Opis:
Sprouts consumption is increasing worldwide due to their nutritional and health benefits. However, they can be sources of foodborne bacterial diseases, including sprout-related outbreaks caused primarily by Shiga toxin-producing Escherichia coli and Salmonella spp. The aim of this study was to determine the efficacy of a dried sea buckthorn fruit extract in sanitization of contaminated radish seeds. The efficacy of 15-min seed treatment and 3-h soaking in the extract solution was evaluated in comparison to the treatment with 20,000 mg/L calcium hypochlorite, recommended by the U.S. Food and Drug Administration. The effect of the sea buckthorn fruit extract on radish seed germination capacity and the profile of phenolic compounds in the extract were also determined. Decontamination effect of the extract after a 15-min seed treatment ranged from 72.1% against Staphylococcus aureus to 93.0% against Listeria monocytogenes and was higher than that of active chlorine against E. coli, S. aureus and Salmonella enterica. Soaking seeds for 3 h in the extract increased the decontamination efficiency only against S. aureus (81.9%). Compared to water, after 4 days of sprouting, no significant differences were found in radish seed germination capacity and the length of roots and hypocotyls of sprouts. The sea buckthorn fruit extract had a high content of phenolic compounds, namely isorhamnetin 3-rhamnosylglucoside (12.99 mg/L), isorhamnetin 3-rutinoside (8.25 mg/L), protocatechuic acid (5.43 mg/L), isorhamnetin 3-glucoside (3.41 mg/L), and gallic acid (3.36 mg/L). The extract can be used as a substance limiting bacterial contamination of radish seeds and, as a result, sprouts, as well as a valuable source of phenolic compounds.

Introduction and aim. The synthesis of heterocyclic compounds containing oxygen and nitrogen is profoundly intriguing due to their mechanistic implications in both research and development within organic chemistry and drug discovery. The primary aim of this study is to fabricate a range of pharmacologically active drugs containing the isoxazole moiety. Material and methods. The synthesis of new derivatives of isoxazole was achieved through a one-pot condensation reaction of 2-[(Substituted phenyl)hydrazono]malononitrile (1) and 3-[(Substituted phenyl)azo]-2,4-Pentanedione (2) with sodium acetate and hydroxylamine hydrochloride (1:1) in ethanol. All the compounds were screened for their in vitro antibacterial activity, in vitro antioxidant and anticancer activity. The synthesized compounds underwent characterization through FTIR, 1H NMR, and 13C NMR analyses, supported by mass spectral data and elemental analysis. Results. A set of novel isoxazole derivatives was synthesized with a favorable yield. Among compounds 1d, 1e, 2c, 2d, and 2e exhibited notable antioxidant activities. Compounds 1a, 1b, and 1c demonstrated significant anticancer potential against prostate cancer [PC3] cell lines compared to normal HEK cell lines, while 2a displayed the highest inhibitory zone against Escherichia coli. Conclusion. Novel compounds with multifaceted biological activities have been successfully designed, and a synthetic route to create isoxazole derivatives has been devised and verified.

Silver nanoparticles have special plasmonic and antibacterial characteristics that make them efficient in a variety of commercial medical applications. According to recent research, chemically synthesized silver nanoparticles are harmful even in low concentrations. It was crucial to identify appropriate synthesis methods that may have low costs and be nontoxic to the environment. Zingiber officinale (ginger) extracts used to prepare silver nanoparticles were inexpensive and environmentally friendly, and the best physicochemical characteristics were analyzed. Silver nanoparticles were characterized by using UV-visible spectroscopy, Scanning electron microscopy (SEM), and X-ray diffraction (XRD). The surface Plasmon resonance peak at 425 nm was observed using UV-Visible spectroscopy. Scanning electron microscopy observed that the nanoparticles were spherical and ranged in size from 5 to 35 nm. The XRD pattern values of 2θ: 38.2° , 46.3° , and 64.58° are used to determine the planes (111), (200), and (220). The silver nanoparticle’s existence was verified by the face-centered cubic (FCC). Silver nanoparticles were found to have antibacterial efficacy against gram-positive Staphylococcus and gram-negative bacteria such as Pseudomonas aeruginosa, Klebsiella Aerogenes, Salmonella, Staphylococcus and Escherichia coli. The antibacterial activity of silver nanoparticles was observed using the agar well diffusion (AWD) method at three different concentrations (100 μg/ ml, 75 μg/ml, and 50 μg/ml). The zone of inhibition measured against the bacterial strains pseudomonas Aeruginosa, Klebsiella aerogenes, Escherichia coli, Salmonella and Staphylococcus which were (18.4±1.25 mm, 16.9±0.74 mm, 14.8±1.25 mm), (16.8±0.96 mm, 14.6±0.76 mm, 14.0±1.15 mm), (19.7±0.76 mm, 18.2±0.66 mm, 15.4±1.15 mm), (16.6±0.67 mm, 14.2±0.23 mm, 12.8±0.78 mm) and (12±0.68 mm, 10±0.20 mm, 08±0.15 mm). These nanoparticles’ potent antibacterial properties may enable them to be employed as nanomedicines for a variety of gramnegative bacterial illness treatments.

W artykule przedstawiono wyniki badań in vitro dotyczące cytotoksyczności i działania bakteriobójczego materiału do stosowania w endodoncji – cementu typu MTA (ang. Mineral Trioxide Aggregate) o nowym składzie chemicznym. Cement wytworzono na bazie krzemianu trójwapnia wzbogaconego ZnO i płynu będącego 15% roztworem CaCl2 , a jako czynnik wprowadzający kontrast RTG zastosowano ZrO2 . Wytworzony cement MTA oceniono poprzez scharakteryzowanie właściwości fizykochemicznych obejmujących: czas wiązania, wytrzymałość na ściskanie, a także kontrast RTG i bioaktywność w roztworze symulującym osocze (SBF). Do zbadania wpływu ziaren ZrO2 na ilość i wielkość porów w cemencie wykorzystano technikę mikrotomografii komputerowej (µCT). Działanie cytotoksyczne cementów oceniano przez zastosowanie referencyjnej linii komórkowej L-929. Warunki hodowli komórkowej w kontakcie z badanymi materiałami lub ekstraktami z cementów oceniano za pomocą analizy obrazu lub testu kolorymetrycznego MTT. Do badania aktywności przeciwbakteryjnej wykorzystano dwa szczepy paciorkowców: Streptococcus mutans i Streptococcus sanguinis. Opracowany cement typu MTA charakteryzuje się odpowiednimi właściwościami użytkowymi i spełnia wymagania zawarte w normie PN-EN ISO 10993-5:2009 „Biologiczna ocena wyrobów medycznych, część 5: Badania cytotoksyczności in vitro”. Jednak, aby mógł być rozważany jako bezpieczny wyrób medyczny do wypełniania wstecznego kanałów korzeniowych, wymaga dalszych badań.
The article presents the results of in vitro cytotoxicity and bactericidal activity of MTA-type cement (Mineral Trioxide Aggregate) for endodontics with a new chemical composition. The cement was made on the basis of ZnO-enriched tricalcium silicate and a liquid being a 15% CaCl2 solution. ZrO2 was used as an X-ray radiopacity agent in the cement. The produced MTA-type cement was assessed by characterizing the physicochemical properties including: setting time, compressive strength, as well as X-ray radiopacity and bioactivity in a simulated body fluid (SBF). Microtomography technique (µCT) was used to investigate the effect of ZrO2 grains on the number and size of pores in the cement. The cytotoxic activity of the cements was assessed by using the reference L-929 cell line. Cell culture conditions in contact with test materials or cement extracts were assessed by image analysis or MTT colorimetric test. Two strains of streptococci were used to test the antibacterial activity: Streptococcus mutans and Streptococcus sanguinis. The developed MTA cement has appropriate functional properties and meets the requirements of the PNEN ISO 10993-5:2009 standard "Biological evaluation of medical devices, Part 5: In vitro cytotoxicity tests". However, in order to be considered a safe medical device for retrograde root canal filling, it requires further research.

Background. During the recent years, appropriate attention has been paid to the oxidative stress which damages the body’s cells, proteins, and DNA. Therefore, the need of antioxidants becomes a therapeutic and preventive priority. In addition, microbial infections also constitute a public health problem. Objective. To find efficient, reliable and safe alternatives sources to synthetic antioxidants, antibiotics and antifungals drugs. Materials and methods. Extract and essential oil of Psidium guajava were screened for their antioxidant and antimicrobial activities against gram positive bacteria (Enterococcus faecalis and Staphylococcus aureus), gram negative bacteria (Citrobacter freundii, Escherichia coli and Pseudomonas sp) and fungi (Candida albicans,Candida tropicalis and Cryptococcus neoformans), as well as to determine the functional groups of phytochemicals present in the essential oil by Fourier transform infrared spectroscopy (FTIR). Results. The results indicate that P. guajava leaves extract demonstrated very high antioxidant activity and P.guajava essential oil showed the highest polyphenols content. The antioxidant capacity showed a significant negative linear correlation to total polyphenolic content (TPC) with Pearson’s correlation coefficients. P. guajava essential oil shows high antibacterial and antifungal activity against all the studied bacteria and fungi.The FTIR analysis of P. guajava essential oil showed the presence of several functional groups (ethers, esters, ketones, terpenes, alkanes, aldehydes, aromatic hydrocarbons, alcohols, and phenols). The relationship between the chemical composition and antimicrobial activity of P.guajava essential oil suggests that the attribution of its antimicrobial activity to a particular compound or a synergistic effects between its different constituents remains difficult. Conclusions. The present study demonstrated that Psidium guajava is a valuable source of active compounds with anti- oxidant and antimicrobial activities. This finding suggests the new use of the fruits and the leaves extracts of this plant in the treatment of bacterial and fungal infections, as well as for the extraction of new antioxidants. Therefore, it is necessary to be carried out in another study to identify the active(s) compound(s) in P.guajava essential oil with respect to their mechanisms and synergistic actions.

Because of side effects and increasing resistance to antibiotics, considerable attention hasbeen given to extract sand biologically active compounds isolated from medicinal plants. In the present study, phytochemical screening and antibacterial activities of different concentrations of various extracts of Onosma chlorotricum were evaluated and compared with those of vancomycin and amikacin. Total phenolic and flavonoid contents and the antibacterial activity of methanol, n-hexane, and aqueous extracts (5 mg/ml to 0.156 mg/ml final concentration) of O. chlorotricum against Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli were evaluated using disc diffusion and microdilution methods. Total phenolic and flavonoid contents were determined according to the Folin-Ciocalteu and aluminum chloride colorimetric assays, respectively. The results showed that the total phenolic and flavonoid contents of these extracts ranged from 56.10 ± 0.13 to 74.12 ± 0.05 mg GAE/g dry extract and from 19.3 ± 0.6 to 23.20 ± 0.41 mg QE/g dry extract, respectively. The methanol extract with the highest phenolic and flavonoid content showed the highest antibacterial activity against all the tested bacterial strains, with the highest inhibition zone of 21 ± 0.7 mm and the lowest MIC and MBC values of 78.12 μg/ml for S. aureus. The antibacterial effects and the total phenolic content of O. chlorotricum were remarkable. The results suggest that the effects of methanol extract on wound healing could be tested in an animal model.

Opis:
Rothmannia wittii, which is found in Binh Chau-Phuoc Buu Nature Reserve, Bung Rieng ward, Xuyen Moc District, Ba Ria-Vung Tau Province, Vietnam, is a rare species of Rubiaceae family. The present study aimed to determine the phytochemical composition and antimicrobial activity of the ethanolic extract of the R. wittii trunk. A total of 10 compounds were identified by liquid chromatography-mass spectrophotometry (LC-MS), including 1) benzene, 1-(1,5-dimethyl-4- hexenyl)-4-methyl, 2) diphenylsulfone, 3) 6β-hydroxy-10-O-acetylgenipin, 4) cinnamyl-cinnamate, 5) genipin, 6) megastigmatrienone, 7) ethyl p-methoxy-trans-cinnamate, 8) rothalazepane, 9) 1,3,6-trimethylpyrano[2,3-d ]imidazole-2,5(1H,3H)-dione, and 10) shanzhiol. The extract also exhibited an antibacterial activity against six bacterial strains: Bacillus cereus, Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Salmonella typhimurium and Salmonella enteritidis, as determined via disk diffusion agar method.

Nowadays coming out of bacterial resistance poses a significant clinical problem. Hence, the aim of this study was to explain the current susceptibility patterns of methicillin-resistant Staphylococcus aureus (MRSA).As well as to find out antimicrobial characteristics in the different organic solvents with increasing polarity viz., hexane, chloroform, ethyl acetate, and methanol extracts of Padina tetrastromatica, Padina gymnospora marine macro algae belonging to the family Phaeophyta were studied. Their crude extracts were tested against Staphylococcus aureus (MTCC 737 & 7443), and three clinical isolates of MRSA were tested and has been shown to exhibit antibacterial activity against methicillin methicillin-resistant Staphylococcus aureus (MRSA). The Minimum inhibitory concentrations (MIC) and Minimum Bactericidal Concentrations (MBC) were determined. The ethyl acetate extracts of the seaweeds showed the presence of Photochemical, terpenoids, tannins, phenolic compounds and steroids strongly than the other solvent extracts. The highest activity was recorded in the ethyl acetate extract of Padina tetrastromatica than the other extract tested. The mean zone of inhibition produced by the extracts in disc diffusion assays against the tested bacterial strains ranged from 7.1 to 26.5 mm. The lowest MIC (62.5 µg/ml) and MBC (125 µg/ml) values were observed in the ethyl acetate extract of P. tetrastromatica against S. aureus (737 & 7443), MRSA1 and MRSA3. Further separation of active principle from the potential seaweed extract as a source of antibacterial compound useful for the control of Methicillin resistant S. aureus is under progress.

In this study, optimal conditions to form cellulose-MgO nanocomposite with antibacterial properties were evaluated. Applying the Taguchi method, 9 experiments were designed and the effects of different concentrations of biopolymers cellulose (0.5, 1 and 2 mg/ml), MgO nanoparticles (2, 4 and 8 mg/ml) and stirring times (30, 60 and 90 min) on antibacterial activity of synthesized nanocomposites were assessed. The characterizations of products were investigated by dynamic light scattering (DLS), raman spectroscopy, scanning electron microscope (SEM), thermogravimetric analysis (TGA) and differential thermal analysis (DTA). The results showed that the nano-composite produced in the conditions of experiment 9 (MgO 8 mg/ml, cellulose 2 mg/ml and stirring time of 60 min) has the strongest antibacterial activity. The outcomes of both methods of colony forming units (CFU) and disc diffusion indicated that the antibacterial activity of cellulose-MgO nanocomposite was significantly higher than its components (P <0.05). Thermal analysis indicated improvement in the thermal stability of the cellulose biopolymer after the formation of the nanocomposite. Due to the improvement of the antibacterial properties of cellulose-MgO nanocomposite compared to its components, we can use it as a new antibacterial agent in the fields of pharmaceutical, medicine and dentistry.

In the GC-MS analysis, 81 bioactive phytochemical compounds were identified in hexane extracts of Cirsium creticum and Cirsium italicum. Terpenoids constituted the main fractions of C. italicum (70.95%), while hydrocarbons were most abundant compounds of C. creticum (41.11%). The antibacterial activity and antifungal activity of extracts from two Cirsium species was tested using microdilution methods. According to the results of antibacterial activity, the highest inhibition effect of C. creticum was found on B. subtilis, E. coli and P. aeruginosa. The highest inhibition effect of C. italicum was found on B. subtilis. In the results of antifungal activity, the highest inhibition effect of C. creticum was found on P. chrysogenum and the highest inhibition effect of C. italicum was found on C. krusei. The present work is the first report on hexane extracts compounds of two Cirsium species as well as antibacterial and antifungal activities.

Opis:
Essential oils (EOs) of Gaultheria procumbens were obtained by hydrodistillation with the yield of 1.30% ± 0.05 (v/dw) for leaves and 2.68% ± 0.08 (v/dw) for fruits. The GC-FID/MS analysis led to the identification of 64 volatile components, among which 27 were found in leaf EO, 49 in fruit EO, and 59 analytes were detected for the first time in G. procumbens. Methyl salicylate, which is known as a potent anti-inflammatory agent, was the dominant component making up to 97.5% and 99.8% of the total fruit and leaf EOs, respectively. The remaining volatiles of the leaves included mainly monoterpenes (α-pinene, β-pinene, limonene), methyl-o-anisate, massoilactone, spiro[4,5]decane-1-one, and aliphatic alcohols (heptan-2-ol, octan-1-ol), while the fruit EO contained apart from the dominant aliphatic and aromatic alcohols (benzyl alcohol, heptan-2-ol, p-cymen-7-ol) also carboxylic acids (3-hydroxyphenylacetic acid, heptanoic acid, octanoic acid) and aldehydes (hexanal, furfural, pent-4-enal). The antibacterial activity of the EOs was evaluated using a microdilution broth method against twelve reference strains, as well as clinical and environmental isolates. Differences between activity parameters of both EOs were not statistically significant (p > 0.05) for most tested bacteria. On the other hand, both EOs were significantly more effective against Gram-negative (MIC, 8.2-10.0 mg/mL) than Gram-positive bacteria (MIC, 13.5-16.7 mg/mL). Leaves and especially fruits of G. procumbens cultivated in Poland were proved valuable sources of methyl salicylate-rich EOs of moderate antibacterial activity, which, therefore, could partly explain the traditional use of the plant materials and the EOs in the treatment of bacterial infections-related inflammatory disorders.

Essential oils from needles, fruits and bark was extracted from Pinus pinea L. (stone pine) grown wildly in Jordan. The chemical composition, antibacterial activity, antioxidant activity of essential oils was evaluated. The chemical compositions were identified using Gas-Chromatography-Mass spectrometry (GC-MS) and retention Indices (Van den Dool & Kratz). The results show that the essential oil obtained from needles composed mainly of Guaiol (12.7%), limonene (11.42%), and β-caryophyllene (7.61%), while fruit contains limonene (32.56%), and α-pinene (6.78%). The essential oils from barks were rich in β-caryophyllene (16.51%), limonene (14.83%), caryophyllene oxide (11.83%), and longifolene (7.51%). In vitro, antibacterial activity of the essential oil samples was evaluated using agar-well diffusion method against three different strains of bacteria (Gram¬-positive bacteria: Staphylococcus aureus and Gram-negative bacteria: Klebsiella pneumoniae and Escherichia coli). The results showed that essential oil showed appreciable antibacterial activity against S. aureus. The essential oil from fruit exhibited weak antibacterial activity against E. coli and K. pneumoniae. Essential oils of P. pinea showed appreciable antioxidant activity in-vitro.
