INTRODUCE The aim of the study was to determinate the effect of low X-ray doses on the percentage and number of peripheral blood lymphocytes T (CD3+), T helper (CD4+), T cytotoxic (CD8+), lymphocytes B (CD19+), activated T lymphocytes (CD3+CD25+) and activated T helper lymphocytes (CD4+CD25+) as well as serum immunoglobulins (IgM, IgG and IgA) concentration in workers of an interventional cardiology ward. MATERIAL AND METHODS The study was performed among a group of 40 workers (28 women and 12 men) and a control group of 35 persons (25 women and 10 men). Flow cytometry was used for the analysis of lymphocytes and the turbidimetric method for assessing serum immunoglobulin concentrations. RESULTS The study showed a decreased percentage and number of CD3+CD25+ and CD4+CD25+ cells as well as IgG concentration in interventional cardiology ward staff in comparison to the control group. A negative correlation between the length of employment and percentage of CD3+CD25+ was found. CONCLUSION The study indicates suppressive impact low doses of ionizing radiation on activated T cells, activated T helper cells as well as IgG production that is adverse in relation to the existing risk of infections caused by different microorganisms.
WSTĘP Celem pracy była ocena oddziaływania niskich dawek promieniowania jonizującego na odsetek i liczbę limfocytów T (CD3+), limfocytów T pomocniczych (CD4+), limfocytów T cytotoksyczych (CD8+), limfocytów B (CD19+), aktywowanych limfocytów T (CD3+CD25+) i aktywowanych limfocytów T pomocniczych (CD4+CD25+) oraz stężeń immunoglobulin IgM, IgG i IgA u pracow-ników oddziału kardiologii interwencyjnej. MATERIAŁ I METODY Badanie przeprowadzono w grupie 40 zatrudnionych (28 kobiet i 12 mężczyzn) oraz w grupie kontrolnej 35 osób (25 kobiet i 10 mężczyzn). Badania limfocytów krwi obwodowej przeprowadzono z wykorzystaniem cytometrii przepływowej, a stężenia immunoglobulin w surowicy oznaczono metodą turbidymetryczną. WYNIKI W badanej grupie pracowników – w porównaniu z grupą kontrolną – wykazano znamienne obniżenie liczby i odsetka limfocytów CD3+CD25+ i CD4+CD25+ oraz stężeń immunoglobuliny IgG. Wykazano także istnienie negatywnej korelacji między okresem zatrudnienia badanych pracowników a odsetkiem limfocytów CD3+CD25+. WNIOSEK Badanie wskazuje na supresyjne oddziaływanie niskich dawek promieniowania jonizującego na aktywowane limfocyty T, aktywowane limfocyty T pomocnicze i produkcję IgG, co nie jest korzystne w odniesieniu do istniejącego ryzyka zakażeń drobnoustrojami.

Regulatory T lymphocytes CD4+CD25+FOXP3+ or Treg cells play a major role in immune system surveillance and tolerance. Treg cells are critical for controlling the immunological system because they inhibit the cytotoxic response. Moreover, it has been found that Treg cell recruitment into the tumor microenvironment reduces the chances of survival in cancer patients and that an increase in Treg cells in the peripheral blood correlates with the progression of ovarian cancer. In our study we addressed Treg cell population changes in cases of ovarian cancer where the patients were treated with chemotherapy. Using flow cytometry we determined the levels of CD4+CD25+FOXP3+ lymphocytes in the peripheral blood of the patients prior to chemotherapy, after 3 courses of chemotherapy, and then after 6 courses (the respective regimens were: TK – 13, CP – 1, TPT – 1 patient). We observed that the Treg lymphocyte levels of ovarian cancer patients decreased after 3 courses of chemotherapy though the difference (p=0.3) was not statistically significant. After 6 chemotherapy courses, however, levels did increase to a statistically significant degree (p=0.02). We have therefore concluded that a measurement of the Treg cell population could be helpful in estimating the impact of chemotherapy on a patient’s host immune system during the systemic treatment of ovarian cancer.
Limfocyty regulatorowe T CD4+CD25+FOXP3+ (komórki Treg) odgrywają istotną rolę w mechanizmach nadzoru i tolerancji immunologicznej, hamując odpowiedź cytotoksyczną układu odpornościowego. Wykazano, że w trakcie rozwoju choroby nowotworowej liczebność limfocytów Treg wzrasta. Obecność nacieków z limfocytów Treg w mikrośrodowisku guza wiąże się z gorszym rokowaniem, a wzrost populacji komórek Treg we krwi obwodowej koreluje z progresją raka jajnika. W niniejszym badaniu sprawdzano wpływ chemioterapii stosowanej u chorych z rakiem jajnika na liczebność populacji Treg we krwi obwodowej. W tym celu odsetek Treg we krwi oznaczano przed, po 3 oraz po 6 kursach chemioterapii (TK – 13, CP – 1, TPT – 1 chora) za pomocą cytometrii przepływowej. Podczas pierwszych 3 cykli chemioterapii zaobserwowano początkowe zmniejszenie odsetka limfocytów T CD4+CD25+ FOXP3+ wśród limfocytów T CD4+, niemniej jednak różnica ta nie była znamienna statystycznie (p=0,3). Następnie w miarę kontynuowania chemioterapii odsetek limfocytów T CD4+CD25+FOXP3+ wśród limfocytów T CD4+ wzrastał i po 6. cyklu w stosunku do aktywności po 3. cyklu chemioterapii był statystycznie znamiennie większy (p=0,02). Autorzy wnioskują, że badanie populacji Treg może być przydatne do oceny wpływu chemioterapii na układ odpornościowy gospodarza podczas leczenia systemowego chorych na raka jajnika.

Normal human T lymphocytes growing in culture undergo replicative senescence. Previously, we have shown that in our conditions polyclonal T cells cease proliferation after about three weeks (Radziszewska et al., 1999, Cell Biol. Int. 23, 97-103). Now we present results of a more detailed analysis of in vitro growth as well as phenotypic changes of T cells. Cell cycle analysis showed that about 20% of cells were in the S phase untill the 17th day of culture (young cells). The highest number of mitotic cells (phase G2/M; 10%) was observed during the first week of culture. All not dividing senescent cells were stopped in the G1 phase (after the 30th day of culture). The sub-G1 fraction which represents apoptotic cells did not exceed 8% during the whole period until the 30th day of culture. During in vitro T-cell growth, a rather rapid selection to CD3+CD8+ cells occurs. In the presenescent (between the 17th and 30th day) and senescent populations the majority of cells (above 90%) were CD8 positive. We also have checked the expression of α-chain interleukin-2 (IL-2) receptor (CD25). In young and presenescent cells about one third of cells was CD25 positive, but only 15% in the pool of senescent cells. Immunoblotting analysis of p16 protein recognized previously as a marker of senescent T cells, showed its highest and transient expression in presenescent cells. A critical review of the polyclonal T cell replicative senescence model is presented.

WC1+ cells in cattle exhibit both regulatory and effector activities. However, it has not been elucidated whether they are so plastic that both activities co-exist in one cell or there are separate subpopulations of effector and regulatory cells. Since the production of IFN-γ and IL-10 seems to be related to WC1+ cells’ effector and regulatory function, respectively, the main aim of this study was to determine whether those cytokines are produced by separate subpopulations of WC1+, or are co-produced by the same cells. Due to increasingly frequent emphasised role of consumption of IL-2 in the mechanism of suppressor action of mouse CD25+CD4+ T regulatory cells, expression of the receptor’s α chain for interleukin 2 (CD25) on WC1+ lymphocytes has been evaluated. An average of 5.21% of WC1+ cells obtained from PBMCs of 12-month-old heifers show constitutive expression of the CD25 molecule, with CD25highWC1+ and CD25lowWC1+ cells accounting for 1.05% and 4.10% of WC1+ lymphocytes, respectively. For detection of intracellular cytokine production, PBMCs were stimulated with concanavalin A. Both IFN-γ- and IL-10-producing cells within the CD25-WC1+ and CD25+WC1+ subpopulations were mainly separate subpopulations. The average percentage of IFN-γ+IL-10-, IFN-γ-IL-10+ and IFN-γ+IL-10+ cells among CD25-WC1+ lymphocytes was 4.03%, 2.67% and 0.51%, respectively. A positive correlation was observed between the presence of the CD25 molecule on WC1+ lymphocytes and production of IL-10 and TGF-β, because the average percentage of IFN-γ-IL-10+ and IFN-γ+IL-10+ among CD25+WC1+ lymphocytes was 3 and 4.5 times higher as compared to the corresponding cells in the CD25-WC1+ subpopulation, whereas the percentage of IFN-γ+IL-10- cells in both the subpopulations was not significantly different. The percentage of TGF-β+ cells within the CD25+WC1+ subpopulation was 2.72 times as high as that of CD25-WC1+ lymphocytes. Therefore, with respect to the production of IFN-γ, IL-10 and TGF-β, CD25+WC1+ lymphocytes turn out to have a more suppressor profile than CD25-WC1+.

Cytotoxic activity is one of the major functions of Natural Killer (NK) cells and is a critical effector mechanism of innate immune responses against infected or cancer cells. A variety of assays have been developed to determine NK cell cytotoxic activity, however a receptor-based screening tool is still lacking. Here, we propose the CD25 receptor as a candidate for NK cell cytotoxicity marker. We have verified that there is a correlation between classic target cell induced cytotoxicity markers and the CD25 expression on NK cells. Non-adherent lymphocyte fractions pre-stimulated with Escherichia coli O55:B5 lipopolysaccharide were co-cultured with settled HeLa targets in a four hour long cytotoxic assay. The cytotoxic effect was evaluated by MTT reduction assay and quantification of soluble cytotoxicity markers (granzyme B, FasL, caspase-8, IFN-γ and IL-2) was done by ELISA. Lymphocytes were stained with anti-CD3-Cy-5, anti-CD56/CD16/Nkp46-FITC and anti-CD25-PE antibodies and analyzed by flow cytometry. We observed that the CD25 expression exclusively on the CD3-CD56+CD25+ NK cells was positively correlated with their cytotoxic function evaluated by the MTT test (r = 0.68), the upregulation of granzyme B (r = 0.89), IL-2 (r = 0.78) and IFN-γ (r = 0.57), however, it was not positively correlated with FasL and caspase-8. We conclude that the CD25 expression might serve as an in vitro receptor-based screening tool for NK cell activity.

The ferromagnetic/diamagnetic semiconductor superlattices, EuS/PbS and EuS/YbSe, were studied by neutron reflectivity. In order to determine the strength of the interlayer coupling, the intensity of the first magnetic Bragg peak vs. applied external magnetic field was measured. Additionally, the in-plane anisotropy and the domain structure were studied by polarized neutron reflectivity. The dependence of the intensity of the antiferromagnetic neutron reflectivity peak vs. magnetic field was simulated using a Stoner-Wohlfarth model. To reproduce the observed spectra it was necessary to take into account the presence of fluctuations of the nonmagnetic layers thickness, by assuming a Gaussian spread of the interlayer coupling constant $J$. For both EuS/PbS and EuS/YbSe superlattices, the best fit was obtained for the directions of the in-plane easy axes, which agree with those determined by polarized neutron reflectivity.

Absorption of electromagnetic waves by graphene-magnetic semiconductor multilayered structure has been investigated for the Voight geometry with taking into account the dissipation processes. The possibility of control of electrodynamic properties of the structure by an external magnetic field, changing in number of periods of the structure, chemical potential of the graphene layers and temperature has been shown. Electrodynamics of the structure is also sensitive to polarization of the incident wave. The structure shows to be especially controllable at terahertz frequencies.

We show how to compute electro-optical spectra of semiconductor superlattices in the region of interband electronic transitions. The method uses the microscopic calculation of eigenvalues and eigenfunctions and the macroscopic real density matrix approach to compute the electromagnetic fields and susceptibilities. The electron-hole screened Coulomb potential is adapted and the valence band structure is taken into account in the cylindrical approximation, thus separating light- and heavy-hole motions. We calculate the electro-optical functions, including the optical Stokes parameters and ellipsometric functions for the case of oblique incidence. Results are given for Zn$\text{}_{1-x}$Cd$\text{}_{x}$Se/ZnSe superlattices and a good agreement with experiments is obtained.

Opis:
Synthetic analogs of vitamin D for potential use in differentiation therapy should selectively regulate genes necessary for differentiation without inducing any perturbations in calcium homeostasis. PRI-1906, an analog of vitamin D2, and PRI-2191, an analog of vitamin D3 bind nuclear vitamin D receptor (nVDR) with substantially lower affinity than 1,25-dihydroxyvitamin D3 (1,25-D3), but have higher differentiation-inducing activity as estimated in HL-60 leukemia cell model. To examine how their increased differentiation-inducing activity is regulated we tested the hypothesis that membrane-mediated events, unrelated to nVDR, take part in the differentiation in response to PRI-1906 and PRI-2191. The induction of leukemia cell differentiation in response to the analogs of vitamin D was inhibited by LY294002 (phosphatidylinositol 3-kinase inhibitor), PD98059 (inhibitor of MEK1,2, an upstream regulator of extracellular-signal regulated kinase) and rapamycin (p70S6K inhibitor) pointing out that activation of signal transduction pathways unrelated to nVDR is necessary for differentiation. On the other hand, inhibition of cytosolic phospholipase A2 accelerated the differentiation of HL-60 cells induced by either 1,25-D3 or by the vitamin D analogs suggesting possible existence of a feedback loop between extracellular-signal regulated kinases and phospholipase A2.
