Starch can be found in microalgae, the raw material for the third generation of bioethanol production. One of them is C. pyrenoidosa. This study was conducted to analyze the effect of α-amylase enzyme concentration on the glucose contents produced and the effects of fermentation time on bioethanol contents produced. The hydrolysis process of this study was conducted using an α-amylase enzyme produced by A. niger. Several analyses in this research were carried out, including the analysis of enzyme activity using the Fuwa method, the analysis of glucose levels from enzymatic hydrolysis using the DNS method, and the analysis of bioethanol contents using the density method and GC-MS. The highest glucose content was 0.67 mg/mL, which was obtained from the addition of 40% (v/w) α-amylase enzyme, and the yield of bioethanol content from the sample treated 40% (v/w) α-amylase enzyme and fermented for 9 days was the optimum, which produced 28.07% of bioethanol content.

Due to urban sprawl, the demand for land has increased for the purpose of construction. It is unlikely that soil available at different construction sites will be suitable for designed structures. For improving the load-bearing capacity of the soil, different soil binders are used, which are present in distinct states. In this review, the authors have collected details about various binders, which are generally used in the soil stabilization, and their effect as a binding agent on the soil. In this article, the authors tried to review different traditional binders. After studying various research articles, the authors found that lime, ground-granulated blast slag (GGBS) polypropylene, polyurethane grouting, and asphalt mix are frequently used binders. However, the authors also gathered information about the negative environmental impact of these traditional soil binders, which led to the need for alternatives to these commonly used soil binders. To diminish this issue, different alternate hydraulic and non-hydraulic binders are discussed. The authors found alternatives to cement and lime with the alkali-activated material consisting of Na2O and silica modulus and belite-calcium sulfoaluminate ferrite, which is also known as "Aether™." According to the research, both alternatives emit 20–30% less CO2 into the environment and also improve the compressive strength of the soil. The various studies promotes bitumen modification. Incorporating 20-mesh crumb rubber and bio-oil into the bitumen reduces its viscosity and improves its fatigue value. When waste oil is mixed with asphalt, it revitalizes the bitumen, improves fatigue resistance, and increases compressive strength. The soil particles treated by Eko soil are held together by enzymes, which give them the same strength as cement. Apart from that, low-carbon binders such as basic oxygen furnace slag, bamboo fiber, enzyme-based soil treatment, zebu manure for stabilization, and lignin-contained biofuels and coproducts are discussed. Replacing these traditional binders helps with energy savings. All waste products are recycled, and energy is saved by not manufacturing traditional binders. Additionally, energy is saved, which is required to avoid the detrimental effects of these conventional binders, making them energy-efficient alternate binders. The authors also summarize the methods used, impacts, and changes that occur in soil properties after using substitutes in place of traditional binders. From the review, the authors determined that different binders have various properties in terms of chemical and physical compositions, and they show different variations in terms of strength when added to soil with low bearing capacity or poor stability.

Opis:
The study focused to develop an enzyme hand peeling. The first stage of the investigation dependent on selecting the qualities that the cosmetic should have. When creating the product, the assumption was made that it would have an exfoliating, moisturizing, firming and even-toning effect. The peeling was developed in several stages starting with the consideration of the cosmetic formulation, the selection of suitable raw materials and the execution of the formulation according to the adopted method. For the preparation of the peeling, the emollients used were so-called oily emollients, including sweet almond oil, Shea butter, coconut oil, oil/water (O/W) emulsifier, stabilizers, moisturisers, rheology modifier, filler, active substances, preservative and solvent. The peeling was prepared on the principle of O/W emulsion. The formulation was made using the classic "hot" method. More than a dozen trials were performed to obtain a product with the desired properties. Subsequently, the peeling was subjected to sensory and hedonistic analysis. Its density, viscosity and stability were evaluated. The formulation was also tested for pH. The final scrub was characterized by an intense but pleasant fragrance, very easy spreading on the skin, light texture, pleasant sensation after use. Hedonistic research has shown that obtained enzyme peeling was fully acceptable in terms of application, color, consistency and feelings after use. The final cosmetic product was also characterised by poor moisturisation and too strong abrasiveness.

Arsenic is a prevalent and pervasive environmental contaminant with varied amounts in drinking water. Arsenic exposure causes cancer, cardiovascular, liver, nerve, and ophthalmic diseases. The current study aimed to find the best conditions for eliminating arsenic from simulated wastewater and their effect on biomarkers of hepatic in mice. Adsorption tests including pH, contact duration, Al-kheriat dosage, and arsenic concentrations were evaluated. Seventy-two healthy albino mice (male) were accidentally allocated into nine groups (n = 8), the first group was considered as healthy control, the second group (AL-Kheriat), and other groups received AL-Kheriat and arsenic 25, 50, 75, 100, 125, 150 and 175 mg/kg, respectively. Next 10 days, the following were examined: LD50 level, ALP (alkaline phosphatase), ALT (alanine aminotransferase), and AST (aspartate aminotransferase), besides the histological condition of the liver. The results showed that the best time for arsenic removal was 4 hours, pH 8, Alkheriat dose 1 gram, and 50 ppm of pollutants. The level of alkaline phosphatase ALP, alanine transaminase ALT, and aspartate transaminase AST was increased to 150.96 (U/L), 143.1(U/L), and 32.8(U/L), respectively, in Al-Khriet and arsenic exposed population than the healthy control group, When the appropriate dose of Al-Khriet and arsenic mixture is used, it can aid in the selection of a safe way of disposing of the adsorbed residue. Additionally, it can serve as a low-cost rodent pesticide, increasing the commercial viability of this removal strategy.

The structure-bioactivity relationship of Tremella fuciformis polysaccharides (TFPs) in anti-aging in vivo is rarely reported. In the present study, a purified TFP, named HM, mainly composed of mannose, fucose, xylose, and glucose in a molar ratio of 4.14:0.98:0.81:0.62, was obtained from the fruiting body of T. fuciformis. Subsequently, two differentially degraded TFPs, named MM and LM, respectively, were prepared by a combined method of ultrasonic irradiation (US) and H2O2 treatment. Their structural properties, scavenging activities against free radicals in vitro, and anti-aging effects on D-galactose-induced aging of mice were determined. The average molecular weight of HM, MM, and LM was 58.3×106, 4.68×106, and 3.14×105 Da, respectively. All three TFPs were devoid of triple helix conformation and exhibited concentration- and molecular weight-dependent scavenging activity against radicals. The TFPs markedly relieved skin aging, effectively attenuated oxidative stress, and significantly decreased inflammation in D-galactose-induced aging mice. MM exhibited the best anti-aging effect among the TFPs. Additionally, TFPs partially restored the alterations in pH and the total content of short-chain fatty acids (SCFAs) in the colon but exhibited various impacts on the content of the individual SCFAs. These findings would provide rational guidance for a better application of TFPs in anti-aging foods and expand our understanding of the structure-function relationship of mushroom polysaccharides.

Enzyme activities in soils are influenced by soil characteristics and pollutants and could be indicator of soil quality. This study was undertaken to determine the relationship between physicochemical characteristics, heavy metals contents and enzyme activities (EA) in agricultural soils in the Saiss plain (Morocco). The analysis of six agricultural soil samples (sites 1–6), collected from urban and periurban agricultural soils of Fez region, showed that soils are generally alkaline with high CaCO3 that rich 46%, EC (525–703 μS/cm), rich in organic matter (3.14–5.80%). The Cr, Cu, Ni, Pb and Zn concentrations in the studied area are generally greater than the Upper Continental Crust, with a deceasing order: Zn> Cr> Pb> Cu> Ni. Geo-accumulation index showed that soils are unpolluted to moderately polluted except site 5 and site 6 that are moderately to strongly polluted by respectively Cu, Pb, Zn and Pb. Potential ecological rіsk factors were below 40, which means low potential ecological rіsk except site 5 that has moderate potential ecological risk by Cu and Pb. Soil potential ecological risk indices were found <150 indicating low ecological risk. According to this index, Cu caused more serious pollution than the other elements. The enzyme actіvities of the sіx soil samples showed almost the similar ranges of values. These EA showed the highest values in site 5.The sensіtivity of soil enzyme to heavy metals were observed UREA>PHOS>GALA. Pearson’s correlation showed significant posіtive correlatіon between studied soil EA and between these EA and heavy metals (Cu, Pb and Zn), and significant negative correlatіon between EA and heavy metals (Cr and Ni). The results of this study enrich and provide data base of the impact of heavy metals on soil enzyme actіvities in agrіcultural soils in the Saiss plain.

Due to their nutritional and sensorial characteristics, tropical fruits like guava, mamey sapote, and passion fruit are regularly incorporated into daily diets. Their by-products, especially peels, are approximately 10 to 35% of their weight and possess an important content of bioactive compounds, such as dietary fiber and phenolics. The nutritional, technological, physio-functional properties and phenolic compound compositions of guava, mamey sapote, and passion fruit peels were studied. Peels had promising techno- and physio-functional characteristics, good dietary fiber contents (45.18-61.42 g/100 g), and phenolic profiles with ferulic acid, gallic acid, p-coumaric, and catechin as the main compounds. Peel powders also showed excellent DPPH radical scavenging activity (125.3–252.4 µmol TE/100 g) and Trolox equivalent antioxidant capacity, TEAC (369.2–656.8 µmol TE/100 g). The α-amylase and lipase inhibitory activity varied from 28.15 to 51.4% and 30.89 to 57.15%, respectively. Higher values of α-glucosidase inhibition capacity were found, ranging from 51.64 to 70.32%. The chemical composition and properties reported in the present work suggest that peel powders of these guava, mamey sapote, and passion fruit could be used as constituents in different foods, such as bakery and meat goods, with beneficial health effects like control of hyperglycemia, improved intestinal function, and control of overweight; however, more studies are necessary for animal models and humans to confirm these bioactivities conclusively.

This research aimed to investigate the potential screening of wood degrading fungi to degrade the textile dye. Nine fungi were selected based on their ability to remove the turquoise blue textile dye on liquid potatoes dextrose médium. The fungi which have a high color removal efficiency were identified and analyzed qualitatively in terms of the manganese peroxidase and lignin peroxidase enzymes and their enzyme activity was determined. The selected fungi were investigated for their ability to degrade turquoise blue under variations in pH, dye concentration, and contact time. The results showed that three of the nine strains of wood-degrading local fungi, identified as Trametes hirsuta, Microporus xanthopus, and Ganoderma applanatum, were able to degrade textile dye. The color removal efficiency of dye treatment was monitored under the initial pH, dye concentration, and contact time variation. The study analyzed that the optimal color removal efficiency of the turquoise blue textile dye with a concentration of 30 mg/L in an aqueous solution was achieved at 78.50 and 85.84% at pH 6 for Microporus xanthopus and Ganoderma applanatum, while the color removal efficiency was 82.17% at pH 5 for Trametes hirsuta with seven days of incubation time.

Nutritional value and chemical composition, including the content of vitamins, fatty acids, 5’-nucleotides and nucleosides and amino acids, as well as biological activities, including antioxidant, angiotensin converting enzyme (ACE) inhibitory and cytotoxic activity of black trumpet (Craterellus cornucopioides (L.) Pers.) were tested in vitro. C. cornucopioides was low in energy, fat and carbohydrate contents, but rich in dietary fibre, especially β-glucan as well as niacin and α-tocopherol. The content of essential and non-essential free amino acids was 1.49 and 5.48 mg/g dry weight (dw). The nucleosides and 5’-nucleotides were determined at 1.84 and 3.99 mg/g dw, respectively. The share of unsaturated fatty acids (UFAs) was 75.92% with oleic acid as the major UFA. Cyclohexane and dichloromethane extracts expressed significant cytotoxic activity against selected cell lines, human epithelial cervical cancer cells (HeLa), adenocarcinomic human alveolar basal epithelial cells (A549), colorectal cancer cells (LS174) and normal MRC-5 human embryonic lung fibroblast cells (IC50 of 78.3–155.6 μg/mL). ACE inhibitory activity of the aqueous extract was strong with an IC50 of 0.74 μg/mL. It can be concluded that black trumpet is a good source of nutrients, such as vitamins, dietary fibres, amino acids, nucleotides and fatty acids, which contribute to the overall nutritional value of this fungus with potential for ACE inhibitory activity and use in anti-hypertensive diet.

A novel avian orthoreovirus (N-ARV) variant characterized with obvious arthritis and synovial inflammation, was isolated from Shandong, China in May 2016. It caused chicken poor growth and enormous economic losses to the poultry industry of China. However, there are few effective methods for detecting the antibody levels of N-ARV. In this study, a viral structural protein σC was expressed using the prokaryotic expression vector pET32a (+). The target protein was obtained by inducing for 6 hours at an IPTG concentration of 0.6mM. The optimal dilution of the coating antigen and serum antibody were determined to be 1000 fold and 10 fold respectively. A specificity test showed that there was no positive reactivity between N-ARV and other pathogens, and when the positive serum was diluted 100 times detection results were still checkable. The repeatability of this method was determined by the inter assay and intra assay tests with variability ranging from 4.85% to 7.93%. In conclusion, this indirect enzyme linked immunosorbent assay (ELISA) will be useful for large-scale serological surveys and monitoring antibody levels in N-ARV infection.

The purpose of this study was to evaluate in detail both the in vivo and in vitro efficacy of the enzyme agents, ZYMOX® Plus Otic (ZYMOX-P), in the treatment of canine otitis externa (OE). Eight dogs with a diagnosis of non-seasonal severe chronic OE were recruited for the study. ZYMOX-P was administered for 2-4 weeks. The Otitis Index Score (OTIS3) and bacteria or yeast colony growth were measured. Also, minimum biofilm (BF) formation inhibition concentration (MBIC) and BF bactericidal concentration (BBC) were measured in vitro. OTIS3 showed a statistically significant reduction after treatment (88.2%, p<0.001; pre-treatment = 11.0 ± 0.9; post-treatment = 1.3 ± 0.4, mean ± SEM). The individual OTIS scores, erythema, edema, erosions/ ulcerations, exudate and pruritus showed significant reduction (85.7%, 95.7%, 83.3%, 80.0%, and 89.3%, respectively). Microscopic examination revealed the presence of BF exopolysaccharide in all 8 ear samples when stained with alcian blue. Seven of the 8 dogs (87.5%) showed a reduction in colony growth. ZYMOX-P was effective at 34-fold and 16-fold dilutions on MBIC and BBC, respectively. These findings indicate that ZYMOX-P has efficacy against BF-related infection and is beneficial when used for the management of canine OE.

The diastase, xylose, cellulase, pectinase, and lipase enzymes were used for the pre-treatment of fresh turmeric rhizome to obtain a higher yield by rapturing biological cells. The quantitative and qualitative analysis of turmeric oil was performed by GC–MS. The various influencing parameters for the extraction of turmeric oil such as an enzyme, incubation/pre-treatment time, distillation time have been studied in the present work. The obtained turmeric oil by enzymatic pretreatment process is richer in bioactive/medicinal components than in the other traditional methods. The maximum yield was obtained with cellulase enzyme, which is 25–27% higher than the yield obtained by the traditional hydro distillation process. The detailed qualitative and quantitative analyses are also presented. The present method can be considered energy-efficient, effective, economical, and eco-friendly.

Opis:
Although pear is one of the most preferred fruits globally due to its high nutrient content, its juice products have not received equal consumer acceptability due to their undesirable haziness and turbidity. However, enzymatic treatment of juice has been noted to improve the overall acceptability of juices better than traditional decantation, filtration, and heating methods, which are either expensive or detrimental to the product quality. In this study, pear juice was effectively clarified using partially purified polygalacturonase from an entomopathogenic fungal endophyte Beauveria bassiana SAN01. The optimization of the juice clarification process led to 137% improvement in clarification compared to control, under optimal conditions of 39.37 U/mL enzyme load, 36.87°C temperature and 2.75 h treatment time. Results also showed that the polygalacturonase treatment resulted in a significant decrease in viscosity and an increase in the pseudoplasticity of the pear juice as the rheological data of the juice was observed to fit adequately into the power law model (R2>0.9). Furthermore, the enzyme-assisted juice clarification was found to reduce browning index (-14.34%) and turbidity (-19.72%) while increasing the reducing sugar content (9.55%). Unlike some conventional juice treatments, the polygalacturonase treatment preserved the antioxidant potential and the total phenolic contents of the pear juice as no significant changes were observed after the treatment. This study thus demonstrates the efficacy of a fungal polygalacturonase in improving pear juice quality as well as the potential applicability of B. bassiana enzymes in the food industry.

Opis:
The essential oil of celery (Apium graveolens) is characterized by exceptionally high content of alkylphthalides. The mentioned compounds exhibit a number of biological effects (including hypotensive, lipid-lowering, neuroprotective, and cytotoxic) and are also responsible for distinctive aroma of the plant. In the current work, parameters of conventional hydrodistillation (HD) and simultaneous distillation-extraction (SDE) were optimized to obtain phthalide-enriched fractions of celery seeds. A positive correlation was shown between hydrodistillation time and improved essential oil and phthalide yields. The 6-h HD of comminuted seeds yielded essential oil (2.9%) with a higher total phthalide content (51%), as compared to the samples collected after 1.5-3.0 h, which gave 2.4-2.7% of oil containing 24.6-39.2% of total phthalides. The oil contained sedanenolide (36.7%), 3-n-butylphthalide (13.1%), and sedanolide (1.1%). A further increase in the total phthalide content was achieved by omitting the size reduction step prior to hydrodistillation (68.8%) and utilization of the salting-out effect (84.3%). Enzyme pretreatment had a negligible effect on essential oil and phthalide yields. The change of distillation mode from HD to SDE significantly increased the oil yield of whole seeds (from 2.8 to 5.8% for 6 h processing) while maintaining its high phthalide content (62.5%), which translated to an increase in the total phthalide yield from 19.4 to 36.0 g/kg.
