Opis:
New hydrazone derivatives were synthesized from 4-chloro-2-(methylthio)pyrimidine-5-carbohydrazide. Synthesized compounds were characterized by IR, 1H-NMR, 13C-NMR, mass spectral data and elemental analysis. All compounds were investigated for analgesic and anti-inflammatory activity with hot plate test and biochemical assay, respectively. The results of anti-inflammatory activity showed that compound 2t had maximal Lipoxygenase (LOX) inhibition (66.30%) whereas inhibitions of 2a, 2c, 2i, 2k, 2l, 2m, 2p, 2s and 2u were observed between approximately 45-15 %. Our results also indicated that 15 compounds of 21 had analgesic effect in hot plate test. Analgesic effect of 2l, 2s, 2t began earlier than others while effect of 2i began at latest time. According to the activity results, compound 2t have significant analgesic and anti-inflammatory effects.

Opis:
Ononis spinosa (spiny restharrow) is a perennial herb that belongs to the Fabaceae family. Among several uses, the aerial parts of O. spinosa are used in traditional medicine for toothache. In this study, analgesic activity of the methanolic extract of leaves was studied. In writhing test, the 300 mg/kg and 600 mg/kg doses of O. spinosa extract produced 90.1% and 94.5% inhibition of abdominal cramps, respectively compared to 23.6% inhibition produced by 30 mg/kg diclofenac sodium. O. spinosa methanolic extract reduced the time of licking in early and late phases of formalin test. The inhibitory effect of O. spinosa extract was more pronounced in late phase. The lowest dose tested (300 mg/kg) of O. spinosa extract produced 81.0% inhibition compared to 50.5% inhibition in 30 mg/kg diclofenac sodium in the late phase of formalin test. This effect was not abrogated by glibenclamide, naloxone or atropine indicating that the action of O. spinosa in formalin test is not mediated by ATP-sensitive K+ channel, opioid or muscarinic receptors. In tail-flick and hot-plate tests, O. spinosa increased the latency time at 30 min and 150 min. Naloxone antagonized the action of O. spinosa in both tests suggesting an interaction with opioid receptor as a possible mechanism of O. spinosa in thermal pain models at the spinal and supraspinal levels. Phytochemical screening indicated the presence of phenolics, saponins, flavonoids, and terpenoids. Alkaloids, sterols, and anthraquinone glycosides were absent. HPLC analysis confirmed the presence kaempferol, apigenin and myricetin in the extract.

Analgesic and anti-inflammatory activities of four transition metal complexes of a Schiff base derived from salicylaldehyde and glycine, viz. [N-salicylideneglycinato diaqua cobalt(II) dimer] (SGCo)2, [N-salicylideneglycinato-di-aqua-nickel(ll)dimer] (SGN)2, [N-salicylideneglycinato-aqua-copper(II)] (SGC) and [N-salicylideneglycinato diaqua zinc(II) dimer] (SGZ)2 were studied in swiss albino mice. 10 mg/kg (p.o) dose for each compound were selected throughout the research work. The analgesic activity was determined by acetic acid induced writhing and tail immersion methods. All such data were compared with standard drugs at the dose of 10 mg/kg (p.o. The anti-inflammatory activity of the test compounds was determined by the carragenan induced mice paw edema inhibition method.). Analgesic and anti-inflammatory activities of the test compounds at 10 mg/kg (p.o.) were quite comparable to those of standard drugs at 10 mg/kg (p.o.). All these compounds can be considered as potent analgesic and anti-inflammatory agents.

Opis:
The electromagnetic fields (EMFs) have been shown to alter animal and human behavior, such as directional orientation, learning, pain perception (nociception or analgesia) and anxiety-related behaviors. The aim of this study was to evaluate the influence of electromagnetic fields of high-frequency microwaves on pain perception and anti-nociceptive activity of tramadol (TRAM) - analgetic effective in the treatment of moderate to severe acute and chronic pain states. Electromagnetic fields exposures of a) 1500 MHz frequency and b) modulated, 1800 MHz (which is identical to that generated by mobile phones) were applied. Paw withdrawal latency (PWL) to thermal stimulus was measured in vehicle or tramadol (TRAM) treated animals before and after 30, 60 and 90 minutes from injections. The differences in the level of pain (PWL) between control group and rats exposed to EMF alone in three measurements, were not observed. Tramadol alone significantly increased PWLs to thermal stimulus in comparison to vehicle results at 30 (p < 0.001) and 60 minutes (p < 0.05) after drug injection. EMF exposure of both frequencies transiently suppressed analgesic effect of tramadol, significantly reducing paw withdrawal latency in animals treated with this drug at 30 minutes from the drug injection.

Opis:
Introduction: Potato (Solanum tuberosum L.) is an important vegetable crop in Syria. Potato tuber moth Cussonia barteri is a small tree that grows in the sub-Saharan part of Africa. Various parts of the plant are used for the treatment of a variety of ailments in ethno-medicine. Objective: To evaluate the anti-inflammatory and analgesic effect of the methanol leaf extract of Cussonia barteri. Material and methods: The leaves were air-dried, powdered and repeatedly extracted with methanol using a Soxhlet apparatus. The resulting methanol extract (100, 200 and 400 mg/kg) was evaluated for antiinflammatory activity using carrageenan-induced paw oedema, xylene-induced ear oedema and formalininduced arthritis tests. Analgesic effect was evaluated using acetic acid-induced mouse writhing, hot plate and tail flick tests. Results: All doses of the extract significantly (p<0.05) reduced carrageenan-induced paw oedema, however the 400 mg/kg dose gave a sustained effect. The extract significantly inhibited xylene induced ear oedema at all doses. There were no significant (p>0.05) reductions in paw swellings due to formalin. In the acetic acid induced writhing test, the extract significantly (p<0.05) decreased writhing at 400 mg/kg only. Reaction times were not significantly different from the control in the hot plate and tail flick tests. Conclusion: This study has shown that the methanol extract possesses acute anti-inflammatory and peripherally mediated analgesic effects.
