Background. The aim of the study was to determine the role of game animals as reservoirs of Anaplasma phagocytophilum, a bacteria species transmitted by Ixodes ricinus ticks, from north−western Poland (Zachodniopomorskie vovoidship). The area under question is endemic for A. phagocytophilum. Material and methods. Blood and spleen samples were taken from 72 roe deer between April and December 2003. Animals culled during winter did not harbour ticks, on the other hand 155 individuals of Ixodes ricinus were collected from 35 of 43 animals taken during spring. We tested all samples for A. phagocytophilum by PCR amplification of the msp2 gene. An individual was considered infected if pathogens were detected in at least one isolate (blood or a tissue sample). Results. DNA from A. phagocytophilum was not found in isolates from ticks collected from the animals. The general level of infection for the roe deer was 31.94% (23/72). DNA of A. phagocytophilum was most commonly detected in blood samples; only in three cases was anaplasma DNA detected in spleen and not in blood. Ruminants seem to be the most competent reservoir for A. phagocytophilum in north−western Poland.

Opis:
The study was aimed at determining the cause of the diseases in three horses exhibiting symptoms of fever, ataxia, mucus membrane paleness, haematuria and thrombocytopenia. The PCR technique revealed the presence in the blood of 18S RNA Babesia/Theileria spp. genetic material. DNA amplification using primers RLB F2 and RLB R2 produced 430 bp size products. The sequences of these PCR products demonstrated a 95.6–97.5% similarity with the sequence of the fragment of 18S RNA Babesia equi, gene number DQ287951 in the GenBank. The treatment utilizing the subcutaneous application of the imidocarb resulted in gradual recovery of the diseased animals.

Introduction. Ticks (Acari: Ixodida) are vectors and/or reservoirs of many pathogens, i.e. Borrelia burgdorferi sensu lato, Anaplasma phagocytophilum and Babesia microti. These pathogens are ethiological agents of such diseases as Lyme borreliosis, human granulocytic anaplasmosis and human babesiosis. Objective. The aim of the study was to evaluate the role of the Ixodes ricinus in the transmission of Borrelia burgdorferi sensu lato, Borrelia afzelii, Borrelia garinii, Borrelia burgdorferi sensu stricto, Anaplasma phagocytophilum and Babesia microti in Opolskie Province in Poland. Materials and method. DNA from 222 ticks was isolated by the ammonia method. The pair of primers specific to the flagelline gene was used to detect of B. burgdorferi s. l. To detect of genospecies of this spirochete, three pairs of internal primers were used. In turn, two pairs of primers specific to the 16S rDNA gene and the 18S rRNA were used, respectively, for the detection of A. phagocytophilum and B. microti. Borrelia burgdorferi s. l., A. phagocytophilum, and B. microti were detected in 4.5%, 2.7% and 5.4% of examined ticks, respectively. Results and conclusions. Of the ten ticks infected with B. burgdorferi s. l., B. afzelii was found in seven, undefinied genospecies in two, and mixed infection with B. afzelii and B. burgdorferi s. s. in one. The study demonstrated the potential risk of exposure of humans and animals to infections of B. burgdorferi s. l., A. phagocytophilum and B. microti in the examined area of Poland.

A total of 684 Ixodes ricinus ticks (321 nymphs, 184 males, and 179 females) were collected by fl agging lower vegetation in 6 forest districts located on the territory of Lublin province (eastern Poland). Ticks were examined by polymerase chain reaction (PCR) method for the presence of Anaplasma phagocytophilum DNA with two pairs of primers: EHR521/EHR747 for detecting 16S rRNA gene, and LA6/LA1 for detecting ankA gene. To study the relationship between infection in ticks and people occupationally exposed to tick bite, blood serum samples of 261 forestry workers employed in the same forest districts were examined by immunofl uorescence method for the presence of specifi c antibodies against A. phagocytophilum. A total of 70 ticks out of 684 examined (10.2%) showed the presence of A. phagocytophilum 16S rRNA gene. The prevalence of infection was signifi cantly dependent on tick’s stage (χ2=49.2, p<0.00001) and geographical locality (χ2=34.4, p<0.00001). The percentage of I. ricinus females infected with A. phagocytophilum (24.6%) was signifi cantly greater compared to males (6.5%) and nymphs (4.4%) (p<0.00001). Only 19 ticks out of 684 examined (2.8%) showed the presence of A. phagocytophilum ankA gene, signifi cantly less compared to 16S rRNA gene (p<0.00001). The prevalence of infection demonstrated by the presence of ankA gene was also signifi cantly dependent on tick’s stage (χ2=23.6, p<0.00001) but not on locality (χ2=9.8, p=0.082). A signifi cant correlation was found between the presence of A. phagocytophilum 16S rRNA gene in I. ricinus female ticks from the particular forest districts and the serologic response to A. phagocytophilum of forestry workers employed in the same districts (p<0.05). No signifi cant correlation was found between the presence of A. phagocytophilum ankA gene in I. ricinus ticks and serologic response of exposed workers. In conclusion, detection of A. phagocytophilum infection in ticks by PCR with the use of EHR521/EHR747 primers detecting 16S rRNA gene is signifi cantly more sensitive compared to LA6/LA1 primers detecting ankA gene.

Background: The objective of the study was the evaluation of the infection of ticks with pathogenic microorganisms at various workplaces (timber acquisition, forest growing, forest cultivation, forest protection). Material and Methods: Eight hundred sixty one Ixodes ricinus ticks collected from 4 workplaces were examined for the presence of Borrelia burgdorferi, Anaplasma phagocytophilum and Babesia microti by polymerase chain reaction (PCR). Then, a comparative analysis of the relative density and infection of ticks at individual workplaces was done. In the statistical analysis, Chi² test, and Pearson’s test for correlation were applied. Results: The differences in infection (15.9–50%) of ticks with B. burgdorferi between the examined workplaces were highly significant, with the highest percentage observed at forest growing. The percentages of infection of ticks with A. phagocytophilum at individual workplaces ranged from 1.1–3.7%, and differences were statistically insignificant. The percentages of infections of ticks with Babesia microti at individual workplaces fluctuated from 3.6–4.4% and differences were also insignificant. Co-infections of ticks with 2 or 3 pathogens were rare. Conclusions: Co-infections with B. burgdorferi and B. microti showed a significant relationship with the workplaces, while those with B. burgdorferi and A. phagocytophilum did not show such a dependence. No significant positive correlation was found between the relative density of ticks and the frequencies of infections with B. burgdorferi, A. phagocytophilum and B. microti. Med. Wstęp: Celem badań była ocena zakażenia kleszczy występujących na różnych miejscach i stanowiskach pracy w leśnictwie (uzyskiwanie drewna, hodowla lasu, uprawa lasu, ochrona lasu) patogenami przenoszonymi przez kleszcze. Materiał i metody: Z 4 miejsc pracy zebrano 861 kleszczy Ixodes ricinus, które zbadano na obecność Borrelia burgdorferi, Anaplasma phagocytophilum i Babesia microti przy pomocy łańcuchowej reakcji polimerazy (polymerase chain reaction – PCR). Następnie przeprowadzono analizę porównawczą względnej gęstości kleszczy i ich zakażenia w poszczególnych miejscach pracy. W analizie statystycznej zastosowano test Chi² i test korelacji Pearsona. Wyniki: Różnice w zakażeniach kleszczy B. burgdorferi (15,9–50%) zebranych z różnych miejsc pracy były statystycznie wysoce istotne, a najwyższe zakażenie kleszczy zaobserwowano w miejscu hodowli lasu. Odsetki zakażeń kleszczy A. phagocytophilum i B. microti na poszczególnych miejscach pracy wahały się odpowiednio od 1,1% do 3,7% i od 3,6% do 4,4%, a różnice były nieistotne statystycznie. Współzakażenia kleszczy 2 lub 3 patogenami występowały rzadko. Wnioski: Wykazano istotną zależność koinfekcji B. burgdorferi i B. microti od stanowiska pracy, natomiast nie stwierdzono jej dla współwystępowania B. burgdorferi i A. phagocytophilum. Nie wykazano korelacji między względną aktywnością a zakażeniem kleszczy B. burgdorferi, A. phagocytophilum i B. microti. Med Pr 2014;65(5):575–581

The aim of the study was to determine the prevalence of Ehrlichia canis, Anaplasma platys and Babesia spp. in dogs. It describes the practice of veterinarians in detecting tick-borne diseases in Nueva Ecija, Philippines. Seventy blood samples were collected and were subjected to multiplex PCR for the detection of E. canis, Babesia spp. and A. platys. The prevalence of babesiosis is the highest in Cabanatuan City (2/10), while a 10% prevalence (1/10) was observed in Science City of Muñoz, Talavera and Sta. Rosa. E. canis were only detected in Cabanatuan City. However, no anaplasmosis was detected in any area. The prevalence of babesiosis and ehrlichiosis in Nueva Ecija is 7.14% (5/70) and 2.85% (2/70) respectively. In addition, 70% (7/10) of the Nueva Ecija veterinary practitioners encountered cases of suspected ehrlichiosis in their practice. The diagnosis of ehrlichiosis is based primarily on presented clinical signs and complete blood counts, which include a platelet count. Of the 10 respondents, half utilized test kits while 90% interpreted blood samples. Meanwhile, only 60% of the respondents used an ELISA test kit for ehrlichiosis. For some practitioners, the main reason for not utilizing a kit is the high cost. None of the respondents had previously attended cases of suspected anaplasmosis. Only one respondent diagnosed a case of babesiosis by blood smear microscopy.

Introduction and objective. Previous studies on tick-borne pathogens in the avian population have focused mainly on the detection of the agents in ticks collected from birds, but data about the presence of tick-borne bacteria in these animals are scant. The aim of the presented study was to verify the exposure to some zoonotic tick-borne bacteria, in particular, Anaplasma phagocytophilum, Borrelia burgdorferi sensu lato, Coxiella burnetii, and the Rickettsia species of the Spotted Fever Group (SFG), in pheasants (Phasianus colchicus) living in a central Italy area, characterized by conditions favourable for the diffusion of the ticks. Materials and method. Blood serum samples from 276 farm-reared pheasants were examined by indirect immunofluorescence antibody test to detect antibodies against the 4 reported pathogens. Results. A total of 124 (44.92%) birds resulted as seropositive: in particular 3 (1.08%) to C. burnetii, 31 (11.23%) to A. phagocytophilum, 46 (16.67%) to B. burgdorferi s.l., 49 (17.75%) to SFG Rickettsia spp. antigens. Three pheasants resulted positive both to A. phagocytophilum and B. burgdorferi s.l. antigens, whereas 2 – both to B. burgdorferi s.l. and SFG Rickettsia spp. antigens. The antibody titers varied from 1:40–1:320. Conclusions. The obtained results show that the pheasants had developed antibodies to the investigated tick-borne agents. For this reason, they seem to be involved in the epidemiology of the studied tick-borne bacteria.

The presence of Anaplasma phagocytophilum, a tick-transmitted zoonotic pathogen, has been reported in Italy in humans and several animal species, including dogs, but data concerning its prevalence in the Italian canine population are lacking. The aim of this study was to determine the prevalence of A. phagocytophilum infection in hunting dogs which live in geographic areas of central Italy where the infection had been previously detected in wild animals. Sera from 215 hunting dogs were tested by indirect immunofluorescence antibody test (IFAT) to detect antibodies to A. phagocytophilum. Buffy coat samples from the same 215 animals were submitted to a polymerase chain reaction (PCR) assay specific for the A. phagocytophilum 16S rRNA gene. Thirty-two (14.8%) dogs proved to be seropositive with antibody titres from 1:40–1:2,560. The highest seroprevalence was observed in dogs 6–10-year-old. Two seropositive (0.9%) animals were also PCR positive. Sequencing of PCR products revealed gene sequences of A. phagocytophilum in both cases. These results confirm the presence of A. phagocytophilum in the Italian areas studied; thus, the zoonotic potential of this agent should be considered particularly for people, as hunters, at risk of exposure to tick bites.
