Polyglutamine (polyQ) diseases comprise a group of nine genetic disorders that are caused by the expansion of the CAG triplet repeat, which encodes glutamine, in unrelated single genes. Various oligonucleotide (ON)-based therapeutic approaches have been considered for polyQ diseases. The very attractive CAG repeat-targeting strategy offers selective silencing of the mutant allele by directly targeting the mutation site. CAG repeat-targeting miRNA-like siRNAs have been shown to specifically inhibit the mutant gene expression, and their characteristic feature is the formation of mismatches in their interactions with the target site. Here, we designed novel single-stranded siRNAs that contain base substitutions and chemical modifications, in order to develop improved therapeutic tools with universal properties for several polyQ diseases. We tested these ONs in cellular models of Huntington's disease (HD), spinocerebellar ataxia type 3 (SCA3) and dentatorubral-pallidoluysian atrophy (DRPLA). Selected siRNAs caused the efficient and selective downregulation of the mutant huntingtin, ataxin-3 and atrophin-1 levels in cultured human fibroblasts. We also prove the efficiency of novel ONs, with chemical modification pattern mainly containing 2'-fluoro (2'F), in HD mouse striatal cells.

Mitochondrial DNA polymerase gamma (POLG) is the only DNA polymerase involved in maintaining the mitochondrial genome. Recent studies demonstrated an association of CAG repeat polymorphism in the second exon of POLG gene with the risk of cancer. We investigated the CAG repeat variability in the POLG gene in tumor and non-tumor tissues from colorectal cancer patients and in DNA samples isolated from blood obtained from age-matched healthy persons. Somatically occuring CAG-repeat alterations in cancer tissues have been observed in 10% of patients, but no association has been found between the CAG repeat variants in the POLG gene and colorectal cancer risk.

Numerous human transcripts contain tandem repeats of trinucleotide motifs, the function of which remains unknown. In this study we used the available gene expression EST data to characterize the abundance of a large group of these transcripts in different tissues and determine the mRNAs which had the highest contribution to the observed levels of transcripts containing different types of the CNG repeats. A more extensive characteristics was performed for transcripts containing the CUG repeats, and those encoding the repeat-binding proteins. The scarcity of double-stranded CUG repeats as well as various proportions of the single-stranded and double-stranded CUG repeat-binding proteins were revealed in the studied transcriptomes. The observed correlated levels of transcripts containing single-stranded CUG repeats and of proteins binding single-stranded CUG repeats may imply that in addition to transcripts which only provide binding sites for these proteins there may be a substantial portion of the transcripts whose metabolism is directly regulated by such proteins. Our results showing a highly variable composition of triplet repeat-containing transcripts and their interacting proteins in different tissues may contribute to a better understanding of the mechanism of RNA-mediated pathogenesis in triplet repeat expansion diseases.

Introduction. Huntington’s disease is a rare neurodegenerative disease, inherited in an autosomal dominant manner. Every child in the family whose parent is a carrier of the mutant gene has a 50% risk of inheriting the disease. Genetic tests unambiguously confirm whether a person at risk is ill or not. Symptoms include movement, neuropsychiatric and cognitive disorders. Currently, the disease is incurable and there are no effective methods for its treatment. Aim. The aim is to present information about Huntington’s disease, its inheritance, symptoms and pathologies, as well as to draw attention to its unique impact on patients and their families. Material and methods. A literature review of the following databases has been conducted: PubMed, Science Direct, EBSCO, Springer Link. Results. Huntington’s disease, due to the autosomal dominant inheritance, disturbs the whole family system. Over several generations, a family can struggle with the problems of taking care of several patients at the same time, providing children with information about the risk of falling ill, making decisions about genetic testing, and starting a family or having children. Conclusion. Huntington’s disease is a challenge for healthcare professionals who are not always prepared to solve unique, multi-generational problems in families with Huntington’s disease.

24th Colloquium of Geological Society of Africa covered a wide range of fields in geology, from basic research and mineral resources geology to hydrogeology and geotourism. Presentations delivered at the colloquium confirmed vast scale of mineral wealth of Africa, showing at the same time that any substantial further progress in identifying new resources is hampered by the lack of good geological maps at appropriate scales. Some major problems in exploitation of mineral resources in Africa are related to artisanal and small scale mining (ASM). On one hand, ASM provides an income that is sufficient to cover basic needs for tens of millions of Africans, and on the other hand-its environmental impact may be very harmful, especially in the case of mining and processing of gold-bearing ores. Geological research in Africa is usually supported scientifically and financially by EU countries, USA, Canada, Australia as well as China. This is due to the lack of local specialists and sufficiently equipped laboratories practically everywhere except for RSA. Also two papers presented by the Author and his team gave results of research carried out in Angola and Zambia mainly thanks to assistance projects financed by the Polish Aid Programme of the Ministry of Foreign Affairs of the Republic of Poland.

INTRODUCTION: : Coronary artery disease (CAD) leads to mortality and morbidity globally. Approximately, 18 million deaths occurred due to cardiovascular diseases. Objective: To determine the association between knowledge about coronary angiography and anxiety level among patients undergoing coronary angiography. MATERIAL AND METHODS: Analytical cross-sectional study was conducted in two major public-sector hospitals between September 2017 to March 2018. A total of 264 patients were recruited in the study Rusing a non-probability convenience sampling technique. A modified questionnaire about coronary angiography was distributed among participants and the HADS scale was used to assess the anxiety level of the participants. SPSS version 22 was used for data analysis. RESULTS: The mean knowledge score of the participants about angiography was 17.88 ± 4.047. The total knowledge score of both hospitals was good knowledge (6%), fair knowledge (42%) and had poor knowledge (52%). Regarding anxiety level, the participants of both hospitals experienced a high level of anxiety i.e. mild (20%), moderate (34%) & sever (10%). A significant association was found between knowledge and anxiety levels among the participants of both hospitals with a p-value of <0.001. CONCLUSIONS:This study found a significant association between patient’s knowledge about coronary angiography and anxiety level among patients undergoing coronary angiography. The study puts forwards that nurses shall assess the patient’s knowledge before coronary angiography procedure. The provision of accurate knowledge renders categorical purpose to this procedure for salubrious patients’ outcomes.

The article describes the role and types of cooling lubricants used in grinding processes. Conventional methods of coolants delivery into the grinding zone were described, such as: flood method, high-pressure jet method, centrifugal coolant provision method as well as methods method to minimize coolant flow rate: minimum quantity lubrication (MQL), minimum quantity cooling (MQC) and minimum quantity cooling lubrication (MQCL). Against this background, a methods were presented to completely eliminate grinding fluid from the grinding process (dry grinding) like: impregnated grinding wheels and cold air guns (CAG).
