Opis:
On account of the interesting pharmacological properties of hydantoin and 2-thiohydantoin derivatives, and other purposes such as textile printing, catalysts for polymerizations, their use in the production of resins and plastics, they have been the subject of chemical and biological studies. In this review article, we focused our interest on the most important methods for the synthesis of hydantoin and 2-thiohydantoin derivatives, different reactions and different application of them. We explored their antitumor, antimicrobial, antiviral, anxiolytic, antidiabetic, anticonvulsant activities, their utilities in treatment of vascular dysfunction, using them as an inhibitor of fatty acid amide hydrolase, using in hyperpigmentation applications, and utilities of them in the production of antimicrobial polyurethane coatings.

This study analyzed chemical composition of the essential oils (EOs) of Thymus vulgaris L., harvested in two regions of northwest Algeria (Tlemcen and Mostaganem), and evaluated their antimicrobial and cytotoxic activity. Fifty one compounds representing 99.99% and 100% of total thyme oil of Mostaganem (EO.TM) and Tlemcen (EO.TT), respectively were identified by GC-FID and GC-MS. Carvacrol was a dominant compound in both oils (89.15 and 82.42%). The antimicrobial activity, demonstrated by disc diffusion and broth macrodilution methods, showed a strong inhibitory effect of the oils on seven reference strains tested and particularly on Bacillus cereus ATCC 10876 and Candida albicans ATCC 10231. A cytotoxicity assay for two normal and two cancer cell lines indicated the highest sensitivity of canine B-cell lymphoma cell line (CLBL-1) for which IC50 values were 35.39 ± 9.0 (EO.TT) and 84.51± 3.2 (EO.TM). Murine fibroblast cell line (3T3), with IC50 116.97 ± 2.7 for EO.TT and 146.29 ± 0.5 for EO.TM was the most resistant line.

Opis:
An important trend in current cosmetic industry is increasing demand for new, biologically active compounds and preservatives of natural origin. These products constitute a major ingredients of natural (organic) cosmetics and usually may also be used in typical cosmetics as functional additives. This work summarizes the perspectives of the use of essential oils as active ingredients and preservatives in cosmetic products and as biopesticides. Brief characteristics of essentials oils, their preparation and biological activity is provided. Literature data suggests that essential oils exhibit broad therapeutic effects including antibacterial, antiseptic, antifungal and antioxidant activity, they can be also used as transdermal enhancers. On the other hand, in essential oil have been found compounds which can be use as a biopesticides. The use of essential oils in cosmetic products is possible, but requires a detailed knowledge regarding their compatibility, active concentration as well as toxicological and skin irritant characteristics. The literature review, presented in this paper, shows the great potential of essential oils as a biologically active preservatives and antioxidants, repellents and transepidermal enhancers.

Opis:
The aim of this study is to exploit a suitable chitosan extraction method from the chitin of indigenous shrimp shells by employing different physicochemical treatments and to improve different bioactive properties of this extracted chitosan (CS) by applying gamma radiation. Chitin was prepared from shrimp shell by pretreatment (deproteination, demineralization and oxidation). Chitosan was extracted from chitin by eight different methods varying different physicochemical parameters (reagent concentration, temperature and time) and assessed with respect to the degree of deacetylation, requirement of time and reagents. The method where chitin was repeatedly treated with 121°C for 30 min with 20 M NaOH, produced the highest degree of deacetylation (DD) value (92%) as measured by potentiometric titration, with the least consumption of time and chemicals, and thus, selected as the best suitable extraction method. For further quality improvement, chitosan with highest DD value was irradiated with different doses (i.e., 5, 10, 15, 20 and 50 kGy) of gamma radiation from cobalt-60 gamma irradiator. As the radiation dose was increased, the molecular weight of the wet irradiated chitosan, as measured by the viscosimetric method, decreased from 1.16 × 105 to 1.786 × 103 , 1.518 × 103 , 1.134 × 103 , 1.046 × 103 and 8.23 × 102 dalton, respectively. The radiation treatment of chitosan samples increased the antimicrobial activity signifi cantly in concentration dependent manner on both gram-positive (Staphylococcus aureus) and gram-negative (Escherichia coli) bacteria, as determined by the well-diffusion method. Four to fi ve percent wet chitosan treated with a radiation dose range of 5.0–10.0 kGy rendered the highest antimicrobial activity with least energy and time consumption. Solubility, water binding capacity (WBC) and fat binding capacity (FBC) also improved due to irradiation of chitosan.

Human neutrophil peptides (HNPs, alpha-defensins) are a group of six defensins being considered as new antimicrobial drugs because of their multifunctional efficiency against bacteria, viruses and fungi. Regardless of the unique biological properties alpha-defensins are unstable compounds and their activity depends on many physical and chemical factors as well as on the kind of the used cultivation media. This leads to research difficulties and obstructions in their therapeutic application. The purpose of this study was to determine antibacterial activity of alpha-defensin 2 dissolved in pharmacopeia buffers and in parallel to develop selective and accurate analytical tests for identification and assay of alpha-defensin 2 in the course of study. The antibacterial effect of alpha-defensin 2 was determined against the strain Escherichia coli (ATCC 25922). It was found that 10 mg/l of Human neutrophil peptide-2 (HNP-2, alpha-defensin 2) dissolved in pH 9.0 buffer caused 90% inhibition of the bacterial respiratory activity. This buffer was considered as a suitable environment for deploying the antibacterial activity of the alpha-defensin. А selective MS analysis method for identification of alpha-defensin 2 in sample mixtures was developed. Also HPLC method with alternative selectivity was elaborated for identification and assay of mixtures containing alpha-defensin 2 and aminoacids in Mueller Hinton broth. The procedure includes development of system suitability test determination.

In this research antimicrobial activity of nanoparticles ZnO on perilous bacteria such as Pseudomonas aeruginosa was evaluated. P. aeruginosa is important pathogen that caused burn wound infections as it is multi-drug resistant and has several virulence factors. Fifteen samples of P. aeruginosa were collected from patients who suffering from Burn infections in Al-Hilla teaching hospital burn unit with the age range between (7-80) years old for both genders. After collecting burn samples, the diagnosis and characterization were performed by culturing and biochemical tests. ZnO NPs were synthesized by chemical method, Zinc oxide nanoparticles are well-known to be one of the multifunctional inorganic compounds which are widely used in medical applications. This study aims to prepare ZnO nanoparticles with particle size ranging from 23-29  nm. In the present study, surface modification of ZnO nanoparticles was performed, and influence of modification of the structure and morphological properties was investigated. The resulting nanoparticles were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM) and atomic force (AFM). Zinc oxide nanoparticles with the average diameter of about 29 nm were modified with an oleic acid to exert more compatibility. From the results obtained it is suggested that modified ZnO-nanoparticles could be used effectively in safety environmental and medical applications. Antibacterial activity for nanoparticle ZnO against P. aeruginosa isolates was measured by: Agar Diffusion Technique and Minimum Inhibitory Concentration (MIC)/Minimum bactericidal Concentration (MBC) with microdilution. The best zone of inhibition was (35.5mm) at a concentration of 40 μg/ml of nano-ZnO in one strain of P. aeruginosa while the lowest inhibition zone was (16 mm) at a concentration of 20 μg/ml of nano ZnO in one strain also. In addition, all P. aeruginosa isolates were completely inhibited at the concentration of 3.7 μg/ml of nano-ZnO (MIC) but no significant antibacterial activity was observed at concentrations less than 1.8 μg/ml of nano-ZnO and the (MBC) was same as MIC (3.7 μg/ml) for all P.aeruginosa isolates.

Five divalent transition metals Cu(II), Co(II), Ni(II), Mn(II) and Zn(II) complexes have been synthesized using 3-hydroxy-4-[N-(2-hydroxynaphthylidene)-amino]-naphthalene-1-sulfonic acid (H3L) Schiff base as a ligand derived from the condensation reaction between 4-amino-3-hydroxynaphthalene-1-sulfonic acid and 2-hydroxy-1-naphthalde-hyde. The synthesized complexes were characterized using microanalytical, conductivity, FTIR, electronic, magnetic, ESR, thermal, and SEM studies. The microanalytical values revealed that the metal-to-ligand stoichiometry is 1:1 with molecular formula [M2+(NaL)(H2O)x].nH2O (where x = 3 for all metal ions except of Zn(II) equal x = 1; n = 4, 10, 7, 4, and 6 for Cu(II), Co(II), Ni(II), Mn(II) and Zn(II), respectively). The molar conductivity result indicates that all these complexes are neutral in nature with non-electrolytic behavior. Dependently on the magnetic, electronic, and ESR spectral data, octahedral geometry is proposed for all the complexes except to zinc(II) complex is tetrahedral. Thermal assignments of the synthesized complexes indicates the coordinated and lattice water molecules are present in the complexes. SEM micrographs of the synthesized complexes have a different surface morphologies. The antimicrobial activity data show that metal complexes are more potent than the parent ligand.

The aim of the research was the solvent-based impregnation of poly(lactic acid) (PLA) withThe present work developed a new and simple method for producing engineered nylon yarns by two-step coating. The nylon yarns were first immersed in a solution containing a silane coupling agent (3-Aminopropyltriethoxysilane) to improve the adhesion properties. Then the modified samples were heat-treated in a solution containing cupric nitrate, hydroxylamine sulfate, and sodium pyrosulfite. Microstructural imaging of the coated surfaces shows a homogeneously formed coating layer without any microscopic cracks or discontinuity in the sample surface. Results showed that this two-step coating can produce nylon yarns with good electrical conductivity and anti-microbial properties. The coated samples displayed very good light fastness and washing fastness to multiple washes in terms of electrical conductivity changes and anti-microbial activity.
W ramach badań opracowano nową i prostą metodę wytwarzania specjalnych przędz nylonowych poprzez powlekanie dwuetapowe. W celu poprawy właściwości adhezyjnych przędze nylonowe zanurzono w roztworze zawierającym silanowy środek sprzęgający (3-aminopropylotrietoksysilan). Następnie zmodyfikowane próbki poddano obróbce cieplnej w roztworze zawierającym azotan miedzi, siarczan hydroksyloaminy i pirosiarczyn sodu. Obrazowanie mikrostrukturalne powlekanych powierzchni pokazało jednorodnie utworzoną warstwę powłokową bez mikroskopijnych pęknięć lub nieciągłości na powierzchni próbki. Wyniki wykazały, że ta dwustopniowa powłoka może wytwarzać przędzę nylonową o dobrej przewodności elektrycznej i właściwościach przeciwdrobnoustrojowych. Pokryte próbki wykazywały bardzo dobrą odporność na światło i odporność na wielokrotne pranie pod względem zmian przewodności elektrycznej i aktywności przeciwdrobnoustrojowej.

Origanum dubium is a valuable wild oregano species of the natural flora of Antalya, Turkey. In this study, we extracted essential oil (EO) by hydro-distillation of the aerial parts of selected O. dubium chemotype with highest EO content, and analyzed by gas chromatography/mass spectrometry. Carvacrol was the primary component (85.9%) among 24 different compounds in the EO. The volatile test showed minimum inhibitory effect of the EO against Xanthomonas axonopodis pv. vesicatoria, Clavibacter michiganensis subsp. michiganensis, Xanthomonas axonopodis pv. phaseoli, Xanthomonas axonopodis pv. phaseoli var. fuscans, and Pseudomonas syringae pv. tomato, at 309, 303, 318, 254, and 901 µL/mL, respectively. A 1-hour treatment of bean and tomato seeds in the volatile phase of the EO effectively sterilized the seeds from bacterial pathogens without inhibiting their germination. To the best of our knowledge, this is the first study on the potential for the use of O. dubium EO as a seed protectant against bean and tomato seed-borne bacterial pathogens.

The synthesis of a novel xanthene derivatives bearing dimedone as an excellent precursor has been achieved by applying one pot three component Hantzsch type condensation. The newly synthesized compounds were characterized by spectral and elemental analyses. All synthesized compounds undergo docking studies and biological screening for antimicrobial activity against Gram-positive bacteria, Gram-negative bacteria and fungal species. Among all the tested compounds, it was found that compound 3c, 3d, 3g and 3h revealed better activities against the Gram-positive rather than the Gram-negative bacteria whereas results of docking studies revealed that compounds 3b, 3g and 3i showed best binding affinity towards ATP binding pocket of Human PIM1 kinase receptor through steric favorable and H-bond interactions.

Opis:
Some new 3-[3-(1-phenyl-3-aryl-1H-pyrazol-4-yl)acryloyl]coumarins 3a-f were synthesized (coumarin chalcones) by the condensation of various 3-acetyl coumarins 1 and appropriate 1-phenyl-3-aryl-1H-pyrazole-4-carbaldehyde 2. These coumarin chalcones 3a-f were then employed for the synthesis of pyrazolyl bipyridinyl substituted coumarins 7a-f, 8a-f, and 9a-f under Krohnke’s reaction condition. The characterization of all the synthesized compounds was carried out by elemental analysis, IR, 1H-NMR, 13C-NMR, DEPT-135 and mass spectral analysis. In addition to that, in vitro antimicrobial competency of the title compounds was assessed against selected pathogens. Compounds 3b, 3e, 7b, 8b, 8c and 9b exhibited excellent antimicrobial activity and said to be the most proficient members of the series.

Opis:
The civilization diseases of the 21st century are non-infectious disorders, affecting a large part of modern society. They are associated with the significant development of industry and technology, and hence with environmental pollution and an unhealthy lifestyle. These factors have led to the development of many civilization diseases, which currently include: cardiovascular diseases, respiratory diseases, diabetes, obesity, malignant tumors, gastrointestinal diseases, mental disorders and allergic diseases. The development of technologies, including modern therapies and new drugs, resulted in increase in life expectancy. This creates a global problem of an aging population with an increasing number of diseases of the old age, i.e. dementias. In addition, sedentary lifestyles and changing diets are the reasons why more and more people develop metabolic diseases, as well as neurological and cognitive disorders characterized by progressive damage to nerve cells and dementia. Currently, problem on a global scale is also the growing resistance to existing antimicrobial drugs. Therefore, the scientists face many challenges related to searching for the causes of these diseases, their diagnosis and treatment. Scientific research conducted at the Department of Biomedical Chemistry at the Faculty of Chemistry of the University of Gdańsk is part of this research trend. In this publication, we discuss various research topics with the long-term aim of solving the problems associated with the diseases mentioned above. The following chapters are dedicated to (i) looking for new effective fluorophores with diagnostic and anti-cancer activity; (ii) designing of new compounds with antibacterial and antiviral activity and their synthesis; (iii) investigating the mechanisms of amyloid deposit formation by human cystatin C and possibilities of inhibition of this process; (iv) designing and studies of compounds activating the proteasome with the potential to suppress the development of neurodegenerative diseases; (v) designing peptide fibrils and hydrogels as drug carriers; (vi) searching for peptide inhibitors of immune checkpoint as potential drugs for immunotherapy; (vii) studying the mechanism of action of selected herpesviruses by determining the structure of viral proteins and (viii) studying the composition of natural glycans and glycoconjugates in order to better understand the mechanisms of interaction of bacteria with the environment or with the host.
