Opis:
The aim of the study was to evaluate the megakaryocyte lineage of bone marrow and coagulation parameters in fallow deer during the last month of pregnancy. The animals were managed in the barn-feeding system. Twenty female fallow deer, aged 2-3 years, divided into 2 groups were used in the study. Group 1 comprised the females in the last month of pregnancy, and the non-pregnant females were used as the control. All the animals were clinically healthy. Coagulation parameters were measured in all the deer: thrombin time (TT), prothrombin time (PT), activated partial thromboplastin time (APTT), and plasma concentrations of fibrinogen, D-dimer, and antithrombin III. A quantitative assessment of bone marrow was carried out for the erythroblastic, myeloid, lymphoid, monocyte-macrophage, and megakaryopoietic cell lines. A detailed analysis of megakaryocyte lineage was performed after whole blood and platelet count. There were no significant differences in the erythroblast, granulocyte, monocyte-macrophage and lymphoid systems between the animal groups. Thrombocyte count in the pregnant deer was lower than that found in the control group. Bone marrow smears revealed a slightly decreased megakaryocyte count, while the megakaryoblast and promegakaryocyte counts were unchanged. The analysis of coagulation parameters showed increased levels of fibrinogen, thrombin time, prothrombin time and activated partial thromboplastin time in the pregnant animals. The study suggested a hyperactivation of the coagulation system with a slight reduction in the megakaryocyte count in bone marrow, and a reduction in platelet count in peripheral blood at the end of pregnancy.

Opis:
Iron is one of the most abundant elements on Earth, a vital component of every living organism, and an essential micronutrient for all animal species. The primary source of iron for calves and other new-born ruminants is milk or a milk replacer, while for adults it is forage. Drinking water can contain various amounts of Fe ions and can be a source of this element as well. Systemic iron homeostasis in vertebrates is mostly regulated by hepcidin (HEP), a peptide synthesized and secreted by hepatocytes. The largest amounts of iron in the body are incorporated into proteins, mainly haemoglobin and myoglobin. This element participates in several biochemical processes including blood production, transport of oxygen, energy metabolism and immune processes. Iron deficiency can result in so-called iron deficiency anaemia, mostly occurring in young calves due to low iron content in cow’s milk and high demand for this element by calf’s organism. Iron poisoning mostly occurs due to excess iron intake caused by an inappropriate dose of iron given to animals (injection or per os administration) or accidental consumption of high doses of supplements. Excess iron intake can cause overload and damage of internal organs. Oxidation of ferrous iron in haemoglobin can result in methemoglobinemia and an inability of erythrocytes to carry oxygen. In recent years, scientists have implicated some questions regarding iron and ruminants that need re-evaluation or further investigation, e.g. the upper tolerable concentration of Fe in drinking water, the relationship between inflammation and iron status, serum Fe as an indicator of inflammation, or ferritin as a predictive factor for iron deficiency anaemia in cattle.

Opis:
Long-distance endurance races are a big challenge for a horse’s body. The prolonged exercise and dehydration associated with it, in adverse circumstances, may dysregulate many metabolic processes. The aim of this study was to assess changes in electrolyte balance in horses during endurance races, and to explain the potential need for electrolyte supplementation. The experiment was performed on fifty-six horses aged 9-15 years. Haematological and biochemical parameters of horse blood were tested before, during and after the completion of a 120 km endurance race. Electrolyte status (Na+, K+, Cl-) was assessed by analysing test results in horses that completed the race without any clinical symptoms of metabolic disorders and received no additional mineral supplements before the race. The following haematological parameters were determined: red blood cell and white blood cell counts, haemoglobin levels, haematocrit and red blood cell indices (MCV, MCH, MCHC). Enzymatic tests involved the serum levels of lactate dehydrogenase and creatine kinase. A significant increase was found in the levels of haemoglobin, lactate dehydrogenase and creatine kinase activity, but the values of these parameters were within the normal physiological ranges. Other parameters (WBC, RBC, Ht, MCV, MCH, MCHC, Na+, K+, Cl-) changed slightly, but were also within the ranges characteristic of sporthorses. The results suggest that horses well prepared for effort do not require additional supplementation of minerals during endurance races under conditions of the Polish climate.

Opis:
Selenium is an essential nutrient, which is crucial for proper body function. Its role is complemented by vitamin E. Nutritional muscular dystrophy (NMD) is one of the main disorders caused by a selenium deficiency. NMD most often affects calves at the age of 4 to 6 weeks. The study was performed on 40 Holstein-Friesian (HF) calves divided into two groups of 20 animals each. Control group calves were administered an IM injection of selenium and vitamin E on the second day of life. The experimental group comprised calves with symptoms of NMD. Samples of the biceps femoris muscle were collected from six animals in each group for histopathological analyses to confirm changes in muscle parameters. Blood samples were obtained from all animals on three different dates. The following blood parameters were determined in laboratory analyses: pH, pCO 2 , pO 2 , HCO 3 – , BE, O 2 SAT, the concentrations of Na + , K + , Cl – , and Ca and P levels. A drop in pH, an increase in the partial pressure of carbon dioxide, a decrease in the partial pressure of oxygen, a significant decrease in bicarbonate ion concentrations and hemoglobin oxygen saturation as well as a base deficit were reported in the group of calves demonstrating symptoms of NMD. The above changes point to the development of uncompensated metabolic acidosis due to increased levels of pyruvic acid and lactic acid produced as a result of anaerobic processes that accompany muscle fiber degeneration. Minor fluctuations in sodium and chloride levels were observed throughout the experiment, but their concentrations remained within the norm in animal groups. Potassium levels were significantly higher in the experimental group than in the control group. The serum concentrations of inorganic phosphorus and calcium were within the reference range in both groups. In calves, NMD leads to disruptions in the acid-base equilibrium and the electrolyte balance, which are manifested by uncompensated metabolic acidosis and hyperkalemia. Significant changes in calcium and phosphorus levels are not observed in the blood serum of calves affected by NMD.

Opis:
Minerals, in particular micronutrients such as copper, manganese, zinc, selenium and chromium, deliver health benefits for high-yielding dairy cows. The availability of nutrients is particularly important during the transition period (between 3 weeks prepartum to 3-4 weeks postpartum), which is a highly vulnerable time in the production cycle that determines the health status of lactating cows, their milk yield and reproductive performance. In cows, prolonged mineral deficiency can produce subclinical symptoms of nutrient deficiency and, if untreated, can lead to a clinical presentation of the disorder. Less severe deficiencies, in particular micronutrient deficiencies, are more difficult to identify because they induce only minor functional and structural changes in the body, but together with energy metabolism disorders, they can lead to oxidative stress and immunosuppression, in particular in the transition period. Micronutrient supplements reduce the harmful effects of free radicals because the majority of micronutrients form the active centers of antioxidant enzymes. Dietary supplementation promotes reproductive performance, healthy fetal development and immunity. Micronutrients alleviate bacterial infections associated with mastitis, lower somatic cell counts in milk and minimize the risk of metabolic diseases caused by a negative energy balance. This study discusses the role of the major micronutrients in cattle nutrition and their effect on the health status of transition cows.

Opis:
The purpose of this study was to investigate possible alterations in acid-base balance parameters and the coagulation profile in neonatal diarrheic calves. Twenty neonatal diarrheic and 20 clinically healthy neonatal calves aged between 1 week to 10 days were used. All blood samples were taken on the third day from the onset of diarrhea symptom. Venous blood samples were collected from each animal to determine platelet numbers, pH, pCO₂, pO₂, HCO₃⁻, BE, O₂SAT, ctCO₂ and electrolytes (K⁺, Na⁺ and Cl⁻). Plasma samples were collected from each animal for the measurement of prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT), the concentrations of fibrinogen, D-dimer and the activity of antithrombin III (AT III). Blood pH (7.19), BE (-10.6 mmol/l), HCO₃⁻ (25.15 mmol/l), pO₂ (3.33 kPa), O₂SAT (24.12 %) were significantly lower and serum concentration of K⁺ (6.55 mmol/l) was significantly higher in diarrheic calves. These changes indicate the state of uncompensated metabolic acidosis with accompanying hyperkalemia. TT (32.05s) and APTT (39.9s) values were more prolonged in calves with diarrhea than in the control group. D-dimer (587.25 μg/l) concentrations were significantly increased while a visible drop in AT III (103.75%) activity and platelets counts (598 x10⁹/l) were observed in diarrheic group of calves. The results suggest that a consumptive type of disseminated intravascular coagulation (DIC) developed in diarrheic calves.

Opis:
The aim of this study was to determine the presence of cocaine- and amphetamine-regulated transcript-like immunoreactive (CART-LI) neurons and co-localisation of CART with vesicular acetylcholine transporter (VAChT), neuronal nitric oxide synthase (n-NOS), vasoactive intestinal polypeptide (VIP), substance P (SP) and leu-enkephalin (LENK) in the enteric nervous system of the porcine gastric cardia by using a double-labelling immunofluorescence technique. CART-LI neurons were observed in the myenteric plexus (18.2}2.6%). A dense network of CART-LI nerve fibers was mainly observed in the muscular layer. CART showed co-localization mainly with VAChT, n-NOS, VIP and to a lesser degree with LENK and SP. Distribution of CART and its co- ocalization with other neurotransmitters suggest that this peptide plays an important role in gastric motility in the pig.
