Opis:
A simple one-step procedure for synthesis of 1-methoxy-1-oxoalkan-2-yl salicylates and 1-methoxy-1-oxoalkan-2-yl 2-[(1-methoxy-1-oxoalkan-2-yl)oxy]benzoates by reaction of salicylic acid with several methyl 2-bromoalkanoates was developed. The reactions were carried out in N,N-dimethylformamide (DMF) in the presence of anhydrous potassium carbonate. Conditions for regioselective synthesis of target compounds were established. The developed procedure could be easily applied in the industrial production process. The new salicylic acid derivatives were obtained with satisfactory yields and were characterized by MS and 1H NMR spectra. The fungicidal activity of the prepared compounds was tested in vitro against seven species of plant pathogenic fungi. The best results were observed for 1-methoxy-1-oxoalkan-2-yl salicylates which showed moderate or good activity against Botrytis cinerea and Rhizoctonia solani.

QSAR analysis of a set of previously synthesized 2,5,6-trisubstituted benzoxazole, benzimidazole and 2-substituted oxazolo(4,5-b)pyridine derivatives tested for growth inhibitory activity against Candida albicans, was performed by using the computer-assisted multiple regression procedure. The activity contributions for either heterocyclic ring systems or substituent effects of these compounds were determined from the correlation equation and the predictions for the lead optimization were described. The resulting QSAR revealed that the oxazolo(4,5-b)pyridine ring system with the substitution of a benzyl moiety at position 2 was the most favourable structure among the heterocyclic nuclei. Moreover, the fifth position in the fused ring system is found more significant than the other positions in improving the activity.

An oral cavity hygiene plays a key role in prophylactic and therapeutic measures to prevent pathological changes caused by different viruses, bacteria, fungi and protozoa. It is important to maintain proper oral hygiene and assist the removal of potent pathogens; use of the mouthrinses can be one of method providing to these goal. The purpose of this study was to investigate the effects of selected mouthrinses on the standard strains of Candida presented in the oral cavity. Eight reference strains of fungi were investigated: C. albicans (CBS 2312), C. albicans (L 45), C. albicans (ATCC 24433), C. dubliniensis (CBS 7987), C. glabrata (CBS 862), C. krusei (CBS 573), C. parapsilosis (CBS 10947) and C. tropicalis (CBS 2424). Thirteen mouthrinses were used in the study, including pure chlorhexidine (CHX), and 12 commercially available varieties: Azulan, Colgate Plax Complete Care Sensitive, Corsodyl 0,2%, Curasept ADS 205, Dentosept, Dentosept A, Eludril Classic, Listerine Total care, Octenidol, Oral-B Pro-Expert Clinic Line, Sylveco and Tinctura salviae. The present study used a qualitative diffusion in agar gel-well plate method to evaluated the antifungal properties of mouthrinses. Among the 12 commercially available mouthrinses examined in the study, the following were not found to show antifungal activity: Azulan, Dentosept, Eludril Classic, Listerine Total care, Tinctura salviae. The largest inhibition zones were produced by Dentosept, Chlorhexidine and Colgate. The smallest inhibition zones were produced by Octenidol and Curasept. With regard to mouthwash type, statistically significant differences in growth inhibition zone diameter were found between the following pairs of fungi: C. albicans and C. krusei for Colgate without dilution and with 1:2 dilution; C. albicans and C. glabrata for Corsodryl without dilution and with 1:2 dilution; C. albicans and C. dubliniensis for Dentosept A without dilution; C. glabrata and C. parapsilosis with 1:2 dilution; C. dubliniensis and C. tropicalis for Sylveco without dilution, 1:2 dilution and 1:4 dilution; C. dubliniensis and C. parapsilosis for Sylveco without dilution, 1:2 dilution and 1:4 dilution. The lowest MIC values calculated from the Iinear regression equation, indicating the strongest potential activity, were obtained for Dentosept A, followed by chlorhexidine; the lowest activity, was calculated for Curasept and for Octenidol. Some of the tested mouthrinses have antimycotic properties at commercially available concentrations. In spite of the fact that chlorhexidine is thought to be the gold standard for mouthrinse use, Dentosept has stronger antimycotic activity and acts on a wider spectrum of fungi species. Chlorhexidine and Colgate do not appear to act against C. tropicalis, neither does Curaspet against C.dubliniensis; therefore, the determination of the fungus species is necessary.

Leather products provide an ideal environment for microbial growth. In order to prevent the appearance and spread of various types of microorganisms, it is treated with selected biocides and even essential oils. The aim of this paper was to investigate the antifungal activity of selected essential oils applied to finished chromium tanned leather which is intended for leather fancy products. Antifungal activity was evaluated according to the PN-EN 14119: 2005 standard. Four finished chromium tanned leathers were tested, each of which was soaked in the following essential oils: tea tree essential oil at a concentration of 1, 2 and 5% and thyme essential oil at concentration of 1, 2 and 5%. The results obtained indicated that leather finishing with thyme essential oil showed the antimicrobial activity against Aspergillus niger, Chaetomium globosum and Candida albicans strains. The application of 5% thyme essential oil to leather samples inhibited the growth of all strains tested. Using tea tree essential oil on leather samples showed a antimicrobial effect only against Candida albicans. The application of essential oils in the leather finishing process may be an alternative to biocides used in the tanning industry.

In an effort to develop antimicrobial agents, a series of substituted (E)-1-(4-fluoro-3-methylphenyl)-3-phenylprop-2-en-1-one compounds were synthesized by crossed-aldol condensation reaction of 4-fluoro-3-methyl acetophenone with various substituted benzaldehydes in the presence of sodium hydroxide. The synthesized compounds were characterized by means of their UV, IR and NMR spectral data. The synthesized substituted (E)-1-(4-fluoro-3-methylphenyl)-3-phenylprop-2-en-1-one compounds were tested for their antibacterial and antifungal activities by the Bauer-Kirby disc diffusion method.

The aim of the study was to estimate the effect of actinomycetes of genus Streptomyces on the growth of the antagonistic fungus Trichoderma koningii. 150 strains of Streptomyces spp. isolated from two potato cropping systems were used to the tests. Analyses were conducted experimentally in vitro on PDA medium with pH 6 and 7. The results obtained clearly indicate the inhibitory effect of actinomycetes on the fungus T. koningii. Of the tested population of actinomycetes only two strains did not inhibit the growth of the tested fungus. The strains from the rhizosphere of potato cultivated under the ecological system were predominant in the group of actinomycetes with a very strong effect. pH value appeared to be a crucial factor influencing the inhibition zone. High antagonistic activity in relation to the useful fungus T. koningii noted with numerous strains of Streptomyces spp. is unfavorable in respect of biological control.

Three series of some novel chalcone based urea, thiourea and acetamide derivatives were designed, synthesized and screened for their antimicrobial and antifungal activities. All the synthesized compounds are first reported. The structures of the compounds were elucidated with the aid of elemental analysis and spectral methods including IR, 1H-NMR spectral data. The prepared compounds were evaluated for antibacterial activity against two Gram-positive bacteria (Staphylococcus aureus, Staphylococcus pyogenus), two Gram-negative bacteria (Pseudomonas aeruginosa, Escherichia coli). The title compounds were also investigated for their antifungal activity using the broth micro dilution method. The bioassay results showed that compounds a few compounds showed good to superior in vitro antibacterial and antifungal activity.

Background. During the recent years, appropriate attention has been paid to the oxidative stress which damages the body’s cells, proteins, and DNA. Therefore, the need of antioxidants becomes a therapeutic and preventive priority. In addition, microbial infections also constitute a public health problem. Objective. To find efficient, reliable and safe alternatives sources to synthetic antioxidants, antibiotics and antifungals drugs. Materials and methods. Extract and essential oil of Psidium guajava were screened for their antioxidant and antimicrobial activities against gram positive bacteria (Enterococcus faecalis and Staphylococcus aureus), gram negative bacteria (Citrobacter freundii, Escherichia coli and Pseudomonas sp) and fungi (Candida albicans,Candida tropicalis and Cryptococcus neoformans), as well as to determine the functional groups of phytochemicals present in the essential oil by Fourier transform infrared spectroscopy (FTIR). Results. The results indicate that P. guajava leaves extract demonstrated very high antioxidant activity and P.guajava essential oil showed the highest polyphenols content. The antioxidant capacity showed a significant negative linear correlation to total polyphenolic content (TPC) with Pearson’s correlation coefficients. P. guajava essential oil shows high antibacterial and antifungal activity against all the studied bacteria and fungi.The FTIR analysis of P. guajava essential oil showed the presence of several functional groups (ethers, esters, ketones, terpenes, alkanes, aldehydes, aromatic hydrocarbons, alcohols, and phenols). The relationship between the chemical composition and antimicrobial activity of P.guajava essential oil suggests that the attribution of its antimicrobial activity to a particular compound or a synergistic effects between its different constituents remains difficult. Conclusions. The present study demonstrated that Psidium guajava is a valuable source of active compounds with anti- oxidant and antimicrobial activities. This finding suggests the new use of the fruits and the leaves extracts of this plant in the treatment of bacterial and fungal infections, as well as for the extraction of new antioxidants. Therefore, it is necessary to be carried out in another study to identify the active(s) compound(s) in P.guajava essential oil with respect to their mechanisms and synergistic actions.

The aim of the research was to assess a potential biological activity of cell-free supernatants (CFS) obtained from 4, 6, 8, 10 and 24-hour culture of Bacillus subtilis against 4 pathogenic strains of Rhizoctonia solani marked as R1, R2, R3 and R4. The antagonistic properties of metabolites were assayed by the dual-culture technique on PDA medium. Fungistatic activity of B. subtilis was determined on the rate of mycelia growth inhibition. On the basis of obtained results, it has been proved that fungistatic activity of B. subtilis is varied and depends on the age of the bacterial culture and susceptibility of the fungus. Taking into consideration all the analyzed parameters, R. solani R1 was the most sensitive but R3 lest sensitive to the metabolites produced by B. subtilis. The highest inhibition of the growth rate was obtained for R. solani R1 and the decrease of index amounted between 72-87%, whereas a lower inhibition was noted for strain R3 (44-78%), depending on the age of the bacterial cultures. B. subtilis may find a wide range of application in the process of plant protection against diseases caused by R. solani.
Celem badań była ocena biologicznej aktywności supernatantów otrzymanych z 4-, 6-, 8-, 10i 24-godzinnej hodowli Bacillus subtilis wobec 4 szczepów Rhizoctonia solani oznaczonych jako R1, R2, R3 i R4. Antagonistyczne właściwości metabolitów B. subtilis oznaczono metodą hodowlano-płytkową na podłożu PDA i oceniono na podstawie indeksu tempa wzrostu. Z uzyskanych wyników badań wykazano, że fungistatyczna aktywność B. subtilis jest zróżnicowana i zależy od wieku hodowli bakteryjnej oraz wrażliwości grzybów. Biorąc pod uwagę wszystkie analizowane parametry, szczep R. solani R1 był najbardziej wrażliwy, natomiast R3 najmniej na metabolity produkowane przez B. subtilis. Największe zahamowanie indeksu tempa wzrostu, w wysokości 72-87%, stwierdzono w przypadku R. solani R1, natomiast najmniejsze w przypadku szczepu R3 (44-78%) w zależności od wieku hodowli bakterii. B. subtilis może znaleźć szerokie zastosowanie w ochronie roślin przed chorobami wywołanymi przez R. solani.

The aim of the study was to assess the fungistatic activity of supernatants obtained from 4, 6, 8, 10 and 24-hour culture of Bacillus KF2 and Bacillus BK2 against Fusarium spp. The antagonistic activity was evaluated on the basis the rate index of fungal growth on Czapek-Dox and PDA media. The rate index of Fusarium spp. growth on PDA medium was 5-fold and 3.5-fold slower than in the control, after application of supernatants obtained from the 24-hour culture (respectively for Bacillus KF2 and Bacillus BK2). Similarly, a high antifungal activity of the tested strains was observed on Czapek-Dox medium. The growth of Fusarium spp. was 6-fold and 3.5-fold slower after application of supernatants obtained from 6-hour culture, respectively, for Bacillus KF2 and Bacillus BK2. Supernatants obtained from the culture of both strains showed fungistatic activity against Fusarium spp., although the Bacillus KF2 strain showed a stronger impact than Bacillus BK2 strain. The inhibitory properties of Bacillus species was depended on the age of the bacterial culture and/or strains of Bacillus, and the composition of the medium. The experimental results exhibit the fungistatic activity of Bacillus strains and indicate the possibility of use their as antifungal agents against Fusarium spp.
Celem podjętych badań była ocena aktywności fungistatycznej supernatantów otrzymanych z 4-, 6-, 8-, 10- i 24-godzinnych hodowli Bacillus KF2 i Bacillus BK2 wobec Fusarium spp. Ocenę właściwości antagonistycznych metabolitów bakteryjnych przeprowadzono metodą hodowlano-płytkową z zastosowaniem podłoży Czapka i PDA. Tempo wzrostu Fusarium spp. na podłożu PDA było 5-krotnie oraz 3,5-krotnie wolniejsze niż w próbce kontrolnej, po zastosowaniu supernatantów uzyskanych z 24-godzinnej hodowli, odpowiednio dla Bacillus KF2 i Bacillus BK2. Podobnie dużą aktywność przeciwgrzybową badanych szczepów stwierdzono na podłożu Czapka. Indeks tempa wzrostu Fusarium spp. był 6-krotnie oraz 3,5-krotnie wolniejszy po zastosowaniu supernatantów z 6-godzinnej hodowli, odpowiednio dla Bacillus KF2 i Bacillus BK2. Supernatanty otrzymane z hodowli obu szczepów wykazały działanie fungistatyczne wobec Fusarium spp. przy czym silniejszy wpływ wykazał Bacillus KF2. Stopień zahamowania wzrostu Fusarium spp. zależał od gatunku bakterii, wieku jej hodowli, rodzaju podłoża i czasu trwania hodowli. Uzyskane wyniki badań wykazały aktywność fungistatyczną szczepów Bacillus spp. i wskazują na możliwość wykorzystania ich jako środków przeciwgrzybiczych wobec Fusarium spp.

In the process of biological control of plants pathogens, special biological preparations containing cells of microorganisms or their metabolites are applied. The aim of conducted research was to assess the effect of Bacillus subtilis culture or its supernatant on the mycelial growth of Fusarium culmorum. The antagonistic properties of B. subtilis were assessed with the culture-plate method. The research was conducted on two different media, containing glucose or sucrose, in order to consider the effect of the source of carbon on the inhibitory process. The culturing process was conducted at 25±2ºC for 7 days and every 2-3 days the diameter of the mycelium was measured while the fungistatic activity of B. subtilis was determined against the mycelial growth rate index. The obtained results showed that the measured values of the growth rate index in the control trials were similar regardless of the carbon source in the medium. In tested trials no significant inhibition of the mycelial growth have been noted in the presence of glucose. Measured values of the growth rate index in tested trials were lower of 0.2-1.7 units compared with the control trials. The growth inhibition in the cultures on the medium with sucrose obtained 20% with an addition of the supernatant and 40% with an addition of B. subtilis culture.
W metodach biologicznej walki z fitopatogenami roślin stosuje się między innymi biopreparaty, które jako substancję czynną zawierają żywe komórki mikroorganizmów lub ich metabolity. Celem przeprowadzonych badań było określenie wpływ hodowli bakteryjnej Bacillus subtilis oraz jego supernatantu na wzrost grzybni Fusarium culmorum. Ocenę właściwości antagonistycznych B. subtilis przeprowadzono metodą hodowlano-płytkową. Badania przeprowadzono na dwóch różnych podłożach, zawierających glukozę lub sacharozę, uwzględniając w ten sposób wpływ źródła węgla na proces inhibicji. Hodowle prowadzono w temperaturze 25±2ºC przez 7 dni, dokonując co 2-3 dni pomiaru średnicy grzybni, a aktywność fungistatyczną B. subtilis określono w oparciu o indeks tempa wzrostu grzybni. Na podstawie uzyskanych wyników stwierdzono, iż w próbach kontrolnych indeks tempa wzrostu był na zbliżonym poziomie niezależnie od źródła węgla zawartego w podłożu. W próbach badanych nie odnotowano znaczącego zahamowania wzrostu grzybni w obecności glukozy. Wartości indeksu tempa wzrostu dla prób badanych były niższe jedynie o 0,2-1,7 jednostki w porównaniu do prób kontrolnych. In this newly approached method a novel series of Transition metal complexes were synthesized by the reaction of 5-bromo-2-fluorobenzaldehyde (1) react with hydroxylamine in ethanolic solution at room temperature. Mononuclear complexes were synthesized by reaction with Cu(II) , Zn(II) and Hg(II) chloride salts. The complexes were characterized by elemental analysis, Ultraviolet, Infra -red and 1HNMR spectral studies. The antimicrobial activities of the ligand and its metal complexes were estimated.

A new series of 2-azetidinone compounds 5(a-g) were synthesized. The structures of all the synthesized compounds were confirmed by chemical and spectral analyses such as IR, 1H NMR and 13C NMR. The compounds were screened for their antimicrobial activity against Aspergillus flavus, Aspergillus niger, Penicillim chryogenum, Trigoderma veride & Fusarium oxysporum and the zone of inhibition was determined by disc diffusion technique. All the synthesized compounds exhibited promising antidungal activity against the studied set of microorganisms. However, the activity was less than that of standard drugs used in this study.

Actinomycetes are considered to be the biggest producer of bioactive compounds which are expected to have antifungal activity for controlling many fungi such as Rhizoctonia solani. The objective of this study was to obtain potential soybean rhizosphere actinomyc- etes as a biocontrol agent for R. solani which cause damping-off disease both in vitro and in vivo, including their ability to produce siderophore, chitinase, and HCN. Out of 26 isolates, 18 (56%) showed diverse antifungal activities against R. solani with percentages of inhibi- tion radial growth (PIRG) from 18.9 to 64.8%, as evaluated by a dual culture method. Ten isolates with the strongest antifungal activity were numbered for further characterization. All the tested isolates were not antagonistic towards Bradyrhizobium japonicum. These iso- lates were able to suppress damping-off disease caused by R. solani in the greenhouse exper- iment. Isolate ASR53 showed the highest disease suppression, 68% and 91% in sterile and non-sterile soil, respectively. Based on 16S rRNA sequence analysis this isolate belonged to Streptomyces violaceorubidus LMG 20319 (similarity 98.8%) according to GenBank data base available at www.ncbi.nlm.gov.nih. Furthermore, isolate ASR53 had significantly longer roots and shoots, as well as greater fresh and dry weights of seedlings than the con- trol. Crude extract derived from ASR53 isolates contained 10 dominant compounds that were biologically active against fungal pathogens. Thus, this study suggests that the applica- tion of potential actinomycetes of the soybean rhizosphere can act as a promising biocon- trol agent against damping-off disease caused by R. solani.

N3-(4-Methoxyfumaroyl)-L-2,3-diaminopropanoic acid (FMDP) and 2-amino-2-deoxy-D-glucitol-6-phosphate (ADGP) are strong inhibitors of the essential fungal enzyme, glucosamine-6-phosphate synthase, but their antifungal activity is poor, due to slow penetration of these agents through the cytoplasmic membrane. In the present studies we have exploited the possibility of enhancement of ADGP and FMDP antifungal activity by improving their transport properties. It has been found that membrane-permeabilising polyene macrolides amphotericin B (AMB) and its N-methyl-N-fructosyl methyl ester derivative (MF-AME), at subinhibitory concentrations, facilitate diffusion of ADGP through the fungal cell membrane, thus allowing a decrease of its minimal inhibitory concentration (MIC). Synergistic effects have been observed for combinations of ADGP with AMB or MF-AME. Fractional inhibitory concentration (FIC) indexes, determined against a number of Candida spp., have been in the 0.18-0.81 range. Weak antifungal synergistic effects have been found for combinations of FMDP with AMB or MF-AME. ADGP can be easily encapsulated into unilamellar lipid vesicles. Liposomal preparations of ADGP demonstrated stronger antifungal activity against some fungal strains than free ADGP.
