Opis:
Microcystis blooms and the related toxin known as microcystin-LR (MC-LR) put the safety of human water consumption and global irrigation practices in jeopardy. MC-LR is widely distributed in various environments, including water, sediments, plants, and other aquatic organisms. The use of water-containing microcystins for agricultural purposes may have to be restricted despite the limited availability of clean water resources. Accordingly, the present work aimed to determine the MC-LR concentrations and recognize the environmental parameters that initiate the growth of toxic cyanobacteria and MC-LR occurrence in 20 irrigation ponds in the Jordan Valley area. The irrigation ponds studied were found in a hypereutrophic condition, with high levels of N:P ratio and low transparency. These cause inseparable effects such as cyanobacterial bloom and MC-LR occurrence. The investigated ponds were classified as hypereutrophic according to General Quality Index (GQI), with two different types of algae covering the surface. The first was the Lemna sp. or duckweeds (Family Araceae) which are free-floating masses, and the second was the cyanobacteria algal bloom. Unpaired t-tests were performed and showed that the concentrations of MC-LR in pond water abundant with cyanobacteria algal bloom in September 2021 were significantly higher (P = 0.7906) than in June for the same year (0.3022 ± 0.0444 and 0.1048 ± 0.0171 ppb, respectively). Two methods for extracting MC-LR were used and showed a significant difference in MC-LR concentration in ponds with an abundance of cyanobacteria algal blooms (0.2273 ± 0.0356 ppb) compared to the ponds with an abundance of Lemna sp. or duckweeds collected in June 2021 (0.1048 ± 0.0171 ppb). Despite all of the efforts made by Jordan Valley farmers to prevent or limit the mass growth of cyanobacteria and its consequences for the eutrophication process in their irrigation ponds through the use of fish breading and chemicals such as copper sulfate, this environmental problem is still harming their crops and irrigation methods and requires immediate government assistance.

Phosphatized biomorphs, resembling modern and ancient bacteria, were identified for the first time in phosphate nodules, present at the base of the Bílá Hora Formation (uppermost Cenomanian - lower Turonian), and in phosphate coprolites at the base of the Teplice Formation (upper Turonian) in the Bohemian Cretaceous Basin. They are present in colonies as filaments, coccoids, strings, rods and outgrowths, associated with the phosphate as part of the rock constituents and display the characteristics of fossilized bacteria. Two types of bacteria were identified: chemotrophic, sulphur-reducing bacteria in the phosphate nodules and phototrophic cyanobacteria in the phosphate coprolites. Microanalysis of some of the fossil bacteria revealed a fluoride-rich calcium phosphate composition, compatible with the composition of bulk samples, in which carbonate-fluorapatite is the main mineral in the phosphate nodules and coprolites. The environmental indications of these fossil bacteria support the interpretation of an anoxic environment of phosphogenesis in the latest Cenomanian - earliest Turonian and variable redox conditions of coprolite phosphatization in the late Turonian. The potential microbial role in phosphogenesis in the former may have involved the suboxic breakdown of P-rich organic matter by sulphur-reducing bacteria and the release of phosphorus in the pore water, leading to the biochemical precipitation of phosphate. The latter involved initial P-storage by phototrophic bacteria in an oxic environment, followed by P-release below the sediment-water interface under suboxic conditions and subsequent phosphatization of the coprolites.

Cyanobacteria constitute a rich source of biologically active and structurally diverse compounds. The pharmacological potential of these compounds resides among others in their ability to control the proliferation and growth of cancer cell lines and potent disease-causing microbial agents. Despite recent scientific advances, the way these compounds interact with the body’s molecular structure are still unclear and science still has to discover how the cyanobacterial metabolites interact with cell structures and how cells react to them. In this project, we will study yet unexamined cyanobacterial metabolites, especially the compounds which act as chemical ligands for microRNA (miRNA) -binding sites, making them promising regulators (inhibitors) of gene networks that are involved in various diseases. We will first develop a stable cell line that constitutively expresses a unique miRNA reporter system. Then, we will conduct a screen on chemical compounds discovered in Baltic cyanobacteria to identify small molecules with inhibitory activity and specificity to MIR92b-3p, which has a significant impact on liver cell behavior in humans. We assume that a successful MIR92b-3p inhibitor will bind to the precursors of MIR92b-3p miRNA, disabling the action of either of the two processing enzymes involved in the biogenesis of any miRNA in a cell (Drosha or Dicer), thus affecting the MIR92b function. The discoveries made with these inhibitory chemical molecules could provide insight into the role of the MIR92 pathway in liver diseases and cancer, and possibly, if promising results appear, they may facilitate a strategy for treating some human diseases in the future.

In aquatic ecosystems, allelopathic activity depends on the production and secretion of allelopathic compounds and their effective dispersal in the environment. In addition, macroalgae have been found to produce active metabolites that affect other organisms that compete with them for nutrients. However, the allelopathic activity of Baltic red and brown macroalgae on filamentous cyanobacteria is still insufficiently understood. Therefore, the main objective of this study was to demonstrate and compare the allelopathic effects of macroalgae Fucus vesiculosus L. and Coccotylus brodiei (Turner) Kütz. on the growth and photosynthetic activity of two Baltic cyanobacteria Aphanizomenon sp. and Nostoc sp. It was found a stimulating effect of different concentrations (5, 25, and 50 µL mL-1) of the aqueous extract obtained from C. brodiei on the number of cells of Nostoc sp. which constituted 108%, 140%, and 147%, respectively, relative to the control treatment. On the other hand, extracts obtained from F. vesiculosus had no statistically significant effect on the number of cells of the cyanobacteria Aphanizomenon sp. and Nostoc sp. Moreover, the C. brodiei extracts had no significant impact on the growth of Aphanizomenon sp. Furthermore, Baltic macroalgae F. vesiculosus and C. brodiei was able to exert allelopathic effects on photosynthesis performance of Nostoc sp. and Aphanizomenon sp. and compounds produced by them had inhibitory, stimulatory, or no significant effect on the maximum PSII quantum efficiency (Fv/Fm) and the effective quantum yield of PSII photochemistry (ΦPSII). The results obtained in this work constitute an important contribution to the knowledge on the allelopathic activity of Baltic red and brown algae on certain bloom-forming species of filamentous cyanobacteria.

Allelopathy is a prevalent natural phenomenon in aquatic ecosystem. We reported the effects of the green macroalga Ulva intestinalis L. collected from estuaries of the Baltic Sea (Poland) on the growth and chlorophyll fluorescence of common filamentous cyanobacterium Nostoc sp. It was found that the addition of 50 µL mL-1 extracts obtained from U. intestinalis inhibited growth of cyanobacterium and after one week of exposition the reduction was 35% of initial amount of Nostoc sp. In addition, we demonstrated that on the seventh day of the exposition, the values of Fv/Fm of target cyanobacterium after addition of 100 µL mL-1 extracts obtained from U. intestinalis was reduced to 49%, compared to control treatment. These results showed for the first time the allelopathic activity of U. intestinalis on Baltic filamentous cyanobacteria Nostoc sp.
Allelopatia jest powszechnym zjawiskiem występującym w ekosystemach wodnych. W artykule opisano wpływ zielenicy bentosowej Ulva intestinalis L. (ulwa kiszkowata), zebranej z Morza Bałtyckiego (Zatoka Gdańska); na wzrost i fluorescencję chlorofilu u nitkowatej sinicy Nostoc sp. Na podstawie uzyskanych wyników badań stwierdzono, że po dodaniu ekstraktów uzyskanych z U. intestinalis, najmniejszy wzrost Nostoc sp. obserwowano w siódmym dniu eksperymentu. Dodanie 50 μL mL–1 ekstraktu z U. intestinalis zahamowało rozwój sinicy Nostoc sp. i po 7 dniu ekspozycji jej wzrost osiągnął poziom 35% w stosunku do warunków kontrolnych. Ponadto największy spadek wartości parametru fluorescencji Fv/Fm dla Nostoc sp. obserwowano po tygodniu trwania eksperymentu. Stwierdzono, że po 7 dniach ekspozycji wartość Fv/Fm dla badanej sinicy po dodaniu 100 μL mL–1 ekstraktu z U. intestinalis spadła o 49%. Przeprowadzone badania po raz pierwszy wykazały, że U. intestinalis wpływa allelopatycznie na bałtyckie sinice nitkowate z rodzaju Nostoc.

Macroalgae have been found to produce active allelochemicals that inhibit of growth other organisms that compete with them for light and space. However, their allelopathic activity on Baltic cyanobacteria is still insufficiently recognised. Therefore, this study aimed to demonstrate the allelopathic effects of Baltic macroalga thallus (Ulva intestinalis) on the growth and photosynthetic activity of three bloom-forming cyanobacteria: Aphanizomenon sp., Nodularia spumigena, and Nostoc sp. This study investigated the cell count of the analysed cyanobacteria (N 105 mL-1), the maximum quantum yield of the second photosystem (PSII) in the dark (Fv/Fm), and the real quantum yield of PSII in the light (ΦPSII) (in the control and the experiments). After 7 days of exposure, the following were added: 0.01, 0.05, and 0.1 g mL-1 of U. intestinalis fresh thallus. It was found that thallus obtained from U. intestinalis had no statistically significant effect on the number of cells of the cyanobacterium Aphanizomenon sp. (at 0.05 and 0.1 g mL-1) and Nostoc sp. (at concentrations of 0.01 and 0.05 g mL−1). On the other hand, it was examined a stimulating effect of 0.01 g mL-1 of the fresh thallus on the number of Aphanizomenon sp. cells which constituted 168%, relative to the control. It was shown that the fresh thallus addition resulted in a decrease in the number of N. spumigena cells (45%, 27%, and 46% after addition of 0.01, 0.05, and 0.1 g wet weight mL−1 of fresh thallus, respectively). In experiments with Nostoc sp., the addition of U. intestinalis thallus has been a negative effect on cyanobacterial growth at 0.1 g mL−1 and constituted 97% of control. It was also found, that U. intestinalis had no allelopathic effect on fluorescence parameters of N. spumigena. All tested concentrations of thallus U. intestinalis (0.01, 0.05, and 0.1 g wet weight mL-1) stimulated the values of Fv/Fm or ΦPSII of cyanobacteria Aphanizomenon sp. and Nostoc sp. compared to the control. These studies help define the role of U. intestinalis allelopathy as a biological factor in the distribution of bloom-forming cyanobacteria in the coastal Baltic Sea region.
Makroglony mają zdolność do produkowania aktywnych związków allelopatycznych, które wpływają na inne organizmy, konkurujące z nimi o światło i przestrzeń. Jednakże ich działanie allelopatyczne na bałtyckie sinice jest wciąż niedostatecznie rozpoznane. Dlatego głównym celem niniejszej pracy było wykazanie aktywności allelopatycznej plechy bałtyckiej zielenicy, ulwa (lub taśma czy błonica) kiszkowata - Ulva intestinalis, na wzrost i aktywność fotosyntetyczną i wzrost trzech sinic bałtyckich, tworzących masowe zakwity: Aphanizomenon sp., Nodularia spumigena oraz Nostoc sp. W niniejszej pracy badano liczebność komórek analizowanych sinic (N 105 mL-1), maksymalną wydajność kwantową drugiego fotosystemu (PSII) w ciemności (Fv/Fm) oraz rzeczywistą wydajność kwantową PSII w świetle (ΦPSII) (w kontroli oraz w eksperymentach). Po 7 dniach ekspozycji, dodawano do nich: 0,01, 0,05, i 0,1 g mL-1 świeżej plechy U. intestinalis. Wykazano, że obecność plech U. intestinalis nie miała istotnego wpływu na liczebność komórek Aphanizomenon sp. w ilości (0,05 i 0,1 g mL-1) oraz Nostoc sp. w ilości plechy (0,01 i 0,05 g mL−1). Wykazano także stymulujący wpływ plechy dodawanej w najmniejszej ilości 0,01 g mL-1 na liczebność komórek Aphanizomenon sp., która wynosiła 168%, w stosunku do kontroli. Dodanie 0,01, 0,05 oraz 0,1 g mL−1 świeżej plechy powodowało obniżenie liczebności komórek N. spumigena (o odpowiednio: 45%, 27%, i 46%, w stosunku do kontroli). Dodatkowo, dodanie 0,1 g mL−1 plechy U. intestinalis powodowało zahamowanie wzrostu Nostoc sp. o 97%. W pracy wykazano także, że plecha U. intestinalis nie miała wpływu na wartość parametrów fluorescencji u N. spumigena. Plecha tego gatunku (w każdym testowanym stężeniu: 0,01, 0,05 oraz 0,1 g mL-1) stymulowała wartość parametrów Fv/Fm lub ΦPSII u sinic Aphanizomenon sp. i Nostoc sp. Wyniki uzyskane w niniejszej pracy definiują rolę allelopatii U. intestinalis jako ważnego czynnika biologicznego, wpływającego na występowanie zakwitów sinic w przybrzeżnych rejonach Morza Bałtyckiego.

The use of Arthrospira platensis for food is increasingly relevant as its components (proteins, lipids, minerals and vitamins) have substantial potential to be competitive with the same components from other food sources, and Arthrospira platensis-based food production is relatively low in price and holds medicinal properties.

Under some conditions the growth of toxic cyanobacteria must be controlled by treatment with algicidal compounds. Hydrogen peroxide has been proposed as an efficient and relatively safe chemical which can remove cyanobacteria from the environment selectively, without affecting other microorganisms. However, the uncontrolled release of secondary metabolites, including toxins may occur after such a treatment. Our proposal presented in this paper concerns fast biodegradation of microcystin released after cell lysis induced by hydrogen peroxide. The effectiveness of both, Sphingomonas sp. and heterologously expressed MlrA enzyme, in the removal of the toxin from Microcystis aeruginosa culture was investigated. The results indicate that neither Sphingomonas cells nor MlrA are affected by hydrogen peroxide at the concentrations which stop the growth of cyanobacteria. A several-fold reduction in microcystin levels was documented in the presence of these agents with biodegradation ability. Our results provide evidence that such a combined treatment of water reservoirs dominated by microcystin-producing cyanobacteria may be a promising alternative which allows fast elimination of both, the bloom forming species and toxins, from the environment.

Opis:
The main goal of this study was to determine the distribution of potentially toxic cyanobacteria in 39 selected Polish water bodies. From the water bodies with blooms and also from those in which blooms were not visible 87 samples were investigated. For the first time samples from ponds localized in villages with high agricultural activities were included. Lakes for which microcystin concentrations had been determined before were included as a reference for the research. The detection of cyanobacteria was conducted by microscopic observation as well as by PCR amplification of the rpoC1 gene fragment. Cyanobacteria were present in 75 out of 87 samples. The presence of potentially toxic cyanobacteria was detected by amplification of the mcyB and mcyE genes, which are involved in the biosynthesis of microcystins. Both genes were detected in 7 out of 9 blooms investigated. In the case of samples collected from water bodies in which blooms were not observed, the mcyB and mcyE genes were detected in 20 out of 36. In order to identify the cyanobacteria occurring in selected reservoirs, 16S plus ITS clone libraries were constructed. The method allowed distinguishing 18 different genotypes. After sequence analysis, cyanobacteria belonging to genera Microcystis, Planktothrix, Anabaena, Pseudanabaena, Synechocystis, Synechococcus and Woronichinia were identified. Results confirmed the usefulness of the rpoC1 and mcy genes for monitoring water bodies and detection of potentially toxic cyanobacteria. Application of molecular markers allowed detecting potentially toxic cyanobacteria before the bloom was visible. This is the first comprehensive study concerning cyanobacteria present in different types of Polish water bodies performed using molecular markers.

Opis:
The aim of the paper is to present the hydrodynamic, mass transfer and illumination characteristics of a laboratory helical-tube photobioreactor Biostat PBR-2S, commercially available and used in many laboratories in Poland and worldwide. The investigated hydrodynamics parameters were: mean liquid circulation rate, liquid velocity/residence time in the tubular part of the apparatus and mixing time, measured in the wide range of rotary speed of the circulation pump. The influence of the aeration intensity on these parameters was also checked. The volumetric oxygen and carbon dioxide transfer coefficients in the liquid phase and their dependency on the liquid circulation rate and gas inflow rate were determined. The experiments were performed in tap water and then in a real three-phase cultivation broth at the end of thermophilic cyanobacteria T. synechococus growth. For the final evaluation of the tested PBR there were series of test cultivations run under different conditions of illumination. The highest final concentration of the biomass of tested cyanobacteria reached the relatively high value of 4.38 g/dm3 of the dry biomass, although the process conditions were not fully optimized. The laboratory photobioreactor PBR-2S proved to be a good tool for investigations of microalgae cultivation processes. The presented results and practical observations may help to analyze and understand the mutual influence of the specific process parameters in the described PBR, especially during autotrophic organism cultivations.

Celem przeprowadzonych badań było zastosowanie mikroorganizmów fotoautotroficznych jako biokatalizatora w redukcji -ketoalkilofosfonianu dietylu. Przeprowadzone badania przesiewowe wskazały szczepy cyjanobakterii Arthrospira maxima CCALA 027 oraz Nodularia sphaerocarpa CCALA 114 jako zdolne do redukcji 2-oksopropanofosfonianu dietylu do odpowiedniego 2-hydroksypropanofosfonianu dietylu. W trakcie 7-dniowego procesu biokonwersji stopień przereagowania substratu w przypadku zastosowania wymienionych szczepów wynosił odpowiednio 26,4% oraz 12,9%, zaś czystość optyczna produktu w obu przypadkach wyniosła ponad 99%.
The aim of the study was applying of autotrophic microorganisms as a biocatalyst in the reduction of oxoalkylphosphonates. Cyanobacteria strains Arthrospira maxima CCALA 027 and Nodularia sphaerocarpa CCALA 114 are capable to reduce diethyl 2- oxopropylphosphonate to the corresponding diethyl 2- hydroxypropylphosphonate. The degree of conversion of the substrate was respectively 26.4% and 12.9%, and the optical purity of the product in both cases was over 99%.

W normalnym stanie wód powierzchniowych liczebność komórek sinic (Cyanophyta) w 1 cm3 wody waha się od kilkuset do kilku tysięcy, przy czym w okresach intensywnego zakwitu może dochodzić do kilkuset tysięcy na 1 cm3 wody. Intensywne zakwity mogą powodować zmianę barwy i nieprzyjemny zapach wody, a także uwalniają duże ilości neurotoksyn alkaloidowych i hepatotoksyn o wysokiej toksyczności. W pracy przedstawiono wyniki badań dotyczące skuteczności oczyszczania wody powierzchniowej w obecności komórek sinic i mikrocystyny LR w układzie oczyszczania wody w Tomaszowie Mazowieckim w 2004 r. Wykazano, że zastosowanie koagulacji bez wstępnej dezynfekcji chlorem lub dwutlenkiem chloru powodowało wzrost zawartości mikrocystyny LR w wodzie (śr. o 176%), w wyniku rozpadu (lizy) komórek sinic pod wpływem koagulantu. Ozonowanie wody dawką 3,0÷5,5 gO3/m3 było najefektywniejszym etapem usuwania mikrocystyny LR z wody. Końcowa efektywność usuwania mikrocystyny LR z wody zmieniała się w zakresie 49,8÷100,0%.
Normally, the number of cyanobacterial cells (Cyanophyta) in surface water varies from several hundred to several thousand per cubic centimeter. While blooming, their number may rise to several hundred thousand per cubic centimeter of water, and the water may take a specific hue and an unpleasant smell. In addition, during blooming cyanobacteria produce large amounts of alkaloid neurotoxins and oligopeptide hepatotoxins of high toxicity. The study reported on in this paper was carried out at the Water Treatment Plant of Tomaszów Mazowiecki in 2004. It aimed at determining the efficiency of their treatment train when the surface water to be treated contained cyanobacterial cells and microcystin LR. The investigations have produced the following findings: coagulation without prechlorination (with chlorine or chlorine dioxide) promoted a rise in microcystin LR concentration by 176% on average, as a result of cell lysis due to exposure to the coagulant; ozonation (with a 3.0 to 5.5 gO3/m3) was the most efficient unit process for microcystin LR removal. The final efficiency of microcystin LR removal from the water ranged from 49.8 to 100%.
