Opis:
Punch-through is a major threat to the jack-up unit, especially at well sites with layered stiff-over-soft clays. A model is proposed to predict the spudcan penetration resistance in stiff-over-soft clays, based on the random forest (RF) method. The RF model was trained and tested with numerical simulation results obtained through the Finite Element model, implemented with the Coupled Eulerian Lagrangian (CEL) approach. With the proposed CEL model, the effects of the stiff layer thickness, undrained shear strength ratio, and the undrained shear strength of the soft layer on the bearing characteristics, as well as the soil failure mechanism, were numerically studied. A simplified resistance profile model of penetration in stiff-over-soft clays is proposed, divided into three sections by the peak point and the transition point. The importance of soil parameters to the penetration resistance was analysed. Then, the trained RF model was tested against the test set, showing a good prediction of the numerical cases. Finally, the trained RF was validated against centrifuge tests. The RF model successfully captured the punch-through potential, and was verified using data recorded in the field, showing advantages over the SNAME guideline. It is supposed that the trained RF model should give a good prediction of the spudcan penetration resistance profile, especially if trained with more field data.

Opis:
His296 of Zymomonas mobilis levansucrase (EC 2.4.1.10) is crucial for the catalysis of the transfructosylation reaction. The three-dimensional structures of levansucrases revealed the His296 is involved in the substrate recognition and binding. In this study, nine mutants were created by site-directed mutagenesis, in which His296 was substituted with amino acids of different polarity, charge and length. The substitutions of His296 with Arg or Trp retained partial hydrolysis and transfructosylation activities. The positively charged Lys substitution resulted in a 2.5-fold increase of sucrose hydrolysis. Substitutions with short (Cys or Ser), negatively charged (Glu) or polar (Tyr) amino acids virtually abolished both the activities. Analysis of transfructosylation products indicated that the mutants synthesized different oligosaccharides, suggesting that amino acid substitutions of His296 strongly affected both the enzyme activity and transfructosylation products.

Opis:
Purpose: This study aimed to analyze the changes of the stress distributions in TMJs for the pre- and postoperative patients with mandibular prognathism under unilateral occlusions, a frequent occlusion in mastication. Methods: Pre- and six-mouth postoperative cone-beam computed tomography images of thirteen patients diagnosed with mandibular prognathism were scanned and used to construct complete maxillofacial models, assigned as the Pre and Post group, respectively. Another ten asymptomatic individuals were defined as the Control group. The inhomogeneous properties were assigned to the models. The muscle forces and boundary conditions corresponding to left and right unilateral occlusions were applied on the models. The analysis of variation (ANOVA) was chosen for the comparison among the groups. Results: The results showed that the Pre group had abnormal stress distributions ang higher stress level in TMJs, compared with those of the Post and Control groups. Moreover, from clinical cases, symptoms of temporomandibular disorders (TMDs) always followed with increased stresses. Conclusion: Generally, orthognathic surgeries could improve the stress distribution in TMJs of the patients with mandibular prognathism under the unilateral occlusions. However, the postoperative complications, especially symptoms of TMD, were closely related to changes of stress for patients with mandibular prognathism after orthognathic surgeries. Individual virtual surgery and finite element analysis should be conducted to prevent complications in TMJ.

Opis:
Energy levels, line strengths, oscillator strengths, radiative decay rates and fine structure collision strengths are presented for sixteen-times ionized iron (Fe XVII). The atomic data are calculated with the AUTOSTRUCTURE code, where relativistic corrections are introduced according to the Breit-Pauli distorted wave approach. The calculations of atomic data for 89 fine-structure levels generated from eleven configurations 2s²2p⁶, 2s²2p⁵ (3s, 3p, 3d), 2s²2p⁵ (4s, 4p, 4d, 4f) and 2s¹2p⁶ (3s, 3p, 3d) of the Ne-like Fe ion are presented. Fine structure collision strengths for transitions from the ground and the first four excited levels are presented at four electron energies: 75, 125, 175, and 250 Ry. These atomic structure data are compared with the available experimental and theoretical results.

Opis:
With the increasing number of electric vehicles (EVs), the disordered charging of a large number of EVs will have a large influence on the power grid. The problems of charging and discharging optimization management for EVs are studied in this paper. The distribution of characteristic quantities of charging behaviour such as the starting time and charging duration are analysed. The results show that charging distribution is in line with a logarithmic normal distribution. An EV charging behaviour model is established, and error calibration is carried out. The result shows that the error is within its permitted scope. The daily EV charge load is obtained by using the Latin hypercube Monte Carlo statistical method. Genetic particle swarm optimization (PSO) is proposed to optimize the proportion of AC 1, AC 2 and DC charging equipment, and the optimal solution can not only meet the needs of users but also reduce equipment investment and the EV peak valley difference, so the effectiveness of the method is verified.

Opis:
Classical swine fever (CSF) and porcine reproductive and respiratory syndrome (PRRS) are responsible for major economic losses and represent a threat to the swine industry worldwide. Routine surveillance serology for CSF and PRRS viruses is critical to maintaining the health status of sow farms in Hunan Province, which is one of the top pig production provinces in China. The aim of our study was to investigate the serological statistics of CSF virus (CSFV) and PRRS virus (PRRSV) in Hunan Province. The cohort serum samples were collected from vaccinated and unvaccinated pigs. Our findings showed that the average rates of CSFV and PRRSV antibody seropositivity were 82.2% (95% CI: 80.1-84.3) and 84.8% (95% CI: 82.5-87.1), respectively, in the immunized group and that these rates were higher than those in the unvaccinated group (58.6% for CSFV and 47.8% for PRRSV). Additionally, the level of CSFV antibody in piglet serum declined gradually with age, whereas PRRSV-specific antibody level increased initially (1 to 2 weeks old) and then declined with age (2 to 4 weeks old). In summary, we investigated the difference in CSFV/PRRSV antibody levels among piglets at various weeks old (1 to 4 weeks) to further establish the duration of maternal immunity in piglets. In addition, routine monitoring of CSFV/PRRSV antibodies in immunized pigs was carried out to evaluate the efficacy of vaccination.
