Introduction and aim. The synthesis of heterocyclic compounds containing oxygen and nitrogen is profoundly intriguing due to their mechanistic implications in both research and development within organic chemistry and drug discovery. The primary aim of this study is to fabricate a range of pharmacologically active drugs containing the isoxazole moiety. Material and methods. The synthesis of new derivatives of isoxazole was achieved through a one-pot condensation reaction of 2-[(Substituted phenyl)hydrazono]malononitrile (1) and 3-[(Substituted phenyl)azo]-2,4-Pentanedione (2) with sodium acetate and hydroxylamine hydrochloride (1:1) in ethanol. All the compounds were screened for their in vitro antibacterial activity, in vitro antioxidant and anticancer activity. The synthesized compounds underwent characterization through FTIR, 1H NMR, and 13C NMR analyses, supported by mass spectral data and elemental analysis. Results. A set of novel isoxazole derivatives was synthesized with a favorable yield. Among compounds 1d, 1e, 2c, 2d, and 2e exhibited notable antioxidant activities. Compounds 1a, 1b, and 1c demonstrated significant anticancer potential against prostate cancer [PC3] cell lines compared to normal HEK cell lines, while 2a displayed the highest inhibitory zone against Escherichia coli. Conclusion. Novel compounds with multifaceted biological activities have been successfully designed, and a synthetic route to create isoxazole derivatives has been devised and verified.

Dietary fibers and phenolic acids are recognized for their various health benefits; thus, cellulose was selected as a carrier polymer of phenolic acids, including gallic acid and caffeic acid. Dried cellulose particles were prepared through the complexation of different amounts of cellulose (2.5, 5, 7.5 and 10%, w/v) with a constant amount of phenolic acids. Additionally, the complexation time was 15 or 60 min with the goal to determine an impact of the duration of complexation on the adsorption of phenolic acids onto cellulose. The prepared particles were stored at ambient temperature for 12 months to assess their storage stability. Cellulose particles were assessed for the amount of adsorbed phenolic acids and antioxidant activities. For both phenolic acids, the same correlation was established, i.e., reduction of adsorption of phenolic acids occurred with the increase of cellulose amount during complexation. The duration of complexation was not a significant factor in the adsorption of phenolic acids. Antioxidant activity generally followed the trend that was obtained for the amount of adsorbed phenolic acids. Comparing both phenolic acids, cellulose had a higher affinity for caffeic acid (4.665 g/kg) than for gallic acid (3.399 g/kg). However, greater stability of gallic acid/cellulose particles was observed throughout the storage. After storage, the content of gallic acid in cellulose particles slightly decreased (up to 3%), while that of caffeic acid decreased from 10 to 20%. The complexation of phenolic acids with cellulose was proven by recording infrared spectra. Formulated cellulose particles can be a valuable tool for the preparation of plant-based functional additives which can be used for the enrichment of products with phenolic acids in order to improve their antioxidant potential and stability.

Due to their nutritional and sensorial characteristics, tropical fruits like guava, mamey sapote, and passion fruit are regularly incorporated into daily diets. Their by-products, especially peels, are approximately 10 to 35% of their weight and possess an important content of bioactive compounds, such as dietary fiber and phenolics. The nutritional, technological, physio-functional properties and phenolic compound compositions of guava, mamey sapote, and passion fruit peels were studied. Peels had promising techno- and physio-functional characteristics, good dietary fiber contents (45.18-61.42 g/100 g), and phenolic profiles with ferulic acid, gallic acid, p-coumaric, and catechin as the main compounds. Peel powders also showed excellent DPPH radical scavenging activity (125.3–252.4 µmol TE/100 g) and Trolox equivalent antioxidant capacity, TEAC (369.2–656.8 µmol TE/100 g). The α-amylase and lipase inhibitory activity varied from 28.15 to 51.4% and 30.89 to 57.15%, respectively. Higher values of α-glucosidase inhibition capacity were found, ranging from 51.64 to 70.32%. The chemical composition and properties reported in the present work suggest that peel powders of these guava, mamey sapote, and passion fruit could be used as constituents in different foods, such as bakery and meat goods, with beneficial health effects like control of hyperglycemia, improved intestinal function, and control of overweight; however, more studies are necessary for animal models and humans to confirm these bioactivities conclusively.

Introduction: Spice plants are a rich source of biologically active compounds. Processes of drying, limits the development of microorganisms and biochemical reactions in raw herbs, at the same time modifies their chemical composition. Objective: The aim of this work was to evaluate the effect of drying process of selected plants on their antioxidant potential and the content of biologically active compounds. Methods: The herbal materials thyme herb (Thymus vulgaris L.), sage leaves (Salvia officinalis L.) and rosemary leaves (Rosmarinus officinalis) were dried using two methods: convective (at 30° C), and freeze-drying. In both, fresh and subjected to drying processes herbs, changes in total polyphenol and antioxidant potential (with the DPPH radical), vitamin C (in the form of ascorbic acid) and chlorophyll pigments contents, were studied. Results: The highest content of polyphenols and vitamin C was recorded in rosemary, while sage was characterized by the highest content of chlorophylls. It was found that the drying processes caused large losses of vitamin C (ascorbic acid) and dyes in comparison to the fresh material. The highest losses of vitamin C occurred in convection dried sage (95%), while most was preserved in lyophilised thyme (losses 74%). Drying had also influenced the reduction of the content of chlorophylls with the largest loss of these compounds, at 94% observed in the freeze-dried rosemary. However, the content of polyphenols increased, underscoring a large impact on the antioxidant activity of herbs. Conclusions: Drying processes resulted in the release of phenolic compounds, so their content increased three to four times in the case of rosemary and sage. The largest increase was observed in the case of lyophilised thyme. Simultaneously, the antioxidant potential was significantly increased. Herbal droughts proved to be a rich source of antioxidant compounds with promising applications as a food additive.

Opis:
The purpose of this study was to evaluate the photochemical, antioxidants and anticancer activity of the medicinal plant Aerva javanica. This plant belongs to the Amaranthaceae family. Locally it is called “bui”. It is a shrub with a long tap root that grows all over India in the wild. The plant extracts were prepared using ethanol, methanol and distilled water as solvents. The antioxidant activity was determined using DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) free radical scavenging activity and IC50 was determined. The total flavonoids compounds found in Aerva javanica ethanolic extract were (0.90 ±0.16) while the total phenolic contents found in ethanolic extract were (0.78 ±0.16), followed by the methanolic and aqueous extract. The antioxidant results of methanolic extract of Aerva javanica showed 0.78 ±0.18 percent inhibition and SCV 49.10% at concentration of 1.5 mg/ml, ethanolic extract showed 0.54 ±0.12 percent inhibition with 64.28% SCV. Phytochemical analysis of best result oriented Aerva javanica extract was done with Gas Chromatography-Mass Spectrometry (GC-MS) technique. The results revealed the presence of different compounds predominantly Acetone (1.18%), Ethyl Acetate (38.95%), (20.77%), n-Propyl acetate (4.09%), Isobutyl acetate (2.71%), (3.84%), isoquinoline,1-[(3,4-diethoxyphenyl)methyl]-6,7-diethoxy- (3.36%), Cyclohexanone (1.43%), 1,1-Diisobutoxy-isobutane (2.02%), n-Hexadecanoic acid (5.61%), Phytol (3.57%), 9-Octadecenoic acid, 1,2,3-propanetriyl ester (10.72%), Octadecanoic acid (1.78%), Bis(2-ethylhexyl) phthalate (3.48%), Squalene (1.40%), 2,2-Dimethyl-3-(3,7,16,20-tetramethyl (1.12%) and 1,6,10,14,18,22-Tetracosahexaen-3-ol (1.195%). The study concludes that extensive research is required to detect more novel compounds in order to develop effective management approaches that significantly reduce the impact of the pathogens on human health as well as on environment.
Celem pracy była ocena fotochemicznej, antyoksydacyjnej i przeciwnowotworowej aktywności rośliny leczniczej Aerva javanica. Ta roślina należy do rodziny Amaranthaceae. Lokalnie nazywa się „bui”. Jest to krzew o długim korzeniu palowym, który dziko rośnie w całych Indiach. Ekstrakty roślinne przygotowano przy użyciu etanolu, metanolu i wody destylowanej jako rozpuszczalników. Aktywność przeciwutleniającą oznaczono za pomocą DPPH (2,2-difenylo-1-pikrylohydrazyl-hydrat) neutralizując wolne rodniki i wyznaczając IC50. Całkowita zawartość związków flawonoidowych znaleziona w ekstrakcie etanolowym Aerva javanica wynosiła (0,90 ±0,16), podczas gdy całkowita zawartość fenoli znaleziona w ekstrakcie etanolowym wynosiła (0,78 ±0,16), a następnie w ekstrakcie metanolowym i wodnym. Wyniki antyoksydacyjne ekstraktu metanolowego z Aerva javanica wykazały 0,78 ±0,18 procent inhibicji, a SCV 49,10% przy stężeniu 1,5 mg/ml, ekstrakt etanolowy wykazał 0,54 ±0,12 procent inhibicji z 64,28% SCV. Analizę fitochemiczną ekstraktu Aerva javanica przeprowadzono techniką chromatografii gazowej ze spektrometrią mas (GC-MS). Wyniki wykazały obecność różnych związków, głównie acetonu (1,18%), octanu etylu (38,95%), (20,77%), octanu n-propylu (4,09%), octanu izobutylu (2,71%), (3,84%), izochinoliny, 1-[(3,4-dietoksyfenylo) metylo]-6,7-dietoksy- (3,36%), cykloheksanon (1,43%), 1,1-diizobutoksy-izobutan (2,02%), kwas n-heksadekanowy (5,61%), fitol (3,57%), kwas 9-oktadecenowy, ester 1,2,3-propanotriylowy (10,72%), kwas oktadekanowy (1,78%), ftalan bis(2-etyloheksylu) (3,48%), skwalen (1,40%), 2,2-dimetylo-3-3,7,16,20-tetrametyl (1,12%) i 1,6,10,14,18,22-tetrakozaheksaen-3-ol (1,195%). Potrzebne są badania mające na celu wykrycie większej liczby nowych związków w celu opracowania skutecznych metod zarządzania, które znacznie zmniejszą wpływ patogenów na zdrowie człowieka, a także na środowisko.

Opis:
Encapsulation of sinigrin in liposomes with the proliposomal method was performed in order to evaluate the effect of this process on in vitro simulated digestion and antioxidant potential of sinigrin. The recovery of sinigrin after simulated gastric and duodenal digestion of its free and liposomal forms was determined with HPLC-UV using human digestive juices. The antioxidant potential of sinigrin and sinigrin-loaded liposomes was determined with the Rancimat test as their ability to prolong oxidative stability of edible oil . The efficiency of 62% was obtained by encapsulating sinigrin in liposomes. The values of mean diameter, polydispersity index and zeta potential showed satisfactory size uniformity and physical stability of the liposomes containing sinigrin. Liposomes were shown to inhibit the digestion of sinigrin in both human gastric and intestinal juices, clearly enabling its prolonged release. Moreover, sinigrin in the liposomal form significantly prolonged the induction time of edible oil oxidation compared to its free form. The results obtained are encouraging from the point of view of a possible incorporation of the sinigrin-loaded liposomes in real functional food systems or their use as nutraceuticals.

Arabinoxylans are an essential component of dietary fiber, and their health-promoting properties are determined mainly by the content and structural features their biopolymers. Rye bread is particularly rich in these compounds; their unique features in the context of content and chemical structure of rye arabinoxylans make it a valuable component od daily diet. Long-term studies have shown the positive effect of these compounds in the aspect of prevention of civilization diseases such as type 2 diabetes, obesity, and cardiovascular diseases. Among the description of the physicochemical properties and diversity of arabinoxylans, the article contains a collection of the most important reports regarding the health-promoting effects of these polymers, as well as their metabolism in the human body.
Arabinoksylany są niezbędnym składnikiem błonnika pokarmowego, a ich właściwości prozdrowotne są determinowane głównie przez zawartość i cechy strukturalne tych biopolimerów. Chleb żytni jest szczególnie bogaty w te związki; ich unikalne cechy w kontekście zawartości i struktury chemicznej arabinoksylanów żyta sprawiają, że jest cennym składnikiem codziennej diety. Długoterminowe badania wykazały pozytywny wpływ tych związków w aspekcie zapobiegania chorobom cywilizacyjnym, takim jak cukrzyca typu 2, otyłość i choroby sercowo-naczyniowe. Wśród opisu właściwości fizykochemicznych i różnorodności arabinoksylanów w artykule zawarto zbiór najważniejszych raportów dotyczących zdrowotnych efektów działania tych polimerów, a także ich metabolizmu w organizmie człowieka.

The aim of this work was to determine the total phenolic content (TPC) and the antioxidant activity of methanol extracts of Hericium erinaceus, Hericium americanum and Hericium coralloides, including free radical scavenging method (DPPH), ferric reducing antioxidant power (FRAP) and radical cation scavenging method (ABTS). Hericium spp exhibited moderate to high antioxidant activity. The highest TPC (3.27 ±0.01 mg GAE g–1) and antioxidant activity values (17.0 ±0.68 mmol TE g–1 in FRAP; EC 50 = 4.12 ±0.12 mg mL –1 in DPPH •; EC 50= 2.83 ± 0.10 mg mL–1in ABTS•+ ) were found for methanol extracts of H. coralloides. The TPC and antioxidant activity of H. erinaceus isolates varied from strain to strain. H. americanum possessed considerably lower total phenolic content (2.31 ±0.01 mg GAE g–1) and antioxidant activity (10.5 ±0.59 mmol TE g–1 in FRAP; EC 50 = 7.82 ±0.09 mg mL –1 in DPPH•; EC 50 = 6.36 ± 0.12 mg mL–1 in ABTS •+) than H. coralloides and H. erinaceus. A high correlation was determined between TPC and ABTS •+( r2= 0.855), DPPH•(r2= 0.969) and FRAP (r2 = 0.942). According to results obtained in the present study, Hericium spp., especially H. coralloides and some of H. erinaceus isolates, might be promising natural source of antioxidants for food and pharmaceutical industry.

Decalepis hamiltonii Wight & Arn., is a plant species that is endemic to southern parts of India. The aim of this study is to explore the influence of habitat heterogeneity on total phenolics, flavonoids, flavor compound 2-hydroxy-4-methoxy benzaldehyde (2H4MB) and antioxidant potential of tubers. The flavor metabolite 2H4MB was quantified by HPLC using isocratic solvent system (methanol : acetonitrile : water : acetic acid 47 : 10 : 42 : 1) that indicates obvious difference in 2H4MB content of tubers with a maximum of 96.4 ±2.6 and 92.6 ±1.2 mg 100 g–1 dry weight basis (DW) in samples from B.R. Hills and Mysore area of Karnataka, followed by samples from Tirumalai Hills and Kurnool from Andhra Pradesh (89.02 ±0.9 mg 100 g–1 DW), Tamil Nadu (81.6 ±2.4 mg 100 g–1 DW) and Kerala (80.18 ±1.1 mg 100 g–1 DW) of tubers. There was variation in total phenolics, total flavonoids and 2H4MB content of root samples collected from different habitats. Also significant variation in free radical scavenging potential of methanol root extracts was noticed, which is directly proportional to the phenolics, and flavonoids content. Overall, there was 10–16% difference in content of 2H4MB in D. hamiltonii tubers that were collected from different natural habitats, and this habitat heterogeneity has to be considered vital, while using such tubers for edible purposes and food formulations.

In vivo and ex vivo impact of nutritional xenobiotics – acrylamide and sodium nitrates – on plasma antioxidant properties

INTRODUCTION: The thiol (SH) groups present in human blood plasma play an important role in the oxidative/antioxidative homeostasis of the organism. They are susceptible to the adverse actions of different exo- and endogenous factors. Chronic exposure to different xenobiotics, e.g. nitrogen-containing compounds commonly occurring in food, is especially important. The aim of this study was to investigate the effect of acrylamide (ACR) and sodium nitrates (SN) – (V) and (III) – on the plasma antioxidant properties, as reflected by changes in the SH group levels. MATERIAL AND METHODS: The concentration of SH groups was measured by Ellman’s method in blood plasma derived from 62 young people (in vivo model; time t0), and after 1 hour of blood plasma incubation with appropriate ACR and SN (III) concentrations (ex vivo model; time t1). The concentrations used corresponded with their daily intake (DIA – daily intake of acrylamide, and DIN – daily intake of sodium nitrates (V) and (III), respectively), estimated on the basis of a nutritional questionnaire. RESULTS: In both models, acrylamide and nitrates caused a significant decrease in SH group concentrations, but ACR induced stronger changes. The women consumed a greater amount of these nitrogen-containing compounds compared to the men, probably due to their different dietary habits. CONCLUSIONS: The obtained results indicate that these nitrogen-containing xenobiotics are important agents lowering antioxidative plasma potential, hence their intake should be controlled.
WSTĘP: Grupy tiolowe (SH) obecne w osoczu krwi odgrywają ważną rolę w oksydacyjno-antyoksydacyjnej homeostazie organizmu. Są one podatne na niekorzystne działanie różnych czynników egzo- i endogennych. Szczególnie istotnym problemem jest długotrwałe narażenie na różne ksenobiotyki, np. związki zawierające azot, powszechnie występujące w żywności. Celem naszych badań była ocena wpływu akrylamidu (acrylamide – ACR) i azotanu sodu (sodium nitrate – SN) – (V) i (III) – na właściwości przeciwutleniające osocza poprzez pomiar stężenia grup SH. MATERIAŁ I METODY: Stężenie grup SH w osoczu uzyskanym od 62 młodych osób (model in vivo; czas t 0) oraz w próbkach poddanych godzinnej inkubacji z odpowiednim stężeniem ACR i SN (III) (model ex vivo; czas t1) mierzono metodą Ellmana. Stężenia ACR i SN (III) w modelu ex vivo odpowiadały ich dziennemu spożyciu (odpowiednio DIA – daily intake of acrylamide – i DIN – daily intake of sodium nitrates (V) i (III)), oszacowanemu na podstawie kwestionariusza żywieniowego. WYNIKI: W obu modelach akrylamid i azotany spowodowały znaczny spadek stężenia grup SH, ale ACR spowodował silniejsze zmiany. Kobiety spożywały większą ilość związków zawierających azot w porównaniu z mężczyznami, prawdopodobnie z powodu odmiennych nawyków żywieniowych. WNIOSKI: Uzyskane wyniki wskazują, że ksenobiotyki zawierające azot są ważnymi czynnikami obniżającymi potencjał antyoksydacyjny osocza, a ich spożycie powinno być kontrolowane.

In recent years, research of antitumor activity of natural compounds isolated from plant material has increased. Polyphenols have gained significant attention due to their proapoptotic abilities and their involvement in migration and inhibition of metastasis processes. The anticancer effects of polyphenolic extracts of Viburnum opulus fruit against human breast (MCF-7) and cervical (HeLa) cancer cell lines have been confirmed in this study. It was demonstrated that the tested preparations (methanol – M and acetone – A from pomace, juice – J and juice after extraction to the solid phase SPE – PF) show cytotoxic activity and regulate the migration process of cancer cells. The degree of inhibition of cell migration was measured at two times - 24 h and 48 h after addition of the tested preparations. The highest toxicity towards both cell lines was demonstrated by the polyphenol fraction obtained after juice purification SPE (IC50 values at concentration of 63,541 and 19,380 μg/mL for HeLa and MCF cell lines, respectively). At the same time, the same preparation inhibited cell migration the most (nearly 70% compared to controls at both times at the concertation of 15 and 30 μg/mL). All preparations showed the antioxidant ability, but the Viburnum opulus juice (200 and 350 μg/mL) and the preparation after its purification (15 and 30 μg/mL) have larger ability to inhibit the intracellular oxidative stress (30-40%) than preparation obtained from pomace (nearly by 20% at concentration of 20 and 50 μg/mL of M and A). Despite the antioxidative capacity of the preparations, they simultaneously decreased cellular mitochondrial potential. The results obtained indicate the high potential of components of Viburnum opulus polyphenolic compounds can be used in the production of innovative dietary supplements or pharmacological preparations for people with an increased risk or inclination towards developing breast or cervical cancer.

W pracy przedstawiono wyniki badań nad możliwością zastosowania rodnika DPPH do oznaczania właściwości antyoksydacyjnych, stosując chromatograficzne rozdzielanie różnych jego form. Pomiary wykonywane były w układzie faz odwróconych HPLC z detekcją UV-DAD. Zbadano CPA związków o właściwościach przeciwutleniających – flawonoidów, jak również całkowity potencjał antyoksydacyjny wybranych naparów z ziół. Wyniki otrzymane za pomocą RP-HPLC-UV skorelowane zostały z wynikami otrzymanymi w pomiarze fotometrycznym.
The paper presents the results of the possibility of using the DPPH radical to determine antioxidant properties using chromatographic separation of its various forms. The measurements were performed in the RP-HPLC with UVDAD detection. The TAP values were tested for antioxidants - flavonoids, as well as the total antioxidant potential of selected herbs. The results obtained with RP-HPLC-UV were correlated with the results obtained using photometric measurement.
