Babesiosis is an emerging, tick-transmitted disease caused by the intraerythrocytic parasite Babesia microti. In immunocompetent individuals, B. microti infection quickly resolves after antibabesial treatment. Immunocompromised patients and those of advanced age experience chronic and relapsing babesiosis, accompanied by severe complications and often, a fatal outcome. In these individuals, B. microti infection may persist despite multiple courses of treatment with antiprotozoal drugs. The increasing incidence of human babesiosis caused by B. microti, coupled with a growing number of immunosuppressed people who do not respond to standard antibabesial therapy, emphasises the need for new therapeutics for this protozoan infection with more effective mechanisms of action. Plasmodione, namely 3-[4-(trifluoromethyl)benzyl]-menadione, acts as a redox cycler and disrupts the redox homeostasis of Plasmodium-infected erythrocytes. The present study was designed to evaluate the potential inhibitory effect of this novel antimalarial compound against intraerythrocytic stages of B. microti in mice. Our results demonstrate that plasmodione did not reduce the level of parasitemia in B. microti-infected mice, indicating that interfering with the parasite redox balance is not an effective strategy to restrict the division of this protozoan. The mechanism of parasite resistance to plasmodione may be based on the differences in the oxidative metabolisms of Babesia and Plasmodium parasites inside infected erythrocytes. The significance of our results is discussed in relation to the development of novel antibabesial drugs based on redox-active benzylmenadiones.

The course of babesiosis in humans is characterized by various intensity levels − from a subclinical level to the severe one − associated with multiple organ failure, which leads to death. The aim of this study was to evaluate the effect of 21-day and 6-month invasion of B. microti on Wistar rats spleen. Histological changes in the rats’ spleen were characterized by swelling of splenic tissue, especially the tissue adjacent to the capsule. In the structure of the white pulp in some rats, high concentrations of lymphocytes occurred. The boundary between the white pulp and red pulp was blurred. In the red pulp structure of rats, a lot of macrophages and extracellular deposits of bilirubin were present. The submicroscopic studies showed that the nuclear matrix was slightly shrunken. In the red pulp fragments of the damaged cells were located in the intercellular spaces. Near these areas, many thrombocytes were visible. The ultrastructural observation also revealed thickened endoplasmic reticulum membranes, local cellular swelling filled with amorphous substance, and digested erythrocytes. B. microti invasion affects the splenic morphology and ultrastructure in rats. The immunological hyperactivity and signs of inflammation indicate an important role of spleen in a fight against parasites.

Introduction and objective. It is easier and non-invasive to obtain faecal samples compared with blood samples. Molecular techniques may enable detection of parasites even in tiny amounts of blood-containing faeces. We aimed to compare the sensitivity of detection of three Babesia species and Hepatozoon canis in blood and faecal samples, including samples derived from naturally infected hosts. Materials and method. Three groups were involved: 1) Nine BALB/c mice infected with Babesia microti sampled during acute (n=3), post-acute (n=3) and chronic phases of infection (n=3); 2) Eight dogs with symptoms of babesiosis; 3) Six red foxes infected with B. vulpes, one fox infected with B. canis, four foxes infected with H. canis. Genomic DNA was extracted from blood and faeces by use of commercial kits and amplified with genus-specific primers in one-step or nested PCR reactions. Selected PCR products were sequenced. Results. No positive results for faecal samples were obtained from H. canis-positive foxes in contrast to Babesia spp. infections. Positive results from PCRs were obtained for all BALB/c mice (100%), five dogs (62.5%) and four of seven foxes (57.1%). Successful sequencing was obtained for six selected murine samples (B. microti), four canine samples (B. canis) and for one fox sample (B. vulpes). The success of B. microti detection in murine faecal samples from acute, post-acute and chronic phases was identical (100%). Conclusions. Detectability of Babesia spp. infections was lower in naturally infected dogs and foxes, compared to experimentally infected mice. Detection of DNA in faecal samples can be useful in the detection of Babesia infection in populations from which blood samples are hard to obtain, but due regard must be given to the possibility that prevalence of infection may be severely underestimated.

The aim of present study was to evaluate acquisition risk of babesiosis in human population exposed to ticks /xodes ricinus by examination of Babesia microti DNA occurrence in ticks of all development stages. The polymerase chain reaction (PCR) was applied to estimate the occurrence of DNA Babesia microti in Ixodes ricinus. The Bab1 and Bab4 primers were used to amplify fragment, 238bp in length, of 18S rRNA gene for small ribosomal subunit. Amplicons were electroforeticaly separated in agarose gels. Ticks were collected in year 1999 and 2000, twice in each year in spring-summer (May-July) and summer-autumn (August-October) seasons from Goleniów Forest and Pobierowo. These places have been classified as people attendance and tourist areas. The 716 /. ricinus ticks were collected in 1999 with 61.3% of nymphs, 17.8% larvae, 10.9% females and 9.9% males. Highest range of infection was observed in females — 28.8% studied, than males — 18.3%, nymphs — 7.7% and larvae — 3.1%. The total number of 416 /. ricinus was collected in year 2000 with 64% of nymphs, 13.4% males, 11.9% females and 10.7% larvae. The infection with Babesia microti occurred only in three nymphs, which was 0.7% of studied population.

In this study we report the usefulness of nested PCR for screening of the persistent B. microti infections in rodent hosts. Female BALB/c mice were inoculated with 100 μl of donor blood infected with B. microti. Infections were detected using microscopic examination of Giemsa-stained blood smears. To determine whether B. microti DNA was present in blood and/or spleen tissue, nested PCR was performed targeting a specifi c fragment of the gene encoding the 18S rRNA. Blood was sampled every 10 days post-infection (dpi) until day 30, after which mice were sampled every 30 days until the end of experiment at 360 dpi. The most extensive parasitaemia (39% of infected erythrocytes) was observed at 10 dpi. Between 20-60 dpi, less then 1% of infected erythrocytes were detected in blood smears, and from 90 dpi onwards, infected erythrocytes were no longer observed. B. microti DNA was successfully amplifi ed from the blood of mice from 10 dpi until 180 dpi, as well as from spleens of infected mice at 10 and 20 dpi. The presented results show that nested PCR is the method of choice for monitoring infections of B. microti in the blood of rodent hosts, and could therefore be a tool for environmental monitoring of naturally infected rodents which are the predominant source of infection for tick vectors.
