Opis:
In transgenic plant development, the low transformation efficiency of Agrobacterium with exogenous DNA is the major constraint, and hence, methods to improve its transformation efficiency are needed. Recently, nanoparticlemediated gene transfer has evolved as a key transformational tool in genetic transformation. Since silver nanoparticles (AgNPs) can induce pores on the cell membrane, their efficacy in the improvement of conventional calcium chloride freeze-thaw technique of transformation of Agrobacterium was explored in this study. Agrobacterium cells in the exponential growth phase were exposed to different concentrations of AgNPs (0.01, 1, 5, 10, and 20 mg/l), and the half-maximal effective concentration (EC50) was determined via Probit analysis using the SPSS software. Transformation efficiency of AgNPs alone and in combination with calcium chloride was compared with that of the conventional calcium chloride freeze-thaw technique. AgNPs at a concentration of 0.01 mg/l in combination with calcium chloride (20 mM) showed a ten fold increase in the transformation efficiency (3.33 log CFU (colony-forming unit/microgram of DNA) of Agrobacterium tumefaciens strain EHA 105 with plasmid vector pART27 compared with the conventional technique (2.31 log CFU/μg of DNA). This study indicates that AgNPs of size 100 nm can eliminate the freeze-thaw stage in the conventional Agrobacterium transformation technique, with a 44% improvement in efficiency. The use of AgNPs (0.01 mg/l) along with 20 mM calcium chloride was found to be an economically viable method to improve the transformation of Agrobacterium with exogenous plasmid DNA.

Opis:
The aims of this study are to model past (LIG, LGM and Mid-Holocene), present and future (2050 and 2070) distributions of the Quercus vulcanica Boiss. & Heldr. ex Kotschy by using Maximum Entropy, and to predict suitable areas for the conservation of the species for future planning. MaxEnt distribution modeling was used to model distributions. Results for past bioclimatic conditions show that the distribution area of the species expanded and then contracted (LIG to LGM and LGM to HOL). The modelling shows that the distribution range of the species will be narrower in the future. The species will be facing extinction towards 2070. Therefore, the conservation status of the species should be evaluated according to the present findings. Although the largest population of the Q. vulcanica is found in Isparta and Afyonkarahisar Provinces located in Southwestern Turkey, this area will not be suitable for the growth and survival of the species in the future. For this reason, a new nature reserve area should be established in a more suitable climate.

Opis:
This research was conducted to investigate the effects of phytostimulators application (Messenger, Crop-Set, ISR-2000) on yield and morphological parameters of common bean cultivars grown under four irrigation regimes [25% (I₂₅), 50% (I₅₀), 75% (I₇₅) and 100% (I₁₀₀)]. Phytostimulators reversed the negative effect of drought on plant growth. Significant interaction was determined for all parameters except stem diameter and stomatal conductivity between phytostimulator and drought applications. The best effect on stomatal conductivity was provided from ISR-2000 (23.5% reduction) application. The highest yield was obtained from the 25% water deficiency applied with 1.91 ton per hectare. It was determined that the best results were obtained from Messenger in Efsane and Asya cultivars and ISR-2000 in the Öz Ayşe cultivar on yield. Therefore, it was concluded that the use of phytostimulators under drought stress is important for the effective use of water.

Opis:
A unique functional adaptation to herbivory within early ray-finned fishes is exemplified by the late Permian actinopterygians within the family Eurynotoidiidae with policuspid teeth strongly modified with respect to the primitive actinopterygian conditions. Here we report additional finds of multidenticulated teeth from the fluvial latest Permian deposits of Russia. The teeth belong to the members of endemic Eurynotoidiidae and show rather high morphological diversity. We confirm that the Russian forms are the earliest known ray-finned fishes with substantial modifications of teeth adapted to the processing of food. These finds confirm some previous suggestions that the adaptation to herbivory first developed in freshwater fishes, not marine. We found very similar dental adaptations in some groups of Recent freshwater teleosts, especially in characiforms and cichlids. It suggests that sympatric species of Permian Eurynotoidiidae explored various herbivorous niches like modern fish in East African lakes. Apparently, this first pulse of adaptive radiation in ray-finned fishes was probably caused by diversification of Permian aquatic vertebrate community.

Purpose: To determine effect of working in the night shift system on nutritional status, anthropometric measurements, and risk of cardiovascular disease of health staff. Materials and Methods: This cross-sectional study was conducted between August-November 2017 at Samsun Ondokuz Mayıs University among 111 health staff. Data was collected with a questionnaire form including questions about demographic characteristics, nutritional status and anthropometric measurements. SPSS 21.0 statistical package program was used for statistical analysis and p<0.05 was considered statistically significant. Results: Based on waist circumference, 14.3% of men and 31.1% of women were at high risk, and based on waist/hip ratio, 4.8% of men and 33.3% of women were at risk of cardiovascular diseases. It has been shown that eating patterns are disrupted during shifts (89.2%), and 73.9% of participants cannot eat because their meals cool down during shifts. The difference in the numbers of main and snack meals consumed by health staff during the night shift was statistically significant (p<0.05). Body mass index, waist circumference, and hip circumference measurements decreased with increasing shift time. However, body mass index, waist circumference, and hip circumference increased as time spent in the profession increased. Conclusion: It was seen that night shift health staff are at risk of cardiovascular disease due to insufficient and unhealthy nutrition. It is clear that nutrition education programs are required for health staff working night shifts to reduce excess weight and obesity in this population.

Opis:
Sauropterygia, one of the main clades of Mesozoic marine reptiles, diversified shortly after the Permo-Triassic biotic crisis and afterwards remained one of the major components of Early Triassic and later Mesozoic marine ecosystems. On the other hand, actual specimens of marine reptiles of Olenekian age are still rare in the fossil record, coming only from a few localities worldwide. Here we describe associated remains of a larger marine reptile of around 4 m body length, with nothosauroidean affinities from the Sulphur Mountain Formation exposed at the L cirque locality of Wapiti Lake area in British Columbia. Although the specimen records only scattered parts of the posterior vertebral column, some gastral ribs and most notably, the proximal portion of one hind limb together with a fan-shaped ischium, it represents one of the oldest records of Sauropterygia and larger representatives of aff. Nothosauroidea specifically, as well as the northernmost occurrence of such animals in the Triassic. As such, the new specimen is important for understanding the biogeography and early evolution of the group and that of Sauropterygia, in general.

Opis:
Erythrosuchidae is a clade of early archosauriform reptiles that were large-bodied, hypercarnivorous, possibly apex predators in late Early and Middle Triassic ecosystems following the Permo-Triassic mass extinction. Chalishevia cothurnata from the late Middle Triassic (Ladinian) of Russia, is the stratigraphically youngest known erythrosuchid species, but the holotype and referred material of this taxon has received little study. Here, we provide the first detailed anatomical description of C. cothurnata, including comparisons to other erythrosuchids. Although known from relatively fragmentary material, the anatomy of C. cothurnata is distinctive, including an autapomorphic strongly slanted ventral border of the antorbital fossa. The presence of a large accessory opening (the “accessory antorbital fenestra”) in the skull between the premaxilla, nasal and maxilla, together with the inferred presence of a narrow postnarial process of the premaxilla that articulated with a slot on the nasal, provides strong evidence for a sister taxon relationship between C. cothurnata and the erythrosuchid Shansisuchus shansisuchus from the early Middle Triassic (Anisian) of China. The inferred basal skull length of C. cothurnata was approximately 80 cm, making it one of the largest erythrosuchids known.

The study aimed to determine the most appropriate irrigation scheduling for mini-type cucumber plants grown as a spring-summer cycle crop. The research was conducted in a greenhouse structure in the fields of the Research Institute in Kirklareli. Marathon cucumber plants were used as the plant material in the trials. The split plot design experimental layout in 3 replications was used in the research. Four different plant–pan coefficients (0.75, 1.00, 1.25 and 1.50) and two irrigation intervals (2 and 4 days) were applied as subplots and main plots of the study, respectively. As a result of the 3-year investigation, it was determined that yields, number of cucumber fruits, and fresh cucumber fruit weight, length and diameter increased with the increase in the irrigation water amount. The highest average total soluble solids (Brix) value of 5.0 was recorded under conditions of most severe stress imposed on the plots with lowest water application rates of Kcp 0.75. While the lowest values of 4.0–4.1 were obtained for plants growing under more favorable moisture conditions in plots with application of Kcp 1.25 and 1.50. Statistically significant positive linear relationships were obtained for irrigation water amount on one hand, and fruit number, fruit mean weight, fruit length and diameter on the other, while the relationship between water applied and TSS (Brix) was negative.

Sacisaurus agudoensis is the only silesaurid known from the Triassic beds of the Santa Maria Supersequence and the correlation of its type locality to the other Triassic deposits of south Brazil has always been controversial. In an attempt to improve this, a handful of dinosaur and cynodont remains found associated to S. agudoensis are here described and compared. The anatomy of the sauropodomorph is more similar to that of Norian forms such as Pantydraco caducus and Unaysaurus tolentinoi than to that of Carnian taxa such as Saturnalia tupiniquim and Pampadromaeus barberenai. The cynodonts recovered based on isolated teeth include a brasilodontid and a Riograndia-like form. This assemblage is consistent with a Norian age, as is also suggested by local stratigraphic correlation, which positions the site in the Caturrita Formation.

Opis:
Phenolic compounds are naturally occurring substances in plants. Basil (Ocimum basilicum L.) belongs to the family Lamiaceae, is a good source of phenolic compounds and natural antioxidants. Elicitors can activate specific genes involved in secondary metabolite biosynthesis. Chitosan, as elicitor, is a natural biopolymer modified from chitin, which is the main structural component of squid pens, cell walls of some fungi and shrimp and crab shells. On the other hand, water deficit stress is one of the most abiotic stress, which effects on the levels of secondary metabolites. To evaluate the effect of chitosan and different irrigation regimes on essential oil yield, total phenol content and antioxidant activity of extracts from green and purple basil (Ocimum basilicum L.), an experiment was conducted at Shahrekord, southwestern Iran. Treatments comprised control, 0.0, 0.2, and 0.4 g·L–1 chitosan applied to plants under normal irrigation, slight and mild drought stress conditions. Results indicated that the different levels of chitosan and irrigation had significant effects on the essential oil yield, total phenol content, and the antioxidant activity of the extracts. Foliar-applied chitosan in particular 0.4 g·L–1 increased total phenolic content in the basil as compared to untreated plants. In conclusion, it is suggested that the foliar application of chitosan as an elicitor could be a promising material used to increase biological activity and pro-health functional value of basil plants.
