 ± 7.854 years). The majority of genotypes in both silicosis and non-silicosis was GG. When analyzing the concentration of TNF-α in the study participants' blood, it is shown that the average concentration of TNFα in the case group was higher than that in the control group. The genotype AG in the case group was 1.368 times higher than that in the control group. The percentage of all A alleles in the case group with silicosis was 1.342 times higher than the control group without the disease, similar to previous studies. Conclusion: The majority of genotypes in both groups was GG. The average concentration of TNF-α in blood, genotype AG, and the percentage of all A alleles in the case group was higher than that in the control group.

The aim of this study was to establish the effect of the genotype on performance of chickens. The experimental material included 1,320 slow-growing chickens with two genotypes Hubbard JA 957 - available on the Polish market hybrid with a declared slow growth and non-commercial hybrid CxGP, for creation of which a light native breed Greenleg Partridge hens were used. Chickens were maintained up to 63 days of age. During the experiment body weight, feed intake and health of the birds were under control. Based on the observations the growth rate and the feed conversion ratio (FCR) were determined. It was found, that chickens CxGP were characterized by lower body weight, slower rate of growth and higher feed conversion ratio compared with the chickens Hubbard JA 957, but chickens CxGP had a higher health.
Genotyp - czynnik warunkujący wyniki produkcyjne w stadach kurcząt rzeźnych. Celem badania było określenie wpływu genotypu na wyniki produkcyjne kurcząt. Badania przeprowadzono na 1320 kurczętach o dwóch genotypach Hubbard JA 957 - dostępnym na rynku polskim mieszaniec o deklarowanym wolniejszym tempie wzrostu oraz CxZk - mieszaniec, do którego wytworzenia wykorzystano kury rasy lekkiej zielononóżki kuropatwianej. Kurczęta utrzymywano do 63. dnia życia. W czasie odchowu kontrolowano masę ciała, spożycie paszy i zdrowotność ptaków. Na podstawie prowadzonych obserwacji określono tempo wzrostu oraz wskaźnik zużycia paszy. Stwierdzono, że kurczęta CxZk charakteryzowała mniejsza masa ciała w porównaniu z kurczętami Hubbard JA 957 oraz wolniejsze tempo wzrostu. Mniejsze wykorzystanie paszy na przyrost masy ciała stwierdzono dla kurcząt Hubbard JA 957. Kurczęta CxZk charakteryzowała natomiast wyższa zdrowotność.

The objective of this discussion is an attempt to recall that already in Polish modernism, in the work of Tadeusz Miciński (1873-1918), the idea of modifying the human phenotype was signaled. Contemporary times offer the possibility of its partial (for the time being) realization. That is why it is worth confronting this idea with the foundations of religious ethics represented by the teaching of the Roman Catholic Church. It is also worth recalling the concerns that Jürgen Habermas (b. 1929) raises about the possibility of the unrestrained development of genetic engineering. Being a secular thinker, the German philosopher analyzes the ethical dangers of liberal eugenics and, in this way, opens up the search for a consensus with religious ethics.

Introduction: This research aims to detect possible associations between the ACTN3 R577X, ACE I/D, BDKRB2 +9/-9 genetic polymorphisms, and selected anaerobic performance indicators among elite and sub-elite fencers. Methods: The sample of participants included a group of 20 fencers (males, age 25.5 ± 6.9 years; height 185.1 ± 5.8; weight 78.3 ± 9.8). We obtained genotype analysis for selected polymorphisms (ACTN3 R577X, ACE I/D, BDKRB2 +9/-9) through buccal swabs. 30-second Wingate test was used for the anaerobic performance where the following variables were monitored: the maximal anaerobic performance - Pmax [W], the anaerobic capacity - AnC [kJ], the total number of revolutions - TR [n], the peak blood lactate concentration - LApeak [mmol·l-1]. Results: Within ACTN3 R577X polymorphism, no differences were found in the observed variables between genotypes. However, ACE I/D heterozygotes reached higher Pmax, AnC, and LApeak values than homozygotes. Furthermore, BDKRB2 +9/-9 polymorphism homozygotes with -9 allele reached higher Pmax and AnC values than homozygotes with +9 allele. No statistical differences were found in all monitored variables between genotypes of monitored polymorphisms. Conclusions: The research findings might serve as another useful piece of knowledge related to the existing statements in the field of genetics, focusing on speed-strength performance problems. It should be highlighted that current studies are only the first steps in helping us better understand the associations between genetic factors and performance in sports.

Celem pracy było zbadanie wpływu pochodzenia lisów polarnych na cechy fizyczne włosów pokrywowych i gęstość okrywy włosowej. Materiał do badań stanowiły próbki włosów pobrane od samic lisa polarnego odmiany: polskiej, duńskiej, fińskiej oraz czterech grup mieszańców tych odmian. Próbki włosów z okrywy zimowej pobierano przyżyciowo z partii grzbietowej. W każdej próbce policzono wszystkie włosy pokrywowe i podszyciowe oraz określono stosunek między nimi. Na włosach wykonano pomiary ich długości, wytrzymałości i wydłużenia. Pochodzenie lisów nie wpływało istotnie na cechy fizyczne włosów pokrywowych. Udział genów odmiany fińskiej wpłynął pozytywnie na jakość okrywy włosowej mieszańców. Natomiast udział genów odmiany duńskiej nie miał tak pozytywnego wpływu na poprawę cech okrywy włosowej.
The aim of this study was to asses the influence of arctic fox origin on coat hair physical traits and density of cover. The hair samples taken from arctic fox vixens of varieties: Polish, Danish, Finish and four groups of crossbreds of these varieties were studies. The hair samples were taken from the back of live animals in winter coat. In every hair sample coat and undercoat hairs were counted and the ratios were determined. The length, elongation and strength of hair were measured. The origin of the foxes did not influence significantly physical traits of coat hair. Crossbreds after Finish foxes had cover of better quality. Such a positive effect was not observed in case of crossbreds after Danish foxes, however.

Opis:
Strains of the genus Acanthamoeba are usually assigned to sequence types or genotypes according to pair-wise similarity values of the nuclear gene for the small subunit of ribosomal RNA. This classification system was established by comparing full or nearly full gene sequences, > 2000 bp. For practical reasons, diagnostic fragments of smaller lengths have been identified and used for rapid and economic identification of large number of strains. While the use of these small fragments in diagnostics applications remains valid when and only if the reference full sequence-type is available, we contest their use to identify and describe new genotypes. We report herein the case of a new genotype described on the basis of solely a small partial sequence and discuss the poor reliability of this fragment to correctly infer phylogenetic relationships, and its limits in the description of new genotypes of Acanthamoeba.
