Opis:
Techniques for measuring fluid flow have been known since the 19th century. The first solutions based on the use of pressure only allowed relatively slow changes to be observed. It was not until measurement techniques based on the use of electronic components and the phenomenon of thermo-transfer, combined with a method enabling fast signal recording (A/C converters), that it became possible to analyse the flow of a medium (e.g. air) in detail. Although flow sensors based on measuring changes in resistance have been known for many years, new solutions are still being developed. This paper presents the results of a study using three sensors. Their response to laminar airflow was investigated for different velocities (1.2 - 2.6 m/s). The flow forcing was implemented using an axial fan and the signals were measured simultaneously for all the sensors tested. The results showed which sensors had the smallest dispersion of results (PAN and WA sensors) and confirmed that for the investigated velocity variations (0.14 m/s pitch) the results are unambiguously interpretable.. It should be noted that sensor research is related to the need to develop a device to measure flow as accurately as possible, while at the same time ensuring the comfort of the test person during the measurements. Therefore, the search was for a sensor that is small in size and at the same time resistant to damage and operation in a harsh humid environment.

Opis:
In this paper we describe our own construction of a tuneable light source based on a set of light emitting diodes covering the visible spectrum using a homogenizing rod instead commonly used low energy-efficient integrating spheres. The expected prime application of the source is a medical endoscopic system, however it is possible to use it also for other purposes requiring both multispectral operation and a tuneable white light source. We describe the construction of the source and include precise characterization of the output white light - distribution of CCT, Duv, ∆u’v’ and colour rendering indexes (Ra, R9, Rf, Rg) of light in several planes located at various distances. The obtained results prove that our source is characterized by very good colour rendition according to the Ra and Rf method for various correlated colour temperatures (2700-6500) K. As an example of application images of the Macbeth colour chart registered with an RGB camera included in the laboratory measurement stand are presented. The obtained results prove that, after whole system calibration, this source can be used in many applications, where evaluation of objects requires precise analysis of their colour and multispectral procedures.

The legislator placed mediation in administrative and court-administrative proceedings. The normative regulation of this institution is mainly similar to the solutions applicable in civil proceedings. Mediation is voluntary and its conduct is entrusted to the mediator. The practical use of mediation in administrative or court-administrative proceedings makes an amicable formula that makes resolving administrative cases real. Considering the fact that in 2017 the legislator amended the administrative procedure by embedding mediation in the administrative law system, the authors of the study undertook the evaluation of the mediation institution in administrative and court-administrative proceedings.

Opis:
Pathogenic microbes can recruit to their cell surface human proteins that are components of important proteolytic cascades involved in coagulation, fibrinolysis and innate immune response. Once located at the bacterial or fungal surface, such deployed proteins might be utilized by pathogens to facilitate invasion and dissemination within the host organism by interfering with functionality of these systems or by exploiting specific activity of the bound enzymes. Aim of the study presented here was to characterize this phenomenon in Candida parapsilosis (Ashford) Langeron et Talice - an important causative agent of systemic fungal infections (candidiases and candidemias) in humans. We have investigated the interactions of fungal surface-exposed proteins with plasminogen (HPG) and high-molecular-mass kininogen (HK) - the crucial components of human fibrinolytic system and proinflammatory/procoagulant contact-activated kinin-forming system, respectively. After confirming ability of the fungal surface-exposed proteins to bind HPG and HK, four of them - two agglutinin-like sequence (Als) proteins CPAR2_404780 and CPAR2_404800, a heat shock protein Ssa2 and a moonlighting protein 6-phosphogluconate dehydrogenase 1 - were purified using ion-exchange chromatography, gel filtration and chromatofocusing. Then, their affinities to HPG and HK were characterized with surface plasmon resonance measurements. The determined dissociation constants for the investigated protein-protein complexes were within a 10-7 M order for the HPG binding and in a range of 10-8-10-9 M for the HK binding. Detailed characterization of adsorption of these two important plasma proteins on the fungal cell surface may help to increase our understanding of molecular mechanisms of C. parapsilosis-dependent candidiasis.

Opis:
Candida tropicalis is one of the most frequent causes of serious disseminated candidiasis in human patients infected by non-albicans Candida species, but still relatively little is known about its virulence mechanisms. In our current study, the interactions between the cell surface of this species and a multifunctional human protein - high-molecular-mass kininogen (HK), an important component of the plasma contact system involved in the development of the inflammatory state - were characterized at the molecular level. The quick release of biologically active kinins from candidal cell wall-adsorbed HK was presented and the HK-binding ability was assigned to several cell wall-associated proteins. The predicted hyphally regulated cell wall protein (Hyr) and some housekeeping enzymes exposed at the cell surface (known as "moonlighting proteins") were found to be the major HK binders. Accordingly, after purification of selected proteins, the dissociation constants of the complexes of HK with Hyr, enolase, and phosphoglycerate mutase were determined using surface plasmon resonance measurements, yielding the values of 2.20 × 10-7 M, 1.42 × 10-7 M, and 5.81 × 10-7 M, respectively. Therefore, in this work, for the first time, the interactions between C. tropicalis cell wall proteins and HK were characterized in molecular terms. Our findings may be useful for designing more effective prevention and treatment approaches against infections caused by this dangerous fungal pathogen.

Opis:
In the course of infections caused by pathogenic yeasts from the genus Candida, the fungal cell surface is the first line of contact with the human host. As the surface-exposed proteins are the key players in these interactions, their identification can significantly contribute to discovering the mechanisms of pathogenesis of two emerging pathogens from this genus, C. parapsilosis and C. tropicalis. Therefore, the aim of the present study was to identify the cell wall-attached proteins of these two species with the use of cell surface shaving and a shotgun proteomic approach. Different morphological forms of C. parapsilosis and C. tropicalis cells obtained after growth under various conditions were subjected to this treatment. This allowed to indicate the most abundant cell surface proteins on the basis of the normalized spectral abundance factors. In case of yeast-like forms these were, among others, proteins similar to a chitinase, glyceraldehyde-3-phosphate dehydrogenase and an inducible acid phosphatase for C. parapsilosis, and a constitutive acid phosphatase, pyruvate decarboxylase and glyceraldehyde-3-phosphate dehydrogenase for C. tropicalis. In case of pseudohyphal forms, proteins similar to a cell surface mannoprotein Mp65, chitinase and glycosylphosphatidylinositol-anchored transglycosylase Crh11 were identified at the cell surface of C. parapsilosis. The Rbt1 cell wall protein, a hyphally regulated cell wall protein and proteins from agglutinin-like sequence protein family were found as the most abundant on C. tropicalis pseudohyphae. Apart from the abovementioned proteins, several additional covalently bound and atypical cell wall proteins were also identified. These results extend the current knowledge regarding the molecular basis of virulence of these two non-albicans Candida species.

Opis:
T he paper presented the origin and utilization of linseed form, taking into consideration the field and yield production area in the world, Europe and Poland. Linseed forms allow for climate conditions and farmers modulated utilization features. In Poland condition more common was fiber form plant ‘Flax’ since the oldest times till 20th century. Originally, flax was cultivated in gardens, and than relocated into the field without alternative fiber plant. Significant decreasing of flax cultivated in polish field in the middle of XIX was caused by wider spreading of cotton utilization in farms and decreasing of domestic fiber production. Flax products in those days were exchanged by cotton and Australian wool. At present fiber from Flax is exchanged into synthetic fiber or fiber mix with cotton. However bigger which acceptation for flax fiber clothes in states with moderate climate is observed including Poland, may stimulate flax area in the same level. Flax in our soil as well as weather conditions allow to produce higher quality of fiber. Recently seed flax has been utilized for linseed oil production. The assumption for linseed oil output was stable and cheapest by unique composition with α-linolenic acid source. Enrichment of wide sort of groceries with essential unsaturated acids allowe to improve food products. Effective enrichment with NNKT, as a F wit amine, is determined mainly by oilseed yield area, which significance will increase in the future. After 2000 mostly linseed form was cultivated. However, lower yield level of linseed should be determined by better cultivation technology conform to yellow and brown linseed varieties. Increasing of linseed production results to deliver from 1 to 1,5-2g α-linolenic acid into groceries per day, which is believed due to nutrition and health issues. Discovering and popularization of novel and others linseed utilization allow to maintain species in the field.

The rapidly developing measurement techniques and emerging new physical methods are frequently used in otolaryngological diagnostics. A wide range of applied diagnostic methods constituted the basis for the review study aimed at presenting selected modern diagnostic methods and achieved diagnostic results to a wider group of users. In this part, the methods based on measuring the respiratory parameters of patients were analysed. Respiration is the most important and necessary action to support life and its effective duration. It is an actual gas exchange in the respiratory system consisting of removing CO2 and supplying O2. Gas exchange occurs in the alveoli, and an efficient respiratory tract allows for effective ventilation. The disruption in the work of the respiratory system leads to measurable disturbances in blood saturation and, consequently, hypoxia. Frequent, even short-term, recurrent hypoxia in any part of the body leads to multiple complications. This process is largely related to its duration and the processes that accompany it. The causes of hypoxia resulting from impaired patency of the respiratory tract and/or the absence of neuronal respiratory drive can be divided into the following groups depending on the cause: peripheral, central and/or of mixed origin. Causes of the peripheral form of these disorders are largely due to the impaired patency of the upper and/or lower respiratory tract. Therefore, early diagnosis and location of these disorders can be considered reversible and not a cause of complications. Slow, gradually increasing obstruction of the upper respiratory tract (URT) is not noticeable and becomes a slow killer. Hypoxic individuals in a large percentage of cases have a shorter life expectancy and, above all, deal with the consequences of hypoxia much sooner.
