Opis:
Sirtuins (type III histone deacetylases) are an important member of a group of enzymes that modify chromatin conformation. We investigated the role of sirtuin inhibitor, GPI 19015, in double strand break (DSB) repair in CHO-K1 wt and xrs-6 mutant cells. The latter is defective in DNA-dependent protein kinase (DNA-PK)-mediated non-homologous end-joining (D-NHEJ). DSB were estimated by the neutral comet assay and histone γH2AX foci formation. We observed a weaker effect of GPI 19015 treatment on the repair kinetics in CHO wt cells than in xrs6. In the latter cells the increase in DNA repair rate was most pronounced in G1 phase and practically absent in S and G2 cell cycle phases. The decrease in the number of histone γH2AX foci was faster in xrs6 than in CHO-K1 cells. The altered repair rate did not affect survival of X-irradiated cells. Since in G1 xrs6 cells DNA-PK-dependent non-homologous end-joining, D-NHEJ, does not operate, these results indicate that inhibition of sirtuins modulates DNA-PK-independent (backup) non-homologous end-joining, B-NHEJ, to a greater extent than the other DSB repair system, D-NHEJ.

We compared three methods usually applied in biological dosimetry for estimation of radiation-induced DNA damage in human T and B lymphocytes: alkaline comet assay, micronucleus (MN) test and formation of histone gamma-H2AX foci. Human peripheral blood lymphocytes were fractionated using T cells and B cells isolation kits. Cells were irradiated with doses in the range of 0-1 Gy of X-rays. Induction of DNA damage was assessed by the standard alkaline comet assay, MN test and histone gammaH2AX foci immunofluorescence assay. Notwithstanding different end-points measured by the applied methods, all tests revealed a similar induction of DNA damage in B lymphocytes as compared with T lymphocytes. The results indicated that all three tests detect DNA damage with similar sensitivity, the lowest dose being approximately 0.3 Gy. The difference between irradiated and control cells was expressed as the ratio of the value obtained for irradiated cells (1 Gy) to that for control cells. The highest ratio was obtained for formation of gammaH2AX foci and was 6.2 for T and 13.8 for B lymphocytes, whereas those for comet assay and micronucleus test were 3.5; 3.6 and 5.6; 4.8, respectively.

Opis:
Adaptive response (AR), an enhanced resistance to a high dose of ionising radiation acquired after pretreatment with a very low dose, was estimated in normal human lymphocytes. The question posed was whether the extent of radioadaptation, assessed by micronucleus test, would be related to the rate of DNA double-strand break (DSB) rejoining. Phytohemagglutinin-stimulated G1-lymphocytes from 5 healthy male volunteers were pre-treated (or not) with an adaptive (5 cGy) dose of X-rays, followed by a higher (5 or 10 Gy) challenge dose after 20-22 h. DSB rejoining after the challenge dose was monitored with the use of two methods: neutral comet assay, modified to reduce the contribution of single-strand breaks (SSBs) and thermolabile sites, and pulse-field gel electrophoresis (PFGE), specific for DSBs. At the level of micronuclei, an AR was observed in lymphocytes of 3 of 5 donors. Up to 60 min, comet assay showed no statistically significant differences in DNA break rejoining between adapted and non-adapted lymphocytes, independently of AR appearance. PFGE gave similar results, although in three donors it revealed secondary increases in DSBs levels at 30 min and/or 60 min post-irradiation in the adapted vs. the non-adapted samples. Failure to demonstrate changes in DSBs rejoining rate in the adapted lymphocytes could be due to diversity of AR intensity/timing at the level of DNA repair in not fully homogenous cell populations. Also, “rare” DNA cuts characteristic of early apoptosis/necrosis could overlap the process of DNA break rejoining.

Comet assay under neutral conditions allows the detection of DNA double-strand breaks, considered to be the biologically relevant radiation-induced lesion. In this report we describe modifications of the neutral comet method, which simplify and facilitate its use for estimation of DNA double strand breaks in human lymphocytes irradiated with doses of 60Co gamma-rays (from 10 to 100 Gy). The analysis carried out according to this protocol takes less time than those published so far. Also, the use of lysis at 50°C is avoided; this is important in view of the presence of heat-labile sites in the DNA of irradiated cells, recently reported by Rydberg [12]. The comets have well defined, sharp limits, are suitable for computer image analysis and chromatin of the control cells remains condensed.

Opis:
Nanoparticles (NPs) defined as particles having at least one dimension below 100 nm have been applied in the last decade in industry and medicine. Recently, there is an increased concern about the biohazard aspect of the presence of NP in consumer goods and in the environment. Silver NP (Ag NP) cause oxidative stress in mammalian cells in result of generation of reactive oxygen species (ROS). This results in genotoxicity and mutagenicity, disturbed mitochondrial respiration, slowed proliferation and cell death. Using the alkaline comet assay, we examined the effect of combined treatment with Ag NP 20 nm or 200 nm and X-rays (2 Gy) in HepG2 cells. In addition, combined treatment with X-rays and titanium dioxide NP (TiO2 NP) 21 nm was also studied. No effect of NP pre-treatment on X-ray induced initial deoxyribonucleic acid (DNA) damage levels was observed for all three NP. In contrast, Ag NP treatment preceding exposure to X-rays caused a marked decrease in the rate of single strand break rejoining. The effect was particularly strong for Ag NP 20 nm. TiO2 NP pre-treatment had no effect on DNA repair.

The article is devoted to the text of the sermon delivered by the Dominican Fabian Sierakowski on the holiday of Pentecost in Lublin on 24 May 1665; its text was published in the same year. The subject of the sermon was the appearance of a strange light over Lublin on that day, that was defined as a „comet” (a meteor shower?). The phenomenon caused an alarm in the inhabitants of the town who were convinced that the „comet” would bring some disaster to the town. In his „Prediction from the Comet” Fabian Sierakowski shows that the prodigium forecasts happy events – but in the spiritual sense. He bases his prediction on an interpretation of Chapter Two of the Acts of the Apostles (Descent of the Holy Ghost). The text is an interesting example of Baroque homilies. The author sees a parallel between the events that occurred on the day of Quinquagesima Sunday and on the feast of Pentecost in 1665. Sierakowski also refers to the situation of the Polish Republic (Jerzy Lubomirski's rebellion); among others, he presents an analogy between the state and the members of the human body.

The article analyses the struggle of Anglo-American relations connected to slaves and maritime rights on the sea from 1831 to 1842. The study is based on monographs, reports, treaties and correspondences between the two countries from the explosion of the Comet case in 1831 to the signing of the Webster–Ashburton treaty in 1842. This study focuses on three fundamental issues: the appearance of Comet, Encomium, Enterprise, Hermosa and Creole as international incidents with regards to British-American relations; the view of both countries on the abolition of slavery, maritime rights as well as the dispute over issues to resolve arising from these incidents; the results of British-American diplomacy to release slaves and maritime rights after the signing of the Webster–Ashburton treaty. The study found that the American slave ships were special cases in comparison with the previous controversies in bilateral relations. The American slave vessels sailed to the British colonies due to bad weather conditions and a slave rebellion on board. In fact, Great Britain and the United States had never dealt with a similar case, so both sides failed to find a unified view regarding the differences in the laws and policies of the two countries on slavery. The history of British-American relations demonstrated that under the pressures of the border dispute in Maine and New Brunswick, the affairs were not resolved. In addition, it could have had more of an impact on the relationship between the two countries, eventually p the two countries into a war. In that situation, the diplomatic and economic solutions given to the abolition of slavery and maritime rights were only temporary. However, the international affairs related to the American slave vessels paved the way for the settlement of maritime rights for British-American relations in the second half of 19th century.
W artykule zostały przeanalizowane nabrzmiałe konflikty w anglo-amerykańskich relacjach dotyczących kwestii niewolnictwa i praw morskich w okresie od 1831 do 1842 r. Są one oparte na analizie monografii, raportów, postanowień traktatowych oraz korespondencji amerykańskich i brytyjskich dyplomatów od momentu tzw. incydentu Kormeta (Cormet) w 1831 r. do podpisania traktatu Webstera–Ashburtona w 1842 r. W artykule zostały poruszone trzy najważniejsze sprawy, mianowicie kwestie statków: Kormeta, Encomium, Enterprise, Hermosa i Kreola jako międzynarodowych problemów w relacjach brytyjsko-amerykańskich, poglądy obu państw na kwestię zniesienia niewolnictwa, prawa morskie, jak również dyskusja na temat tego, jak rozwiązać powstałe konflikty morskie, które związane były bezpośrednio z kwestią przewożenia niewolników. W artykule ukazano, że przewóz niewolników na amerykańskich statkach stał się powodem do podjęcia nadzwyczajnych przedsięwzięć, które nie miały miejsca w dotychczasowych bilateralnych stosunkach. Gdy po raz pierwszy amerykańskie statki z niewolnikami dopłynęły do brytyjskich kolonii z uwagi na złe warunki pogodowe i bunt niewolników na pokładzie, to z uwagi na to, że do tej pory ani USA, ani Wielka Brytania nie miały do czynienia z podobnymi przypadkami, w efekcie żadnej ze stron nie udało się znaleźć jednolitego poglądu. Dopiero pod presją sporu granicznego dotyczącego Maine i Nowego Brunszwiku podjęto rozwiązania, które uchroniły oba kraje przed rozpoczęciem konfliktu zbrojnego. Podjęto tymczasowe rozwiązania dotyczące niewolników i praw morskich, ale sprawy międzynarodowe związane z amerykańskimi statkami przewożącymi niewolników przyczyniły się do otworzenia drogi regulującej prawa morskie w stosunkach brytyjsko-amerykańskich. Była to nagląca potrzeba.

W artykule przedstawiono nowe podejście do rozwiązywania wielokryterialnych problemów decyzyjnych, polegające na identyfikacji ekspertowego modelu decyzyjnego w przestrzeni stanu problemu. Metoda obiektów charakterystycznych identyfikuje model decyzyjny z wykorzystaniem stałych punktów odniesienia oraz teorii zbiorów rozmytych. Metoda ta jest całkowicie odporna na zjawisko rank reversal, czyli odwracania rankingów przy dodaniu nowej alternatywy lub w momencie usunięcia alternatywy ze zbioru już rozpatrywanych obiektów. Za pomocą metody obiektów charakterystycznych identyfikowany jest model oceny ryzyka wystąpienia ataku serca u pacjenta w okresie najbliższych 10 lat, w celu lepszego zobrazowania działania metody COMET.
Ahe paper presents a new approach to solving Multi-criteria decision-making problems. The presented approach identifies an expert decision-making model in the state of the problem. The characteristic objects method identifies a model by using fuzzy set theory and characteristic objects as reference objects. This method is complete free of the rank reversal phenomenon. It means that if new alternatives are added or removed from alternatives set then rank existing order will be not changed. The characteristic objects method will be used to identify the multi-criteria model of 10-year risk of having a heart attack.

Multi-criteria decision-making (MCDM) methods are associated with the ranking of alternatives based on expert judgments made using a number of criteria. In the MCDM field, the distance-based approach is one popular method for receiving a final ranking. One of the newest MCDM method, which uses the distance-based approach, is the Characteristic Objects Method (COMET). In this method, the preferences of each alternative are obtained on the basis of the distance from the nearest characteristic objects and their values. For this purpose, the domain and fuzzy numbers set for all the considered criteria are determined. The characteristic objects are obtained as the combination of the crisp values of all the fuzzy numbers. The preference values of all the characteristic object are determined based on the tournament method and the principle of indifference. Finally, the fuzzy model is constructed and is used to calculate preference values of the alternatives. In this way, a multi-criteria model is created and it is free of rank reversal phenomenon. In this approach, the matrix of expert judgment is necessary to create. For this purpose, an expert has to compare all the characteristic objects with each other. The number of necessary comparisons depends squarely to the number of objects. This study proposes the improvement of the COMET method by using the transitivity of pairwise comparisons. Three numerical examples are used to illustrate the efficiency of the proposed improvement with respect to results from the original approach. The proposed improvement reduces significantly the number of necessary comparisons to create the matrix of expert judgment.

Opis:
The function of acetylcholinesterase (AChE) is the rapid hydrolysis of the neurotransmitter acetylcholine (ACh), which is involved in the numerous cholinergic pathways in both the central and the peripheral nervous system. Therefore, AChE measurement is of high value for therapy management, especially during the course of intoxication with different chemicals or drugs that inhibit the enzyme. Pyridinium or bispyridinium aldoximes (oximes) are able to recover the activity of the inhibited enzyme. Since their adverse effects are not well elucidated, in this study the efficiency of HI-6 oxime in protection and/or reactivation of human erythrocyte AChE inhibited by the antineoplastic drug irinotecan as well as its cyto/genotoxicity in vitro were investigated. HI-6 was effective in protection of AChE and increased its activity up to 30%; the residual activity after irinotecan inhibition was 7%. Also, it reactivated the enzyme previously inhibited by 50% irinotecan (4.6 µg/ml) applied at ¼ of the IC50 value. The tested concentrations of HI-6 exhibited acceptable genotoxicity towards white blood cells, as estimated by the alkaline comet assay, DNA diffusion assay and cytogenetic endpoints (structural chromosome aberrations and cytokinesis-block micronucleus assay). The results obtained warrant the further investigation of HI-6 in vivo, as well as its development for possible application in chemotherapy.

This study aimed to evaluate the antidotal potency of tenocyclidine (TCP) that probably might protect acetylcholinesterase (AChE) in the case of organophosphate poisoning. TCP was tested alone as a pretreatment or in combination with atropine as a therapy in rats poisoned with ¼ and ½ of LD50 of soman. Possible genotoxic effects of TCP in white blood cells and brain tissue were also studied. Results were compared with previous findings on the adamantyl tenocyclidine derivative TAMORF. TCP given alone as pretreatment, 5 min before soman, seems to be superior in the protection of cholinesterase (ChE) catalytic activity in the plasma than in brain, especially after administration of the lower dose of soman. Plasma activities of the enzyme after a joint treatment with TCP and soman were significantly increased at 30 min (P < 0.001) and 24 h (P = 0.0043), as compared to soman alone. TCP and atropine, given as therapy, were more effective than TCP administered alone as a pretreatment. The above therapy significantly increased activities of the enzyme at 30 min (P = 0.046) and 24 h (P < 0.001), as compared to controls treated with ¼ LD50 of soman alone. Using the alkaline comet assay, acceptable genotoxicity of TCP was observed. However, the controversial role of TCP in brain protection of soman-poisoned rats should be studied further.

There is increasing evidence for the existence of an association between the presence of etoposide phenoxyl radicals and the development of treatment-related acute myeloid leukemia (t-AML), which occurs in a few percent of patients treated with this chemotherapeutic agent. The most common side effect caused by etoposide is myelosuppression, which limits the use of this effective drug. The goal of the study was to investigate the influence of antioxidant querectin on myelosuppression and oxidative DNA damage caused by etoposide. The influence of quercetin and/or etoposide on oxidative DNA damage was investigated in LT-12 cell line and bone marrow cells of rats via comet assay. The effect of quercetin on myelosuppression induced by etoposide was invetsigated by cytological analysis of bone marrow smears stained with May-Grünwald-Giemsa stain. Etoposide caused a significant increase in oxidative DNA damage in bone marrow cells and LT-12 cell line in comparison to the appropriate controls. Quercetin significantly reduced the oxidative DNA damage caused by etoposide both in vitro and in vivo. Quercetin also significantly protected against a decrease in the percentage of myeloid precursors and erythroid nucleated cells caused by etoposide administration in comparison to the group treated with etoposide alone. The results of the study indicate that quercetin could be considered a protectively acting compound in bone marrow cells during etoposide therapy.

A population study is reported in which the DNA damage induced by γ-radiation (2 Gy) and the kinetics of the subsequent repair were estimated by the comet and micronucleus assays in isolated lymphocytes of 82 healthy donors and patients with head and neck cancer before radiotherapy. The parameters of background and radiation-induced DNA damage, rate of repair, and residual non-repaired damage were measured by comet assay, and the repair kinetics for every donor were computer-fitted to an exponential curve. The level of background DNA damage before irradiation measured by comet assay as well as the level of micronuclei were significantly higher in the head and neck cancer patient group than in the healthy donors, while the parameters of repair were widely scattered in both groups. Cancer patient group contained significantly more individuals, whose irradiated lymphocytes showed high DNA damage, low repair rate and high non-repaired DNA damage level. Lymphocytes of donors belonging to this subgroup showed significantly lower inhibition of cell cycle after irradiation.
