Opis:
Proteomics including the studies of the structure, function and dependences between proteins is more and more extensively applied in human medicine and veterinary medicine. The analysis of protein profiles of tissues and body fluid from healthy and ill individuals allows to identify diagnostic, prognostic and predictive markers in various pathological states in people and animals. This paper presents preparation of urine samples for analysis in the mass spectrometer MALDI-TOF (Ultraflextreme, Bruker, Bremen, Germany) by means of two methods: liquid chromatography based on the system Nano-LC (PROTEINER FC II, Bruker Daltonics, Bremen Germany). and two-direction electrophoresis 2DE (GE Healthcare, United Kingdom). Both methods enable separation of the mixture under consideration into individual fractions of high purity indispensable for obtaining readable mass spectra. The purpose of this paper is to determine applicability of these methods in analysis of protein composition of urine samples.

Opis:
In this study, we present investigations of morphological changes in the inflorescence of bolting Allium sativum L., which forms umbel-like inflorescences with small, inconspicuous flowers, bulbils, and leaf-like membranous bracts. Particular attention was focused on the architecture of a single flower, and the full sequence of developmental events and the differentiation of flower elements and inflorescence were traced. During the several-year long investigations of the garlic species, we observed that all inflorescence elements emerged chaotically, taking a completely random place in the inflorescence receptacle and it was impossible to identify the sequence of formation of the individual inflorescence elements. The absence of regular phyllotaxis and the specific sequence of formation of inflorescence elements are characteristic for inflorescence reversion. Since domesticated garlic has lost the capacity for sexual reproduction, it has probably evolved alternative reproduction modes in its developmental strategy, which is reflected by the formation of numerous bulbils (vegetative buds, topsets) in the inflorescence. This is one of the forms of reproductive strategy of the species, which through domestication progresses from sexual to asexual reproduction.

Opis:
Traditional data storage systems provide access to user’s data on the "best effort" basis. While this paradigm is sufficient in many use cases it becomes an obstacle for applications with Quality of Service (QoS) constraints. Service Level Agreement (SLA) is a part of the contract agreed between the service provider and the client and contains a set of well defined QoS requirements regarding the provided service and the penalties applied in case of violations. In the paper we propose a set of SLA parameters and QoS metrics relevant to data storage processes and the management methods necessary for avoiding SLA violations. A key assumption in the proposed approach is that the underlying distributed storage system does not provide functionality for resource or bandwidth reservation for a given client request.

Thrips nigropilosus Uzel is a polyphagous species occurring mainly in temperate climates. Its life cycle depends on photoperiodic and temperature conditions. T. nigropilosus feeds on different plant species, but it is considered one of the most serious pests of pyrethrum plants causing serious economic problems. However, several additional agricultural host plants have been affected by T. nigropilosus, including spearmint, cucumber, and lettuce, indicating that this insect can significantly widen its habitats and occurs especially frequently in greenhouses. We report that T. nigropilosus massively attacked Mentha × piperita L. cultivated in greenhouses in central Poland and destroyed the entire mint crops within a short time. The study provided insight into the harmful effect of the thrips and showed that the length of the thrips developmental cycle was reduced with temperature increases from 18 to 26.6°C. The lower threshold temperatures were 13.7, 10.2, 5.0, and 10.1 for eggs, larvae, pupae, and total development, respectively, and the thermal constant for the same developmental stages was 65.9, 90, 132.5, and 284.9-degree days. Both parameters were estimated by linear regression analysis. During our experiment, T. nigropilosus developed by thelytokous parthenogenesis. The morphological and anatomical changes in damaged plants were associated with the fact that the insect began feeding on the lower lamina surface close to the leaf midribs, but no damage to vascular bundles and glandular cells was observed.

The aim of this study was to use a rapid and easy DNA-based test, the loop-mediated isothermal amplification (LAMP), for diagnosis of Babesia canis canis infections in dogs. 10 DNA samples of 18S RNA-A and 10 DNA samples of 18S RNA-B of B. canis canis were used in the study. LAMP method could successfully detect DNA in all examined samples down to 0.1 pg dilution. Obtained results suggest that this method has high specificity and sensitivity and can be applied in analytical laboratories in diagnosis of canine babesiosis.

The aim of the present study was to investigate the occurrence of Borrelia burgdorferi sensu lato DNA in a group of 120 wild bison (Bison bonasus) from the Bialowieza Primeval Forest in eastern Poland. The PCR technique revealed the presence of 16S RNA of Borrelia burgdorferi sensu lato in the blood of 16 out of 120 examined animals. DNA amplification by means of primers SC1 and SC2 gave a product with a size of 300-bp. The sequences of the PCR products obtained showed 100% homology with each other and 100% homology with B. burgdorferi s.l. 16S RNA gene DQ111061. Results of this study suggest that wild bison are important in maintaining agents of Lyme borreliosis, and that studies of reservoir competence of this species are indicated.

Jerusalem artichoke (Helianthus tuberosus Linne) is a tuberous perennial plant of the Asteraceae family, which originates from North America, and is also known as wild sunflower or topinambur (TPB). It is characterized by good tolerance to frost, drought and poor soil, strong resistance to pests and plant diseases. For ages it was cultivated due to being both an edible tuber and having healing properties. In folk medicine, TPB leaves are used for the treatment of bone fractures and pain. TPB tubers are rich in sugar and have therefore been used for the production of functional food ingredients, such as inulin. Moreover, TPB is one of the potential crops for bioenergy production, such as bioethanol, biobuthanol and biodiesel, and chemicals (lactic acid, butyric acid). A number of bioactive compounds from the above ground parts of this plant have been isolated which have demonstrating antifungal, antioxidant and anticancer activities. In recent years, a number of animal experiments have been carried out to assess the health properties of TPB. Obtained results show that TPB possess a wide spectrum of medical applications, e.g. reduction in the levels of plasma glucose, total cholesterol and triglyceride. Interestingly, TPB has been shown to be a valuable alternative source of prebiotic compounds. This review article presents recent scientific reports on the chemical and biological properties of TPB and its potential use as a prebiotic diet supplement.
