To develop a sensitive, specific, and rapid approach for the detection Getah virus (GETV), a set of primers targeting the conserved region of the E1 gene was created. The TaqMan-based real-time PCR method for GETV detection was developed by optimizing the reaction conditions. The method demonstrated excellent specificity, and amplification did not occur with the causative agents of all prevalent swine viral infections (CSFV, PRRSV, PRV, PEDV, PTV, and JEV), except GETV. Additionally, upon assessing the sensitivity of the method, the minimum detection limit for GETV was found to be 5.94 copies/μL, which is 10 times higher than that of the traditional PCR approach. Further, the intra- and inter-assay variation coefficients were less than 1%, demonstrating good repeatability. Moreover, GETV was found in 10 of the 20 field serum samples using real-time PCR but only in three of the samples using traditional PCR. Consequently, the first GETV TaqMan-based real-time PCR approach based on the E1 gene was developed for GETV pathogenic diagnoses, and this exhibited high specificity, sensitivity, and repeatability. This assay is practical for the pathogenic diagnosis and epidemiology of GETV.

Chlorocresol nanoemulsion disinfectant (CND) is an environmental disinfectant prepared with nanoemulsion as its drug carrier. This study aimed to investigate the bactericidal effect of CND on Staphylococcus aureus ( S. aureus) and its effect on bacterial ultrastructure. The neutralizing effect of CND against S. aureus was first screened by suspension quantitative evaluation experiment procedure of neutralizer. Disinfection performance was evaluated by the determination of Minimal Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC), quantitative bactericidal experiment, and comparative experiment of disinfection performance between 0.1% CND and 0.1% chlorocresol aqueous solution. Meanwhile, the effect of CND on the ultrastructure of S. aureus was investigated with scanning electron microscope (SEM) and transmission electron microscope (TEM) to preliminarily explore the bactericidal mechanism. The results showed that 3% Tween-80 in PBS could be screened as the neutralizer of CND against S. aureus. MIC and MBC were 100 μg/mL and 200 μg/mL, respectively. The bactericidal rates were all 100% when 0.06% and 0.08% disinfectant acted for 15 and 5 min, respectively. Furthermore, compared with 0.1% chlorocresol aqueous solution, the bactericidal effect of 0.1% CND was significantly enhanced (p<0.01). After treatment with CND for 10 min, SEM observation showed that the morphology of S. aureus cells were changed and the integrity destroyed. TEM observation showed that the cell shape changed, and the structures of the cell wall, cell membrane and cytoplasm were damaged in varying degrees. CND showed the strong bactericidal effect on S. aureus and could cause ultrastructure alterations of S. aureus.

A novel avian orthoreovirus (N-ARV) variant characterized with obvious arthritis and synovial inflammation, was isolated from Shandong, China in May 2016. It caused chicken poor growth and enormous economic losses to the poultry industry of China. However, there are few effective methods for detecting the antibody levels of N-ARV. In this study, a viral structural protein σC was expressed using the prokaryotic expression vector pET32a (+). The target protein was obtained by inducing for 6 hours at an IPTG concentration of 0.6mM. The optimal dilution of the coating antigen and serum antibody were determined to be 1000 fold and 10 fold respectively. A specificity test showed that there was no positive reactivity between N-ARV and other pathogens, and when the positive serum was diluted 100 times detection results were still checkable. The repeatability of this method was determined by the inter assay and intra assay tests with variability ranging from 4.85% to 7.93%. In conclusion, this indirect enzyme linked immunosorbent assay (ELISA) will be useful for large-scale serological surveys and monitoring antibody levels in N-ARV infection.

Opis:
The relationships among sections and species within each section in the genus Populus have attracted much attention from taxonomists and biologists. In this paper, we provide an overview of the current state of Populus phylogenetics and discuss the origin of some hybrid species. There has been some controversy surrounding the origin of the genus Populus in North America and subsequent dispersal and differentiation in other parts of the world. We present the most plausible scenario based on phylogenetic analysis and fossil records. Sections Abaso, Turanga and Populus are all monophyletic, whereas sections Aigeiros, Tacamahaca and Leucoidesshow some degree of polyphyly. Sections Abaso and Turanga are relatively primitive lineages in Populus. With the accumulation of sequencing data and new analytical tools, it will be possible to investigate more complicated issues concerning genus Populus within a phylogenetic framework.

Opis:
Establishing a fatigue life prediction model considering the machined surface state is of great significance to improve fatigue life prediction accuracy. Fatigue tests with different machined surface states of GH4169 alloy were conducted firstly. The influence of surface state parameters on the fatigue life was analyzed. Then, the machined surface stress concentration factor and residual stress were introduced into the fatigue crack initiation and propagation models. Finally, the fatigue life prediction model of GH4169 material considering the surface states was established. The prediction results based on the proposed model are almost within a factor of two scatter band of the test results.

Opis:
Wenjiang (China) is the area which undertakes high-intensity planting activities. Additionally, the soil environmental capacity has been a hot area of research as it plays a key role in environmental protection planning, environmental impact assessment and sustainable development. In this paper, the static model of soil environmental capacity is employed to investigate the distribution of residual soil environmental capacity in Wenjiang. The results show that the soil environmental capacity of mercury is the largest for industrial land while it is the lowest in garlic-rice planting areas; the soil environmental capacity of arsenic is the largest in city construction land while it is the lowest in ecological conservation zone; the soil environmental capacity of lead is the largest in city construction land while it is the lowest in garlic-rice planting areas; and the soil environmental capacity of chromium is the largest in city construction land while it is the lowest in garlic-rice planting areas.

This article is the result of treatments on ground fissures for environmental protection and scientific shallow coal seam mining. In the southwestern mining area of China, the traditional longwall mining method has caused a large area of surface sinkhole, ground fissures, vegetation deterioration and disorderly coal gangue. To solve these problems, an integrated treatment technology that includes ground fissure treatment technology and underground backfilled technology was proposed as a solution. The technical principle and technical process were explained in detail; the ground fissure treatment technology involves a “three-step treatment method”, and the underground backfilled technology adopted a strip mining method with backfilling technology. The compression mechanical behavior of backfilled material, including coal gangue, fly ash and ordinary Portland cement, was studied; the mixed ratio of 1:0.3:0.18 was selected. In addition, the vertical stress, vertical displacement and plastic zone of the coal pillar were determined by FLAC3D numerical simulation, and a rational mining scheme of “11 m mining width, 2 m coal pillar width” was determined to be appropriate because of the lower vertical stress, smaller vertical displacement and better supporting capacity of the coal pillar. The monitoring results of ground sinkhole indicated that the maximal ground sinkhole deformation was 17.3 cm, and the deformation showed few changes after this technology was implemented. The treatment capacity of coal gangue and fly ash reached 821.150 t per year, and the vegetation survival rate of the ground fissure treatment area reached 85%. This integrated treatment technology could effectively control ground fissures and surface sinkhole as well as protect the environment.
Artykuł jest opisem zapobiegania pęknięciom gruntu w celu ochrony środowiska, jak również naukowego podejścia do wydobywania płytko zalegających pokładów węgla. W południowo-zachodniej części górniczej Chin tradycyjna, ścianowa metoda wydobywania węgla, powodowała duży obszar powierzchniowego zapadliska, pęknięcia gruntu, pogorszenie stanu wegetacji roślin. W celu rozwiązania tych problemów zaproponowano zintegrowaną technologię zapobiegania, która obejmuje technologię zapobiegania pęknięciom gruntu i podziemną technologię podsadzki. Zasada i proces techniczny zostały szczegółowo opisane; technologia zapobiegania pęknięciom gruntu obejmuje „trójstopniową metodę zabiegu”, a technologia podsadzki zaadoptowała metodę wydobywania węgla pasami. Badano ściśliwość podsadzki, w tym odpadów węglowych, popiołu lotnego i zwykłego cementu portlandzkiego; wybrano mieszaninę o stosunku 1: 0,3: 0,18. Ponadto pionowe naprężenie, pionowe przemieszczenie i strefa plastyczności filaru węglowego zostały określone za pomocą symulacji numerycznej FLAC 3D. Racjonalny schemat wydobycia „szerokość wydobycia 11 m, szerokość filaru węglowego 2 m” był odpowiedni ze względu na niższą wartość naprężenia pionowego, mniejsze pionowe przemieszczenie i lepszą nośność filaru węglowego. Wyniki monitoringu pęknięć gruntu wskazują, że maksymalna deformacja gruntu wynosiła 17,3 cm i wykazała kilka zmian po wdrożeniu tej technologii. Ilość zagospodarowanych odpadów węglowych i popiołu lotnego wynosiła 821 150 Mg na rok, a wskaźnik przeżycia roślinności w obszarze zapobiegania pęknięciom gruntu osiągnął poziom 85%. Ta zintegrowana technologia zapobiegania może skutecznie kontrolować pęknięcia gruntu i deformację powierzchni, a także chronić środowisko.

The study area in the West Junggar Basin is known to be rich in hydrothermal gold deposits and occurrences, even though there has been minimum exploration in the area. It is here hypothesised that this area could host more gold deposits if mineral exploration methods were to be reinforced. This research is aimed at identifying geochemical anomalies of Au, and determining possible factors and conditions which facilitate the formation of anomalies by referring to As and Hg as gold pathfinders. Geostatistical analyst techniques have been applied to 9,852 stream sediments and bedrock data collected on a total surface of 1,280 km2 of West Junggar, Xinjiang (northwest China). The kriging interpolation and quantile-quantile plot methods, combined with statistical methods, successfully identified both Au and its pathfinders’ anomalies. In the present study, median was considered as background values (10.2 ppm for As, 9.13 ppb for Hg and 2.5 ppb for Au), whereas the 95th percentile were threshold values (28.03 ppm for As, 16.71 ppb for Hg and 8.2 ppb for Au) and values greater than thresholds are geochemical anomalies. Moreover, the high concentrations of these three discovered elements are caused primarily by hydrothermal ore mineralisation and are found to be controlled mainly by the Hatu and Sartohay faults of a northeast-southwesterly direction as well as their related secondary faults of variable orientation, which facilitate the easy flow of hydrothermal fluids towards the surface resulting in the formation of geochemical anomalies. Most of anomalies concentration of Au are found near the mining sites, which indicates that the formation of new Au anomalies is influenced by current or previous mining sites through geological or weathering processes. In addition, the low concentration of gold and its pathfinders found far from active gold mine or faults indicates that those anomalies are formed due to primary dispersion of hosting rock.
