In forest production, there is an emerging tendency towards the planting of fast-growing trees as attractive, renewable energy sources. Hence, efforts were made to develop a method of micropropagation by organogenesis of seven clones of black locust (Robinia pseudoacacia L.) that are resistant to propagation by traditional vegetative methods, as well as one plus tree (no. 9755) at the age of 60, to see if the age of the mother plant is a limitation in the micropropagation of black locust trees. Overall results suggest that Murashige and Skoog medium supplemented with 30 g l−1 sucrose, 0.6 mg l−1 6-benzylaminopurine (BAP) and 0.1 mg l−1 naphthalene acetic acid (NAA) is better for the propagation of each genotype of R. pseudoacacia than Woody Plant Medium with the same growth regulators, and the age of the donor plant does not affect the organogenic potential. Recalcitrance to adventitious rooting from adventitious shoot formation is a major limitation for the clonal micropropagation of forest trees. Our results showed that although the roots were also formed spontaneously in the growth medium without growth hormones for the tested black locust clones, the application of auxin increased the total root length compared to that in the medium with active carbon and control. A significant effect of the additives of hormone and sucrose on the total root length was found. Increasing the sucrose concentration stimulated the induction of roots in each of the tested concentrations (5, 10, 15 or 20 g l−1). Additionally, the change in sugar dose in the rooting medium caused significant differences in total root length.

Opis:
The purpose of this study was to analyze the indolic, phenolic, and fatty acid content and antioxidant activity of garlic sprouts growing in the dark and in the daylight. The pro- or anti-inflammatory properties of the garlic sprout extract were investigated by evaluating the cyclooxygenase-2 (COX-2), prostaglandin E synthase (cPGES), glutathione S transferase (GSTM1), nuclear factor NF-κB, peroxisome proliferator-activated receptors (PPARs), and aryl hydrocarbon receptor (AhR) protein levels in the RAW 264.7 cells activated with lipopolysaccharide (LPS). The highest amount of total indolic (73.56 mg/100 g f.w.) and phenolic compounds (36.23 mg/100 g f.w.) was detected in garlic sprouts grown in the daylight. Studies on antioxidant activity (the FRAP and DPPH method) of garlic sprouts showed that this activity is significantly higher for sprouts grown in full access to light when compared to those grown in the dark. In garlic sprout extracts, α-linolenic acid (ALA) was found to be in greater amount. COX-2 and cPGES level was lower when compared to LPS alone activated cells. After garlic extract treatment, higher level of GSTM1, PPARΥ, cytosolic p50 and p65 protein, as well as a lower NF-ĸB p50/p65 activity was noted in the RAW 264.7 cells which suggested PPARs and AhR transrepression mechanism of NF-ĸB signalling. The obtained results indicate Allium sativum sprouts are a rich source of n-3 fatty acids, indolic and phenolic compounds characterized by anti-inflammatory and antioxidative activity, which may support their high therapeutic and dietary potential.

Mechanical stretching of single proteins has been studied experimentally for about 55 proteins yielding a variety of force patterns and peak forces. Here, we perform a theoretical survey of proteins of known native structure and map out the landscape of possible dynamical behaviors under stretching at a constant speed. We consider 7510 proteins each comprising not more than 150 amino acids. The model used is constructed based on the native geometry. It is solved by methods of molecular dynamics and validated by comparing the theoretical predictions to experimental results. We characterize the distribution of peak forces and correlations with the system size and with the structure classification as characterized by the hierarchical classification of protein domain structures (CATH). Despite the presence of such correlations, proteins with the same CATH index may belong to different classes of the dynamical behavior. We identify proteins with the biggest forces and show that they belong to few topology classes. We determine which protein segments act as mechanical clamps and show that, in most cases, they correspond to long stretches of parallel β-strands, but other mechanisms are also possible.
