Opis:
Interleukin 18 (IL-18) is a pleiotropic cytokine involved in the regulation of innate and acquired immune response. In the milieu of IL-12 or IL-15, IL-18 is a potent inducer of IFN-gamma in natural killer (NK) cells and CD4 T helper (Th) 1 lymphocytes. However, IL-18 also modulates Th2 and Th17 cell responses, as well as the activity of CD8 cytotoxic cells and neutrophils, in a host microenvironment-dependent manner. It is produced by various hematopoietic and nonhematopoietic cells, including dendritic cells and macrophages. In an organism, bioactivity of the cytokine depends on the intensity of IL-18 production, the level of its natural inhibitory protein - IL-18BP (IL-18 binding protein) and the surface expression of IL-18 receptors (IL-18R) on the responding cells. This review summarizes the biology of the IL-18/IL-18BP/IL-18R system and its role in the host defense against infections. The prospects for IL-18 application in immunotherapeutic or prophylactic interventions in infectious and non-infectious diseases are discussed.

Opis:
18O-water obtained from an 2-deoxy-2-[18F]-fluoro-D-glucose (18FDG, Synthera) synthesis unit is used for purification. The purification process consists of ultraviolet (UV) irradiation followed by a distillation in a preevacuated system. After purification, the loss of enriched water was about only 5 per cent. Organic compounds, radioisotopes, trace metals and gases are eliminated efficiently. Results show that there is no significant difference in the production yield of 2-deoxy-2-[18F]-fluoro-D-glucose ([18F]FDG) whether using purified enriched water by the proposed method of purification or using the new enriched water. This method offers the necessary precaution from contamination by tritium during the purification process. The obtained 18FDG or Na-18F using the purified 18O-water is of tritium free.

Opis:
There are many aspects of materials surface processing ranging from simple heat treatments to ion implantation or laser surface treatments, e.g. the laser-plasma-coating formation in nitrogen gas, which is called laser nitriding. These methods are often very complicated, involving many basic processes and they have to be optimised for the desired application. Nuclear methods can be successfully applied for this task. First, Ag/Fe multilayers can be prepared by pulsed laser deposition employing an excimer laser. The Ag/Fe system is immiscible and sharp interfaces are expected for that reason. Nevertheless, it will be presented that some intermixing occurs which can be seen by conversion electron Mössbauer spectroscopy with the additional trick of preparing a sensitive $\text{}^{57}$Fe layer at various distances from the interfaces. Several Ag neighbours to the Fe spy-atom can be resolved, whose abundances vary with the location of the $\text{}^{57}$Fe marker layer. It is found that the intermixing at the bottom and top interface of the Fe layers is asymmetric.

Opis:
Alkylphosphocholines (APCs) are new generation, highly selective antineoplastic drugs, whose mechanism of action is not fully understood. It is known that in contrast to traditional chemotherapeutics, APCs do not induce cell death by apoptosis or necrosis as a result of DNA damage, but target cellular membranes and affect their biophysical properties. However, it is still unknown which membrane component attracts APC molecules selectively to cancer cells. In order to get insight into this issue, systematic investigations on the interactions between APCs and particular membrane components are highly required. Such experiments can be performed with the Langmuir monolayer technique, serving as a biomembrane model. Because of overexpression of gangliosides in tumor progression and the ability of APCs to insert into membrane rafts, two sphingolipids, i.e. sphingomyelin (SM) and ganglioside $GM_1$ have been examined as potential membrane targets. In this respect, their interactions with three alkylphosphocholines, differing in their hydrophobic part: hexadecylphosphocholine (HePC), octadecylphosphocholine (OcPC) and erucylphosphocholine (ErPC) have been studied and the following systems have been analysed: SM(or GM1)/HePC, SM(or GM1)/OcPC and SM(or GM1)/ErPC. It was found that all the investigated APCs show strong affinity to ganglioside in contrast to sphingomyelin. Differences in interaction of APCs with both investigated sphingolipids were studied based on experimental surface pressure (π) versus mean molecular area (A) isotherms, and analyzed qualitatively (with mean molecular area values) as well as quantitatively (with Δ $G^{exc}$ function). The obtained results have also been analysed taking into consideration geometry of interacting molecules. Our results suggest that gangliosides may be molecular targets for APCs, attracting them to tumor cells. Although the interactions with sphingomyelin were found to be unfavourable, further studies on more complex system, containing APCs mixed with sphingomyelin and cholesterol, are required to better understand the role of lipid rafts in the selectivity of APCs.
