Opis:
This paper firstly expands an efficient numerical methodology developed from the source panel method to the added mass calculation of long column systems in fluid. Structures submerged in fluid are considered as two-dimensional and are discretized into a number of source panels. The influence coefficient matrices and the potential function are then calculated and the fluid forces are calculated by the unsteady Bernoulli equation. Finally, we utilize this present method to calculate the mass coefficients of some typical problems, which effectively verifies its feasibility and accuracy. This method takes into account both applicability and computational efficiency. On the one hand, in contrast to the analytical method which is only applicable to specific cross-sections, this method is applicable to arbitrary boundaries with C0 continuity in mathematics. On the other hand, this method requires less mesh and computation than commercial software. This paper extends the application of the source panel method which is widely used in aerodynamics to provide a reference for added mass calculation problems in engineering.

Opis:
Plate-like structures in channel flow are commonly found in engineering. This paper reports a theoretical study on the static aeroelastic instability of an inverted cantilevered plate in an inviscid channel flow through the state space. This study begins with the kernel function of the flow potential determined in the Fourier domain with the help of the mirror image method. Then, the instability equation is derived from the operator theory and transformed in the state space. Finally, with Glauert’s expansion, model functions, and error functions, the instability problem of such a plate has been modeled as a mathematical function approximation problem and solved by the least squares method. The derived instability equation is considered at the continuum level of description, and no approximation appears at the first equation level. The convergence and reliability of the proposed modeling and its solutions approximation are entirely tested, and it can successfully predict the instability boundary, behavior, and the channel effect. Numerical results show that the decreased channel height and asymmetric plate placement in the channel significantly decrease the critical flow velocity. The plate instability modes are close to the plate’s first natural ones and not sensitive to the channel parameters. This conclusion allows further theoretical exploration of a semi-analytical approximation of the instability boundary from the obtained instability equation. The current modeling strategy in a continuum sense may provide a new idea and essential reference for other instability problems.

Opis:
Goose astrovirus (GoAstV) is a novel avastrovirus that typically causes gosling gout and results in 2 to 20% mortality. GoAstV capsid protein is the sole structural protein, which is responsible for viral attachment, assembly, maturation as well as eliciting host antibodies. However, the epitopes within capsid protein have not been well studied. In this study, a monoclonal antibody, named 1D7, was generated against GoAstV capsid protein by hybridoma technology. Western blot results showed that this MAb could react with recombinant capsid protein expressed in E. coli. Also, it recognized the precursor of capsid protein, VP90 and VP70, in GoAstV-infected cells. Besides, excellent specificity of MAb 1D7 was further demonstrated in indirect immunofluorescence assay and immunohistochemical analysis. Epitope mapping results revealed that MAb 1D7 recognized the epitope 33QKVY 36 within Cap protein. Sequence alignment indicated that 33QKVY 36 is a conserved epitope among the isolates of goose astrovirus type 2 (GoAstV-2), suggesting the potential for its use in GoAstV-2 specific diagnostic assay. These findings may provide some insight into a function of the GoAstV capsid protein and further contribute to the development of diagnostic methods for GoAstV infection.

In this study, multiscale advancing contact angles for glycerol/water drops at silica surfaces are reported for millidrops, submicron-drops, and nanodrops. Selected silica surfaces were muscovite, silicon, and talc. The contact angles for millidrops (1–2 mm) were determined by the traditional sessile drop technique. For submicron-drops (0.1–1.0 μm), a hollow tip Atomic Force Microscope (AFM) procedure was used. The contact angles for nanodrops (~7 nm) were examined from Molecular Dynamics (MD) simulation. The results were compared to evaluate the effect of drop size on the contact angle. In the case of the hydrophobic talc surface, the 75° advancing contact angle did not vary significantly with drop size. For the hydrophilic muscovite surface, the water drop wet the surface and an advancing contact angle of about 10° was found for the millidrops and submicron-drops. However, for the MD simulated nanodrops, attachment and spreading of the ~7 nm drop created a 2D film of molecular dimensions, the contact angle of which was difficult to define and varied from 0° to 17°. Perhaps of equal interest from the MD simulation results was that the spreading of the glycerol/water nanodrop at the muscovite surface resulted in crystallographic directional transport of water molecules to the extremities of the 2D film. Such separation and segregation left the center of the film with an increased concentration of glycerol. Based on these results, the line tension, which has been found in other investigations to account for contact angle decrease with a decrease in drop size, does not seem to be a significant factor in this study.

The Guanshan Biota (South China, Cambrian, Stage 4) contains a diverse assemblage of biomineralizing and non-biomineralizing animals. Sitting temporally between the Stage 3 Chengjiang and Wuliuan Kaili Biotas, the Guanshan Biota contains numerous fossil organisms that are exclusive to this exceptional deposit. The Guanshan Konservat-Lagerstätte is also unusual amongst Cambrian strata that preserve non-biomineralized material, as it was deposited in a relatively shallow water setting. In this contribution we double the diversity of radiodonts known from the Guanshan Biota from two to four, and describe the second species of Paranomalocaris. In addition, we report the first tamisiocaridid from South China, and confirm the presence of a tetraradial oral cone bearing small and large plates in “Anomalocaris” kunmingensis, the most abundant radiodont from the deposit. All four radiodont species, and three genera, are apparently endemic to the Guanshan Biota. When considered in the wider context of geographically and temporally comparable radiodont faunas, endemism in Guanshan radiodonts is most likely a consequence of the shallower and more proximal environment in which they lived. The strong coupling of free-swimming radiodonts and benthic communities underlines the complex relationship between the palaeobiogeographic and environmental distributions of prey and predators. This local adaptation of radiodonts to their prey is highlighted by the frontal appendage morphology of the two species of Paranomalocaris, apparently specialised to different feeding modes, while the recognition of the limited geographic range of some radiodont faunas highlights the importance of exploring as many deposits as possible to fully understand this group.

Fleas are highly specialized holometabolic insects. So far, only 16 species of fossil fleas in five families have been reported due to the rare fossil records. At present, the earliest flea fossils are reported from the Middle Jurassic Jiulongshan Formation of Northeastern China. The descriptions of these earliest species pushed back the origin of Siphonaptera by at least 40 million years. It is generally accepted that saurophthirids are the “transitional” taxa from stem Jurassic fleas to living crown groups. Herein, we described a new “transitional” flea species, Saurophthirus laevigatus Zhang, Shih, Rasnitsyn, and Gao sp. nov., from the Lower Cretaceous Yixian Formation of Northeastern China, assigned to Saurophthiridae. The new species provides new evidence to support saurophthirids as a “transitional” group. Sexual dimorphism suggests significant differences in biology of opposite sexes in Saurophthirus. Analysis of described Mesozoic species demonstrates the body size reduction from the Middle Jurassic to the Early Cretaceous. Smaller body size was likely advantageous in reducing the probability of being detected and removed by the host and in minimizing flea’s demand for blood intake and energy input, indicating the adaptation of the ectoparasitic lifestyle of fleas in their early stage of evolution.

The enzyme strictosidine synthase (STR, EC: 4.3.3.2) plays a key role in the biosynthetic pathway of terpenoid indole alkaloid (TIA). It catalyzes the condensation of the tryptamine and secologanin to form 3α(S)-strictosidine, which is the common precursor of all TIAs. In this paper, a STR gene designated as DoSTR (GenBank: KX068707) was first cloned and characterized from Dendrobium officinale with rapid amplified cDNA ends method (RACE). DoSTR has a length of 1380bp with 1179bp open reading frame encoding 392 amino acids. BlastP analyses showed that its amino acid sequence was classified into Str_synth superfamily. qRT-PCR showed that DoSTR was expressed in all tissues tested, with a significantly higher level in flower and the lowest in stem. Four different treatments with MeJA, SA, ABA and AgNO₃, respectively, could induce the DoSTR expression to a different extent. And the effect of MeJA was the most obvious and transcript level of DoSTR induced by MeJA was 20.7 times greater than that of control at 48 hours after treatment. Furthermore, it was found that DoSTR was localized in vacuole through transient expression in tobacco. The characterization and expression of DoSTR can help in further studying the role of DoSTR in the biosynthesis of TIAs in D. officinale. This study may throw light on the alkaloid biosynthesis pathway of D. officinale.

In this study, cubic and cylindrical cement mortar specimens were first subjected to high temperatures, then the cubic and cylindrical specimens were taken out and conducted with uniaxial compressive test and splitting tensile test, respectively. The effect of the length to side ratio on the uniaxial compressive properties and the effect of thickness-to-diameter ratio on the splitting tensile properties of cement mortar specimens after high temperature were studied. Test results show that: (1) With temperature increasing from 25 ºC (room temperature) to 400 ºC, the compressive strength and elastic modulus of cubic specimens with three kinds of side lengths decrease; the decreasing rates of compressive strength and elastic modulus of cubic specimen with side length of 70.7 mm is higher than those of cubic specimens with side length of 100 mm and 150 mm, and the strain at the peak stress of cubic specimens with three kinds of side lengths increase. (2) After the same temperature, the tensile strength of cylindrical specimen decreases with the thickness-to-diameter ratio increasing from 0.5 to 1.0. The decreasing rate of tensile strength of cylindrical specimen with thickness-to-diameter ratio is highest when the temperature is 25 ºC (room temperature), followed by that after the temperature of 200 ºC, and that after the temperature of 400 ºC is the lowest.

Background. Mushrooms are a food that is often considered as an important source of minerals and other nutrients for consumers. There is little data on the minerals in mushrooms processed culinary and on the impact of processing. Objective. The research was aimed at understanding the similarities and differences in the mineral composition (Hg, Li, Mg, Al, Co, Ni, Cu, As, Se, Rb, Sr, Ag, Cd, Cs, Sb, Tl, Pb, U, Ba, Cr, Zn, Mn and V) of white button mushrooms (Agaricus bisporus) processed in industrial conditions. Material and methods. Fungal materials came from six producers. The elements were determined by ICP-MS DRC and CV-AAS using validated methods and QA/QC protocol. The interdependencies of 18 elements and 10 batches of mushrooms examined were tested with the help of principal component analysis. Results. Some significant differences were found in the content and composition of minerals in marinated white button mushrooms depending on producer. Conserved white button mushrooms are poorer in major essential elements but also in toxic Hg, As, Ag, Cd, Sb, Tl or Pb which has been reported for unprocessed mushrooms. Conclusions. The relatively higher levels of Ag in some batches seem to be largely explained by the quality of the substrate used for mushrooms cultivation, while of Li, Rb, Cs, Cr, Al, U, V, As and Mn (in part also of Ba and Sr) largely by the quality of the marinade.
Wprowadzenie. Grzyby jadalne to surowiec spożywczy bogaty w niezbędne pierwiastki metaliczne ale też możliwie źródło narażenia konsumenta na toksyczne metale i metaloidy. Niewiele jest danych na ten temat odnośnie grzybów przetworzonych kulinarnie czy wpływu procesów przetwarzania. Cel. Badania miały na celu poznanie podobieństw i różnic w składzie mineralnym (Hg, Li, Mg, Al, Co, Ni, Cu, As, Se, Rb, Sr, Ag, Cd, Cs, Sb, Tl, Pb, U, Ba, Cr, Zn, Mn and V) pieczarek (Agaricus bisporus) przetworzonych w warunkach przemysłowych. Materiał i metody. Pieczarki pochodziły z sześciu przetwórni. W analizie zastosowano sprawdzone metodyki analityczne łącznie z bieżącą kontrolą i zapewnieniem jakości wyników analizy. Pomiar wykonano technikami ICP-MS-DRC i CV-AAS. Współzależności pomiędzy 18 pierwiastkami i 10 partiami grzybów badano metodą analizy głównych składowych. Wyniki. Wykazano duże różnice w zawartości składników mineralnych w pieczarkach z różnych przetwórni. Konserwowe pieczarki są znacznie uboższe w główne pierwiastki niezbędne ale także w pierwiastki toksyczne takie jak Hg, As, Ag, Cd, Sb, Tl czy Pb w porównaniu z opublikowanymi danymi dla grzybów nieprzetworzonych. Wnioski. Względnie większą zawartość Ag w określonych partiach badanych konserwowych pieczarek wydaje się tłumaczyć jakość podłoża zastosowanego w uprawie grzybów a w przypadku Li, Rb, Cs. Cr, Al, U, V, As i Mn (po części też Ba i Sr) główny wpływ wydaje się mieć jakość użytej zalewy (marynaty).

The morphology and ontogeny of the trilobite Changaspis elongata based on 216 specimens collected from the Lazizhai section of the Balang Formation (Stage 4, Series 2 of the Cambrian) in Guizhou Province, South China are described. The relatively continuous ontogenetic series reveals morphological changes, and shows that the species has seventeen thoracic segments in the holaspid period, instead of the sixteen as previously suggested. The development of the pygidial segments shows that their number gradually decreases during ontogeny. A new dataset of well-preserved specimens offers a unique opportunity to investigate developmental traits after segment addition is completed. The ontogenetic size progressions for the lengths of cephalon and trunk show overall compliance with Dyar’s rule. As a result of different average growth rates for the lengths of cephalon, trunk and pygidium, the length of the thorax relative to the body shows a gradually increasing trend; however, the cephalon and pygidium follow the opposite trend. Morphometric analysis across fourteen post-embryonic stages reveals growth gradients with increasing values for each thoracic segment from anterior to posterior. The reconstruction of the development traits shows visualization of the changes in relative growth and segmentation for the different body parts. The new dataset and growth gradient of the trunk suggest that the thoracic segment growth dynamics of early Cambrian to Silurian trilobites follow the same general continuous, steady-state growth gradient decreasing from posterior to anterior.

Opis:
Ship route planning is one of the key issues in enhancing traffic safety and efficiency. Many route planning methods have been developed, but most of them are based on the information from charts. This paper proposes a method to generate shipping routes based on historical ship tracks. The ship's historical route information was obtained by processing the AIS data. From which the ship turning point was extracted and clustered as nodes. The ant colony algorithm was used to generate the optimize route. The ship AIS data of the Three Gorges dam area was selected as a case study. The ships’ optimized route was generated, and further compared with the actual ship's navigation trajectory. The results indicate that there is space of improvement for some of the trajectories, especially near the turning areas.
