Opis:
Purpose: Today, Enterococcus species are one of the most frequent etiological agents in nosocomial infections. The aim of this study was to determine the susceptibility to antibiotics and the prevalence of high-level aminoglycoside resistance (HLAR) among Enterococcus strains. Materials and methods: The susceptibility of 85 isolates of Enterococcus (47 E. faecalis and 38 E. faecium) was determined using the disk diffusion method. The results were interpreted according to European Committee on Antimicrobial Suscepti-bility Testing (EUCAST) guidelines. PASW Statistics 17.0 was used for statistical analysis. Results: E. faecalis strains showed the highest susceptibility to ampicillin, tigecycline, vanco-mycin, imipenem, and linezolid and E. faecium to linezolid, tigecycline, and quinupristin/dalfopristin. Among all tested strains, high-level gentamicin resistance (HLGR) was found in 4% of E. faecalis and 8% of E. faecium strains, high-level strepto-mycin resistance (HLSR) in 45% and 42%, and HLAR in 50% and 32% of strains, respectively. HLGR was detected only in vancomycin-resistant Enterococcus (VRE)− strains (12%), while HLSR in 76.9% of VRE+ and 24% of VRE− strains, and HLAR in 23.1% of VRE+ and 64% of VRE− strains. The tested strains were also divided into two groups: HLSR+ and HLAR+. In both groups, statistically significant susceptibility differences (p<0.05) were found for ampicillin, imipenem and trimethoprim/sulfamethoxazole. The most frequent antibiotic resistance profile among E. faecalis strains was SR (resistance phenotype to strepto-mycin), and among E. faecium, AMPR, IMPR, CNR, SR, SXTR (ampicillin, imipenem, gentamicin, streptomycin, trimethoprim/sulfamethoxazole). Conclusions: This study showed the slowly increasing prevalence of HLAR and resistance to newer antibiotics (linezolid and tigecycline) among Enterococcus strains. It is necessary to search for new directions in the treatment of enterococcal infections.

Opis:
An increase in the antibiotic resistance among Enterococcus faecium strains has been observed worldwide. Moreover, this bacteria has the ability to produce several virulence factors and to form biofilm that plays an important role in human infections. This study was designed to compare the antibiotic resistance and the prevalence of genes encoding surface protein (esp), aggregation substance (as), surface adhesin (efaA), collagen adhesin (ace), gelatinase (gelE), and hialuronidase (hyl) between biofilm-producing and non-producing E. faecium strains. Therefore, ninety E. faecium clinical isolates were tested for biofilm-forming ability, and then were assigned to two groups: biofilm-positive (BIO+, n =70) and biofilm-negative (BIO-, n = 20). Comparison of these groups showed that BIO+ isolates were resistant to β-lactams, whereas 10% of BIO- strains were susceptible to ampicillin (statistically significant difference, p = 0.007) and 5% to imipenem. Linezolid and tigecycline were the only antibiotics active against all tested isolates. Analysis of the virulence factors revealed that ace, efaA, and gelE genes occurred more frequently in BIO- strains (ace in 50% BIO+ vs. 75% BIO-; efaA 44.3% vs. 85%; gelE 2.9% vs. 15%, respectively), while hyl gene appeared more frequently in BIO+ isolates (87.1% BIO+ vs. 65% BIO-). These differences were significant (p < 0.05). We concluded that BIO+ strains were more resistant to antibiotics than BIO- strains, but interestingly, BIO- isolates were characterized by possession of higher virulence capabilities.

Eighty enterococcal isolates (E. faecium, n=38, E. faecalis, n=42) from diseased farm animals (swine, cattle, poultry) in Lithuania have been studied for the prevalence of antibiotic resistance and for resistance and virulence genetic determinants. 86% of E. faecium and 71% of E. faecalis isolates were multidrug resistant (resistant to three or more unrelated antibiotics). Resistance to aminoglycosides, tetracycline and erythromycin was found most frequently in both species (61%, 69%) and was linked to aph(3’)-IIIa, aac(6’)-Ie-aph(2”)-Ia, ant(6)-Ia (aminoglycoside resistance), tetM, tetL (tetracycline resistance), ermA, ermB (erythromycin resistance) gene combinations, which were supplemented with chloramphenicol resistance genes catA7, catA8 (E. faecalis) and catA9 (E. faecium). All E. faecalis isolates harboured genes coding for virulence factors agg, esp, fsr gelE alone or in combinations with the high prevalence of esp gene in isolates from cattle (63%) and pigs (79%). The origin- dependent incidence of agg gene variants prgB and asp1 was observed. The results indicate the existence of a large pool of potentially virulent and multidrug resistant E. faecalis in diseased farm animals posing risk to humans.

The number of antibiotic resistant bacterial strains found in the human environment has been growing in recent years. Antibiotic resistance has long been linked exclusively with the hospital environment. However, according to the findings of various studies, food can also be a source of antibiotic resistant strains. The ability of Enterococcus species to survive in a broad range of adverse environments causes that these bacteria are present in nearly all kinds of food. In this review we aimed to summarize mechanisms of antibiotic resistance in enterococci, the role of mobile genetic elements played in spreading of the resistance and to characterize antibiotic resistance among enterococci from food, inclusive of ready-to-eat food.
W ostatnich latach obserwowany jest w otoczeniu człowieka systematyczny wzrost liczby szczepów antybiotykoopornych. Przez długi czas zjawisko antybiotykooporności wiązano jedynie ze środowiskiem szpitalnym. Jednakże badania wykazały m.in., że także żywność może być jednym ze źródeł rozprzestrzeniania szczepów opornych na antybiotyki. Zdolność paciorkowców z rodzaju Enterococcus do przeżywania szeregu niekorzystnych warunków powoduje, że są one obecne w niemal każdym rodzaju żywności. Celem niniejszego przeglądu było opisanie mechanizmów oporności u enterokoków, roli mobilnych elementów genetycznych w jej rozprzestrzenianiu oraz scharakteryzowanie antybiotykooporności u enterokoków izolowanych z żywności w tym żywności gotowej do spożycia.

Purpose: Enterococcus faecium strains have been reported worldwide as etiologic factors of many nosocomial infections, which are difficult to manage because of the constantly increasing resistance of these microorganisms to antibiotics and the ability to form biofilm. The aim of this study was to analyze the ability to produce a biofilm in E. faecium strains, depending on the patient’s clinical material. Materials and methods: Sixty-six E. faecium strains were investigated. Identification and susceptibility testing were conducted by the VITEK2 system. The ability to form biofilm was assessed by phenotypic methods. The presence of selected virulence genes was established by PCR followed by gel electrophoresis and sequencing. Results: Among the tested E. faecium isolates, 72.7% were biofilm-positive (BIO+) and 27.3% biofilm-negative (BIO-). Strains were collected mostly from rectal swabs (30.4%) and blood (18.3%). BIO+ strains from infections constituted 31.8% (52.4% isolated from blood) and from colonization 40.9% (48.2% from rectal swabs). 91.7% of the Blood Group strains and 68.5% of the Other Group strains produced biofilm. Strains from the Colonization Group produced biofilm in a proportion similar to the Infection Group (about 75%). There were no statistically significant differences in virulence and resistance, except for vancomycin (more resistant BIO+ Other than the BIO+ Blood Group, and more resistant BIO+ Colonization than BIO+ Infection Group) and teicoplanin (more resistant BIO+ Colonization than the BIO+ Infection Group). Conclusion: The majority of E. faecium isolates carries high levels of resistance to many antimicrobials, is well equipped with virulence genes, and possesses the ability to form biofilm.

This research work assessed the antibacterial properties of probiotic bacterial which was isolated from breast milk. The breast milk has a distinct amalgamation of minerals, proteins, lipids, carbohydrates and various vitamins that endorse the proper development, growth and immunity of the children. That’s the reason behind it’s consideration to be a comprehensive and inclusive food for new born babies. Furthermore, it is also abundant in various bioactive compounds which encourage the maturation of the immune system over and above develop body’s defense against infections. This research used a standard methodology to isolate the bacterial. In the midst of these bioactive agents, probiotic bacteria were properly isolated from human milk in this research work by means of selective MRS media. Five Lactobacillus spp. were isolated from every one of the three breast milk samples and two Enterococcus species, were observed as potential probiotics, and identified using morphological and biochemical tests which includes L. rhamnosus, L. plantarum, L. casei, L. fermentum, L. acidophilus, En. faecalis and En. faecium. The isolated bacteria were facultative anaerobic, catalase negative, gram positive and non-endospore forming. Sugar fermentation arrangements of equally isolated bacteria also significantly different. The adding up of breast milk probiotics to children formulas possibly will be an innovative substitute to mimic some of the purposeful consequences of human milk in children who are not breastfed.

Introduction and Objectives. Nosocomial infections, also known as hospital-acquired infections, has become one of the most important health problems in health care units worldwide. The presented study aims to determine the average amount of microorganism loads and to show that the atmospheres of the two hospitals can be a potential source regarding nosocomial infections. The effect of surface and floor disinfection processes in the two hospitals and the antibiotic susceptibility of the bacterial isolates were also evaluated. Materials and Methods. Microorganisms were isolated from air samples collected from different areas (patient wards, corridors, operating theatres and postoperative units) of the two hospitals in Izmir. Sampling was conducted between December 2006 – March 2007. Results. During the 3-month sampling period, the average number of live microorganisms in the air samples collected from second-class environments in the hospital 1 and the hospital 2 was found to be 224.44 and 536.66 cfu/m3, respectively. The average number of microorganisms in hospital 2 collected before the disinfection process was higher than those after the disinfection process. However, because of the closure of the air-conditioning system and the hepa filters after the disinfection process, this was reversed in hospital 1. In total, 54 and 42 isolates were obtained from hospital 1 and hospital 2, respectively. 49 isolates from hospital 1 and 35 isolates from hospital 2 were identified as Staphylacoccus sp. The remaining isolates were identified as Aerococcus sp. and Enterococcus sp. Pseudomonas sp. was not determined in the air samples of the two hospitals. Conclusions. It was detected that the microbial loads in the atmospheres of the two hospitals studied varied greatly depending on the number of people in the environment. As the results indicate, the total number of microorganisms in the atmospheres of operating theatres in both hospitals does not pose a threat according to the Air Microbe Index.

Opis:
Food preservation using high pressure is a promising technique in food industry as it offers numerous opportunities for developing new foods with extended shelf-life, high nutritional value and excellent organoleptic characteristics. High pressure is an alternative to thermal processing. The resistance of microorganisms to pressure varies considerably depending on the pressure range applied, temperature and treatment duration, and type of microorganism. Generally, Gram-positive bacteria are more resistant to pressure than Gram-negative bacteria, moulds and yeasts; the most resistant are bacterial spores. The nature of the food is also important, as it may contain substances which protect the microorganism from high pressure. This article presents results of our studies involving the effect of high pressure on survival of some pathogenic bacteria - Listeria monocytogenes, Aeromonas hydrophila and Enterococcus hirae - in artificially contaminated cooked ham, ripening hard cheese and fruit juices. The results indicate that in samples of investigated foods the number of these microorganisms decreased proportionally to the pressure used and the duration of treatment, and the effect of these two factors was statistically significant (level of probability, P ≤ 0.001). Enterococcus hirae is much more resistant to high pressure treatment than L. monocytogenes and A. hydrophila. Mathematical methods were applied, for accurate prediction of the effects of high pressure on microorganisms. The usefulness of high pressure treatment for inactivation of microorganisms and shelf-life extention of meat products was also evaluated. The results obtained show that high pressure treatment extends the shelf-life of cooked pork ham and raw smoked pork loin up to 8 weeks, ensuring good micro-biological and sensory quality of the products.

Enterokoki są dobrze przystosowane do funkcjonowania w środowisku sera i wraz z innymi bakteriami mlekowymi stanowią grupę niestarterowych bakterii mlekowych. Ich aktywność proteolityczna i lipolityczna przyczynia się do rozwoju typowych właściwości sensorycznych serów. Dzięki wytwarzaniu bakteriocyn enterokoki wpływają na mikrobiotę sera, z jednej strony ograniczając ewentualny rozwój drobnoustrojów patogennych i powodujących psucie a z drugiej strony stymulują procesy autolityczne innych bakterii mlekowych. Dzięki tym cechom drobnoustroje te mają potencjał do stosowania jako kultury dodatkowe lub ochronne do serów. Niektóre gatunki Enterococcus wykazują cechy wirulencji i antybiotykooporność i z tego powodu rodzaj ten nie posiada statusu QPS (qualified presumption of safety).
Enterococci are well adapted to function in the cheese environment and, along with other lactic acid bacteria, constitute a group of non-starter lactic bacteria. Their proteolytic and lipolytic activity contributes to the development of typical sensory properties of cheeses. By producing bacteriocins, enterococci influence the cheese microbiota, on the one hand, limiting the possible development of pathogenic and spoilage microorganisms, and, on the other hand, stimulating the autolytic processes of other lactic bacteria. Due to these characteristics, enterococci have the potential to be used as adjunct or protective cultures for cheeses. Some Enterococcus species show virulence and antibiotic resistance and therefore the genus does not have qualified presumption of safety status (QPS).

Opis:
The microbial assay for risk assessment (MARA) test was used for acute cesium toxicity evaluation in water solutions. The test contained 11 different microorganisms with a wide spectrum of sensitivity. The resistance of microorganisms to cesium was characterized as follows: microbial toxic concentration (MTC), half maximal inhibitory concentration (IC50), maximal inhibitory concentration (IC100). The sensitivity to cesium was characterized by the lowest observed effect level (LOEL). High levels of sensitivity in the range 3.1–6.3 mM were shown by the following microorganisms: Serratia rubidaea > Pseudomonas aurantiaca, Delftia acidovorans, Citrobacter freundii, Staphylococcus warneri. Lower levels of sensitivity (up to 16 mM) were noted for Comamonas testosteroni, Microbacterium species, Kurthia gibsonii, Pichia anomala, whereas that in the range 24–31 mM for Brevundimonas diminuta > Enterococcus casseliflavus. High resistance to Cs+ was found for E. casseliflavus (MTC 86.9 g/l) > the yeast – P. anomala (MTC 19.3 g/l) > K. gibsoni (MTC 17.4 g/l) > B. diminuta (MTC 13.4 g/l). The phenomenon of resistance of enterococcus and yeast strains was discussed.
