Opis:
Synthetic analogs of vitamin D for potential use in differentiation therapy should selectively regulate genes necessary for differentiation without inducing any perturbations in calcium homeostasis. PRI-1906, an analog of vitamin D2, and PRI-2191, an analog of vitamin D3 bind nuclear vitamin D receptor (nVDR) with substantially lower affinity than 1,25-dihydroxyvitamin D3 (1,25-D3), but have higher differentiation-inducing activity as estimated in HL-60 leukemia cell model. To examine how their increased differentiation-inducing activity is regulated we tested the hypothesis that membrane-mediated events, unrelated to nVDR, take part in the differentiation in response to PRI-1906 and PRI-2191. The induction of leukemia cell differentiation in response to the analogs of vitamin D was inhibited by LY294002 (phosphatidylinositol 3-kinase inhibitor), PD98059 (inhibitor of MEK1,2, an upstream regulator of extracellular-signal regulated kinase) and rapamycin (p70S6K inhibitor) pointing out that activation of signal transduction pathways unrelated to nVDR is necessary for differentiation. On the other hand, inhibition of cytosolic phospholipase A2 accelerated the differentiation of HL-60 cells induced by either 1,25-D3 or by the vitamin D analogs suggesting possible existence of a feedback loop between extracellular-signal regulated kinases and phospholipase A2.

Opis:
Toxicity of Pb2+ and Cd2+ is a widespread issue in the world; however, few studies have been conducted to understand their effect at environmentally realistic concentration in a mixture. In the present study, Crucian carp was exposed to Pb2+ (30 µg·l-1), Cd2+ (100 µg·l-1) and their mixture (30+100 µg·l-1) for 96 h and 21 d period to assess changes in the liver and muscle vitamin A and E content, and hematological and serum biochemical parameters. The results indicated significant decline in the level of antioxidant vitamins A, E and alterations in the hematological and serum biochemical indices. The toxicity revealed anemia, impairment of the liver and kidney with evident responses after 21 d exposure due to additive effect of Pb2+ and Cd2+ in mixture. Moreover, the differential response of vitamins A, E and blood parameters to low levels of waterborne Pb2+ and Cd2+ in freshwater fish can be used as biomarkers for monitoring contamination of aquatic environment.

Opis:
Cyclodextrin (CD) is a cyclic compound with a spatial structure in the shape of a toroid. It is characterized by specific properties. The outer portion of the structure has hydrophilic properties, while the cavity of the toroid is hydrophobic. This enables cyclodextrin to form inclusion complexes with the enclosure of lipophilic molecules in the interior. This paper presents the results of attempts to form a complex of γ-CD/CL-20. This study determined unambiguously that a γ-CD with CL-20 complex in a mole ratio 1:1 is formed as a result of mixing of solutions of γ-CD and CL-20. This conclusion was corroborated by FTIR, 1H NMR, UV-Vis spectroscopic techniques and by measurement of the density of the complex obtained. The resulting complex of γ-CD/CL-20 is characterized by much lower sensitivity to friction and impact than CL-20 itself.

Microbially assisted phytoremediation is considered as the most promising eco-friendly solution for recultivation of heavy metal polluted soils. In vitro experiments can be favorable systems that allow assessing compatibility and efficiency of both partners (e.g. plant-microorganism) which reduces time and space in the initial stages of this technology. The main objective of this study was: (1) to calculate the Cd2+ accumulation factors (e.g. BCF, Ti, AF) using willow (Salix viminalis L.) inoculated with three Streptomyces sp. strains under in vitro conditions and (2) to compare obtained results with that derived from pot experiments by Złoch et al. (2017). Our results reveal significantly increase in Cd2+ accumulation capacity of Streptomyces spp. inoculated wil- low plants, indicating microbial stimulation of phytoextraction. Additionally, inoculated plants showed higher biomass production and lower lipids peroxidation level. The results revealed significant increase of MEA, BCF, Ti, MER by Strep-1 and Strep-2 in the above-ground parts of inoculated plants. Moreover, of the three strains tested, Strep-1 (Streptomyces sp. SIIB-Zn-R8) demonstrated the highest impact on the Cd2+ phytoextraction efficiency. In conclusion, the proposed in vitro model system allowed predicting Cd2+ phytoextraction capacity per- formed using inoculated willow plants with the significant reduction of both time and space.

Introduction. Reports have shown that there is a rise in beta-2-microglobulin (β2M) concentration in patients with HIV infection and that the degree of elevation correlates well with the extent of disease burden and could be an independent prognostic marker for death. However, there is the dearth of information on the interplay between alteration in haematological profile, a common cause of morbidity and mortality in HIV, and β2M. Aim. Changes in selected haematological parameters and β2M in Nigerian HIV patients stratified based on CD4+ T-cells counts were thus assessed in this study. Material and methods. Forty-eight asymptomatic, drug naïve HIV patients were enrolled into this cross-sectional study. Haemoglobin concentration (Hb), packed cell volume (PCV), total and differential white blood cell count, platelet count and CD4+ T-cells count were determined using standard methods while serum levels of β2M were determined using ELISA. Thereafter, the patients were stratified into three groups based on the CD4+ T-cells count. Results. Hb and lymphocyte counts increased with increasing CD4+ T-cells count. In contrast, neutrophils percentage, MCV and MCH reduced with increasing CD4+ T-cells count. The mean lymphocytes percentage was significantly higher while the mean neutrophils percentage was significantly lower in patients with CD4+ T-cells count of 500–800 cells/μl compared with the patients with CD4+ T-cells count <200 cells/μl. Similarly, the mean MCV was significantly lower in patients with CD4+ T-cells count of 500–800 cells/μl compared with patients with CD4+ T-cells count of 200–499 cells/μl and patients with CD4+ T-cells count <200 cells/μl. β2M had significant positive correlation with WBC and neutrophils percentage but had a significant negative correlation with lymphocytes percentage and MCH in patients with CD4+ T-cells count <200 cells/μl. However, β2M had sig nificant positive correlation with PCV, Hb, monocytes and morphology in patients with CD4+ T-cells count of 500–800 cells/μl. Conclusion. It could be concluded from this study that HIV infection is associated with alteration in haematological profile and the alteration is CD4+ T-cells count-dependent. Also, elevation in β2M concentration appears to be a marker of lymphopaenia in patients with low CD4+ T-cells count

Opis:
Przeciwwirusowe terapie dla pacjentów z wirusem HIV są stale ulepszane. Podawanie kombinacji dwóch lub więcej leków powoduje spadek liczby wirusów poniżej poziomu detekcji u wielu pacjentów. W szczególności połączenie wysoce efektywnej terapii antyretrowirusowej (HAART) z aktywatorem immunologicznym (interleukiną 2, IL-2) wydaje się stanowić skuteczną metodę długookresowej kontroli replikacji wirusa  HIV in vivo.W naszym artykule zaproponowaliśmy matematyczny model odpowiedzi odpornościowej dla pacjentów z wirusem HIV przy zastosowaniu połączonej terapii  HAART i IL-2. Zbadaliśmy dynamikę modelu w poszukiwaniu warunków, przy których układ odpornościowy odnawia się dzięki takiej terapii. Z~analitycznego punltu widzenia oznacza to globalną stabilność stanu stacjonarnego odpowiadającego zdrowemu organizmowi. Analiza matematyczna została uzupełniona symulacjami komputerowymi.
Antiviral therapy for HIV-infected patients has greatly improved in recent years. Administration of drug combinations consisting of two or more different drugs can reduce and maintain virus load below detection level in many patients. Cyclic administration of the immune activator interleukin-2 (IL-2) in combination with highly active antiretroviral therapy (HAART) has been suggested as an effective strategy to realize long-term control of HIV replication in vivo. In this article, we formulate a mathematical model of immune response for HIV-infected individual in the presence of HAART and IL-2. We look  for the conditions under which the immune system recovers by applying IL-2 as an immune activator along with HAART. From analytical point of view this means global stability of the disease-free equilibrium. Comprehensive numerical simulations are presented to illustrate the analytical results.
