Opis:
Blakeslea trispora is a good alternative source for producing such carotenoids as lycopene and β-carotene. The objective of this research was to elaborate a method for the simultaneous determination of lycopene and β-carotene in Blakeslea trispora products using a usual UV-vis spectrophotometer. The standard solutions of the mixture of different concentrations of β-carotene and lycopene were measured with the UV-vis method and correlation formula for the extinction coefficients of 1% standard solution of lycopene in the solvent (hexane) and the ratios of the optical densities at the character peaks of 470 and 502 nm was elaborated. This gives a possibility to calculate the concentrations of lycopene and β-carotene in the mixture. The prediction quality of the UV-vis method was sufficient and the obtained results were very close to the ones, being measured with the HPLC technique. The proposed method can be used for both routine industrial work and academic research, providing the rapid analysis for simultaneous measurements of lycopene and β-carotene.

Opis:
Due to the valuable properties of carotenoidsthe interest in new methods of obtaining them is still increasing. For this purpose, since the 60s of the twentieth century, scientists conducted numerous studies to recognize the processes in which carotenoids could be produced . Carotenoids, revealing a strong antioxidant activity, act as free radical scavengers. These compounds can support many beneficial processes such as the stimulation of the immune system, the modulation of intracellular signaling pathways, the regulation of the cell cycle and apoptosis as well as theregulation of growth factors. Carotenoids were determined by spectrophotometric analysis in the following fungal species: Leucopaxillus giganteus, Sarcodon imbricatus, Lactarius piperatus, Lactarius deliciosus, Agaricus arvensis, Agaricus bisporus, Agaricus romagnesii, Agaricus silvaticus, Agaricus silvicola, Hypholoma fasciculare, Calocybe gambosa, Craterellus cornucopioides, Marasmius oreades. High performance liquid chromatography (HPLC) coupled with a UV detector detected β,β-carotene in Agaricus bisporus, Polyporus squamosus, Lepista nuda, Russula delica, Verpa conica, Pleurotus ostreatus and Hypsizgus marmoreus. β-carotene and lycopene were also found in three wild species of edible mushrooms: Leucopaxillus giganteus, Sarcodon imbricatus and Agaricus arvensi. These compounds were isolated in several species of the family Cantharellus as well. One example is Cantharellus cibarius, which contains mainly β,β-carotene and minor amounts of lycopene, α-carotene and other carotenoids, which may be δ – and γ – isomers.

Carotenoids and their metabolites are essential factors for the maintenance of important life processes such as photosynthesis. Animals cannot synthesize carotenoids de novo, they must obtain them via their food. In order to make intensive animal husbandry possible and maintain human and animal health synthetic nature identical carotenoids are presently commercially available at the multi-tonnes scale per year. Synthetically accessible 13C enriched carotenoids are essential to apply isotope sensitive techniques to obtain information at the atomic level without perturbation about the role of carotenoids in photosynthesis, nutrition, vision, animal development, etc. Simple highly 13C enriched C1, C2 and C3 building blocks are commercially available via 99% 13CO. The synthetic routes for the preparation of the 13C enriched building blocks starting from the commercially available systems are discussed first. Then, how these building blocks are used for the synthesis of the various 13C enriched carotenoids and apocarotenoids are reviewed next. The synthetic Schemes that resulted in 13C enriched β-carotene, spheroidene, β-cryptoxanthin, canthaxanthin, astaxanthin, (3R,3'R)-zeaxanthin and (3R,3'R,6'R)-lutein are described. The Schemes that are reviewed can also be used to synthetically access any carotenoid and apocarotenoid in any 13C isotopically enriched form up to the unitarily enriched form.

Carotenoids in eukaryotic phototrophic organisms can be classified into two groups; β-carotene and its derivatives, and α-carotene and its derivatives. We re-examined distribution of α-carotene and its derivatives among various taxa of aquatic algae (17 classes) and land plants. α-carotene and its derivatives were found from Rhodophyceae (macrophytic type), Cryptophyceae, Euglenophyceae, Chlorarachniophyceae, Prasinophyceae, Chlorophyceae, Ulvophyceae, Charophyceae, and land plants, while they could not be detected from Glaucophyceae, Rhodophyceae (unicellular type), Chryosophyceae, Raphidophyceae, Bacillariophyceae, Phaeophyceae, Xanthophyceae, Eustigmatophyceae, Haptophyceae, and Dinophyceae. We also analyzed the chirality of α-carotene and/or its derivatives, such as lutein and siphonaxanthin, and found all of them had only (6'R)-type, not (6'S)-type.

When we investigated the genotoxicity of β-carotene cleavage products (CPs) in primary rat hepatocytes stimulated to proliferate, we observed dose-dependent increases of chromosomal aberrations, sister chromatid exchanges and micronuclei. In contrast to other genotoxic substances, however, this increased genotoxicity was not accompanied by increased cytotoxicity. As a consequence we observed metaphases showing massive chromosomal damage, indicating inhibition of apoptosis by CPs enabling these cells to proceed in the cell cycle. Since proliferative stimulation by growth factors may support this effect, the in vitro toxicological effects of CPs were studied on proliferatively quiescent primary rat hepatocytes. A significant increase of both apoptosis and necrosis was found. Supplementation with antioxidants did not significantly lower the level of apoptosis, while the level of necrosis was significantly reduced by Trolox and N-acetylcysteine at all concentrations tested as well as ascorbic acid (50 µM) and a combination of Trolox (50 µM) and ascorbic acid (50 µM). These observations indicate that a) the cytotoxic potential in combination with the genotoxic potential of CPs may promote the initiation of cells due to compensatory cell division in exposed tissues and may aggravate inflammatory processes under chronic exposure, and b) the applied antioxidants may protect from cytotoxicity primarily via the detoxification of aldehydic β-carotene cleavage products.

Opis:
A role of carotenoids as modulators of physical properties of model and biological membranes has been already postulated. However, there is a lack of information on the influence of these pigments on interactions between the lipids which form such membranes. This paper applies atomic force microscopy (AFM) in to study the effects of β-carotene on the adhesion properties of DPPC multilamellar liposomes. This allowed us to gain, for the first time, a direct insight into the interactions between the components in model systems on a molecular level. We observe that the adhesive forces in DPPC multilamellar liposomes containing 1mol% of β-carotene decrease exponentially with increasing temperature, and that at about 37°C they diminish. In the case of pure liposomes the decline in adhesion is of a different nature and the adhesive forces disappear at 34°C. The adhesive forces are about 5 times higher at 31°C in the presence of β-carotene than in its absence. However, measurements using differential scanning calorimetry (DSC) showed a shift of the lamellar-to-undulled-lamellar phase transition toward lower temperatures by about 0.8±0.2°C in a system containing β-carotene. The enthalpy changes (ΔH) of this transition are similar for both systems. For the main transition, gel-to-liquid crystalline, the peak is shifted by about 0.5±0.1°C, and ΔH decreases by about 30% in liposomes treated with β-carotene in comparison to pure liposomes. Our results suggest increased cooperation between liposome components in a system with enriched β-carotene, which cause a change in phase transition temperatures. Moreover, these interactions are very sensitive to temperature.

Pulses are a vital source of plant-based proteins and amino acids for people around the globe and should be eaten as part of a healthy diet to address obesity, as well as to prevent and help manage chronic diseases such as diabetes, coronary conditions and cancer; they are also an important source of plant-based protein for animals. Pulses provide protein and fibre, as well as a significant source of vitamins and minerals, such as iron, zinc, folate, and magnesium, and consuming half a cup of beans or peas per day can enhance diet quality by increasing intakes of these nutrients. The present attempt deals with comparison antioxidant activities of various sprouts of selected pulses. The β-carotene, ascorbic acid, total antioxidant activity, total phenol and flavonoid content of black gram (Vigna mungo), cowpea (Vigna unguiculata), desi chickpea (Cicer arietinum), and yellow mustard (Brassica alba) seeds and their sprouts (48 h) were determined. The parameters selected reflect the antioxidant capacity with respect to the dietary antioxidants (β-carotene, ascorbic acid) which were noted to be highest in chickpea sprouts and black gram sprouts respectively. The highest antioxidant activity in terms of % DPPH inhibition (49.837 ± 0.61 % ) and flavonoid content ( 211.06 ± 8.17 mg/100g ) was observed in cowpea sprouts. The highest total phenol content was noted in yellow mustard sprouts (58.45 ±6.67 mg/100g). Inclusion of pulses in the diet is a healthy way to meet dietary recommendations and is associated with reduced risk of several chronic diseases including cancer. Long-term randomized controlled trials are needed to demonstrate the direct effects of pulses on the cancer like diseases.

Electron paramagnetic resonance (EPR) spin-labeling methods were used to study the effects of carotenoids on the physical properties of saturated phosphatidylcholine (PC) membranes to evaluate the contribution of the terminal hydroxyl groups of xanthophyll molecules to the carotenoid-membrane interaction. Effects of the dipolar, terminally dihydroxylated carotenoid lutein on membrane phase transition, fluidity, order, and polarity were compared with those of monopolar (β-cryptoxanthin) and nonpolar (β-carotene) carotenoids. These effects were monitored at the membrane center as a function of the amount of the carotenoid added to the sample and as a function of temperature for fluid-phase membranes. PC membranes with different thickness (from 12 to 22 carbons in alkyl chains) were used. Carotenoids shifted to lower temperatures and broadened the main phase transition of PC membranes. They decreased the membrane fluidity and increased the order of alkyl chains. Carotenoids also increased the hydrophobicity of the membrane interior. These effects were the strongest for lutein, significantly weaker for β-cryptoxanthin, and negligible for β-carotene. They decreased with the increase of the membrane thickness. Presented results suggest that anchoring of carotenoid molecules at the opposite membrane surfaces by polar hydroxyl groups is significant in enhancing their effects on membrane properties. This manuscript also shows the ability of EPR spin-labeling methods to monitor different membrane properties that can be applied in biotechnological studies with the use of liposomes.

Long-term exposure to cadmium (Cd) leads to the development of a number of conditions associated with liver and kidney damage, reproductive and cardiovascular disorders, in addition to visual impairment, blindness and hearing loss, among others. Cadmium has been classified as a human carcinogen by the International Agency for Research on Cancer. The toxicity of Cd is related to its pro-oxidant properties and the associated increase in oxidative stress. Antioxidant ingredients may be helpful in preventing the adverse effects of Cd. The effect of well-known antioxidant vitamins (E, C, A and β-carotene) in the prevention of Cd-induced toxicity is presented in this study. Numerous studies in animal models have shown that the effects of vitamins: E, C, A, and β-carotene were effective in reducing Cd concentrations in organs and tissues and reduced Cd-induced changes in liver, kidney, and reproductive, circulatory, nervous, immune, and respiratory systems. In contrast, the limited number of human studies does not allow to accurately determine the role of these nutrients in reducing Cd-induced toxicity, indicating the need for further studies clarifying the role of antioxidant vitamins in reducing Cd-induced toxicity. However, it seems reasonable to promote the consumption of natural food products that are sources of antioxidant vitamins in groups of people with occupational and environmental exposure to Cd.

High doses of synthetic antioxidative vitamins: A, E, C and β-carotene are often used on long-term basis in numerous preventive and therapeutic medical applications. Instead of expected health effects, the use of those vitamins may however lead to cases of hypervitaminosis and even to intoxication. The article points out main principles of safety which are to be observed during supplementation with antioxidative vitamins. Toxic effects resulting from erroneous administration of high doses of those substances on organs and systems of the organism are also discussed. Attention is drawn to interactions of antioxidative vitamins with concomitantly used drugs, as well as intensification of adverse effects caused by various exogenous chemical factors. Moreover, the article presents the evaluation of supplementation with these vitamins, which was performed in large studies.

Background: Oxidative stress is involved in lead toxicity. This suggests that some antioxidants may play a role in the treatment of lead poisoning. In the light of this, the aim of the study was to determine whether β-carotene administration reduces oxidative stress and homocysteine level in workers chronically exposed to lead. Material and Methods: The exposed population included healthy male workers exposed to lead who were randomly divided into 2 groups (mean blood lead level ca. 44 μg/dl). Workers in the 1st group (N = 49, reference group) had no antioxidants, drugs, vitamins or dietary supplements administered, while workers in the 2nd group (N = 33) had β-carotene administered in a dose of 10 mg per day for 12 weeks. Biochemical analysis included markers of lead-exposure and the level of malondialdehyde (MDA), an oxidative stress biomarker. We also measured the level of homocysteine (Hcy) and thiol groups as well as the activity of superoxide dismutase (SOD) and its isoenzyme EC-SOD in serum. Results: After supplementation, the level of MDA significantly decreased, compared to baseline, by 16%, and to the reference group. When compared to the reference group, Hcy level was also significantly decreased. However, the level of thiol groups was significantly higher after supplementation with β-carotene compared to the reference group. Analogically, the activity of SOD and EC-SOD was significantly higher compared to the baseline and to the reference group. Conclusions: Despite some controversies over antioxidant properties of β-carotene, our results indicate that its antioxidant action could provide some beneficial effects in lead poisoning independent of chelation. Wstęp: Ponieważ ołów wywołuje stres oksydacyjny, przypuszcza się, że antyoksydanty mogą mieć zastosowanie w leczeniu zatrucia tym metalem. Celem niniejszej pracy było zbadanie, czy podawanie β-karotenu redukuje stres oksydacyjny oraz obniża stężenie homocysteiny u pracowników narażonych na ołów. Materiał i metody: Grupę badaną stanowili zdrowi pracownicy płci męskiej podzieleni na 2 podgrupy (średnie stężenie ołowiu we krwi: ok. 44 μg/dl). W pierwszej (N = 49, grupa porównawcza) nie stosowano żadnego leczenia, a drugiej podawano β-karoten w dawce 10 mg na dzień przez 12 tygodni. Analiza biochemiczna objęła markery narażenia na ołów i marker stresu oksydacyjnego - dialdehyd malonowy (malondialdehyde - MDA). Ponadto zbadano stężenie homocysteiny (homocysteine - Hcy), grup tiolowych oraz aktywność dysmutazy ponadtlenkowej (superoxide dismutase - SOD) i jej izoenzymu EC-SOD. Wyniki: Po suplementacji w grupie leczonej zaobserwowano obniżenie się stężenia MDA w porównaniu z wartościami wyjściowymi oraz z wynikami w grupie porównawczej. Obniżeniu uległo także stężenie Hcy w porównaniu z wynikami uzyskanymi dla grupy porównawczej. Z kolei stężenie grup tiolowych oraz aktywność SOD uległy zwiększeniu w porównaniu z grupą porównawczą, a w przypadku SOD i EC-SOD także w porównaniu z wartościami wyjściowymi. Wnioski: Wyniki niniejszego badania sugerują, że podawanie β-karotenu w zatruciu ołowiem może przynieść korzyści, takie jak zmniejszenie stresu oksydacyjnego, mierzonego pośrednio jako stężenie MDA, lub obniżenie stężenia aterogennej homocysteiny. The all-trans-β-carotene is a natural pigment used in various industrial fields (food, cosmetics, pharmaceuticals, etc) and possesses the higher provitamin A activity, in respect to other carotenoids. All-trans-β-carotene is produced industrially by chemical and biotechnological means. For β-carotene biotechnological production in industrial scale mated cultures of Blakeslea trispora, a heterothallic fungus, are mainly used. Despite the intense research for β-carotene production by B. trispora, natural substrate utilization has not been extensively studied. Solid agro-food wastes such as cabbage, watermelon husk and peach peels from northern Greece as main carbon source into submerged B. trispora cultures for carotenoids production, was examined. The media containing only the agro-food waste (2-4) gave a biomass accumulation 7.77±0.4 g/L, while a reference medium 1 with glucose (10 g/L) gave 4.65±0.21 g/L. In another experiments series agro-food wastes were used with corn steep liquor and thiamine (media 6-8), giving a biomass accumulation and total carotenoid volumetric production 10.2±2.41 g/L and 230.49±22.97 mg/L, respectively. These are the higher values reported for solid wastes so far in respect to those obtained from a synthetic medium, with higher glucose concentration of 50 g/L where the correspondent values were 9.41±1.18 g/L and 45.63 mg/L respectively. The results support that B. trispora is able to utilize, almost equivalently, different origin agro-food wastes for carotenoids production. Furthermore, β-carotene percentage in all examined cases was over 76%, as it was estimated by HPLC analysis, suggesting that these agro food wastes may be used for high purity, large scale β carotene production.

Opis:
Praca przestawia analizę jakości marchwi po suszeniu konwekcyjnym w stałej i zmiennej (częstość i amplituda) temperaturze powietrza suszącego. Plastry korzenia marchwi o początkowej zawartości wilgoci 88÷90% były suszone konwekcyjnie w stałej temperaturze 70°C i w zmiennej temperaturze, aż do końcowej zawartości wilgoci 9÷12%. Analizowana była kinetyka suszenia, zmiana koloru, aktywność wody i zawartość β-karotenu. Pokazano, że niestacjonarne suszenie prowadzone w zmiennej temperaturze minimalizuje niekorzystne efekty konwekcyjnego suszenia marchwi.
Analysis of carrot quality after convective drying at constant and variabl temperatures (frequencies and amplitudes) of drying air is presented in th paper. Slices of carrot root with initial water content of 88-90% were drie convectively at constant temperature of 70°C and variable temperature u to the final moisture content of 9÷12%. The drying kinetics, color chang( water activity and retention of β-carotene of carrot were evaluated. It was shown that non-stationary drying carried out at variable air temperature minimized unfavorable effects of convective drying of this biomaterial.

Opis:
Essential oils from needles, fruits and bark was extracted from Pinus pinea L. (stone pine) grown wildly in Jordan. The chemical composition, antibacterial activity, antioxidant activity of essential oils was evaluated. The chemical compositions were identified using Gas-Chromatography-Mass spectrometry (GC-MS) and retention Indices (Van den Dool & Kratz). The results show that the essential oil obtained from needles composed mainly of Guaiol (12.7%), limonene (11.42%), and β-caryophyllene (7.61%), while fruit contains limonene (32.56%), and α-pinene (6.78%). The essential oils from barks were rich in β-caryophyllene (16.51%), limonene (14.83%), caryophyllene oxide (11.83%), and longifolene (7.51%). In vitro, antibacterial activity of the essential oil samples was evaluated using agar-well diffusion method against three different strains of bacteria (Gram¬-positive bacteria: Staphylococcus aureus and Gram-negative bacteria: Klebsiella pneumoniae and Escherichia coli). The results showed that essential oil showed appreciable antibacterial activity against S. aureus. The essential oil from fruit exhibited weak antibacterial activity against E. coli and K. pneumoniae. Essential oils of P. pinea showed appreciable antioxidant activity in-vitro.

Aerial parts of Mercurialis annua L. were used in the present study, and fatty acid content in lipid extract was determined using GC-FID. The major fatty acids identified were α–linolenic acid (20.3%), heptadecanoic acid (12.8%), palmitic acid (11.9%), pentadecanoic acid (11.7%), cis-10-pentadecenoic acid (11.2%), linoleic acid (7.7%), tridecanoic Acid (4.6%), stearic (4.4%), cis-11,14-eicosadienoic acid (3.8%), beside of minor fatty acids (palmitoleic acid, cis-13,16-docosadienoic acid, arachidic acid, behenic acid, cis-10-heptadecenoic acid and myristic acid). Antioxidant properties of the extract were determined via DPPH radical scavenging, β-carotene bleaching assay and NO radical scavenging assay. The extract produced significant antioxidant activity in-vitro. The data shown here may broaden our knowledge on composition and antioxidant activity of lipid constituents from Mercurialis annua L.
