The cowpea aphid (Aphis craccivora Koch) is considered a serious insect pest attacking several crops. We carried out biochemical studies to elucidate the role of the metabolising enzymes in conferring resistance to thiamethoxam, in two strains (resistant and susceptible) of the cowpea aphid. Bioassay experiments showed that the thiamethoxam selected strain developed a 48 fold resistance after consecutive selection with thiamethoxam for 12 generations. This resistant strain also exhibited cross-resistance to the tested carbamates; pirimicarb and carbosulfan, organophosphorus (malathion, fenitrothion, and chlorpyrifos-methyl), and the neonicotinoid (acetamiprid). Synergism studies have indicated that S,S,S-tributyl phosphorotrithioate (DEF), a known inhibitor for esterases, increased thiamethoxam toxicity 5.58 times in the resistant strain compared with the susceptible strain. Moreover, the biochemical determination revealed that carboxylestersae activity was 30 times greater in the resistant strain than in the susceptible strain. In addition, the enzyme activity of glutathione S-transferase (GST) and mixed function oxidases (mfo) increased only in the resistant strain 3.7 and 2.7 times, respectively, in relation to the susceptible (the control). Generally, our results suggest that the higher activity of the detoxifying enzymes, particularly carboxylesterase, in the resistant strain of the cowpea aphid, apparently have a significant role in endowing resistance to thiamethoxam, although additional mechanisms may contribute.

In this work, approximate analytical solution is developed using differential transformation method for finding the molar concentration of the native and denatured enzyme in terms of second-order rate constant. Also, the obtained solutions are used to study the kinetics of thermal inactivation of enzyme as applied in biotechnology. The analytical solution was validated with numerical solution using fourth- order Runge-Kutta. Good agreement was established between the numerical and approximated analytical solutions.

Tintinnids (Ciliophora: Spirotricha: Tintinnina) are occasionally the dominant ciliates in the marine plankton. The tintinnid loricae are minute artworks fascinating scientists for more than 230 years, but their chemical composition remained unclear, viz., chitinous or proteinaceous substances were discussed. Since sedimenting loricae contribute to the flux of elements and organic compounds in the oceans, knowledge about their nature is necessary in assessing their ecological role. Previous techniques and new methods, e.g. enzymatic digestion and high-resolution transmission electron microscopy, are applied in the present study. A chitinous nature of the loricae is rejected by the Van-Wisselingh test and failure of chitinase digestion. Only proteins might show a resistance against strong hot bases (KOH at 160°C for ~ 40 min. in tintinnid loricae) similar to that of chitin. Actually, the presence of nitrogen in the EDX analyses and the digestion of at least some loricae by proteinase K strongly indicate a proteinaceous nature. Furthermore, the crystal lattice revealed by high-resolution TEM in Eutintinnus loricae is similar to the proteinaceous surface layer (S-layer) of archaea, and the striation recognizable in transverse sections of Eutintinnus loricae has a periodicity resembling that of the crystalline proteins in the extruded trichocysts of Paramecium and Frontonia. The proteolytic resistance of some loricae does not reject the idea of a proteinaceous nature, as proteins in S-layers of some archaea and in most naturally occurring prions show comparable reactions. The data from the present study and the literature indicate proteins in the loricae of thirteen genera. Differences in the proteolytic resistance and staining properties between genera and congeners are probably due to deviations in the protein composition and the additional substances, e.g. lipids, carbohydrates. At the present state of knowledge, correlations between lorica structure, wall texture, ultrastructure of the lorica forming granules, and the histochemical and enzymatic findings are not evident. Therefore, further studies are required to estimate the taxonomic significance of these features and the ecological role of sedimenting loricae.

This study was conducted to evaluate the effects of higher conductivity of nutrient solution created by nitrate or chloride salts of potassium and calcium on growth characteristics of pepper plants (Capsicum annuum var annuum) during four months of growth period. Two EC5 and EC8 dS/m of Hoagland nutrient solutions were prepared using various salt combinations, namely: KCl + CaCl2, KNO3 + CaNO3, and KNO3 + CaNO3 + NaCl. Hoagland nutrient solution with EC 1.8 dS/m served as a control. Higher conductivity treatments had different effects on pepper plant growth. The most significant reduction in growth parameters of plant height, shoot fresh weight, fruit yield and nutrients uptake was in plants treated with KCl + CaCl2 particularly at EC8. Application of KNO3 + CaNO3 particularly at EC5, showed no difference as compared to the control regarding many growth parameters. Application of KNO3 + CaNO3 at EC5 resulted in higher shoot fresh weight compared to the control. All salinity treatments, except from KNO3 + CaNO3 at EC5, reduced the fruit yield compared to the control. Treatments of KCl + CaCl2 and KNO3 + CaNO3 + NaCl particularly at EC8 of nutrient solution, resulted in higher leaf proline concentration, catalase and peroxidase enzymes activity compared to the control. Other conductivity treatments showed no difference in catalase or peroxidase enzymes activities. Significantly the lowest amount of leaf N, K, Mg and Ca was in KCl + CaCl2 at EC8. On the other hand, the highest leaf macronutrient concentrations were in KNO3 + CaNO3 at EC5 and/or EC8 that showed only higher leaf N and Ca values compared to the control. Leaf micronutrient concentrations were the highest in KNO3 + CaNO3 at EC5 that generally showed no difference with control plants. However, application of KCl + CaCl2 particularly at EC8 and to less extent KNO3 + CaNO3 + NaCl at EC8, reduced the leaf micronutrient concentrations. Application of KNO3 + CaNO3 at EC5 increased and KCl + CaCl2 or KNO3 + CaNO3 + NaCl at EC8 decreased the leaf Fe concentration compared to control plants.

Enzymes play a significant role in controlling the characteristics of various chemical and biochemical reactions. They act as catalysts that increase the rate of reaction without undergoing any change in quantity. Enzymatic reactions occur through the active sites, which combine with the substrates to form intermediate complexes, subsequently leading to products. An enzyme having two active sites can show cooperative phenomena. Against this background, an enzyme-kinetic mathematical model is formulated using fractional order derivatives. Optimal control mechanism has been incorporated into the fractional-order model system to maximize the product output. Euler-Lagrange optimality conditions are derived for the FOCP (fractional order control problem) using maximum principle. Numerical iterative schemes have been developed to solve the fractional order optimal control problem through Matlab.

Arsenic is a prevalent and pervasive environmental contaminant with varied amounts in drinking water. Arsenic exposure causes cancer, cardiovascular, liver, nerve, and ophthalmic diseases. The current study aimed to find the best conditions for eliminating arsenic from simulated wastewater and their effect on biomarkers of hepatic in mice. Adsorption tests including pH, contact duration, Al-kheriat dosage, and arsenic concentrations were evaluated. Seventy-two healthy albino mice (male) were accidentally allocated into nine groups (n = 8), the first group was considered as healthy control, the second group (AL-Kheriat), and other groups received AL-Kheriat and arsenic 25, 50, 75, 100, 125, 150 and 175 mg/kg, respectively. Next 10 days, the following were examined: LD50 level, ALP (alkaline phosphatase), ALT (alanine aminotransferase), and AST (aspartate aminotransferase), besides the histological condition of the liver. The results showed that the best time for arsenic removal was 4 hours, pH 8, Alkheriat dose 1 gram, and 50 ppm of pollutants. The level of alkaline phosphatase ALP, alanine transaminase ALT, and aspartate transaminase AST was increased to 150.96 (U/L), 143.1(U/L), and 32.8(U/L), respectively, in Al-Khriet and arsenic exposed population than the healthy control group, When the appropriate dose of Al-Khriet and arsenic mixture is used, it can aid in the selection of a safe way of disposing of the adsorbed residue. Additionally, it can serve as a low-cost rodent pesticide, increasing the commercial viability of this removal strategy.

Leaf scald, caused by the necrotrophic fungus Monographella albescens, is one of the main threats to rice (Oryza sativa L.) around the world. This disease decreases yields in rice by up to 30% because of dead leaf tissue, damaged seeds, and sterile flowers. Currently, there is limited knowledge about the molecular mechanisms involved in rice plant resistance against this pathogen. For this purpose, six commercial cultivars of rice were primarily screened for M. albescens infection and development. Dasht and Salari were found to be the most resistant and susceptible to M. albescens infection, respectively. The plants were kept in a greenhouse at 29 ± 2°C during the day and 26 ± 2°C at night with a relative air humidity of 85 ± 5%. Forty-five days after sowing, the plants with three biological replications were inoculated by transferring a PDA disc (0.3 cm2) containing M. albescens mycelia to the middle third of the 7th, 8th, and 9th completely open leaves. The leaves were collected 24, 48, 72, 96 and 120 hai. Leaf samples were also collected from the non-inoculated plants (0 h) to serve as controls. Real-time quantitative PCR (RT-qPCR) showed rapid induction and significant accumulation of jasmonic acid (JA) and ethylene (ET) responsive genes such as lipoxygenase (LOX), allene oxide synthase 2 (Aos2), jasmonic acid carboxyl methyltransferase 1 (JMT1) and ACC synthase 1 (ACS1) in the resistant Dasht cultivar after infection with M. albescens. Furthermore, the transcripts of salicylic acid (SA) responsive phenyl alanine ammonia lyase 1 (PAL1) and nonexpressor of pathogenesis-related genes 1 (NPR1) genes were induced in the incompatible interaction. The activities of the defense enzymes superoxide dismutase (SOD), peroxidase (POX) and glutathione reductase (GR) increased strongly in Dasht in response to M. albescens infection. In addition, there was an increase in the H2 O2 levels in the leaves of the Dasht cultivar during the infectious period of M. albescens associated with the enhancement of catalase (CAT) activity as well as higher levels of malondialdehyde (MDA). This is the first study on the interaction between rice and M. albescens at the molecular level. It can contribute to understanding how rice responds to pathogen infection, as well as assist with future research plans of molecular breeding regarding the tolerance to leaf scald disease.

The optimal methodology to prepare the novel modified enzyme, polymer-enzyme complex, was developed to give a high catalytic activity in aqueous solution. The non-covalent complexes of two different enzymes (horseradish peroxidase and glucose oxidase) were prepared with various molar ratios (nD/nE 0,05; 0,1; 1; 5; 10; 15; 20) by using 75kDa dextran. The thermal stabilities of the obtained complexes were evaluated with the activities determined at different temperatures (25, 30, 35, 40, 50, 60, 70, 80°C) applying 60 minutes incubation time for pH 7. The complexes with the molar ratio nD/nHRP: 10 and nD/nGOD: 5 showed the highest thermal stability. Its activity was very high (ca. 1,5-fold higher activity than pure enzyme for HRP-dextran complexes) and almost the same between applying one hour incubation time and without incubation, and could also be measured at high temperatures (70, 80 °C). We finally succeeded in preparing dextran-enzyme complexes which showed higher activity than pure enzyme in aqueos solution at all temperatures for pH 7. In addition, the mentioned complexes at pH 7 had very long storage lifetime compared to purified enzyme at +4 °C; which is considered as a good feature for the usage in practice.

Although pear is one of the most preferred fruits globally due to its high nutrient content, its juice products have not received equal consumer acceptability due to their undesirable haziness and turbidity. However, enzymatic treatment of juice has been noted to improve the overall acceptability of juices better than traditional decantation, filtration, and heating methods, which are either expensive or detrimental to the product quality. In this study, pear juice was effectively clarified using partially purified polygalacturonase from an entomopathogenic fungal endophyte Beauveria bassiana SAN01. The optimization of the juice clarification process led to 137% improvement in clarification compared to control, under optimal conditions of 39.37 U/mL enzyme load, 36.87°C temperature and 2.75 h treatment time. Results also showed that the polygalacturonase treatment resulted in a significant decrease in viscosity and an increase in the pseudoplasticity of the pear juice as the rheological data of the juice was observed to fit adequately into the power law model (R2>0.9). Furthermore, the enzyme-assisted juice clarification was found to reduce browning index (-14.34%) and turbidity (-19.72%) while increasing the reducing sugar content (9.55%). Unlike some conventional juice treatments, the polygalacturonase treatment preserved the antioxidant potential and the total phenolic contents of the pear juice as no significant changes were observed after the treatment. This study thus demonstrates the efficacy of a fungal polygalacturonase in improving pear juice quality as well as the potential applicability of B. bassiana enzymes in the food industry.

The use of sewage sludge in agriculture is known to be environmentally friendly as it is a practical way of recycling municipal waste. A short-term experiment was carried out to study the changes in soil chemical and microbiological properties influenced through the addition of sewage sludge along with saline irrigation under a pearl millet-wheat crop rotation. There were three irrigation (canal water (0.35 dS m-1), 8 and 10 dS m-1 electrical conductivity saline water) and five fertilizer treatments (control-F1, sewage sludge (5 t ha-1)-F2, sewage sludge (5 t ha-1) +50% recommended dose of fertilizer (RDF)-F3, sewage sludge (5 t ha-1)+75% RDF-F4 and RDF-F5). The results showed that soil organic carbon, and available nitrogen, and phosphorus were reduced significantly under saline conditions but there was an increase in available potassium with the increasing salinity levels of the irrigation water. A significant reduction in soil microbial biomass carbon and enzyme activities was caused by 8 and 10 dS m-1 as compared to 0.35 dS m-1 at different growth stages of crops. Treatment F4 attained the highest soil microbial activity at each crop growth stage by a significant margin among all of the fertilizer treatments, which is associated with a substantial build-up of organic carbon and available NPK in the soil. Soil microbial activities followed a particular trend: at 35 and 75 days after sowing>harvest>sowing in pearl millet and wheat, respectively. Sewage sludge dumping is a major problem in India, and the present study suggests that by applying sewage sludge, soil biological health is improved; therefore its use in agriculture is recommended to farmers.

Chitin deacetylase is the only known enzyme which is able to deacetylate N-acetyl-D-glucosamine units in chitin or chitosan chains. As chitin can hardly be dissolved in organic/inorganic solvents, new solvents are still searched. Ionic liquids are promising for that application and for homophase enzymatic deacetylation. The aim of the work was to investigate the influence of selected ionic liquids on activity of chitin deacetylase. It has been shown that [Amim] ionic liquids increase the activity of chitin deacetylase. The highest activity was observed for [Amim][Cl]. Ionic liquids with shorter (ethyl (C2)) and longer side chain (buthyl (C4)) only insignificantly influenced the activity of the enzyme. All tested ionic liquids with [Br] anion increased the activity of chitin deacetylase while the [Emim] and [Bmim] cation in combination with [Cl] anion inhibited the activity of the enzyme.

Opis:
Chromium is one of the most common toxic metals present in the environment that induces various toxic effects in plants. A pot experiment was conducted to determine the effects of chromium on germination percentage, seedling growth, chlorophyll ‘a’, ‘b’ and proline content of Sesbania sesban (L.) Merr. The seedlings were treated with different concentrations of control, 0.10, 0.25, 0.50, 0.75 and 1.00 g kg-1 of chromium. The parameters such as germination percentage, root and shoot length, seedling fresh weight dry weight, chlorophyll ‘a’, ‘b’ and proline content of leaves were measured. Our results indicated that a significant inhibitory effect was observed at all levels of chromium compared to control. Increasing the concentration of chromium to 1.00 g kg-1 showed a significant decrease in seed germination, shoot and root length, fresh weight, dry weight and chlorophyll ‘a’ and ‘b’ content of plant. While proline, catalase and peroxidase contents were increased by increasing Cr concentration. It was also noted that accumulation of chromium in the roots was much higher than the shoots of the seedlings under treatment.
