Opis:
Human rhinoviruses (HRV) are one of the nine genera belonging to a large family of Picornaviridae. They are responsible for the most cases of common cold, as well as one third to one half of upper respiratory tract (URT) infections. However, HRV are also associated with more severe illnesses, like acute otitis media, sinusitis and some lower respiratory tract diseases such as pneumonia, wheezing in children and exacerbations of asthma. Viral infections are associated with the majority of asthma exacerbations both in children (80-85%) and adults (75-80%), and about 60% of these are caused by HRV. However, the exact mechanism of HRV-induced exacerbations of the disease is not well understood, which makes it difficult to establish the effective treatment. There have already been many attempts to develop a sensitive and specific method of HRV detection in clinical samples. Some of them were based on virus cultures followed by acid lability test, whereas others implemented the reverse transcription-polymerase chain reaction (RT-PCR) and amplification of conserved sequences of the rhinoviral genome. As numerous of these sequences are common to both rhinoviruses and enteroviruses (EVs), further analyses were necessary, which made those methods laborious, time-consuming and too difficult to use in routine diagnostics. Steininger et al. established an RT-PCR-based sensitive and specific method of rhinovirus detection in clinical samples, which was tested to amplify 87 different tissue-culturegrown serotypes of HRV. The aim of this study was to evaluate a modified RT-PCR based method of HRV detection in clinical samples obtained from patients with asthma exacerbations. We collected 41 nasal lavages from patients with asthma exacerbations who received hospital treatment either following an admission or in an out-patient clinic. HRV was found in 22 cases (54%), which corresponded well with the published data.

Bronchial asthma is characterized by persistent airway inflammation and airway wall remodeling. Among many different cells and growth factors triggering changes in bronchi structure, transforming growth factor β1-induced fibroblast to myofibroblast transition is believed to be very important. The aim of this study was to evaluate whether theophylline (used in asthma therapy) and two other methylxanthines (pentoxifylline and its active metabolite lisofylline), may affect transforming growth factor β1-induced fibroblast to myofibroblast transition in bronchial fibroblasts derived from asthmatic patients. We show here for the first time that selected methylxanthines effectively reduce transforming growth factor β1-induced myofibroblast formation in asthmatic bronchial fibroblast populations. PTX was found to be the most effective methylxanthine. The number of differentiated myofibroblasts after PTX, LSF and THEO administration was reduced at least twofold. Studies on the use of methylxanthines opens a new perspective in the development of novel strategies in asthma therapy through their two-pronged, anti-inflammatory and anti-fibrotic action. In the future they can be considered as promising anti-fibrotic drugs.

Purpose: The aim of the work was to create an appropriate substrate for organ transplantation using bioactive tissue-based scaffold populated by cells of the graft recipient. The purpose of the modeling was to investigate the mechanical effects of wave loading of aortic and pulmonary tissue material. Methods: The biological properties of tissues of aortic and pulmonary valves were modified by the process of decellularization. The host cells were removed by various physical methods with focus on minimal degradation of the extracellular matrix. Thus, the decellularization process was controlled by histological methods. The tissue decellularization process was simulated by finite element modelling. Results: The mechanical results represented by a displacement at the center of the sample were coherent and the heterogeneity of the distribution of the caves on the surface of the samples was confirmed, both by experiment and in the simulation by the alternate occurrence of local minima and maxima. The latter results from the uneven removal of cells from the effect of the wave causing decellularization were also predicted by the numerical model. Laser radiation had a destructive effect on the components of the extracellular matrix (e.g., collagen and elastic fibers), mainly depending on the fluence and number of pulses in a single exposure. Conclusions: The differences between the valve tissue materials were shown, and the impact of the process of decellularization on the properties of the tissues was analyzed. It should be emphasized that due to low absorption and high scattering, laser radiation can deeply penetrate the tissue, which allows for effective decellularization process in the entire volume of irradiated tissue.
