Background . Melanin finds enormous applications in different industries for its unique photoprotective and anti-oxidant properties. Due to its emerging demand, scientific researchers are putting efforts to unravel more microorganisms with a potential of producing melanin on large scale. Hence, the present study was aimed at the isolation of extracellular melanin producing microorganisms from lime quarries of Karnataka, India. Besides this, the tyrosinase gene governing melanin synthesis in different organisms were compared in silico to understand its evolutionary aspects. Material and methods . Melanin producing microorganisms were screened on tyrosine gelatin beef extract agar medium. Potential isolate was explored for submerged production of melanin in broth containing L-tyrosine. Melanin was characterized by UV-Vis spectroscopy, thin layer and high performance liquid chromatographic techniques. Antibacterial activity of melanin was performed by agar well assay. Comparative tyrosinase gene sequence analysis was performed by using Geneious 2021.1 trial version software. Results . Pseudomonas otitidis DDB2 was found to be potential for melanin production. No antibacterial activity was exerted by the melanin against tested pathogens. The in silico studies showed that the common central domain of tyrosinase protein sequence of selected Pseudomonas sps. exhibited 100% identity with the common central domain of Homo sapiens tyrosinase (NP_000363.1). Conclusions . Our study shows the production of melanin in good quantities by the isolate Pseudomonas otitidis DDB2 which can be explored for scale-up process. Since the melanin formed is of eumelanin type and the tyrosinase gene sequence of several Pseudomonas sp. showed relatedness to humans, this molecule may be further developed for sunscreen formulations.

This paper discloses the development and synthesis of polymer blended nanocomposites filled with nanoclay. The hybridization of epoxy is mixed with vinylester resin (VER) to prepare polymer blend filled with organoclay was studied to enhance mechanical properties of epoxy/VER. Clay loading was done in such a way that with different wt. proportions viz.1, 1.5, 2.0, 2.5, 3.0, 4.0, 5.0, & 7.5 w % ratios. Appropriately cured samples gave excellent mechanical and tribological properties. Results showed that the tensile strength of the composites increased with increase in filler content for the range of filler contents (2.5-4 % vol.). The results indicated that at 4 % wt. of filler concentration the tensile strength obtained is good i.e. 47.79 MPa with density 1.37 gm/cm3 and hardness 45.5. Dry Sliding wear tests were also conducted by following a well-planned experimental schedule based on Taguchi’s design of experiments, considering parameters like Filler content, Normal load, Sliding Velocity and Sliding distance, on a Pin-On-Disc set-up (ASTM G-99 standard, Make: DUCOM Engineers, Bangalore, India). In the experimentation composite pins were worn against a rotating steel disc (Europe Norm) EN-31, (Rockwell C Hardness) HRC 60 and (Roughness Average) Ra 0.02 microns. Control factors like Filler Content, Normal Load were found to be significant factors affecting the Wear rate i.e. the inclusion of nanoclay as filler found to be contributed in improving the wear resistance of the composite. SEM observations are made to probe the wear mechanisms involved.

A numerical model is developed to study the Soret and Dufour effects on MHD boundary layer flow of a power-law fluid over a flat plate with velocity, thermal and solutal slip boundary conditions. The governing equations for momentum, energy and mass are transformed to a set of non-linear coupled ordinary differential equations by using similarity transformations. These non-linear ordinary differential equations are first linearized using a quasi-linearization technique and then solved numerically based on the implicit finite difference scheme over the entire range of physical parameters with appropriate boundary conditions. The influence of various governing parameters along with velocity, thermal and mass slip parameters on velocity, temperature and concentration fields are examined graphically. Also, the effects of slip parameters, the Soret and Dufour number on the skin friction, Nusselt number and Sherwood number are studied. Results show that the increase in the Soret number leads to a decrease in the temperature distribution and to an increase in concentration fields.

Opis:
An analytical solution of an MHD free convective thermal diffusive flow of a viscous, incompressible, electrically conducting and heat-absorbing fluid past a infinite vertical permeable porous plate in the presence of radiation and chemical reaction is presented. The flow is considered under the influence of a magnetic field applied normal to the flow. The plate is assumed to move with a constant velocity in the direction of fluid flow in slip flow regime, while free stream velocity is assumed to follow the exponentially increasing small perturbation law. The velocity, temperature, concentration, skin friction, Nusselt number and Sherwood number distributions are derived and have shown through graphs and tables by using the simple perturbation technique.

This paper presents the study of the tensile properties of Sansevieria trifasciata – fiber (here after called STF) reinforced polyester composites. The composite sample was fabricated with five different fiber lengths of STF (2, 4, 6, 8 and 10 mm). The fabrication was made by hand lay-up technique. Mechanical properties were determined using tensile testing. An interact between fiber and matrix was observed from the SEM (scanning electron microscope) micrographs. The study reveals that the tensile strength increased with fiber length without effecting the elongation at break of the composite.

Opis:
VPS33B, a human Vacuolar Protein Sorting (VPS) protein which mediates the phagolysosomal fusion in macrophage of the eukaryotic organisms. This protein has a great role during the mycobacterial infections, which binds with the Mycobacterium protein tyrosine phosphatase A (PtpA). A single functional domain of PtpA has been identified using SMART domain databases, followed by finding the antigenicity of PtpA using CLC main workbench tool. The protein-protein interaction network predicts the interface of biological functions of proteins, built by using Cytoscape 2.8.3 version tool for manual literature survey of protein sets. According to the literature the specific interactivity of PtpA with VPS33B of human lead to pathogenesis, and provided a good platform to find the structure of VPS33B as it lacks the 3 dimensional structure in PDB. Homology Modelling of VPS33B provides a significant properties to design a specific drug through screening the drug databases (eDrug3D). The modelled protein has been validated through SAVES server maintained by NIH and UCLA with the standard Ramachandran plot with accuracy of 90.7 %. From our findings the interface residues are very crucial points which has been found through docking the modelled protein and Mycobacterium protein and interface residues were selected manually using PyMol software.

The microstructure of explosive particles, as well as formulations, has a significant influence on the shock sensitivity of various energetic formulations. Particle shape and specific size distributions are always important considerations in explosives processing in order to realize better process parameters. The present study aimed to explore crystallization variables required to achieve a specific particle size of Reduced Shock Sensitivity RDX (RSS-RDX). Crystallization process factors such as cooling rate, agitator speed and configuration, etc. have been systematically studied to understand their effect on the particle size. The study also established the crystallization process parameters required to tightly control a specific particle size distribution ranging from 50 to 500 μm by cooling crystallization. Fine particles ranging from 15 to 30 μm were prepared by an ultrasonication technique. Ultrasonic process parameters, including temperature, time and ultrasonic frequency, were studied and the process conditions were optimized. Micromeritic characterization of RSS-RDX revealed the nature of flowability, which in-turn is useful in identifying the ease of processability. Overall, our interest stemmed from an investigation which showed that careful optimization of crystallization variables can lead to a specific particle size distribution.

Novel Sugar Dendritic Gd-DTPA Complexes for MRI Contrast Agents were prepared and evaluated by in vitro and in vivo methods. The sugar dendritic MRI contrast agents have a good blood vesse pool character, and draw blood vessels and liver cancer remarkably clearer than the clinically using Gd-DTPA. Phospha sugar derivatives or phosphorus heterocyclic derivatives provided by functional groups such as epoxide, bromide, etc., were prepared and evaluated by MTT in vitro method. These phospha sugar derivatives showed excellent activities against leukemia cells as well as solid cancer cells in fashions of (i) higher activity, (ii) wider spectra, (iii) higher selectivity and specificity distingushing healthy and cancer cells, etc., compared with the molecular targeting chemotheraputic anti-cancer agent, Gleevec.
