Opis:
Three populations of Norway spruce from ecologically extreme environments in Slovenia and Bosnia and Herzegovina were examined for genetic polymorphism. The spruces there grow in specific forest communities (Sphagno-Piceetum) which represent the remnants of the post-glacial vegetation. The aim of the study was to search for similarities in the genetic variation among populations adapted to such conditions. In total, 10 isoenzyme systems involving 16 gene loci were analysed. The results showed differences in genetic differentiation at loci Got-B, Skdh-A and 6-Pgdh-C between the two Slovenian populations and the Bosnian population, but also indicated an interestingly close relationship between the Slovenian population Pohorje and the Bosnian population Nišići.

Pinus sylvestris (Scots pine) and taxa from the P. mugo (mountain pine) complex hybridize in contact zones producing morphologically-intermediate fertile hybrids. However, the hybrid specimens sometimes express only the P. sylvestris phenotype. Such cryptic hybrids were detected among P. sylvestris and P. uliginosa in the western part of Błędne Skały in the Stołowe Mountains, where the pines grow on the tops of sandstone rocks and phenotypically resemble P. sylvestris, P. uliginosa, and P. mugo. Hybrids with the P. sylvestris phenotype could be potentially present in other relic populations of this species in these mountains. During the present study, the hybrids were identified only in the area of Błędne Skały based on chloroplast and mitochondrial markers, morphological differentiation of various needle and cone traits, and phenotype assessments of the trees during sampling. these hybrids included three cryptic hybrids of P. sylvestris × P. mugo with the P. sylvestris phenotype and one displaying the phenotype of P. uliginosa. The other populations analyzed represented Scots pine with no evidence of hybridization with P. uliginosa and/or P. mugo. Biometric data on the cone and needle morphology also suggest possible hybridization within the P. mugo complex on Błędne Skały. The results indicate that hybridization takes place in this population but not in neighboring populations despite the possible connection by pollen-mediated gene flow.

Divergence in genetic as well as phenotypic structures can be expected in species with disjunctive geographic ranges and restricted gene flow among isolated populations. Dwarf mountain pine has such a disjunctive geographic range in the mountains of Central Europe. We hypothesised that populations of Pinus mugo from the Giant Mts. differ from Alpine and Carpathian populations to a greater extent than differentiation within these regions; furthermore, these differences would be detectable at both the genetic and phenotypic levels. To verify this hypothesis, the diversity and differentiation within and among eleven populations from the Giant Mts., Carpathians and Alps were analysed using 19 isozyme isozyme loci, 17 needle and 15 cone morphological characters. Moreover, the data on 10 chloroplast microsatellites used in the previous study, were reanalysed. The differences between the three regions were greater than among populations within them. The microsatellites and isozymes clearly differentiated between regions, while in the multivariate analyses of cone and needle characters the Alpine and Carpathian populations were intermingled but distinct from those sampled in the Giant Mts. The significant genetic structuring among regions may result from an ancient fragmentation and long lasting geographic isolation between the Giant Mts., Alps and Tatras. The populations from the Giant Mts., the northernmost within the geographic range of P. mugo, presented lower level of genetic variation then those from the Alps and Carpathians. The pattern of genetic structure observed in dwarf mountain pine may be characteristic of wind-pollinated trees with a disjunctive geographic distribution

Three diploid Trifolium (Fabaceae) species, T. campestre, T.fragiferum and T. montanum, were electrophoretically assayed to elucidate the range and organization of genetic variation in relation to the mating system, reproduction mode and longevity of the species. T. campestre is an annual self-pollinated species. T.fragiferum and T. montanum are cross-pollinated perennials. The former species reproduces both sexually and vegetatively, whereas the latter relies solely on sexual reproduction. Several populations of each species collected throughout Poland were surveyed for 15 enzymes. The measures of variation included: proportion of polymorphic loci per population, average number of alleles per locus, average gene diversity (He) and genetic distance (D). To describe the organization of diversity in each species, total genetic variation per locus (Ht) was calculated and partitioned into intra-(Hs) and inter-(Dst) populational components. Both cross-pollinated species had more polymorphic loci, higher numbers of alleles per locus and higher level of diversity than self-pollinated T. campestre. Most of genetic variation of the latter species was allocated among populations unlike in T.fragiferum and T. montanum.

Opis:
The flowering and graft mortality of Norway spruce clones of Istebna origin was only slightly different from clones of other origin in the same seed orchard. The negative effect ofmoving Istebna clones from the south to the north of their origin was probably compensated by moving them from a higher altitude to lowland. There is a danger that the complete loss of several clones, observed in the seed orchard with time,may really decrease the expected level of genetic variation in the progeny. It is recommended that the mortality of grafts and the reduction in the number of clones in the seed orchard should be under permanent observation.

Field elm (Ulmus minor Mill.) is distributed mainly across central and southern Europe. In Poland this species occurs in the lowlands and foothills, where it grows mainly in the floodplain forests along the rivers. U. minor exists in a variety of climatic and ecological conditions. It is capable to tolerate floods as well as drought. Currently, most populations of U. minor are small and fragmented resulting from human activity and Dutch elm disease. Moreover, in the natural field elm populations, vegetative propagation by root suckers or sprouting can be observed. All these factors may affect the level of genetic variation of U. minor populations in Poland. In the present study, we determined the level of genetic variation and the clonal diversity of twelve natural U. minor populations in Poland (407 individuals) using eight nuclear microsatellite loci. The obtained results indicate that the studied field elm populations are characterized by low level of genetic variation (He=0.382; Ho=0.555; A=7.0). Additionally, the high level of clonality in field elm populations was estimated. The clonality level of examined elm populations varied among them, and in some cases was very high. Out of the 407 individuals analysed for clonal structure only 61 multilocus genotypes were identified. Furthermore, only one genotype was identified in the three study populations of field elm, which means that in each of these populations all trees belong to one genet. The values of genotypic richness (R) were heterogeneous among populations, with mean 0.148. The knowledge on the genetic diversity and the clonal structure of U. minor populations is essential to make future decisions regarding conservation of genetic resources of this species in Poland.

Wych elm (Ulmus glabra Huds.) is a rare scattered forest tree species in Poland. First studies on genetic diversity of the species in its natural range in Poland give a good basis for the management and conservation of its genetic resources. In the present study, we determined the level of genetic variation and genetic differentiation of seventeen natural Wych elm populations in Poland using nine nuclear microsatellite loci. The number of analyzed individuals was 601. The level of genetic diversity of Polish populations of Wych elm corresponded with the results of the previous studies on this species in Europe. The populations show quite low level of genetic diversity of the species on the population level and quite high diversity on the interpopulation level. A total of 119 alleles was found, with average number per locus (A) equal 6.0 and allelic richness at medium high level (AR10=4.7). Observed (Ho) and expected (He) heterozygosity reached 0.583 and 0.602 respectively. The genetic differentiation between Polish populations of Wych elm occurred at a low level (Fst=0.089). Inbreeding depression may occur in the next generations (Fis=0.031). Genetic diversity between Wych elm populations in Poland may be the result of unfavorable random processes related to the reduction of population size resulting from elm disease. To better understand the processes related to the genetic diversity of Wych elm populations, research on field elm variation should be undertaken. Such studies may give an answer to the potential influence of introgression between these species on the genetic structure of the Wych elm. Both in situ as well as ex situ conservation measures are highly recommended to preserve genetic resources of this valuable noble hardwood species in Polish landscape.

Deschampsia antarctica Desv. plants collected on King George Islands (Antarctica) at two localities that differ in topographic and nutrition conditions exhibited morphological variation that differentiated plants of both locations. The molecular variation characteristic to individuals of both sites was tested using AFLP approach in order to verify whether morphological variation characteristic to the plants resulted from environmental factors or possibly from differences at the DNA level. Four primer pair combinations were used to generate 339 AFLP fragments, 132 of which were polymorphic and allowed evaluation of genetic relationships among D. antarctica individuals. Chi-square test revealed that only 12 signals were discriminative for the plants from both locations. Cluster analysis conducted on these AFLP fragments demonstrated that plants from the location rich in biogenes were more polymorphic than those from poor one. Our data suggest that the phenotypic variation specific to plants of both locations seem to be the result of adaptation to the environmental conditions like soil and moisture rather than reflect genetic differences.

Cochlearia polonica Fröhl. (Brassicaceae) is one of the rarest species in the Polish and European flora and a taxon endemic to a very small area in southern Poland. Due to industrial activities and subsequent transformation of habitats it was extinct in all natural localities around 1994. The persistence of the species was ensured thanks to the active protection efforts including a series of transplantations based on the material from the last and decreasing natural population. The history of conservation efforts of C. polonica provides a model example of successful active protection in the European flora. Here, we provide a complete review comprising the following aims: (i) outline of the discovery and taxonomic conceptions on C. polonica, (ii) review of conservation efforts aimed at preserving its populations, (iii) description of the existing population resources, and (iv) analysis of the genetic structure of all existing populations based on previously published data and new, supplementary results.

W artykule omówiono zmiany, jakim podlegają patogeny zwierzęce, które na drodze ewolucji stały się chorobotwórcze dla ludzi. Ryzyko zakażenia dotyczy szczególnie osób mających bezpośredni kontakt ze zwierzętami w ramach obowiązków zawodowych – weterynarzy, hodowców, osób pracujących w zakładach zajmujących się ubojem lub przetwórstwem surowców pochodzenia zwierzęcego. W artykule przedstawiono drobnoustroje wskazywane w raportach epidemiologicznych Europejskiego Centrum Zapobiegania i Kontroli Chorób (European Centre for Disease Prevention and Control, ECDC), które od 19 lat gromadzi i publikuje w postaci raportów dane na temat zoonoz ze wszystkich krajów członkowskich Unii Europejskiej. Obecnie ECDC monitoruje występowanie chorób wywoływanych przez 11, uznanych za najważniejsze, czynników etiologicznych zoonoz, z których 7 stanowią patogeny bakteryjne: Salmonella spp., Campylobacter spp., Listeria monocytogenes, Mycobacterium bovis, Brucella spp., Coxiella burnetti oraz Verotoxin-producing E. coli (VTEC) / Shiga-like toxin-producing E. coli (STEC). Za szczególnie ważne uważa się te, które mogą rozprzestrzeniać się za pośrednictwem żywności. W artykule uwzględniono także wybrane nowe zagrożenia, niesione przez gatunki jeszcze niemonitorowane – paciorkowce Streptococcus iniae, S. suis i S. dysgalactiae oraz gronkowce Staphylococcus intermedius i S. pseudintermedius. Drobnoustroje te w ciągu ostatnich lat przełamały barierę międzygatunkową. Przyczyny tego zjawiska nie są znane, ale wiąże się je ze zmiennością drobnoustrojów oraz ich adaptacją do życia w zmieniającym się środowisku, co wynika z łatwości rearanżacji DNA i poziomego transferu genów. Skalę problemu nowych infekcji odzwierzęcych odzwierciedla istotny wzrost liczby publikacji w tej dziedzinie w ostatnich latach. Med. Pr. 2014;65(6):819–829
This article presents the problem of evolutionary changes of zoonotic pathogens responsible for human diseases. Everyone is exposed to the risk of zoonotic infection, particularly employees having direct contact with animals, i.e. veterinarians, breeders, butchers and workers of animal products’ processing industry. The article focuses on pathogens monitored by the European Centre for Disease Prevention and Control (ECDC), which has been collecting statistical data on zoonoses from all European Union countries for 19 years and publishing collected data in annual epidemiological reports. Currently, the most important 11 pathogens responsible for causing human zoonotic diseases are being monitored, of which seven are bacteria: Salmonella spp., Campylobacter spp., Listeria monocytogenes, Mycobacterium bovis, Brucella spp., Coxiella burnetti and Verotoxin- producing E. coli (VTEC) / Shiga-like toxin producing E. coli (STEC). As particularly important are considered foodborne pathogens. The article also includes new emerging zoonotic bacteria, which are not currently monitored by ECDC but might pose a serious epidemiological problem in a foreseeable future: Streptococcus iniae, S. suis, S. dysgalactiae and staphylococci: Staphylococcus intermedius, S. pseudintermedius. Those species have just crossed the animal-human interspecies barrier. The exact mechanism of this phenomenon remains unknown, it is connected, however, with genetic variability, capability to survive in changing environment. These abilities derive from DNA rearrangement and horizontal gene transfer between bacterial cells. The genetic similarity of three species: Septoria tritici, Stagonospora nodorum and Stagonospora avenae f. sp. triticea - important pathogens in many cereal production areas worldwide was assessed by random amplified polymorphic DNA (RAPD) assay. In preliminary research DNA of 14, 9, and 7 monopyenidios- pore isolates of S. nodorum, S. tritici, and S. a. tritícea, respectively, were amplified by PCR with four primers. Afterwards the research was focused on three mono- pyenidiospore isolates from each species studied. The isolates of each species selected for the study varied in pathogenicity and were diverse geographically. PCR with the set of 14 selected primers resulted in 99 different bands, ranged from 180 to 2500 base pairs in length. Most primers in PCR (especially RAD11, RAD31, RAD32, RAD33) revealed uniform bands for isolates of S. a. tritícea, that allow to identify this species among the others. The cluster analysis using Unweighed Pair-Group Method with Averaging (UPGMA) revealed interspecies disagreement among the isolates ranging from 32 to 53%. The intraspecies disagreement ranges were 17-20%, 38-43%, 42-44% for S. avenae f. sp. triticea, S. nodorum and S. tritici, respectively. Cluster analysis classified isolates into three homogeneous clusters. Each cluster grouped isolates of one species according to their current taxonomie ranks based on spore size, colony morphology and host ranges. In addition, two of the clusters represented by isolates of S. nodorum and S. a. tritícea were distinctly separated at a lower linkage distance from the third one comprising isolates of S. tritici. A slight inconsistency found in grouping some isolates indicates that such groupings should be done with caution. The present study indicates that the PCR- RAPD assay is of a potential use in taxonomy of Stagonospora spp. and Septoria tritici as well as in molecular identification of casual disease agents.

Opis:
The genus Nicotiana is one of the largest in the Solanaceae family and includes more than 80 species. The most well-known and widespread species of the genus Nicotiana is tobacco (Nicotiana tabacum), within which there are numerous cultivars. Tobacco is one of the most important industrial plants in Poland and worldwide. The great diversity within the genus makes it an excellent source of variation in a narrowing gene pool and can be used in breeding programmes. Studies of Nicotiana species also concern mechanisms of polyploidisation and evolution. There are also model species within the genus. However, in order to make full use of the collected germplasm resources, a detailed knowledge of the collection materials is necessary. While there are various reports in the international literature describing specific issues, the aim of this paper is to indicate the diversity of species in the genus Nicotiana as a whole on the basis of our own research and available studies. This review covers the characterisation of the genus Nicotiana in terms of origin and geographical distribution, as well as cytogenetic and molecular differences between species. An important aspect is the presentation of the morphological diversity of Nicotiana accessions and the variation in the most important tobacco alkaloids. A very important issue is the resistance of Nicotiana species to bacterial, fungal and viral diseases, which allows their use in resistance breeding.
