Opis:
The effective piezoelectric coefficients of BaTiO₃ ferroelectric films epitaxially grown on different single crystal substrates with finite thickness have been theoretically analyzed. The effective longitudinal converse piezoelectric coefficients d₃₃ of film and "film-substrate" heterostructure all monotonously increased with increase of the film thickness fraction k, and the latter is always larger than the former at the range of 0 < k < 1. Meanwhile, we also found that the effective piezoelectric coefficients d₃₃ were affected by the substrates due to different elastic constants. These results show that the elastic deformation and clamping effect of substrate have significant impacts on the piezoelectric behavior of bilayer heterostructure.

Opis:
Objectives To investigate the influence of the anti-tumor necrosis factor-α infliximab (IFX) in the case of rats with silicosis. Material and Methods Forty-eight Wistar rats were randomly divided into 3 groups. The study group (N = 16) – silicosis was induced by intratracheal instillation of 50 mg silica on day 1, and IFX was subcutaneously administered at a dose of 15 mg/kg of body weight from day 2 to day 6, the vehicle group (N = 16) – silica used as the study group but without IFX, the sham group (N = 16) – 1 ml of saline was intratracheal-used. Eight rats in each group were euthanized on day 7 and on day 14, respectively. Lung tissue sections were stained with hematoxylin and eosin or Masson’s trichromedye. The nuclear factor-κB p65 (NF-κB p65) positioning in the lung tissues were determined by immunohistochemical staining. Levels of tumor necrosis factor α (TNF-α) in rat serum and bronchoalveolar lavage fluid were measured with enzyme linked immunosorbent assay. The inducible nitric oxide synthase (iNOS) mRNA in the lung tissues was measured by quantitative real-time polymerase chain reaction, as well as inhibitor protein-κB (I-κB) and NF-κB p65 expression were measured quantitatively by western blotting. Results Silica installation increased the lung tissues inflammation reaction, oxidative stress and pulmonary fibrosis. Infliximab treatment significantly improved silica-induced lung pathological changes (inflammatory cells, collagen deposition), decreased the TNF-α inhibited NF-κB signaling (I-κB, NF-κB p65) as well as oxidant status (iNOS). Conclusions Infliximab may improve silica-induced pulmonary inflammation by decreasing the TNF-α, inhibiting NF-κB signaling (I-κB, NF-κB p65) as well as oxidant status (iNOS), which suggest that IFX has potential role in the treatment of silica-induced lung damage. Through taking the cold rolling process as the research object, the three-dimensional finite element model of the strip rolling process is established by using ANSYS/LS-DYNA software. The actual rolling product data has strong consistency with the finite element simulation results. The rolling process is dynamically simulated, and the distribution curves of important rolling parameters such as equivalent stress, control efficiency coefficient, transverse rolling pressure, lateral thickness and work roll deflection is obtained. Based on summarizing the influence of rolling parameters on rolling deformation, the research results of this paper can play an important role in the actual rolling process control. The research results have certain guiding significance for the development and optimization of the rolling control system.

Opis:
The Kuroshio Extension front (KEF) considerably influences the underwater acoustic environment; however, a knowledge gap persists regarding the acoustic predictions under the ocean front environment. This study utilized the high-resolution ocean reanalysis data (JCOPE2M, 1993–2022) to assess the impact of the KEF on the underwater acoustic environment. Oceanographic factors were extracted from the database using the Douglas-Peucker algorithm, and acoustic propagation characteristics were obtained using the Bellhop raytracing model. This study employed a backpropagation neural network to predict the acoustic propagation affected by the KEF. The depth of the acoustic channel axis and the vertical gradient of the transition layer of sound speed were identified as the fundamental factors influencing the first area of convergence, with correlations between the former and the distance of the first convergence zone ranging from 0.52 to 0.82, and that for the latter ranging from −0.42 to −0.7. The proposed method demonstrated efficacy in forecasting first convergence zone distances, predicting distances with less than 3 km error in >90% of cases and less than 1 km error in 68.61% of cases. Thus, this study provides a valuable predictive tool for studying underwater acoustic propagation in ocean front environments and informs further research.

Opis:
The extracellular domain of the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) may function as a soluble cytokine to selectively kill various cancer cells without toxicity to most normal cells. We used a high-biosafety plasmid pVAX1 as a vector and constructed a recombinant plasmid expressing the extracellular domain (95-281 aa) of human TRAIL fused with signal peptides of human IgGγ, designated as pVAX-sT. Transduction of human BEL7402 liver cancer cells with pVAX-sT led to high levels of sTRAIL protein in the cell culture media and induced apoptosis. The therapeutic potential of pVAX-sT was then evaluated in the BEL7402 transplanted naked mouse model. Subsequent intratumoral administration of naked pVAX-sT resulted in the expression of soluble TRAIL in the sera and the tumor site, as well as effective suppression of tumor growth, with no toxicity to liver. In conclusion, the successful inhibition of liver cancer growth and the absence of detectable toxicity suggest that pVAX-sT could be useful in the gene therapy of liver cancer.

Opis:
Layouts of bus networks in cities are always irrational currently, transport service frequencies also need to be optimized according to the real network layouts, operation conditions and travel experience of passengers, so it is essential to optimize bus transit network layouts and transport service frequencies systematically. Different stakeholders are involved in the optimization of urban bus transit network layouts like the government, operators and passengers, whose interests are always contradictory. In order to optimize transit network layout and service frequencies from the view point of operators and utilizers, this research constructs a multi-objective model and proposes a solution algorithm. The proposed multi-objective model is established from the perspective of operators with the goal of minimizing total operating costs for one day, and from the perspective of the utilizers to minimize the total travel time, respectively. Also with the application of electric bus in cities, buses in this research are electric buses all for green travel. Moreover, a solution algorithm is proposed in this research to solve the proposed multi-objective model with simulated annealing algorithm and genetic algorithm. Simulated annealing algorithm is used as the main framework of the solution algorithm from the perspective of operators to minimize operating costs, while genetic algorithm is used as the subroutine of simulated annealing algorithm to optimize total travel time. Verification of the proposed model and the solution algorithm is based on an intuitive network. The application results of a numerical experiment verified that the proposed optimization model and the solution algorithm are able to optimize the network layout and service frequencies at the same time.

Opis:
On a global basis, cash cropping is usually identified as an important enterprise undertaken by farmers to increase incomes. However, the responses of farmers to these new enterprises vary greatly. Through a case study of a Dai village in Xishuangbanna, China, this paper examines how farmers make decisions about adopting new cash crops by focusing on the farm economy and land conditions. The results show that farmers did not adopt watermelons due to poor irrigation and accessibility conditions, and then they did not adopt bananas due to a transient collapse of banana market, induced by a rumor suggesting that eating bananas causes cancer. Consequently, although these non-adopters benefited from commercial exchanges with external businessmen in terms of ecological experiments and management diversification, and leasing lowlands to external businessmen is a livelihood choice that is based on the outcome of the farmer’s trade-off between profitability and risk, they missed opportunities to substantially increase incomes through cash cropping, as evidenced by the success of the farmers who adopted the cash crops. These findings suggest that the government ought to design tailored extension programs for villages, implement efficient refutation strategies to prevent rumor-induced market collapse and promote extension services as early as possible in the initial stages of transition to cash cropping.

Caspase-8 is a member of the cysteine-aspartic acid protease (caspase) family which plays a central role in apoptosis and development. We screened caspase-8 interacting proteins from mouse T-cell lymphoma and 7.5-day embryo cDNA libraries by yeast two-hybrid system and obtained eleven positive clones, including Vacuolar protein sorting 41 (Vps41), a protein involved in trafficking of proteins from the late Golgi to the vacuole. The interaction of Vps41 with caspase-8 was confirmed by co-immunoprecipitation (co-IP) and co-localization studies in HEK293T cells. Co-IP experiments also showed that Vps41 binds to the p18 subunit of caspase-8 through its WD40 region and RING-finger motif. Furthermore, we found that overexpression of Vps41 promotes Fas-induced apoptosis in A549 human lung adenocarcinoma cells. The cleavage of caspase-3, a caspase-8 downstream effector, was increased when cells were transfected with Vps41-overexpressing plasmid. Together, these results suggest a novel interaction of caspase-8 with Vps41 and provide a potential role of Vps41 beyond lysosomal trafficking.

Self-crimp side-by-side bicomponent filaments (SBSBFs) were prepared via melt spinning using two kinds of polyethylene terephthalate (PET) with great disparity of intrinsic viscosity. The influence of the volume ratio on the surface morphology, crystallinity, crimping properties, mechanical properties and shrinkage properties of the bicomponent filaments was investigated using wide-angle X-ray diffraction, a differential scanning calorimetry (DSC), scanning electron microscope, etc. As the proportion of the low-viscosity component increases, the shrinkage in boiling water or hot air, as well as the shrinkage force and the sonic orientation factor of the bicomponent filaments decrease, and the DSC heating curves change from double peaks to a single peak. These phenomena should be ascribed to the high orientation and low crystallinity of the high-viscosity PET component and low orientation and high crystallinity of the low-viscosity PET component. Moreover, the crimp property of the bicomponent filament with a volume ratio of 50:50 is superior to those with other volume ratios.

Opis:
Somatic cell nuclear transfer (SCNT) is currently the most efficient and precise method to generate genetically tailored pig models for both agricultural and biomedical research. However, its efficiency is crucially dependent on the source of nuclear donor cells. In this study, we compared the cloning efficiency by using three lines of donor cells that are derived from fetal, newborn and adult fibroblasts of Chinese indigenous Meishan pig. We showed that cleavage rate and blastocyst formation rate of the reconstructed embryos were not significantly different between the fetal (80.7% and 15.6%) and newborn ear skin (77.5% and 12.3%) fibroblast groups (p>0.05), but in both groups these indices were significantly higher than that found in the adult ear skin (70.5% and 8.8%; p<0.05). Reconstructed embryos derived from fetal, newborn, and adult ear skin fibroblasts were transferred to four surrogates, respectively. For the fetal, newborn, and adult ear skin fibroblasts, the number of pregnancies were two (50.0%), two (50.0%), and one (25.0%), respectively, and the number of deliveries were two (50.0%), one (25.0%), and zero (0.0%), respectively. Seven and two cloned piglets were obtained from the fetal and newborn ear skin fibroblasts respectively, while no piglets were obtained from the adult ear skin fibroblasts. Two cloned piglets from the newborn ear skin fibroblasts died shortly after birth because of neonatal asphyxia caused by dystocia. The birth weights of the piglets derived from the fetal and newborn ear skin fibroblasts were 1230.5 and 1310.0g, respectively, which were statistically insignificant (p>0.05), but both were significantly higher than that of the control groups (p<0.05). Microsatellite analyses demonstrated that the genotypes of all cloned piglets were identical to their donor cells. Therefore, cloned pigs were successfully produced using two sources of donor cells isolated from the fetal and newborn ear skin fibroblasts of Meishan piglet, and indicating a better cloning efficiency than that obtained from adult fibroblasts. We concluded that the nuclear donor cell lines have significant impact on the developmental competence of cloned embryos as well as on the cloning efficiency of Meishan pig.

Numerous evidences suggested that the hepatitis B virus (HBV) was recognized as an important factor in the development of hepatocellular carcinoma (HCC). Dickkopf-1 (DKK1) recently was reported to be involved in the progress of HCC. HBV may regulate DKK1 expression in hematoma carcinogenesis. Here, we demonstrated that HBV could regulate DKK1 promoter activity which resulted in upregulation of its mRNA and protein expression in several HBV existing cell lines, and HBx played a prominent role in this process. Transcription factor binding site search result showed that there is a SP1 site in DKK1 promoter region. Luciferase assay showed that overexpression of SP1 could increase DKK1 promoter activity in a dose dependent manner. Accordingly, siRNA inhibition of SP1 expression reduced DKK1 promoter activity and decreased the expression of DKK1 protein.

Plants must continually calibrate their growth in response to the environment throughout their whole life cycle. Revealing the regularity of plant early growth and development is of great significance to plant genetic modification. It was previously demonstrated that loss of two key light signaling transcription factors, FHY3 and FAR1, can cause a stunted stature in the plant adult stage, and numerous defense response genes can be continuously activated. In this study, we performed a time-course transcriptome analysis of the early 4 weeks of leaf samples from wild plants and their fhy3 and far1 transcription factors. By comparative transcriptome analysis, we found that during the early 4 weeks of plant growth, plants primarily promoted morphogenesis by organizing their microtubules in the second week. In the third week, plants began to trigger large- scale defense responses to resist various external stresses. In the fourth week, increased photosynthetic efficiency promoted rapid biomass accumulation. Weighted gene coexpression network analysis of FHY3 and FAR1 revealed that the two light signaling transcription factors may be originally involved in the regulation of genes during embryonic development, and in the later growth stage, they might regulate gene expression of some defense-related genes to balance plant growth and immunity. Remarkably, our yeast two-hybrid and bimolecular fluorescence complementation experiments showed that FAR1 interacts with the immune signaling factor EDS1. Taken together, this study demonstrates the major biological processes occurring during the early 4 weeks of plant growth. The light signaling transcription factors, FHY3 and FAR1, may integrate light signals with immune signals to widely regulate plant growth by directly interacting with EDS1.
