Opis:
Coal deposit "Katrishte" located in south-western Bulgaria, Ro = 0.34%, was subjected to geochemical study. By chromatographic and spectral methods, a variety of coal biomarkers in bitumen extract has been revealed. n-Alkane distribution pattern indicated at least two possible sources, lacustrine and terrestrial ones. The aliphatic fraction was strongly dominated by 16a(H)-kaurane and 16a(H)-phyllocladane, a proof for the presence of Pinales in coal-forming palaeoplant community. The kaurane prevalence over phyllocladane could indicate the herbaceous flowering plant contribution. The combination of sesquiterpenoids/diterpenoids pointed to Pinaceae and Araucariceae.As a biomarker assemblage peculiarity, the lack of isoprenoids, tricyclic diterpenoids, and pentacyclic triterpenoids could be indicated.Our data gave us ground to assume existence of bush moor with the presence of herbaceous flowering plants, and subordinate contributions of conifers and sedge-reeds to coal forming plant taxa of "Katrishte"

W pracy omówiono aktywność biologiczną poszczególnych grup przeciwutleniaczy, wskazując na ich rolę w zmniejszaniu częstotliwości występowania nowotworów i chorób układu krążenia. Wskazano, że etiologia chorób nowotworowych obejmuje uszkodzenia oksydacyjne w DNA oraz uaktywnienie prokancerogenów pod wpływem aktywnych rodników. Przyczyną chorób krążenia jest proces miażdżycowy wywoływany przez oksydację frakcji LD-cholesterolu. Oba rodzaje chorób mogą by monitorowane przez szereg biomarkerów przydatnych w ocenie szybkości rozwoju zmian chorobowych. Ocena aktywności prozdrowotnej żywności przenosi się wyraźnie na poziom molekularny. Wyrazem tego są narodziny nutrigenomiki, nauki badającej wpływ żywienia człowieka na funkcjonowanie jego genomu.
In this paper, the biological activity of individual groups of antioxidants is discussed with regard to the prevention of two chronic diseases: cancer and cardiac vascular disease. It is pointed out that the cancer etiology is related to oxidative damages of DNA and to activation of pro-carcinogenic factors by free radicals whereas the cardiac vascular disease is initiated by LDL oxidation. The development of both diseases can be monitored by specific biomarkers that appear useful while estimating the developmental rate of pathogenic changes. For the purpose of appraising biological activity of functional foods, molecular analysis of antioxidative effects is employed. A new science has been established: nutrigenomics, which is the study of the effect of human nutrition on his genome functioning.

The Algerian west coast is the prime recipient ofsev eral forms of pollution; hence, the necessity for an impact assessment of this coastal pollution using a suite ofr ecommended marine biomarkers, including lysosomal membrane stability in living cells by the Neutral Red Retention Time (NRRT) method, the evaluation ofmicron ucleus (MN) frequency, and the determination ofacet ylcholinesterase (AChE) activity in mussels Mytilus galloprovincialis, sampled from the large, polluted Oran Harbour (OH) and the Maˆarouf( Mrf) marine mussel farm between July 2005 and April 2006. The difference in the variations oft he annual physical parameters between OH and Mrfco rresponds to the influence ofthe domestic and industrial sewage discharged by the city ofO ran. The biological data ofthe mussels (condition index, protein content) recorded at both sites were related to their natural reproductive cycle. This indicated that intrinsic variation between the sites due to different mussel development phases was minimal. The variation in the AChE activity of some organs ofO H and Mrfm ussels, with minimal inhibition in July and a higher NRRT recorded in the granular haemocytes in the Mrfthan in the OH mussels during the autumn and spring, depends on the quality ofthe biotope and on generic stress factors. Moreover, the variation in MN frequency, in general reflecting a non-significant seasonal and spatial genotoxic effect ofthe contamination at the two sampling sites, requires further investigations regarding biotic and abiotic variations.

Previous studies in the Gulf of Gdańsk discussed the responses of selected enzymatic biomarkers to the contaminant gradient in fish and mussels. In the present study, flounder muscle and liver tissues were analyzed for polychlorinated biphenyls (PCB congeners: 28, 52, 101, 118, 138, 153 and 180), organochlorine pesticides (HCHs, HCB and DDTs), and trace metals (Pb, Cd, Zn, Cu, Hg, Cr). An attempt was made to identify the relationship between the measured enzymatic biomarker responses (cholinesterases, malic enzyme, isocitrate dehydrogenase, glutathione S-transferase) and contaminant concentrations in selected flounder tissues. The observed differences in enzymatic biomarker levels suggest that chronic exposure to low-concentration mixtures of contaminants may be occurring in the studied area. However, no conclusive evidence was found of a clear link between the biomarker responses and contaminant concentrations in flounder tissues.

Recent behavioural medicine studies have revealed that the human secretory substances change according to his/her mental states. Especially, those substances transiently get increase (or decrease) against the short-term experimental stressors. However, the relation with rather longer stressor, or daily stressor, has not yet well understood or reported discrepant results. One possible reason for this discrepancy might be brought from uncertainty of the psychological evaluation, which is the score of subjective questionnaire. We then introduced an evaluation criterion of attributes based on rough set theory to analyze the relation between the psychological state and a major immune substance. As a result, several items in the questionnaire were extracted as dominant items, while there was still no correlation between them.

This review summarizes the application of Tetrahymena spp. in ecotoxicology, in order to promote a more integrated, multi-level ecotoxicological assessment approach regarding the effects of chemical stressors on several biological levels (from molecule to ecosystem). Such a multi-level testing approach in one species facilitates the establishment of missing causal relationships between biochemical responses and ecological effects. The review illustrates that Tetrahymena spp. represent excellent ecotoxicological test species due to their important role in the microbial foodweb, wide distribution and abundance, sequenced genome in T. thermophila, large background knowledge and scientific publications in cellular biology, ecology and ecotoxicology. Several bioassays have already been developed on different biological organisation levels, such as enzyme assays (biochemical level), behavioral tests (individual level), population growth tests (population level) and microcosms (community level). Moreover, specific mode-of-action based assays are available (e.g. genotoxicity), or are in development (e.g. endocrine disruption and neurotoxicity). Tetrahymena spp. combine traits of (1) a single cell, thus might replace or complement specific cell-line testing approaches, with traits of (2) a whole organism and population, thus allowing to study complete metabolic pathways and its consequences on population growth and genetic adaptation. Assays involving Tetrahymena spp. might easily be adapted for a rapid multi-level in situ or ex situ toxicity biosensor test system for ecologically relevant risk assessment.

Objectives: Obesity, insulin resistance and dyslipidemia are the most significant risk factors of non-alcoholic fatty liver disease (NAFLD) but the role of adipokines in the pathogenesis of this disease is not clear. Assessment of retinol binding protein (RBP-4) seems to be promising because data from animal and human studies suggest its role in the patomechanism of insulin resistance. Therefore, the aim of the study was to evaluate the serum levels of RBP-4 in children with NAFLD. Methods: Fasting serum level of RBP-4 was determined in 42 obese children with suspected liver disease and 20 lean controls. The degree of liver steatosis was graded in ultrasound according to Saverymuttu. The intrahepatic lipid content was assessed noninvasively in a semiquantitative fashion using 1HMR spectroscopy (1.5-T scanner with PRESS sequence). Results: Fatty liver was confirmed in 30 children by ultrasonography (16 of them had also increased alanine transaminase (ALT) activity). Serum concentrations of RBP-4 were significantly higher in obese children with NAFLD compared to controls. Significant correlations were found between RBP-4 level and ultrasonographic grade of liver steatosis, intrahepatic lipid content (1HMRS) and triglycerides level, while the serum level of RBP-4 was not significantly higher in children with advanced liver steatosis (grade 2-3, n = 11) compared to patients with mild steatosis (grade 1, n = 19). The ability of RBP-4 to differentiate children with advanced liver steatosis from those with mild steatosis was not significant. Conclusion: RBP-4 can be considered as a convenient serum marker of intrahepatic lipid content in obese children.

Objective: Our aim is to evaluate the validity of exhaled carbon monoxide (CO) and of a newly-created score as markers of Chronic Obstructive Pulmonary Disease (COPD). Materials and Methods: The CO level was measured in a derivation subsample of a cross-sectional study and linked to COPD diagnosis; its predictors were evaluated, and a scale was constructed. It was evaluated in a validation subsample and in a clinical setting. Results: Individuals with COPD had higher CO levels than healthy individuals. CO level signifi cant predictors were cigarettes per day, waterpipes per week, lower age, male gender, living close to diesel exhaust, heating home with the use of diesel, and having indoor family smokers. A score composed of CO predictors was able to signifi cantly predict COPD (Ora = 4–7.5). Conclusions: Coupled with the clinical judgment of physicians, this scale would be an excellent low-cost tool for screening COPD, in absence of spirometry.

W dobie intensywnego rozwoju chorób cywilizacyjnych, m.in. nowotworów żołądka, niezbędne jest zaprojektowanie markerów, które umożliwią wczesne ich wykrycie, pozwolą na szybkie podjęcie leczenia oraz zapobiegną zwiększeniu liczby zgonów. Taką potencjalną możliwością są mikroRNA, czyli krótkie cząsteczki RNA modulujące aktywność ściśle określonych fragmentów RNA i odgrywające szczególną rolę w wielu procesach fijologicznych i patologicznych. Wpływając na proliferację, cykl komórkowy oraz apoptozę komórek nowotworowych odgrywają znaczącą rolę również w nowotworzeniu. Wykrywalne w osoczu odstępstwa od ich normalnego poziomu, związane z każdym z etapów rozwojowych nowotworu mogą czynić je skutecznymi i nieinwazyjnymi biomarkerami, pozwalającymi nie tylko na detekcję i rozpoznawanie nowotworów, ale także na ich predykcję. Dalsze poznanie ich funkcji i właściwości może owocować odkryciem skutecznych leków do walki z nowotworami.
In the era of intensive development of lifestyle diseases such as stomach cancer, it is crucial to design markers that enable their early detection, allow rapid introduction of treatment and prevent the increase in the number of deceases. Potential possibility is microRNAs, short RNA molecules which modulate the activity of specifi fragments of RNA and play an important role in many physiological and pathological processes. Affcting cell proliferation, cell cycle and apoptosis of cancer cells also play a significant role in neoplasia. Detectable levels of derogation from the regular level associated with each of the stages of tumor development may make them effctive and non-invasive biomarkers, allowing not only for early detection and identifiation of cancer’s type, but also on their prediction. Further understanding of their functions and properties can result in the discovery of effctive methods of cancer treatment.

The present study deals with the estimation of antioxidant levels in blood of sewage and garage workers exposed to sewage water, washing water and vehicular air pollution compared with control group of population. The study areas were selected on the basis of drainage clearance and wards where workers are active as sewage workers. For garage workers, the garage selected was a large and busy one where continuous vehicles washing, repairing etc are carried out. These two exposed groups were compared to non-exposed group of population as control. The blood samples were collected from two groups of exposed population as well as control population and the antioxidant levels were estimated in blood. The present results clearly indicate that there was decreasing trend of the antioxidant level in blood for sewage and garage workers. In the control groups of population the antioxidant levels were found to be within normal range, but for sewage workers (0.14-0.36 mM) and garage workers (0.12-0.36 mM) the antioxidant levels were significantly lower. The range of antioxidant level in human blood is 0.5 to 2.0 mM. It was found to be significantly lower (P < 0.001 or 0.05) in addicted sewage and garage workers when compared with non-addicted groups. All the exposed groups were showed to be having significantly lower antioxidant level when compared to control groups. This study is a preliminary assessment to know the potent biomarkers for oxidative stress and estimation of antioxidant level in blood due to occupational exposure. This study shows a way for easy screening of biomarker assessment but further work is needed in relation to biochemical, enzymological and genetic damage study.

Arsenic (As) is a metalloid that causes severe water pollution due to its extravagant toxicity. Ceriodaphnia dubia, a freshwater crustacean, was selected as a model system to evaluate the degree of time and dosage dependent acute toxicity caused by pentavalent As [As(V)]. C. dubia were collected from a natural pond and treated with different concentrations of As(V) for 24 hours and 48 hours. For both 24 hours and 48 hours treatment periods, the mortality rates were increased significantly ( P< 0.05) with increase in As(V) concentrations. Simultaneously, it was also observed that As(V) - induced mortality in C. dubia also depended on the time of exposure to the metalloid. We propose this model as a low-cost technique towards rapid screening of water quality in relation to As contamination.

Regimen-related mucosal toxicity is extremely common following cytotoxic chemotherapy and radiotherapy. Mucositis is as an important determinant of the inflammatory response and infectious complications in cancer treated patients. Most assessment scales for mucosal damage are focussed on oral mucositis, since it is easy to evaluate. Measuring gastrointestinal musocal damage objectively remains difficult because it cannot be seen directly or readily detected. One of potential non-invasive biomarkers of gastrointestinal mucosal damage is plasma citrulline level. Citrulline is an amino acid produced by small bowel enterocytes. Low concentration of free circulating citrulline signifies severe intestinal mucosal damage in humans with nonmalignant disorders, such as villous atrophy-associated diseases, short bowel syndrome, Crohn's disease, and is used in follow-up after small bowel transplantation. The plasma citrulline level is a reliable and objective biochemical marker of enterocyte mass and function in humans, and therefore can be used to monitor enterocyte toxicity resulting from chemotherapy and radiotherapy during anticancer therapy in patients with severely disturbed gut integrity.

Changes in activity of acid hydrolases in tissues of wild pheasants induced by heavy metals

The aim of the study was to determinate the activity of chosen acid hydrolases in tissues of pheasants (Phasianus colchicus L.) and first attempts to use these enzymes as biomarkers of the effect of birds exposure to heavy metals in the environment. The samples of liver, kidneys and testes were collected from wild pheasants shot in the Podkarapacie region in contaminated (n = 5) and clear (n = 5) areas. Tissues were examined for the cadmium and lead concentrations by AAS method, as well as the activity of five hydrolases. The major site of cadmium accumulation were kidneys, the levels of Cd found in liver and testes were by 5-fold and 30-fold lower (P < 0.05), respectively. Similarly, Pb concentrations observed in the liver and testes were lower than in kidney, by 1.5-fold and 6-fold, respectively. The presence of four glycosidases and arylsulphatase in all examined tissues was demonstrated and N-acetyl-β-D-glucosaminiadase (NAG) was identified as the most active enzyme in all studied tissues. Significant differences (P < 0.05) were observed only in the level of NAG activity in liver of pheasants inhabiting the contaminated area as compared to the birds from the clear region (control group). We conclude that the determination of NAG in birds organs can be used as biomarker of environmental exposure to cadmium.
Celem badań było oznaczenie aktywności wybranych kwaśnych hydrolaz w tkankach bażantów (Phasianus colchicus L.) oraz zastosowanie po raz pierwszy tych enzymów jako biomarkerów narażenia środowiskowego ptaków na metale ciężkie. Próbki wątroby, nerek oraz jąder zostały pobrane z bażantów odstrzelonych na Podkarpaciu w rejonie zurbanizowanym (n = 5) i ekologicznym (n = 5). Analizy tkanek obejmowały oznaczenie zawartości metali ciężkich (Cd, Pb) metodą AAS oraz aktywności kwaśnych hydrolaz. Głównym narządem docelowym akumulacji metali ciężkich były nerki, zawartość kadmu w wątrobie oraz jądrach była niższa odpowiednio 5- i 30-krotnie. Podobną zależność zaobserwowano dla ołowiu, koncentracja tego metalu w jądrach i nerkach była niższa odpowiednio 1.5- i 6-krotnie. We wszystkich badanych tkankach stwierdzono obecność czterech glikozydaz i arylosulfatazy, przy czym najobfitszym źródłem enzymów były jądra, a najbardziej aktywnym enzymem we wszystkich tkankach była N-acetylo-β-D-glukozaminidaza (NAG). Wykazano istotne różnice (P < 0.05) w poziomie aktywności NAG w wątrobie bażantów bytujących na terenie zurbanizowanym w porównaniu do regionu ekologicznego. Aktywność NAG w tkankach ptaków może być przydatnym biomarkerem środowiskowego narażenia na kadm.
