Opis:
Cryptosporidium parvum, an Apicomplexan parasite, is an important cause of diarrheal disease, especially in immunodeficient hosts. Nevertheless, there is no entirely successful therapeutic agent against cryptosporidiosis to date. Hence, this study aims to test the potential prophylactic and therapeutic effect of Camellia sinensis (green tea extract) in dexamethasone immunosuppressed mice versus the nowadays used drug, Nitazoxanide (NTZ). Parasitological and molecular methods were used to characterize Cryptosporidium oocysts before infection. Fifty bred female Swiss Albino mice were divided into 5 groups; group I (GI)(GTP): immunosuppressed and prophylactically treated with green tea extract for 5 days prior to infection, group II (GII)(GTT): immunosuppressed, infected with Cryptosporidium parvum and treated with green tea extract, group III (GIII)(NT): immunosuppressed, infected and treated with NTZ, group IV (GIV)(PC): immunosuppressed and infected (Positive control), group V (GV)(NC): immunosuppressed and non-infected (Negative control). Furthermore, parasitological examination for oocysts in the stool, and histopathological examination for the small intestine and liver specimens were performed for the study groups. Cryptosporidium oocysts used for induction of infection proved to be Cryptosporidium parvum genotype 2. Moreover, a significant oocyst reduction in fecal samples correlated with an improvement of histopathological changes in the small intestinal and liver tissues in GI(GTP), GII (GTT) and GIII(NT) groups. Besides, the GII(GTT) group showed the best improvement in parasitological and histopathological parameters among the test groups. This study revealed that Camellia sinensis (green tea extract) has potential activity against cryptosporidiosis and could serve as a promising prophylactic and therapeutic anti-cryptosporidial agent.

Introduction and objectives. The parasite Cryptosporidium spp. is an intracellular protozoa which has a broad range of hosts and zoonotic potential. It presents a serious health risk for agricultural workers and veterinarians. The aim of the study was to identify the species and subtypes of Cryptosporidium occurring in a veterinary student who came into contact with calves on a farm. Materials and method. The Ziehl-Neelsen staining technique was employed to confirm the presence of Cryptosporidium oocysts. ELISA test was applied to detect coproantigen in faecal specimens. Nested PCR was used to amplify a small ribosomal subunit (SSU rRNA) and sequencing of the GP60 gene served to identify the zoonotic subtypes. Results. The nested PCR allowed to confirm the C. parvum species; subsequently, the IIdA15G1 zoonotic subtype was identified. Conclusion. This is the first confirmed case in Slovakia of human cryptosporidiosis caused by the unique subtype IIdA15G1.

Introduction and Objective. The problem of occupational biohazards is very important, especially in the field of agriculture and in human and veterinary medicine. The aim of the study was to determine the potential sources of infection in veterinary professionals with selected zoonotic agents, including: Toxoplasma gondii, Giardia duodenalis, Leptospira spp., Cryptosporidium spp. and Coxiella burnetii. Materials and Method. A total of 50 air samples from barns, piggeries and veterinary surgeries were examined for the presence of Leptospira spp. and C. burnetii DNA. Serum samples of 86 pigs and 80 cows were tested for the presence of antibodies to Leptospira spp. and to phase I and II C. burnetii antigens. Serum of 70 cats were tested for the presence of antibodies to T. gondii and 65 samples of cat faeces for the presence of T. gondii oocysts. The presence of G. duodenalis and Cryptosporidium spp. were examined in 50 of dog faeces and 50 of bovine faeces samples. Results. DNA of Leptospira spp. was detected in 2 air samples from the piggeries (4%). C. burnetii DNA was not found in any sample. Anti-Leptospira spp. antibodies were detected in 51 (59.3%) of examined pigs. Neither anti-Leptospira spp. nor anti-C. burnetii antibodies were found among samples of bovine serum. Anti-T. gondii antibodies was found in 52 cat serum samples (74.3%). Among samples of cat faeces, no T. gondii oocysts were detected. In one sample of cattle stool (2%), G. duodenalis was detected and in another (2%) – Cryptosporidium spp. G. duodenalis was detected in 7 samples (14%) and Cryptosporidium spp. in 2 samples (2%) of dog faeces. Conclusions. The results of this study demonstrate the potential risk of infection with Leptospira spp. in veterinarians working with pigs. Veterinarians could be also be at risk of infection with T. gondii and G. duodenalis.

The main aim of the study was to determine the risk of two selected zoonosis infections caused by Giardia duodenalis and Cryptosporidium spp. among an occupational group of veterinarians in Poland. Two hundred and ninety-seven samples of stool were tested for the presence of Giardia cysts and Cryptosporidium oocysts using Direct Fluorescent Assay (DFA). There were no positive results for Cryptosporidium. The presence of Giardia cysts was found in two samples of faeces (0.67%). The risk with regard to the parasites Giardia duodenalis and Cryptosporidium spp. seems to be low among the group of veterinarians.

To study the occurrence of Cryptosporidium infection in laboratory rats (Rattus norvegicus) raised for experimental usage, 134 faecal samples were obtained from two rearing houses in Ibadan and examined for the presence of Cryptosporidium oocyst using the modified acid fast staining technique. Cryptosporidium species in 2 samples positive for microscopy were further characterized by a nested polymerase chain reaction (PCR) amplifying the 18S rRNA gene. Two of 134 samples were positive for the Cryptosporidium oocysts. Sequencing of the small-subunit rRNA amplicons identified the species in the two PCR positive samples as Cryptosporidium andersoni and Cryptosporidium rat genotype. These findings showed that laboratory rat is a potential reservoir for diverse Cryptosporidium species and suggests that laboratory rats should be screened for Cryptosporidium infection prior to experiments, especially where pathogen free animals are not available. This the first report to identify Cryptosporidium species infecting laboratory rats in Nigeria.

Various types of biological weapons have been known and practiced throughout history, including the use of biological agents such as microbes and plants, as well as biotoxins and the venoms that can be derived from them. In ancient civilisations, the attempt was to infect and kill enemies by throwing cadavers into water wells. Emperor Barbarossa during the battle of the Italian town, Tortona, in 1155, did the same. In modern times, America and the Soviet Union also undertook biological warfare and anti-biological warfare protection activities. This even intensified after WWII. When the Soviet forces captured and interrogated some Japanese scientists in 1945, they utilized the obtained information in their own biowarfare program and their research accelerated in 1946. Following this, a series of new biowarfare study centres and production facilities was constructed in the 1950s. The Soviet biowarfare program included tularemia, anthrax, brucellosis, plague, glanders, marburg virus, smallpox virus, and VEE virus. During the time of the Korean War, it was believed that biowarfare agents were used by America against Soviet Union. The Americans had began their own program in Fort Detrick (former Camp Detrick) in 1943 and a new production facility at Pine Bluff Arsenal in Arkansas was constructed. The United States of America started producing tons of Brucella suis in 1954. In the peak year of their program, they involved about 3,400 people and a number of agents: Bacillus anthracis, Francisella tularensis, Brucella suis, Coxiella burnetti, Venezuelan equine encephalitis virus, yellow fever, botulin, Staphylococcal enterotoxin, and the anti-crop agents Pyricularia oryzae and Puccinia graminis. Due to public pressure, President Nixon declared a unilateral halt in 1969 to biological weapon projects. The only permitted research was defensive, such as diagnostic, vaccines, and chemotherapies tests – as evidenced in the UK where the base in Porton Down was converted into a defence institution.
