Opis:
The civilization diseases of the 21st century are non-infectious disorders, affecting a large part of modern society. They are associated with the significant development of industry and technology, and hence with environmental pollution and an unhealthy lifestyle. These factors have led to the development of many civilization diseases, which currently include: cardiovascular diseases, respiratory diseases, diabetes, obesity, malignant tumors, gastrointestinal diseases, mental disorders and allergic diseases. The development of technologies, including modern therapies and new drugs, resulted in increase in life expectancy. This creates a global problem of an aging population with an increasing number of diseases of the old age, i.e. dementias. In addition, sedentary lifestyles and changing diets are the reasons why more and more people develop metabolic diseases, as well as neurological and cognitive disorders characterized by progressive damage to nerve cells and dementia. Currently, problem on a global scale is also the growing resistance to existing antimicrobial drugs. Therefore, the scientists face many challenges related to searching for the causes of these diseases, their diagnosis and treatment. Scientific research conducted at the Department of Biomedical Chemistry at the Faculty of Chemistry of the University of Gdańsk is part of this research trend. In this publication, we discuss various research topics with the long-term aim of solving the problems associated with the diseases mentioned above. The following chapters are dedicated to (i) looking for new effective fluorophores with diagnostic and anti-cancer activity; (ii) designing of new compounds with antibacterial and antiviral activity and their synthesis; (iii) investigating the mechanisms of amyloid deposit formation by human cystatin C and possibilities of inhibition of this process; (iv) designing and studies of compounds activating the proteasome with the potential to suppress the development of neurodegenerative diseases; (v) designing peptide fibrils and hydrogels as drug carriers; (vi) searching for peptide inhibitors of immune checkpoint as potential drugs for immunotherapy; (vii) studying the mechanism of action of selected herpesviruses by determining the structure of viral proteins and (viii) studying the composition of natural glycans and glycoconjugates in order to better understand the mechanisms of interaction of bacteria with the environment or with the host.

This research aimed to measure the impact of provenance on the yield, chemical profile and antimicrobial activity of Rosmarinus officinalis essential oil (EO) collected from cultivated and wild plants from two different regions in Morocco (Fez and Figuig). The chemical composition analysis was accomplished utilizing the GC-MS techniques. Sixteen compounds were identified in the EO of both samples, dominated by 1,8-cineole (32.18%), camphor (16.20%) and α-pinene (15.40%) in cultivated type. The α-pinene (51.19) presents the majority compound in the rosemary samples collected from the wild populations.The antimicrobial activity was investigated by using the broth dilution methods against yeast, four bacterial strains and two molds. Rosemary aerial part provided EO with the highest yield and comes from Figuig. The GC-MS analysis demonstrated the existence of two chemotypes of oils: α-pinene as well as 1,8-cineole/camphor/α-pinene. Both EOs showed good antimicrobial activity against all microbial strains. The essential oil produced by Fez plants was the most effective against the selected microorganisms having MIC values of 0.315–2.5 mg/L.

Opis:
Chitosan as a nontoxic, biodegradable, and biocompatible biopolymer with film-forming properties can also be modified to improve its parameters. Modification of polymer films by the addition of nanoparticles is an increasingly common solution due to the higher efficiency of products at the nanoscale compared to the macroscale. In this work, thin chitosan films enriched with biogenic zinc oxide nanoparticles (ZnONPs) from Fusarium solani IOR 825 were obtained by the solvent evaporation method. The influence of nanoadditive on the physicochemical, mechanical, and antimicrobial properties of the polymeric matrix was evaluated. Two different concentrations of ZnONPs were added to the chitosan solution. Spectrometric measurements, mechanical tests, microscopic imaging, and microbiological tests were performed for nanoparticlesmodified and control samples. Analysis revealed that ZnONPs influence the properties of chitosan films. FTIR spectroscopy showed changes that are the result of interactions between polymer matrix and the additive. Modified samples were characterized by increased values of Young’s modulus and tensile strength. SEM analysis combined with energy-dispersive X-ray spectrometry confirmed the presence of zinc in the modified films. The addition of nanoparticles slightly affected the surface morphology of the tested samples, and an increase in roughness was observed. Microbiological tests showed the biostatic activity of the films containing ZnONPs. The obtained films based on chitosan with the addition of ZnONPs can be considered easy-to-obtain biomaterials with potential use as cosmetic and biomedical products.

Celem pracy było zbadanie aktywności przeciwdrobnoustrojowej tkanin bawełnianych przeznaczonych na podszewki do obuwia. Badaniem objęto próbę kontrolną bez wykończenia przeciwdrobnoustrojowego oraz cztery próby tkaniny bawełnianej modyfikowane preparatami zawierającymi w swoim składzie środki nadające materiałom właściwości hydrofobowe i przeciwdrobnoustrojowe. Badania wykonano w odniesieniu do dwóch bakterii: Staphylococcus aureus i Escherichia coli oraz grzybów Aspergillus niger i drożdży Candida albicans. Tkaniny bawełniane modyfikowane preparatami zawierającymi Sanitized T99-19 wykazywały dobry efekt hamujący wzrost bakterii S. aureus i E. coli. Poza tym tkaniny te wykazywały średni lub dobry efekt przeciwgrzybowy wobec grzybów A. niger i drożdży C. albicans. Natomiast tkanina modyfikowana preparatem zawierającym Sanitized TH22-29 wykazała bardzo dobry efekt hamujący wzrost bakterii oraz charakteryzowała się bardzo dobrymi właściwościami przeciwgrzybowymi. Dodatek 4-5% preparatu Sanitized T99-19 lub Sanitized TH22-29 w sposób wystarczający nadaje właściwości przeciwdrobnoustrojowe tkaninom bawełnianym.
The aim of the study was to investigate the antimicrobial activity of cotton fabrics intended for footwear linings. The study included a control sample without antimicrobial treatment and four samples of cotton fabric modified with preparations containing agents that give the materials hydrophobic and antimicrobial properties. The tests were performed for two bacteria: Staphylococcus aureus and Escherichia coli, as well as Aspergillus niger and Candida albicans yeast. Cotton fabrics modified with preparations containing Sanitized T99-19 showed a good inhibitory effect on the growth of S. aureus and E. coli. In addition, these fabrics showed a moderate or good antifungal effect against A. niger and C. albicans. The fabric modified with Sanitized TH22- 29 preparation showed a very good inhibitory effect on the growth of the tested bacteria and was characterized by very good antifungal properties. The addition of 4-5% Sanitized T99-19 or Sanitized TH22-29 is sufficient to impart antimicrobial properties to cotton fabrics.

Chitin is a naturally occurring polymer. Together with its derivatives such as chitosan, it has a wide spectrum of application possibilities, and many properties not yet exploited. Chitosan possesses many features desirable in an ideal antimicrobial polymer. It shows activity against multidrug-resistant bacterial and fungal strains that pose a challenge to modern medicine. Chitosan also shows activity against certain viruses, such as SARS-CoV-2. It might be used as a drug or a vaccine delivery system, is biodegradable, bioavailable and considered safe for medical use. It is important to continue exploring the potential of chitosan, as well as to investigate its sources. Indeed, many sources of this polymer are still not or have been poorly described. In this paper, we compile the current state of knowledge on the antimicrobial properties of chitosan, list alternative sources of chitin to highlight the potential of these two polymers and encourage further research.

This study aims to examine the growth characteristics of B. cereus and B. zhangzhouensis and to see their potential in preventing the growth of Aeromonas hydrophila in vitro. This research was conducted in June 2020 - May 2021 at the Laboratory of Microbiology and Biotechnology, Faculty of Fisheries and Marine Sciences, Padjadjaran University. The method used is the exploratory method and the data obtained were analyzed descriptively. The growth curve was performed using the turbidimetric method. The inhibition test used the disc diffusion method with 5 treatments, chloramphenicol antibiotics (4000 ppm), distilled water, bacterial culture, 100% supernatant and 50% supernatant. The curve results showed that the growth of B. cereus and B. zhangzhouensis was slow. The stationary phase of B. cereus occurred at 40-72 hours, while in B. zhangzhouensis the stationary phase occurred at 48-72 hours. The results of the inhibition test showed that the antimicrobial activity of B. cereus and B. zhangzhouensis was weak with a value range of 0.71 - 1.6mm, besides that the results of the inhibition zone produced by these two bacteria have different inhibitory abilities in each treatment.

This study investigated the phytochemical composition and antimicrobial activities of ethanol and methanol leaf extracts of Lantana camara Linn against some clinical pathogens. The ethanol and methanol extracts were obtained by soaking each of the powdered leaf in each solvent. The soaked powdered leaf was allowed to stand for four days at room temperature and later filtered using Whatman filter paper. The filtrate was further concentrated using rotary evaporator and then freeze-dried. The minimum inhibitory concentration (MIC) of the ethanol and methanol leaf extracts was carried out using agar well diffusion method. The phytochemical analysis was done using standard techniques. Data were analysed using Analysis of Variance (ANOVA) to test for significance. Means were separated using Duncan’s Multiple Range Test (DMRT). The results of the antimicrobial activity revealed that V. cholerae was the most susceptible while E. coli was the most resistant to plant extracts. The phytochemicals present in the plant leaf had antimicrobial properties and may serve as a good substitute for resistant human pathogens.

Olejki eteryczne ze względu na swoje właściwości przeciwdrobnoustrojowe znalazły szerokie zastosowanie w przemyśle kosmetycznym, farmaceutycznym i spożywczym. Z uwagi na powszechną dostępność olejków eterycznych, ich wysoką skuteczność przeciwdrobnoustrojową oraz bezpieczeństwo, związki te są rozpatrywane również jako zielona alternatywa dla środków ochrony garbowanej skóry. Celem pracy było wyznaczenie aktywności przeciwdrobnoustrojowej garbowanych skór bydlęcych zmodyfikowanych olejkiem tymiankowym w stężeniu 5%. Skóry modyfikowane olejkiem eterycznym wywoływały silny efekt przeciwgrzybiczy w odniesieniu do grzybów Ch. globosum oraz zadowalający lub dobry efekt przeciwbakteryjny w odniesieniu do bakterii E. coli. Zastosowanie do modyfikacji skór olejku tymiankowego w postaci 5% może być alternatywą dla biocydów stosowanych w garbarstwie. Potrzeba jednak dalszych badań m.in. w zakresie wpływu badanego olejku na jakość skór w dłuższym okresie czasu.
Due to their antimicrobial properties, essential oils are widely used in the cosmetics, pharmaceutical and food industries. Due to the widespread availability of essential oils, their high antimicrobial efficacy and safety, these compounds are also considered a green alternative to tanned leather protection products. The aim of the study was to determine the antimicrobial activity of tanned cattle hides modified with thyme oil at a concentration of 5%. Skins modified with essential oil caused a strong antifungal effect against Ch. globosum and a satisfactory or good antimicrobial effect against E. coli. The use of thyme oil in the form of 5% for modification of skins may be an alternative to biocides used in tanning. However, further research is needed, e.g. in terms of the influence of the tested oil on the quality of leathers over a longer period of time.

Human neutrophil peptides (HNPs, alpha-defensins) are a group of six defensins being considered as new antimicrobial drugs because of their multifunctional efficiency against bacteria, viruses and fungi. Regardless of the unique biological properties alpha-defensins are unstable compounds and their activity depends on many physical and chemical factors as well as on the kind of the used cultivation media. This leads to research difficulties and obstructions in their therapeutic application. The purpose of this study was to determine antibacterial activity of alpha-defensin 2 dissolved in pharmacopeia buffers and in parallel to develop selective and accurate analytical tests for identification and assay of alpha-defensin 2 in the course of study. The antibacterial effect of alpha-defensin 2 was determined against the strain Escherichia coli (ATCC 25922). It was found that 10 mg/l of Human neutrophil peptide-2 (HNP-2, alpha-defensin 2) dissolved in pH 9.0 buffer caused 90% inhibition of the bacterial respiratory activity. This buffer was considered as a suitable environment for deploying the antibacterial activity of the alpha-defensin. А selective MS analysis method for identification of alpha-defensin 2 in sample mixtures was developed. Also HPLC method with alternative selectivity was elaborated for identification and assay of mixtures containing alpha-defensin 2 and aminoacids in Mueller Hinton broth. The procedure includes development of system suitability test determination.

Nowadays, antimicrobial finishing is a primary requirement for cotton apparel products. Many synthetic antimicrobial agents were used previously, but due to the non-ecofriendly nature of synthetic antimicrobial agents, natural herbal based antimicrobial agents are used these days. The various herbs used in Indian ayurveda medicines, which possess antimicrobial properties, are often applied to textile materials to impart functional properties required for microbicidal effects. In this work, Gloriosa superba, Cyperus rotundus and Pithecellobium dulce were used as antimicrobial agents. The herbs were extracted with different solvent sincluding methanol, ethanol, benzene and water to analyse the influence of these solvents on the efficacy of antimicrobial activity. These extracts were applied on cotton fabrics by the exhaust method and the degree of antimicrobial activity analysed by the zone inhibition method. All the herbs listed in this paper showed good antimicrobial properties against various microbes. However, not all the solvents used in the extraction showed good results in antimicrobial activity against all the microbes. Methanol extracts showed better antimicrobial activity in all three herbs due to their inherent antimicrobial properties. Thus, it is proposed to use methanol as a solvent for the extraction of these herbs to obtain concentrates suitabe for antimicrobial activity in cotton fabrics.
Obecnie wykończenie przeciwdrobnoustrojowe jest podstawowym wymogiem dla produktów z bawełny. Wcześniej stosowano wiele syntetycznych środków przeciwdrobnoustrojowych, ale ze względu na ich nieekologiczny charakter, obecnie stosuje się naturalne środki przeciwdrobnoustrojowe na bazie ziół. Różne zioła stosowane w indyjskich lekach ajurwedyjskich, które mają właściwości przeciwdrobnoustrojowe, są często nakładane na materiały tekstylne, aby nadać im właściwości funkcjonalne wymagane do działania bakteriobójczego. W pracy jako środki przeciwdrobnoustrojowe zastosowano Gloriosa superba, Cyperus rotundus i Pithecellobium dulce. Zioła ekstrahowano różnymi rozpuszczalnikami, w tym metanolem, etanolem, benzenem i wodą, aby przeanalizować wpływ tych rozpuszczalników na skuteczność działania przeciwdrobnoustrojowego. Ekstrakty te nałożono na tkaniny bawełniane metodą czerpaną, a stopień aktywności przeciwdrobnoustrojowej analizowano metodą hamowania strefowego. Wszystkie zioła wymienione w artykule wykazały dobre właściwości przeciwdrobnoustrojowe przeciwko różnym drobnoustrojom. Jednak nie wszystkie rozpuszczalniki użyte do ekstrakcji wykazały dobre wyniki w działaniu przeciwdrobnoustrojowym przeciwko wszystkim drobnoustrojom. Ekstrakty metanolowe wykazywały lepszą aktywność przeciwdrobnoustrojową we wszystkich trzech ziołach ze względu na ich nieodłączne właściwości przeciwdrobnoustrojowe. Dlatego zaproponowano stosowanie metanolu jako rozpuszczalnika do ekstrakcji ziół w celu uzyskania koncentratów odpowiednich do działania przeciwdrobnoustrojowego w tkaninach bawełnianych.

Purpose: The present research aimed to determine whether and how the aluminium chloride – based materials affect the cell line of the bacterial line and fungi. Methods: Cytotoxicity of haemostatic astringents: Alustat (liquid), Alustat (gel), Alustat (foam), Alustin, Hemostat, Racestyptine and Traxodent containing AlCl3 was conducted on L929 cell line with the use of MTT and SRB assays. The antimicrobial activity (CFU and MIC) against C. albicans, S. mutans, L. rhamnosus was determined. Results: In the MTT results, cell viability for all agents were very low. In SRB, the lowest cytotoxicity was demonstrated for Hemostat and Alustat (foam), Traxodent and Racestyptine. Total reduction of the CFU of S. mutans was observed. Alustat (gel) and Alustat (liquid) completely inhibited the growth of C. albicans, S. mutans and L. rhamnosus. Conclusions: The viability of L929 cells obtained in the SRB assay is more reliable than that obtained in the MTT assay, in the case of gingival haemostatic agents.

This research work involved the selection of biocides for the antimicrobial treatment of leathers. The scope of the research work covered microbiological tests of leather samples tanned utilising the same technology, with microbiological treatment made using 9 different preparations, as well as a test of leather without biostabilisation. Commercial biocides taken for inwestigation were as follows: NS 500 – an aqueous solution of nanosilver 500 ppm, ODH – tea tree oil, and OP – orange oil. Preparations prepared by diluting the purchased products were as follows: S27-22 – an aqueous solution containing silver chloride and titanium dioxide, SH 22-27 – an aqueous solution containing zinc pyrithione, ST 99-19 – an aqueous solution containing dimethyltetradecyl [3-(trimethoxysilyl)propyl]ammonium chloride, SU – an aqueous solution containing polyhexamethylene biguanide, and SGP – a saturated solution of aluminum potassium sulfate. Also a preparation was made by combining NS 500 M250 – 500 ppm nanosilver solution and a 250 ppm copper colloid. Evaluation of the effectiveness of biostabilisation of the leather samples and antimicrobial activity of the preparations used was carried out. Staphylococcus aureus and Escherichia coli bacteria and Candida albicans, Aspergillus brasiliensis and Trichophyton mentagrophytes fungi were used in the study. The assessment of microbial growth on samples was carried out according to Polish standards. Assessment of the resistance to bacteria and C. albicans yeast was made according to the PN-EN ISO 20645: 2006 standard – a method of diffusionon an agar plate. Assessment of the resistance to A. brasiliensis and T. mentagrophytes fungi was made according to the PN-EN ISO 14119: 2005 standard. Among the tested preparations to protect leathers against the effects of microorganisms, overall efficacy was obtained for SH 22-27, NS 500 M 250, SU and ODH.
Praca obejmowała dobór preparatów biobójczych do obróbki antymikrobowej skór. Zakres tematyczny obejmował badania mikrobiologiczne prób skór wyprawionych według tej samej technologii z obróbką mikrobiologiczną wykonaną przy użyciu 9 przygotowanych preparatów, a także test skóry bez biostabilizacji. Dokonano oceny skuteczności biostabilizacji skór i aktywności antymikrobowej użytych preparatów. Spośród testowanych preparatów do ochrony skóry przed działaniem drobnoustrojów najkorzystniejszą ocenę uzyskały: SH 22-27 zawierający pirytionian cynku, NS 500 M 250 zawierający nano cząsteczki srebra i miedzi, SU zawierający poliheksametylenobiguanidynę, ODH zawierający olejek melaleuca.

The aim of our study was to evaluate and compare antioxidant and antimicrobial activity of aqueous, methanol and chloroform extracts of leaves of Allium ursinum (A. ursinum). Total phenol (TPC) and flavonoid (TFC) content in extracts and antioxidant activity using DPPH (1,1- diphenyl-2-picrylhydrazyl) assay were determined spectrophotometrically. In vitro antimicrobial activity was tested by microdilution method. The highest TPC and TFC were observed for chloroform extract. The extracts showed different degrees of antimicrobial activity. The intensity of antimicrobial action varied depending on the group of microorganism and the type of extracts. Our results demonstrated rational basis for the traditional uses of A. ursinum in alleviation of oxidative stress and against various pathogenic microorganisms.

Introduction: In order to prevent increased bacterial resistance, it is important to use herbal medicines with less side effects. Objective: In this study, the chemical composition, antimicrobial, antioxidant activities and SEM images of Lavandula officinalis and Origanum onites oils obtained in two different ways were determined. Methods: The antimicrobial activity of the oils was determined against reference and multiple strains of foodborne and pathogenic bacteria. The each essential oil sample were analyzed by GC–MS. Antioxidant activities of the samples were examined through DPPH• and ABTS•+ radical scavenging and FRAP assays. Results: The results indicate that the oils of obtained from Origanum onites and Lavandula officinalis exhibited relatively strong antibacterial and antifungal activity. In addition, the antioxidant activities detected were remarkable. Morphological changes in bacterial cells treated with essential oil samples were demonstrated with SEM images. In addition, the chemical components of the oil samples obtained by both water steam and cold press were revealed by GC-MS analysis and compared. Conclusion: When all the obtained results are evaluated together; these plants could be suitable for using as antimicrobial and antioxidative agents in several industries such as food, cosmetic, etc.
