The aim of the study was to determine the seroprevalence of Leptospira spp. and Borrelia burgdorferi sensu lato in healthy horses living in 7 provinces of central Italy. In the period 2007-2009, sera from 386 horses were tested by microagglutination test (MAT) to detect antibodies to Leptospira spp., employing the following serovars as antigens: Bratislava, Ballum, Canicola, Icterohaemorrhagiae, Grippotyphosa, Hardjo, Pomona, Tarassovi. 3 animals were positive for the serovars Icterohaemorrhagiae, 2 to Bratislava, and 1 to Pomona, for a total 1.5% seroprevalence. All sera were examined by immunofluorence antibody test (IFAT) to reveal anti-B. burgdorferi s.l. antibodies. 94 (24.3%) horses were positive with antibody titres ranging from 1:64 to 1:1,024. The seroprevalence was significantly higher in >10 year-old horses compared to younger subjects. No significant differences in the mean seroprevalence were observed in the respective years. The total mean seroprevalence were strictly related to the environmental conditions of the areas in which the horses lived. No crossreactions between Leptospira and Borrelia were observed. This is the first serological survey on antibodies to B. burgdorferi s.l. in Italian horses.

Opis:
Lyme borreliosis is an infectious disease caused by Borrelia burgdorferi sensu lato spirochetes and is transmitted to humans by the bite of an infected tick from the Ixodidae family, e.g. Ixodes ricinus, Ixodes scapularis, Ixodes persulcatus. The same species are responsible for causing Lyme borreliosis: B. burgdorferi sensu stricto, B. afzelii, B. garinii, B. bissettii and B. spielmanii. The infection develops in stages and has various manifestations in the human body. Lyme disease mainly affects the skin, joints, the nervous system and the heart. Different species are bound with different clinical manifestations: Borrelia afzelii usually cause acrodermatitis chronica athrophicans, Borrelia burgdorferi sensu stricto is a common agent of arthritis, neuroborreliosis can be caused by B. Burgdorferi sensu stricto, B. afzelii and B. garinii. Lyme disease is diagnosed based on symptoms, epidemiological history and laboratory tests, e.g. serological tests of the serum or PCR tests from patient tissues or ticks. The most common diagnostic method is the one recommended by the Robert Koch Institute in Berlin. It is a two-stage procedure based on testing by screening assay in the first stage (EIA, IIFA, ELISA tests) and verifying positive and borderline results by Western blot tests during the second stage. Due to high antigenic variability, Borrelia burgdorferi sensu lato spirochetes have an ability to avoid defence mechanisms of an infected organism. This is the main cause of problems with interpretation of the clinical symptoms, diagnostics results and also with treatment of this infection.
Borelioza jest chorobą zakaźną wywoływaną przez krętki Borrelia burgdorferi sensu lato. Do zakażenia tą bakterią dochodzi zwykle podczas ukąszenia przez zarażonego kleszcza z rodziny Ixodidae, m.in. Ixodes ricinus, Ixodes scapularis, Ixodes persulcatus. Za czynniki etiologiczne boreliozy uznawane są następujące genogatunki: B. burgdorferi sensu stricto, B. afzelii, B. garinii, B. bissettii oraz B. spielmanii. Bakteria ta w organizmie człowieka może atakować różne narządy: skórę, stawy, ośrodkowy układ nerwowy, serce. Poszczególne szczepy wiązane są z rozwojem różnych obrazów choroby: Borrelia afzelii powoduje zazwyczaj rozwój przewlekłego zanikowego zapalenia skóry, Borrelia burgdorferi sensu stricto jest najczęściej odpowiedzialna za rozwój zapalenia stawów, neuroboreliozę może powodować zarówno B. burgdorferi sensu stricto, B. afzelii, jak i B. garinii. Boreliozę diagnozuje się na podstawie objawów klinicznych, wywiadu epidemiologicznego oraz testów laboratoryjnych, takich jak testy serologiczne w surowicy, a także badania metodą PCR tkanek pacjenta lub organizmu kleszcza odpowiedzialnego za ukąszenie. Najczęściej praktykowanym postępowaniem diagnostycznym jest rekomendowana przez Instytut Roberta Kocha w Berlinie tzw. diagnostyka dwuetapowa boreliozy, polegająca na wykonaniu czułego testu przesiewowego w pierwszym etapie (testy EIA, IIFA, ELISA) oraz weryfikacji dodatnich i wątpliwych wyników w drugim etapie (test Western blot). Ze względu na możliwości zmiany swojej struktury antygenowej krętki Borrelia burgdorferi sensu lato mają zdolność do unikania mechanizmów obronnych zainfekowanego organizmu. Jest to główna przyczyna problemów w interpretacji objawów klinicznych, wyników badań laboratoryjnych, a także leczenia zakażenia tą bakterią.

Borrelia burgdorferi sensu lato (s.l.) are tick-transmitted spirochaetes of veterinary and human importance. Molecular epidemiology data on ruminants are still lacking in most countries of the world. Therefore, the aim of this study was to estimate the rate of B. burgdorferi s.l. infection in ruminants from Tunisia. A total of 1,021 ruminants (303 goats, 260 sheep, 232 cattle and 226 camels) from different bioclimatic areas in Tunisia were investigated for the presence of B. burgdorferi s.l. DNA in blood by real time PCR. Prevalence rates were 30.4% (92/303) in goats, 6.2% (16/260) in sheep, 1.3% (3/232) in cattle, and 1.8% (4/226) in camels. Only tick species belonging to Rhipicephalus and Hyalomma genera were found on the investigated animals. In small ruminants, the prevalence of B. burgdorferi s.l. varied significantly according to localities and farms. Goats located in humid areas were statistically more infected than those located in sub-humid areas. Prevalence rates varied significantly according to age and breed in sheep, and age and tick infestation in goats. This study provides the first insight into the presence of B. burgdorferi s.l. DNA in ruminants in Tunisia, and demonstrates that host species such as goats and sheep may play an important role in natural Lyme disease cycles in this country.

Opis:
Occurrence of co-infections with various pathogens in ixodid ticks creates a risk of increased severity of tick-borne diseases in humans and animals exposed to bite of the ticks carrying multiple pathogens. Accordingly, co-infections in ticks were subject of numerous analyses, but almost exclusively with regard to Ixodes ricinus complex whereas potential tick vectors belonging to other genera were much less studied. Taking into consideration the role of Dermacentor reticulatus in the transmission of various pathogens, we carried out for the first time the comprehensive statistical analysis of co-infections occurring in this tick species. An attempt was made to determine the significance of the associations between 6 different pathogens occurring in D. reticulatus (Tick-borne encephalitis virus = TBEV, Anaplasma phagocytophilum, Rickettsia raoultii, Borrelia burgdorferi s. l., Babesia spp., Toxoplasma gondii), using 2 statistical methods: determination of Odds Ratios (ORs) and the Fisher’s exact test. 634 questing Dermacentor reticulatus ticks (370 females and 264 males) were collected in 2011–2013 by flagging the lower vegetation in 3 localities in the area of Łęczyńsko-Włodawskie Lakeland, situated in the Lublin region of eastern Poland. The presence of individual pathogens was detected by PCR. Ticks were infected most often with Rickettsia raoultii (43.8%), less with TBEV (8.5%), and much less with Babesia spp., Toxoplasma gondii, Borrelia burgdorferi s.l., and Anaplasma phagocytophilum (2.5%, 2.1%, 1.6% and 1.1%, respectively). The locality-dependent variability proved to be significant for TBEV (c2=11.063; P=0.004) and Toxoplasma gondii (c2=11.298; P=0.0035), but not for other pathogens. Two hundred seventy (42.6%) of the examined ticks were infected only with a single pathogen, and 54 (8.5%) showed the presence of dual co-infections, each with 2 pathogens. The most common were dual infections with participation of Rickettsia raoultii (7.41%); next, those with participation of the TBEV (5.21%), Toxoplasma gondii (1.58%), Borrelia burgdorferi s.l. (1.26%), Anaplasma phagocytophilum (0.95%), and Babesia spp. (0.63%). On the total number of 15 possible associations, in 9 cases co-infections occurred whereas in 6 cases they were not detected. Introduction. Ticks (Acari: Ixodida) are vectors and/or reservoirs of many pathogens, i.e. Borrelia burgdorferi sensu lato, Anaplasma phagocytophilum and Babesia microti. These pathogens are ethiological agents of such diseases as Lyme borreliosis, human granulocytic anaplasmosis and human babesiosis. Objective. The aim of the study was to evaluate the role of the Ixodes ricinus in the transmission of Borrelia burgdorferi sensu lato, Borrelia afzelii, Borrelia garinii, Borrelia burgdorferi sensu stricto, Anaplasma phagocytophilum and Babesia microti in Opolskie Province in Poland. Materials and method. DNA from 222 ticks was isolated by the ammonia method. The pair of primers specific to the flagelline gene was used to detect of B. burgdorferi s. l. To detect of genospecies of this spirochete, three pairs of internal primers were used. In turn, two pairs of primers specific to the 16S rDNA gene and the 18S rRNA were used, respectively, for the detection of A. phagocytophilum and B. microti. Borrelia burgdorferi s. l., A. phagocytophilum, and B. microti were detected in 4.5%, 2.7% and 5.4% of examined ticks, respectively. Results and conclusions. Of the ten ticks infected with B. burgdorferi s. l., B. afzelii was found in seven, undefinied genospecies in two, and mixed infection with B. afzelii and B. burgdorferi s. s. in one. The study demonstrated the potential risk of exposure of humans and animals to infections of B. burgdorferi s. l., A. phagocytophilum and B. microti in the examined area of Poland.
