Opis:
Stigmas isolated from flower buds of ‘Nakielska’ variety of Sinapis alba were used to develop a micropropagation method suitable for breeding of new cultivars. The origin of adventitious bud regeneration was studied on MS medium, under stimulation by bezylaminopurine (BAP) in combination with 2,4-D - naphtoxy acetic acid (2,4-D). Histological analysis showed the structure of Sinapis stigma (composed from four types of tissue: papillae, transmitting tissue, parenchyma and vascular bundles) and revealed that numerous meristematic centers developed from parenchyma cells in close vicinity of vascular bundles. Buds very quickly appeared on the surface of initial explants and later formed multiplantlets that were easily rooted in the soil.

The study was conducted in 2006 – 2008 at the Production and Experimental Station of the University of Warmia and Mazury in Olsztyn, located in Bałcyny (NE Poland). The objective of this study was to determine the microbial quality of soil after Brassicaceae grown as forecrops for winter wheat. A field experiment was established on grey-brown podsolic soil, and it involved the following forecrops: winter rapeseed, spring rapeseed, white mustard, Chinese mustard, and winter wheat as control. Soil samples for microbiological analyses were collected in the spring, before the sowing of forecrops, and in the autumn, after the harvest of Brassicaceae and ploughing-in crop residues. Bacterial and fungal communities isolated from soil sown with Brassicaceae as forecrops were generally more abundant and diverse. These communities exerted an inhibitory effect on the growth of soil pathogens. Forecrops with the greatest microbiological potential were white mustard and winter rapeseed.

Opis:
A study into the sanitary state of roots and culm base of winter wheat was carried out in 1999-2002 in the Production and Experimental Station in Balcyny near Ostróda. Experimental wheat was cultivated after spring cross plants such as spring oilseed rape (Brassica napus ssp. oleiferus Metz.), white mustard (Sinapis alba L), Chinese mustard (Brassica juncea L.), oleiferous radish (Raphanus sativus var. oleiferus L.), false flax (Camelina sativa L.), crambe (Crambe abbysinica Hoechst.) and after oats (Avena sativa L.) as a control. The other experimental factor was the method of after-harvest residue management, i.e. ploughing in the stubble, ploughing in the stubble and straw, ploughing in the stubble and straw with nitrogen added. The occurrence of root rot and stem base diseases was affected by weather conditions and forecrop species. Winter wheat roots were attacked to the lowest degree when spring rape and radish were used as forecrops, and to the highest degree - when grown after oat. The culm base was most intensely infected with fusarium foot rot (Fusarium spp.). The remaining root-rot diseases occurred every year but with different intensity. The method of utilization of after-harvest residues did not have a clear effect on the intensity of infection of the roots and culm base of winter wheat.

Opis:
Cochlearia polonica Fröhl. (Brassicaceae) is one of the rarest species in the Polish and European flora and a taxon endemic to a very small area in southern Poland. Due to industrial activities and subsequent transformation of habitats it was extinct in all natural localities around 1994. The persistence of the species was ensured thanks to the active protection efforts including a series of transplantations based on the material from the last and decreasing natural population. The history of conservation efforts of C. polonica provides a model example of successful active protection in the European flora. Here, we provide a complete review comprising the following aims: (i) outline of the discovery and taxonomic conceptions on C. polonica, (ii) review of conservation efforts aimed at preserving its populations, (iii) description of the existing population resources, and (iv) analysis of the genetic structure of all existing populations based on previously published data and new, supplementary results.

In Iran, Descurainia sophia, Malcolmia africana, and Thlaspi arvense are abundantly found as importunate weeds in winter cereal. Understanding the timing of seed germination under natural conditions is crucial for learning how to manage these annual weeds. Therefore, this study was conducted to evaluate the effect of soil burial, dry storage, cold stratification, KNO3, GA3, and scarification on the seed dormancy and germination of these three species. Species had significantly different responses to the treatment. In D. sophia, seeds buried at a depth of 10 cm for 60 days (55%), and seeds dry stored at 20°C for 180 days (45%) showed the highest level of germination. In M. africana, the germination percentage reached 95% when seeds buried at a depth of 1 cm were soaked in a GA3 concentration of 150 ppm. T. arvense had the lowest level of germination compared to the other species. The highest percentage of T. arvense germination was obtained in seeds treated with 150 ppm GA3. Potassium nitrate partly increased germinability in seeds of M. africana, which initially were less dormant than those of T. arvense and D. sophia.

The experiment was carried out in the years 2006-2008 in Bałcyny (N=53o35`49”; E=19o51`20”). The aim of this study was to determine the effect of sulfur fertilization on the sanitary state of spring oilseed rape, winter oilseed rape, white mustard and Chinese mustard as well as on the species composition of fungi colonizing their seeds. Sulfur fertilization had a beneficial effect on the health of Brassicaceae plants infested by Alternaria blight, grey mould, Sclerotinia stem rot, Phoma stem canker and Verticillium wilt, but it had a varying effect on the occurrence of powdery mildew. Alternaria alternata and Penicillium spp. were isolated most frequently from Brassicaceae seeds. In general, more fungi (including pathogenic to Brassicaceae) were isolated from the seeds of plants grown in non-sulfur fertilized plots. Pathogens occurred primarily on the seed surface, and their number decreased after surface disinfection of seeds.

Isatis concstricta Davis (an endemic plant of Turkey) suffers from low propagation rates under natural conditions and is threatened due to fast unplanned urbanisation. The study compared the effects of variants of BA + NAA on shoot regeneration on leaf and petiole explants excised from one week old in vitro regenerated seedlings. MS medium containing 1 mg L–1 BA + 1 mg L–1 NAA induced maximum proliferation on petiole and leaf explants with 13.33 and 12.75 shoots per explant repectively. However, leaf explant induced shoots were sturdy and healthier compared to petiole explant induced shoots. These shoots were rooted on MS medium containing 0.5 mg L–1 IBA and the plants were acclimatized in peat moss and sand (v/v). They grew to flowering under ex vitro conditions. This system of regeneration is advantageous for conventional propagation and the results will help in establishment of a powerful and meaningful micropropagation system for I. constricta.

Opis:
Supercritical fluid extraction (SFE) both with CO2 and CO2/hexane has been developed as a method of analysis for lipids in cruciferous oilseed, rapeseed products and lupin. The SFE procedure used for quantitative determination of oil was compared to traditional Soxhlet oil extraction. The quantitative values obtained with SFE were equal to those obtained with the Soxhlet procedure with respect to oil and fat contents. The oil obtained by SFE had a much lower phosphorus content (below 14 mg/kg compared to 200-530 mg/kg in oil obtained with Soxhlet). When adding hexane to the supercritical fluid in concentrations varying from 50 to 150 mL/L CO2, the solubility of oil in the fluid could be raised up to 600%, from about 30 mg/g without addition of modifier to nearly 200 mg/g when using continuous addition of 150 mL hexane/ L CO2. The phosphorus content was still lower than in oil from Soxhlet (20-64 mg/kg). Therefore, extraction of oil with SFE opens the possibility for selective removal of oil before separate extraction and determination of amphiphilic compounds, e.g. phospholipids. The carotenoid and chlorophyl content in different oils from SFE and Soxhlet were evaluated by UV-VIS determinations. The presence of other amphiphilic compounds was verified and sinapic acid was found to be a quantitatively dominating phenolic compound.
