Leaves and internodes from Stevia rebaudiana Bertoni plants growing in different conditions were used for transformation with two strains of Agrobacterium rhizogenes: ATCC 15384 and LBA 9402. Hairy roots formation was observed and the percentage of the transformed explants depended on the type of explant, time of inoculation and inoculum concentration. Inoculation of explants from ex vitro and in vitro plants with LBA 9402 strain led to higher efficiency of transformation than inoculation with ATCC 15384 strain. Growth rate of hairy roots in liquid culture was assessed under light and dark conditions. It was found that the growth of hairy roots decreased significantly under light conditions. Transformation of hairy roots growing in different culture conditions was confirmed at the molecular level using PCR method with primers constructed against rolB and rolC genes from A. rhizogenes.

The article presents the results of model research concerning the change of technology of argon blowing into liquid steel at the ladle furnace, using the dual plug system. The results of numerical simulations were verified with experimental data carried out on the water model device. The verified model was used to perform numerical simulations to predict the impact of using a new gas injection technology - with different flow rates - on the time to achieve the assumed degree of metal chemical homogenization after alloy addition. Simulation results show that argon blowing metal bath in dual plug mode can effectively reduce mixing time compared to conventional technology with the same gas flow rates. Generally, the use of the dual plug system is beneficial for reducing the bath mixing time, however, the assumed optimal proportion of gas blown through individual plug should be followed. Finally, numerical predictions were used to perform experimental melt under industrial conditions. Industrial verification has clearly confirmed the validity of numerical modeling and showed that also in industrial conditions, a shorter time of chemical homogenization was obtained for the dual plug system.

Magnetic measurements, carried out by means of the Epstein frame, have shown that most typical dynamo steel sheets have certain anisotropic properties. In numerical analysis, anisotropic properties are taken into account with the use of the special function of the grain distribution in the given dynamo sheet. For engineering purposes, it is desirable to assess the influence of these properties on the changes of the magnetic flux density in typical dynamo steel sheets, especially during the rotational magnetization. For this purpose, measurements of the flux density changes and field strength changes in the circular-shaped samples of two selected typical dynamo sheets were performed. These measurements were carried out for several values of the current flowing in windings which generated the axial or rotational magnetic field in the test dynamo sheet. The influence of the magnetic anisotropy on the magnetization process was briefly discussed for both types of the magnetization processes.

In the present paper, secondary metallurgical treatment in ladle furnace during smelting the high carbon steel and steel with improved ductility for cold-deforming, under industrial conditions were analyzed. Common features of these steels are high requirements/strict standards imposed for their metallurgical purity; however they are especially exorbitant for improved ductility steels. In addition, it is widely used to specify alloying additives having significant weights- such as carbon and manganese - and explicitly restricted content of nitrogen requiring metal bath cover prior nitriding.

In the southern part of Poland, ground beetle fauna was studied in the first large-scale Bt maize experiment. The aim of this study was to determine the long term impact of the Bt maize cultivar in comparison to conventional plants, on selected non-target arthropods. The DKC 3421 YG cultivar (Bt maize) and the respective isogenic non-Bt varieties (DKC 3420) were cultivated at two locations: (a) Budziszów near Wrocław and in Głuchów near Rzeszów in the south-eastern region of Poland, in the 2008-2010 growing seasons. For comparative analysis, two additional non-Bt cultivars sprayed with a lambda-cyhalotrine insecticide were also included. To monitor population density of surface-active invertebrates of the Carabidae family, eighty pitfall traps were used at each location. The average number of ground beetle populations in the Bt-maize cultivar DKC 3421 YG did not significantly differ from the number of beetles in the conventional ones. Significant differences between the number of beetles occurred on individual dates only. Usually, these differences related to the considerably smaller total number of beetles in the whole replication. Probably, the variation in the number of beetles was caused by climatic factors or the terrain layout, therefore it cannot be related to the cultivar effect.
